草地退化不仅仅是今天中国才发生的事情。历史上曾经发生“黑尘暴”的美国North Dakota州,现在仍然有62.7%土地被用作耕作,林地所占面积仅仅占1%。私有制和种植业较畜牧业的高的利润率,是该州种植业比重高于畜牧业的重要原因。草原保护项目(CRP)得到了美国政府财政的大力支持, 在North Dakota州采用了以草地恢复为主的措施。美国发达的教育体系带来的高流动率,使得North Dakota州自30年代后居住人口稳定在60-70万成为可能,从而避免了我国出现的草原地区不断增加的人口压力。由此可见:草地作为重要的自然资源,不可能完全保护起来;没有其他配套措施,私有制可能会带来新一轮垦殖;草地畜牧业仍然是畜牧业的重要组成部分,畜牧业发展需要结合种植业尤其需要与饲料生产相结合。 依据生态系统服务的理念,首次发展了生态系统服务指数(ESI),试图通过对生态系统服务功能的综合考虑,提出科学的适宜放牧率的评价方法。本文利用美国北达科他州立大学中部草原研究站17年长期放牧试验数据,选取植物多样性Shannon-Wiener指数、地上净初级生产力、土壤表层含水量和单位面积家畜增重等四个指标,通过对不同指标分别赋予不同的权重,计算不同管理目标下ESI及其稳定性,并对单目标管理与多目标管理进行了比较研究。结果表明,对于北美混合普列里(Prairie)草地,围封不利用或建立自然保护区,虽然生态系统比较稳定,但既不能有效的提高植物多样性、初级生产力和土壤水分含量等生态功能,又没有畜产品产出;而在重牧或极重牧处理下虽然获得了较大的畜产品生产,但导致了草地生态系统的退化和较大的系统不稳定性。因此,这两种管理方式在实践中都是不可取的。应用生态系统服务指数综合考虑,认为应该权衡各项生态功能和生产功能,此时轻牧或中牧是最适宜的。因此,ESI的建立避免了单项指标的评价偏差,使得适宜放牧率的确定更加合理。 利用前面构建的多目标权重评价体系,结合内蒙古草原生态系统定位站放牧试验样地的数据,探讨了锡林郭勒盟定位站附近的适宜放牧率为2.67羊/ha,低于单目标条件下的每公顷绵羊4羊/ha。同时在经济学和生态系统系统服务理论的基础上,依据谢高地等人(2001年)的研究,并提出了环境税和生态补偿的标准,分别是每公顷15元和90元。

