Background: Gastrointestinal stromal tumours (GISTs) are the most common primary mesenchymal neoplasia in the gastrointestinal tract, although they represent only a small fraction of total gastrointestinal malignancies in adults (<2%). GISTs can be located at any level of the gastrointestinal tract; the stomach is the most common location (60-70%), in contrast to the rectum, which is most rare (4%). When a GIST invades into the adjacent prostate tissue, it can simulate prostate cancer. In this study, we report on a case comprising the unexpected collision between a rectal GIST tumour and a prostatic adenocarcinoma. Findings: We describe the complexity of the clinical, endoscopic and radiological diagnosis, of the differential diagnosis based on tumour biopsy, and of the role of neoadjuvant therapy using imatinib prior to surgical treatment. Conclusions: Although isolated cases of coexisting GISTs and prostatic adenocarcinomas have reviously been described, this is the first reported case in the medical literature of a collision tumour involving a rectal GIST and prostatic adenocarcinoma components.

Background: There has been a significant growth in the prevalence of allergy, mainly associated to IgE-mediated disorders such as asthma and rhinitis. The identification of atopy in asthmatic patients through the measurement of specific IgE can help to identify risk factors that cause asthmatic symptoms in patients. The development and use of individualized allergen-based tests by the Component Resolved Diagnosis has been a crucial advance in the accurate diagnosis and control of allergic patients. The objective of this work was to assess the usefulness of molecular diagnosis to identify environmental allergens as possible factors influencing the development and manifestation of asthma in a group of asthmatic patients from Iran. Methods: Studied population: 202 adult asthmatic patients treated at the Loghman Hakim Hospital and Pasteur Institute of Teheran (Iran) from 2011 to 2012. Specific IgE determined by the ImmunoCAP system were used to both evaluate the patients' atopic condition and the molecules involved in the allergic sensitization. SDS-PAGE IgE-immunoblotting associated with mass spectrometry was carried out to study the cockroach IgE-binding sensitizing proteins. Results: Forty-five percent of all patients could be considered atopic individuals. Eighty-two percent of atopic patients were sensitized to pollen allergens. The Salsola kali (Sal k 1) and the Phleum pratense (rPhl p 1 and/or rPhl p 5) major allergens were the most common sensitizers among pollens (71% and 18%, respectively). Thirty-five percent of the atopic population was sensitized to cockroach. Four different allergens, including a previously unknown alpha-amylase, were identified in the cockroach extract. No significant associations could be demonstrated between the severity of asthma and the specific IgE levels in the atopic population. Statistical analysis identified the Sal k 1 as the main protein allergen influencing the development and expression of asthma in the studied population. Conclusions: Pollen and cockroach were the most relevant allergen sources in the asthmatic population. The Salsola kali major allergen was the main cause for sensitization in the atopic patients suffering asthma. Using the Component Resolved Diagnosis, it was possible to identify a new Blattella germanica cockroach allergen (Blattella alpha amylase 53 kDa) that could sensitize a relevant percentage of this population.

[ES]El objetivo principal de este proyecto se centra en conseguir las características mecánicas requeridas por los componentes del chasis de los vehículos con una reducción de peso y mejora de productividad, para ellos es necesario simular el proceso de templado que se da durante la estampación en caliente de una pieza de chapa mediante utillajes con conductos de refrigeración. Para ello se ha utilizado el programa de elementos finitos ANSYS hasta obtener un proceso de embutición simulado que represente con una desviación dentro de los límites aceptables el comportamiento real de la chapa en la etapa de refrigeración. Como objetivo secundario se encuentra el afianzamiento de las bases teóricas de ciencia de materiales y la adquisición de más conocimientos relacionados con la transmisión de calor entre cuerpos sólidos, centrándose sobre todo en la distribución de temperaturas sobre la superficie de éstos. En una primera parte se tratarán los conceptos generales de la estampación en caliente y sus posibles variantes. También se explicará la necesidad del uso de nuevos materiales para la industria automovilística, así como la razón por la cual se utilizan conductos de refrigeración. A continuación, se definirá la geometría de la chapa a analizar, tanto las consecuentes geometrías de los utillajes, que tendrán diferentes distribuciones de conductos de refrigeración. Además se establecerán los criterios para realizar el análisis térmico transitorio del conjunto troquel – chapa – matriz. Una vez seleccionado el tipo de análisis se profundizará en su estudio, aplicándolo a los diferentes utillajes ya citados. Se analizarán los resultados obtenidos y los errores y se buscarán posibles alternativas. Finalmente, se procederá a sacar las conclusiones de la simulación realizada y se procederá a comparar los resultados obtenidos con las diferentes distribuciones de conductos de refrigeración en los utillajes.

