本论文对12个N-(取代苯磺酰)吲哚类化合物体外抗HIV活性进行了初步筛选,并选择其中活性最好的化合物N-(间硝基苯磺酰)-6-甲基吲哚,对其抗HIV 活性及作用机制进行深入研究。一、体外检测了12个N-(取代苯磺酰)吲哚类化合物对C8166和MT-4细胞的毒性、对HIV-1IIIB 感染C8166后诱导的合胞体形成的抑制及对HIV-1IIIB感染MT-4 细胞后的保护作用。结果发现多个化合物具有抑制HIV-1复制活性,特别是化合物N-(间硝基苯磺酰)-6-甲基吲哚,其对HIV-1IIIB诱导的合胞体形成的EC50和TI (Therapy index)值分别为0.26 μg/ml和543.78;对HIV-1IIIB急性感染的MT-4细胞也有很好的保护作用(TI值为104.23)。二、选择活性最强的化合物N-(间硝基苯磺酰)-6-甲基吲哚进行深入的抗 HIV活性研究。在细胞水平上,通过观察致感染细胞病变和HIV-1 p24抗原表达抑制实验(ELISA方法),采用3类多株HIV病毒株(实验株、临床分离株、耐药株)和3类多种细胞(人T淋巴细胞传代株、HIV-1慢性感染人T淋巴细胞株、人外周血单个核细胞)对化合物进行体外抗HIV活性进行系统评价,实验结果表明,化合物对不同来源的HIV-1病毒株都显示出很好抗HIV活性。同时,我们也研究了化合物抗HIV-2活性,发现该化合物并不能抑制HIV-2在C8166细胞中复制。三、在细胞毒性实验上,我们检测了N-(间硝基苯磺酰)-6-甲基吲哚对不同的细胞系和人外周血单个核细胞(PBMC)毒性作用,结果显示化合物的细胞毒性较低。四、在作用机制和靶点研究上,检测了化合物对感染与未感染细胞之间融合的抑制、对HIV-1急性感染C8166细胞及对HIV-1慢性感染H9细胞(H9/HIV-1IIIB)中病毒复制的阻断作用。结果显示,N-(间硝基苯磺酰)-6-甲基吲哚对急性感染细胞有很好抑制作用(EC50为0.52 μg/ml);但对感染与未感染细胞的融合(EC50为 46.40 μg/ml)和慢性感染H9细胞中病毒的复制没有抑制作用(EC50大于100 μg/ml)。提示化合物作用于HIV侵入细胞后到HIV DNA整合前这一阶段。其后对HIV-1逆转录酶活性的抑制情况进行了分析,发现N-(间硝基苯磺酰) -6-甲基吲哚对HIV-1逆转录酶(RT)有很好抑制作用。五、构效关系分析。我们对12 个N-(取代苯磺酰)吲哚类化合物进行了初步的构效关系研究,发现在N-benzenesulfonyl 环上连接一个吸电子基团(nitro group)比供电子基团(methyl group)有更强的抗HIV 活性,但当在indoles 环上连接吸电子基团(nitro group),抗HIV 活性反而受到抑制。以上实验数据显示N-(间硝基苯磺酰)-6-甲基吲哚是一个安全有效的抗 HIV-1 候选化合物,具有进一步研究价值。

We investigated the effects of Ginsenoside R-e on human sperm motility in fertile and asthenozoospermic infertile individuals in vitro and the mechanism by which the Ginsenosides play their roles. The semen samples were obtained from 10 fertile volunteers and 10 asthenozoospermic infertile patients. Spermatozoa were separated by Percoll and incubated with 0, 1, 10 or 100 mu M of Ginsenoside R-e. Total sperm motility and progressive motility were measured by computer-aided sperm analyzer (CASA). Nitric oxide synthase (NOS) activity was determined by the H-3-arginine to H-3-citrulline conversion assay, and the NOS protein was examined by the Western blot analysis. The production of sperm nitric oxide (NO) was detected using the Griess reaction. The results showed that Ginsenoside R-e significantly enhanced both fertile and infertile sperm motility, NOS activity and NO production in a concentration-dependent manner. Sodium nitroprusside (SNP, 100 nM), a NO donor, mimicked the effects of Ginsenoside R-e. And pretreatment with a NOS inhibitor N-omega-Nitro-L-arginine methyl ester (L-NAME, 100 mu M) or a NO scavenger N-Acetyl-L-cysteine (LNAC, 1 mM) completely blocked the effects of Ginsenoside R-e. Data suggested that Ginsenoside R-e is beneficial to sperm motility, and that induction of NOS to increase NO production may be involved in this benefit.