陆地生态系统与大气之间的水热碳交换是物质、能量循环的关键过程,一直以来都为研究者们所关注。进入20 世纪以来,特别是随着人们对全球气候变暖的逐步认识,气候变化对水热碳交换过程的影响及其对气候变化的响应研究更加备受关注。本研究以2004~2006 年近三年的涡度相关系统连续观测数据为依托,分析了雨养玉米农田水热碳通量的动态及其影响因子。研究表明,玉米农田水热通量(WHF) 呈显著的单峰型日变化, 日最大值出现在正午12:00~13:00,WHF 变化同步。潜热通量(LE)的季节变化规律与日变化相似,冬季小夏季大,年最大值与最小值分别出现在7 月和1 月。显热通量(Hs) 季节变化也呈单峰型,但年最大值出现在5 月,这主要与降水以及作物生长有关。半小时尺度上,WHF 主要受辐射控制,而日峰值受辐射峰值以及植被生长的双重影响;日尺度上,只要有降水过程,Hs 就会随土壤水分的增大而减小,降水停止后逐渐恢复。而降水对LE 的影响受到可用能量(AE)的干扰,表现出复杂的变化趋势。总的来说,降水持续时间越长AE 越少,对LE 的抑制越大;季节尺度上,WHF 受热量与水分的双重制约。Hs 随着天气回暖后第一次较大降水过程的出现呈现明显下降,而LE 则呈现相反的变化趋势。随着雨季到来和作物的生长,Hs 在7 月出现低谷,而LE 呈现相反的趋势随着降水量的增加而增大;年际间WHF 的分布规律大体一致,但因气象条件等的差异,特别是降水的差异造成年际间WHF 略有不同。在不同水文年型下,水分因子的影响作用有显著差异,且WHF 对热量与水分条件变化的敏感程度也不相同。欠水年,水分因子的作用更显著,是制约WHF 变化的主要控制因子,WHF 对水分的变化更敏感;而丰水年,水分因子的影响减弱,热量的盈亏决定着WHF 变化的主要方向。在不同水文年型下,水热碳通量对水热条件的变化表现出不同的响应方式,为研究生态系统对气候变化的响应提供了参考。 净碳(C)吸收期,玉米农田净碳交换(NEE)呈显著的日变化,在日出以后由CO2 释放转变为CO2 吸收,12:30 左右达到一天中的吸收峰值,日落前出现相反的转换。而净C 释放期内,NEE 均为正值且无明显日变化。NEE 季节变化也呈单峰型二次曲线,在7 月下旬或8 月上旬达到年最大吸收率。根据NEE 的正负,一年分为三个阶段:两个C 排放期与一个C 吸收期。一般C 吸收期从6月开始到9 月结束,此前此后均为C 排放期。在半小时、日时间尺度上,光通量密度(PPFD)与NEE 有着相似的变化规律,是控制NEE 的主要因子;在日、季节尺度上,叶面积指数(LAI)和气孔导度(gs)是影响NEE 的主要生物因子,且gs 的影响程度随着发育期的变化而变化,而不同年份间LAI 对NEE 的影响没有显著的差异。几乎在所有时间步长上,土壤温度(Ts)均为生态系统呼吸(Re)的主要控制因子,时间尺度愈短,二者的相关性愈好。总的来说,在较短时间尺度上,高PPFD 与夏季低温将会促进C 的吸收,有利于C 累积。 玉米农田日最大净C 吸收速率(NEEmax, daily)以及吸收释放转换点(NEE=0)均受PPFD 控制。NEEmax, daily 出现时间与PPFDmax, daily 出现时间几乎完全一致,当PPFD 达到1 日内极大值时,净C 吸收也相应达到了日最大值。但NEEmax, daily的量值还受到其它因子的影响。当水分条件充足时,还将受到LAI、gs 等生物因子的控制。NEE 由正转为负的转换点也是由PPFD 决定。当PPFD 稳定大于PPFD*( PPFD*=100 μmol•m-2s -1)时,净C 吸收开始;当PPFD 稳定小于PPFD*时,净C 吸收由此结束。1 日内,PPFD 稳定通过PPFD*之间的时间间隔决定了日净C 吸收的时间长度。日净C 吸收的时间越长,吸收量也越大,且有明显的季节变化,7 月最长9 月最短。 按照热量水分状况将三年分组,分为I 组(水分状况相似,热量条件不同)与II 组(热量条件相似,水分状况不同)。 I 组年际间PPFD 波动是造成C 交换格局变化的关键原因。而II 组年际间C 交换格局不同是由降水量及其不同分布引起的土壤含水量(SWC)变化是造成。SWC 可以解释年际间NEE 变异的97%,而大气水汽压亏缺(VPD)可以解释30.7%;温度因子通过影响C 收支中的呼吸项,间接影响着生态系统的NEE,它可以解释年际间NEE 变异的73.9%,也是造成年际间C 交换格局不同的原因之一;另外,PPFD 和发育期早晚以及净C吸收期长度等也同样影响着C 交换格局的变化。综合两组情况来看,由水分条件年际变化引起的NEE 的波动大于能量年际变化引起的波动。总之,在较长时间尺度上,NEE 对SWC 变化比其对PPFD 变化更敏感,说明在半干旱地区土壤水分条件仍然是决定C 交换格局的主导因子。 NEE 与LE 呈线性相关,它们之间的相关性主要受温度和NEE 的控制,温度越高,二者的相关性越弱,而NEE 越大二者相关性越好。同时,作物蒸腾与土壤蒸发的比例也是影响NEE 与LE 之间关系的主要因素。蒸腾作用所占的比例越大,二者的线性关系越显著,而土壤蒸发比例越大,二者的相关性越弱。总的来说,NEE 与LE 之间的线性关系有明显的季节变化,生长季好于非生长季,夏天好于冬天。 总之,雨养玉米农田水热碳通量既具有其它农田生态系统共有的动态特征,也具有其特有特征。

