通过网上提供的生物信息学分析软件进行搜索和比对,初步筛选到3个较好的针对小鼠双尾-C(Bicc1)基因的RNA干扰(RNAi)序列。合成这3个干涉序列片段后克隆到pRS-HushshRNA载体中。构建Bicc1基因的真核表达载体pEGFP-C3-Bicc1,将绿色荧光蛋白(GFP)标签标记在Bicc1蛋白上。利用细胞转染技术将pEGFP-C3-Bicc1与3个干涉序列载体共转染至体外培养的HEK-293细胞中,最后通过细胞荧光强度、半定量PCR和Westernblotting鉴定出其中两个序列(pRS-Hush-RNAi-Bicc1-N/-C)能明显降低Bicc1蛋白在HEK-293细胞中的表达水平,为下一步建立起低表达Bicc1的稳定细胞株和研究小鼠Bicc1的功能提供了良好的材料。

为了提高污水处理的质量和效率,将高效的生物处理技术——膜-生物反应器(SMBR)与生态净化技术——复合垂直流人工湿地(IVCW)进行有效复合而成的SMBR-IVCW复合系统应用于高浓度综合污水的高效处理与回用中.通过对不同水力负荷组合条件下SMBR-IVCW系统对污染物净化效果的比较,得到了该复合系统的较好水力组合条件.结果表明,SMBR-IVCW具有良好的净化功效及稳定性,SMBR单元在有机物的去除、硝化作用中起到主要贡献,而IVCW单元在反硝化脱氮、深度除磷等方面进一步补充,两者的有机结合能充分发挥各

以螅状独缩虫(Carchesium polypinumLinne,1785)为研究对象,利用RAPD技术对武汉市南湖、沙湖和东湖3个湖区的螅状独缩虫样本进行了DNA多态性比较分析,以确认种群并期望促进微型生物种群生态学研究。结果如下:(1)从120条随机引物中筛选到31条有清晰扩增条带的,其中20条引物扩增出172条条带清晰、多态性高的条带,片段大小为100~1600bp,而其余11个引物扩增出59条相对多态性差的清晰条带,片段大小为100~1500bp;(2)扩增产物经SPSS12.0与RAPDista

The Yangtze finless porpoise, Neophocaena phocaenoides asiaeorientalis, is all endangered small cetacean that occurs only in the middle and lower reaches of the Yangtze River of China. The establishment of a breeding population of the porpoise in Tian-e-Zhou Baiji National Natural Reserve represents the first attempt at ex situ conservation efforts for a cetacean species. With the goal of effective protection, management, and monitoring of this preserved population, we examined its genetic diversity using 930 bp of mtDNA control region sequences and 13 polymorphic microsatellite loci. A very low level of genetic variation (h = 0.6010 +/- 0.0029 s.d.; pi = 0.0007 +/- 0.0000002 s.d.) in the mtDNA control region sequences and a moderate genetic diversity (Ho = 0.5740 +/- 0.2575 s.d.) in the microsatellites were detected in the population. It is necessary to introduce more individuals with representative genetic variations into the reserve ill order to foml a larger and healthier group structure for long-term survival of the population. Successful establishment of the Yangtze finless porpoise population in the Reserve also provides a useful model for an ex situ conservation programme for other rare and endangered species. 同步操作是并发Java程序非常大的一部分开销.在现有程序分析方法的基础上,提出了一种精确而有效的冗余同步操作的静态删除方法.该方法分为基本处理和线程间时序分析两个阶段,充分考虑了控制流结构和线程交互时序对同步删除的影响.构造了一个Java编译器JTool,并在其上实现了同步删除算法.对于确定的单线程程序,同步删除率达到100%;对于多线程程序,同步删除率高于现有的分析工具.

本文提出了一个三段热化学循环分解水制氢和氧的方法以及将此三段热化学法改为二段热化学循环分解水制氢和氧的新方法。本三段热化学法由下列三个反应组成:{Cr_2O_3+6KoH=2K_3CrO_4+H_2O+2H_2 6K_3CrO_4+5H_2O=Cr_2O_3+4K_2CrO_4+10KOH 4K_2CrO_4+4KOH=4K_3CrO_4+2H_2O+O_2本二段热化学法由下列二个反应组成:{Cr_2O_3+6KOH=2K_3CrO_4+H_2O+2H_2 2K_3CrO_4+3H_2O=Cr_2O_3+6KOH+O_2上面的三步法和二步法分别通过正交试验,讨论了影响各个反应产率的主要因素,利用最小二乘法在计算机上对试验结果进行了回归分析和方差分析,得到了下面几个数学模型。利用x-光射线粉末衍射对反应产物进行了测定,提出了有关反应机理,并对这两种方法的特点给予了评价。三段法的数学模型:(1)y=-0.8295+1.7681*10~(-6)T~2+7.4750*10~(-2)t+0.2753n (2)y=0.2730+0.5301t+3.2600*10~(-4)V (3)y=-0.1785+2.6999*10~(-4)T+3.5259*10~(-3)t~2+0.3370n二段法的数学模型:(1)y=-0.8295+1.7681*10~(-6)T~2+7.4750*10~(-2)t+0.2753n (2)y=-0.1670+4.3999*10~(-4)T+7.2500*10~(-3)t+1.5599*10~(-3)V式中,T:反应温度(℃)V:水的通水量(ml)n:氢氧化钾的实际加入量与其理论量的比y:转化率t:反应时间(hr)

A near-infrared single-photon detection system is established by using pigtailed InGaAs/InP avalanche photodiodes. With a 50GHz digital sampling oscilloscope, the function and process of gated-mode (Geiger-mode) single-photon detection are intuitionally demonstrated for the first time. The performance of the detector as a gated-mode single-photon counter at wavelengths of 1310 and 1550nm is investigated. At the operation temperature of 203K,a quantum efficiency of 52% with a dark count probability per gate of 2. 4 * 10~(-3), and a gate pulse repetition rate of 50kHz are obtained at 1550nm. The corresponding parameters are 43% , 8. 5 * 10~(-3), and 200kHz at 238K.