A new poly(fullerene oxide) thin film material has been fabricated by thermal activation and electron bombardment on hexanitro[60]fullerene (HNF) film deposited on a An substrate, all under vacuum conditions. The reaction products in the polymerization process are analyzed by XPS, UPS, IR, TGA-MS and LDI-MS techniques. It is found that the main effect of thermal and radiation treatments is to induce cleavage of -NO bonds from HNF molecules resulted in the release of nitric oxide gas and the formation of fullerene-bound oxyradicals, C-60-C-6. Spectroscopic evidence strongly suggests that rearrangement of fullerenic nitro moieties into nitrito groups is involved in the HNF decomposition process prior to the generation of reactive oxyradical intermediates. Consequently, the intermolecular coupling reaction of these oxyradicals leads to carbon polymer networks containing oxygen-bridged fullerenes. The thermally generated polymeric thin film is stable up to 900 K. Electron bombardment is also effective in both the decomposition of -NO2 groups and the removal of -OH groups present in HNF films. UV irradiation at 365 nm alone is shown to be not as efficient for the polymer formation. (C) 2003 Elsevier Ltd. All rights reserved.

Chiral ferrocene-based phosphine-imine ligands 1-3 and sulfur-imine ligand 4 were applied in the palladium-catalyzed asymmetric allylic alkylation of cycloalkenyl esters. The results revealed that the substitutents in aryl ring, ferrocenylmethyl and benzyliene position strongly affected the enantioselective induction of phosphine-imine ligands, and the most stereoselective ligand was ferrocenylphosphine-imine 1b with a nitro group in the meta-position of phenyl ring. Under the optimized condition, up to 91% (enantiomeric excesses) e.e. of cyclic alkylation product was obtained by the use of 1b. (C) 2004 Elsevier B.V. All rights reserved.

Novel biodegradable poly(carbonate ester)s with photolabile protecting groups were synthesized by ring-opening copolymerization Of L-lactide (LA) with 5-methyl-5-(2-nitro-benzoxycarbonyl)-1,3-dioxan-2-one (MNC) with diethyl zinc (Et2Zn) as catalyst. The poly(L-lactide-co-5-methyl-5-carboxyl-1,3-dioxan-2-one) (P(LA-co-MCC)) was obtained by UV irradiation Of poly(L-lactide acid-co-5-methyl-5-(2-nitro-benzoxycarbonyl)-1,3-dioxan-2-one) (P(LA-co-MNC)) to remove the protective 2-nitrobenzyl group.

A novel triptycene-based dianhydride, 1,4-bis[4-(3,4-dicarboxylphenoxy)]triptycene dianhydride, was prepared from 4-nitro-N-methylphthalimide and potassium phenolate of 1,4-dihydroxytriptycene (1). The aromatic nucleophilic substitution reaction between 4-nitro-N-methylphthalimide and I afforded triptycene-based bis(N-methylphthalimide) (2), which hydrolyzed and subsequently dehydrated to give the corresponding dianhydride (3). A series of new polyimides containing triptycene moieties were prepared from the dianhydride monomer (3) and various diamines in in-cresol via conventional one-step polycondensation method. Most of the resulting polyimides were soluble in common organic solvents, such as chloroform, THF, DMAc and DMSO. The polyimides exhibited excellent thermal and thermo-oxidative stabilities with the onset decomposition temperature and 10% weight loss temperature ranging from 448 to 486 degrees C and 526 to 565 degrees C in nitrogen atmosphere, respectively. The glass transition temperatures of the polyimides were in the range of 221-296 degrees C. The polyimide films were found to be transparent, flexible, and tough. The films had tensile strengths, elongations at break, and tensile moduli in the ranges 95-118 MPa, 5.3-16.2%, and 1.03-1.38 GPa, respectively. Wide-angle X-ray diffraction measurements revealed that these polyimides were amorphous.