Amphibian skin is a rich resource of antimicrobial peptides, like maximins and maximin Hs from frog Bombina maxima. Novel cDNA clones encoding a precursor protein, which comprises a novel maximin peptide (maximin 9) and reported maximin H3, were isolated from two constructed skin cDNA libraries of B. maxima. The predicted primary structure of maximin 9 is GIGRKFLGGVKTTFRCGVKDFASKHLY-NH2. A surprising substitution is at position 16, with a free cysteine in maximin 9 rather than usual conserved glycine in other reported maximins. Maximin 9, the homodimer form and its Cys(16) to Gly(16) mutant were synthesized and their antimicrobial activities were evaluated. Unlike previously reported maximin 3, the tested bacterial and fungal strains were resistant to maximin 9, its homodimer and the Cys(16) to Gly(16) mutant (with MICs > 100 mu M). On the other hand, interestingly, while eight clinical Mollicutes strains were generally resistant to maximin 9 homodimer and its Cys(16) to Gly(16) mutant, most of them are sensitive to maximin 9 at a peptide concentration of 30 mu M, especially in the presence of dithiothreitol. These results indicate that the presence of a reactive Cys residue in maximin 9 is important for its antimycoplasma activity. The diversity of antimicrobial peptide cDNA structures encountered in B. maxima skin cDNA libraries and the antimicrobial specificity differences of the peptides may reflect well the species' adaptation to the unique microbial environments. Antimicrobial peptides secreted by the skin of many amphibians play an important role in innate immunity. From two skin cDNA libraries of two individuals of the Chinese red belly toad (Bombina maxima), we identified 56 different antimicrobial peptide cDNA sequences, each of which encodes a precursor peptide that can give rise to two kinds of antimicrobial peptides, maximin and maximin H. Among these cDNA, we found that the mean number of nucleotide substitution per non-synonymous site in both the maximin and maximin H domains significantly exceed the mean number of nucleotide substitution per synonymous site, whereas the same pattern was not observed in other structural regions, such as the signal and propiece peptide regions, suggesting that these antimicrobial peptide genes have been experiencing rapid diversification driven by Darwinian selection. We cloned and sequenced seven genes amplified from skin or liver genomic DNA. These genes have three exons and share the same gene structure, in which both maximin and maximin H are encoded by the third exon. This suggests that alternative splicing and somatic recombination are less likely to play a role in creating the diversity of maximins and maximin Hs. The gene trees based on different domain regions revealed that domain shuffling or gene conversion among these genes might have happened frequently.

Partial sequences of the mitochondrial cytochrome b gene of the Korean hare (Lepus coreanus) were analyzed to determine the degree of genetic diversity. Nine haPlotyes were observed, and the maximum Tamura-Nei nucleotide distance among them was 2.8%, indicating that genetic diversity of L. coreanus is moderate. In order to clarify the Korean hare's taxonomic status and relationship with the Manchurian hare (L. mandshuricus) and the Chinese hare (L. sinensis), these nine haplotypes of the Korean hare were compared with 13 haplotypes from five other species of eastern Asian Lepus including L. mandshwicus and L. sinensis. The Korean hare was distinct in its cytochrome b gene, and it is confirmed that L. coreanus is a valid species, as noted by Jones and Johnson (1965, Univ. Kansas Publ. (Mus. Nat. Hist.) 16:357). Further analyses of mtDNA cytochrome b gene with additional specimens of L. coreanus from North Korea and other species of Lepus from eastern Asia are needed to clarify the taxonomic status of the divergent mtDNA clades of L. mandshuricus and L. sinensis.

The cryopreservation of oocytes has been only marginally successful with any of the current protocols, including slow cooling, rapid cooling and vitrification. We wished to test the hypothesis that oocytes from a single mouse strain would freeze successfully by 1 of the 3 mentioned protocols. Unfertilized Kunming mouse oocytes obtained 14 h after PMSG/hCG administration were randomly assigned to be cryopreserved after slow cooling, ultra rapid cooling and vitrification. Oocytes were thawed by straws being placed into 37 degrees C water, and their morphological appearance and in vitro fertilization capability were compared with that of oocytes that had not undergone cryopreservation. Survival of oocytes was indicated by the absence of darkened ooplasm or by broken membranes or zona pellucida. Functional integrity was evaluated by the formation of a 2-cell embryo after IVF. Survival rate of slow cooled oocytes did not differ from that seen in vitrified oocytes (55.1 vs 65.9%) but was significantly lower in the rapidly cooled oocytes (24.2%; P<0.01). The results of NF of slow cooled and vitrified oocytes were similar to those of the control group (72 and 73 vs 77%; P>0.05). It appears that Kunming mouse oocytes can be successfully cryopreserved using the slow cooling method with 1,2-propanediol and vitrification, which contains both permeating and nonpermeating cryoprotectants. 用维生素C含量为38、364和630mg/kg的三种饲料饲喂初体重为(23.19±0.58)g的长吻8个月,实验结果表明,38mg/kgVC饲料组饲喂的长吻表现出了典型的VC缺乏症状:体色发黑,脊椎侧弯,鳍边由开始的萎缩直到腐烂、贫血、特定生长率显著降低(p<0.05)。并且38mg/kgVC饲料组长吻的生理指标也受到影响:血清溶菌酶活性显著下降(p<0.05),肝脏的SOD活性及MDA含量均显著升高(p<0.05),血清皮质醇没有显著差异(p>0.05),但各处理组长吻肝脏中均未检测到诱导型HS

在本研究中,将福尔马林灭活的柱状黄杆菌菌苗(FKG4)经腹腔注射免疫草鱼,以注射灭菌PBS作对照,分别在免疫后1、7、15和28d,提取受免鱼和对照鱼肝脏、脾脏和头肾3种组织中的总RNA并反转录成cDNA,利用Real-time PCR方法对不同组织中C-反应蛋白(CRP)、主要组织相容性复合体I(MHCⅠ)、肿瘤坏死因子(TNFα)、白介素1(IL-1β)、白介素8(IL-8)、Ⅰ型干扰素(IFNⅠ)等6种免疫相关基因的表达进行定量分析。结果发现CRP在受免鱼肝脏中的表达于免疫后1、7d显著高于对照鱼,

将柱状黄杆菌胞外多糖(EPS)溶液经腹腔注射免疫草鱼,以注射灭菌生理盐水作对照。免疫一周后,分别提取受免鱼与对照鱼肝脏、脾脏、体肾和头肾4种组织中的总RNA,并通过RT-PCR半定量的方法检测不同组织中C-反应蛋白(CRP)、白介素1(IL-1)、主要组织相容性复合体I(MHC I)、免疫球蛋白M(IgM)、干扰素(IFN)等5种免疫基因的表达情况。结果显示,CRP在受免鱼肝脏中的表达显著上升;MHC I在受免鱼脾脏、体肾中的表达显著下降;IgM在受免鱼4种组织中的表达皆显著上升;IL-1在受免鱼4种组织

本文以多氯联苯(PCBs)模拟水样为实验样品,优化了杯[4]芳烃探头顶空固相微萃取(HS SPME)的分析条件,并将杯[4]芳烃探头和商用PDMS探头对PCBs的萃取效果进行了比较。利用杯[4]芳烃 HS SPME法与USEPA608法同时测定了模拟水样和典型污染地区环境样品中的多氯联苯。研究结果表明:杯[4]芳烃SPME探头对样品中高氯代PCBs的萃取效果优于商用PDMS探头;杯[4]芳烃 HS SPME和USEP A608法对同一样品的检测结果一致;该法对水样中PCBs的检出限达1~6ng/L,比US

运用顶空固相微萃取与色谱联用方法(HS-SPME-GC)对水中的残留有机氯化合物进行了分析.对影响HS-SPME-GC分析灵敏度的各种实验因素如涂层种类、萃取温度、平衡时间、离子浓度等进行了讨论,并将该方法与固相萃取法(SPE)、液液萃取法(LLE)作了对比;同时考察了常见环境共存污染物直链烷基苯磺酸钠(LAS)对几种方法的影响.