A series of dianhydride monomers, 2,2'-disubstituted-4,4',5,5'-biphenyltetracarboxylic dianhydride (substituents = phenoxy, p-methylphenoxy, p-tert-butylphenoxy, nitro, and methoxy) were synthesized by the nitration of an N-methyl protected 3,3',4,4'-biphenyttetracarboxylic dianhydride (BPDA) and subsequent aromatic nucleophilic substitutions with aroxides (NaOAr) or methoxide. These dianhydrides were polymerized with various aromatic diamines in refluxing m-cresol containing isoquinoline to afford a series of aromatic polyintides. The effects of varying 2,2'-substituents of the dianhydride (BPDA) moiety on the properties of polyimides were investigated. It was found that polyimides from the dianhydrides containing phenoxy, p-methylphenoxy, and p-tert-butylphenoxy side groups possessed excellent solubility and film forming capability whereas polyimides from 2,2'-dinitro-BPDA and 2,2'-dimethoxy-BPDA were less soluble in organic solvent. The soluble polymers formed flexible, tough and transparent films. The films had a tensile strength, elongation at break, and Young's modulus in the ranges 102-168 MPa, 8-21%, 2.02-2.38 GPa, respectively. The polymer gas permeability coefficients (P) and ideal selectivities for N-2, O-2, CO2 and CH4 were determined for the -OAr substituted polyimides. The oxygen permeability coefficient (P-O2) and permselectivity of oxygen to nitrogen (PO2/N-2) of the films were in the ranges 3.4-11.3 barrer and 3.8-4.6, respectively.

The orientational behavior of liquid crystalline polymers with para-nitro azobenzene as side chains under electric field was studied by UV-visible spectroscopy. The results showed that lambda(max) of the poled polymer films was around 394nm, compared to that of the unpoled films, the absorption decreased due to poling. The orientational parameters increased linearly with the increase of the electric field. The temporal stability of the poled polymer film is good at room temperature. This kind of materials showed promise application as nonlinear optical component in photorefractive polymers.

The compounds of [Z]-1-[2-(triphenyl stannyl) vinyl] cyclooctanol (1) and [Z]-1-[2-(tri-p-tolyl stannyl) vinyl] cyclooctanol (2) were synthesized by the reactions of triphenyltin hydride and tri-p-tolyltin hydride with 1-ethynyl cyclooctanol. The crystal structure of compound 1 was determined. The reactions of compound 1 and 2 with IC1, Br-2, I-2 formed nine organotin halides. The organotin oxide or hydroxide were prepared by the reactions of [Z]-1-[2-(phenyl dibromo stannyl) vinyl] cyclooctanol (6) and [Z]-1-[2-(diphenyl monobromo stannyl) vinyl] cyclooctanol (5) with KOH. Three complexes were obtained by the reactions of [Z]-1-[2-(phenyl diiodide stannyl) vinyl] cyclooctanol (8) with three ligands (2,2'-bipyridyl,5-nitro-1,10-phenanthroline,8-Hydroxyquinoline). The sixteen new compounds synthesized in this paper were characterized by means of elemental analysis, IR, H-1 NMR. The reaction mechanism of triphenyltin hydride and tri-p-tolyltin hydride with 1-ethynyl cyclooctanol were also proposed.

Polyacrylates containing para-nitro azobenzene have been synthesized by free radical polymerization. The influence of the length of the spacer of the homopolyacrylates (HPn, n=3,4,6), content of methyl acrylate in the copolyacrylates (CP6) with para-nitro azobenzene groups on the thermal properties, such as liquid crystallinity, Tg and Tm, was studied by DSC, WAXD and polarized optical microscopy. Among the polymers studied, only the homopolyacrylate (HP6)with six carbon atoms in the spacer exhibited a nematic phase. The second-harmonic generation (SHG) signal of the poled HP6 film was detected qualitatively by Maker-fringer method.

Quatemized chitosan: N-(2-hydroxyl-phenyl)-NN-dimethyl chitosan (NHPDCS), N-(5-chloro-2-hydroxyl-phenyl)-NN-dimethyl chitosan (NCHPDCS), N-(2-hydroxyl-5-nitro-phenyl)-NN-dimethyl chitosan (NHNPDCS) and N-(5-bromic-2-hydroxyl-phenyl)-NN-dimethyI chitosan (NBHPDCS) were synthesized and their antifungal activities against Botrytis cinerea Pers. (B. cinerea Pers.) and Colletotrichum lagenarium (Pass) Ell.et halst (C. lagenarium (Pass) Ellet halst) were investigated. The results indicated that the quaternized chitosan derivatives had better inhibitory effects than chitosan, and the antifungal activities should be affected by the cation in these compounds. Ferrocenylphosphine-imine ligands 6 derived front (R,S)-PPFNH2-R 5 and a variety of benzaldehydes were applied in the Pd-catalyzed asymmetric allylic alkylation of 1,3-diphenylprop-2-en-1-yl acetate 7a or pivalate 7b with dimethyl malonate. The substituent effects on the catalytic reaction were investigated, and 96% e.e. with 99% yield was achieved when the m-nitro substituted ligand 6k was used. (C) 2002 Elsevier Science Ltd.