[ES]El objetivo de este trabajo es programar y mecanizar un molde de inyección de aluminio, con el objetivo de obtener un pedal de batería. Para ello, previamente se necesitará diseñar en CAD el molde, para posteriormente exportarlo al programa Surfcam V4.0, donde se definirán las trayectorias de las herramientas teniendo en cuenta su geometría. Una vez definidas en el programa, éste se ejecutará en un centro de mecanizado sobre un tocho de aluminio.

Bordetella pertussis causes whooping cough, a respiratory infectious disease that is the fifth largest cause of vaccine-preventable death in infants. Though historically considered an extracellular pathogen, this bacterium has been detected both in vitro and in vivo inside phagocytic and non-phagocytic cells. However the precise mechanism used by B. pertussis for cell entry, or the putative bacterial factors involved, are not fully elucidated. Here we find that adenylate cyclase toxin (ACT), one of the important toxins of B. pertussis, is sufficient to promote bacterial internalisation into non-phagocytic cells. After characterization of the entry route we show that uptake of "toxin-coated bacteria" proceeds via a clathrin-independent, caveolae-dependent entry pathway, allowing the internalised bacteria to survive within the cells. Intracellular bacteria were found inside non-acidic endosomes with high sphingomyelin and cholesterol content, or "free" in the cytosol of the invaded cells, suggesting that the ACT-induced bacterial uptake may not proceed through formation of late endolysosomes. Activation of Tyr kinases and toxin-induced Ca2+-influx are essential for the entry process. We hypothesize that B. pertussis might use ACT to activate the endocytic machinery of non-phagocytic cells and gain entry into these cells, in this way evading the host immune system.

[ES] El estudio de la flora alóctona en hábitats urbanos está adquiriendo cada vez un mayor interés por parte de la comunidad científica. Este estudio se llevó a cabo en el municipio de Basauri, situado en la provincia de Bizkaia, donde por un lado, se realizó un catálogo de la flora vascular alóctona y por otro, se realizaron muestreos de vegetación en siete hábitats diferentes, con el objetivo de evaluar el nivel de invasión en cada uno de ellos. Se identificaron un total de 50 especies alóctonas (exóticas) en el municipio, la mayor parte de ellas de origen americano, introducidas como ornamentales y que muestran comportamiento invasor. A partir de los datos de los inventarios de vegetación, se estimó que aproximadamente un 18,2% de las especies eran exóticas, entre las que destacan las que pertenecen a las familias Asteraceae, Fabaceae y Poaceae. Además, se observó que los hábitats más invadidos fueron aquellos sometidos a mayores perturbaciones, como los hábitats ruderales y los riparios, también se observó una disminución de la diversidad total de especies con el aumento de la cobertura de especies alóctonas.

Background: Intratumor heterogeneity may be responsible of the unpredictable aggressive clinical behavior that some clear cell renal cell carcinomas display. This clinical uncertainty may be caused by insufficient sampling, leaving out of histological analysis foci of high grade tumor areas. Although molecular approaches are providing important information on renal intratumor heterogeneity, a focus on this topic from the practicing pathologist' perspective is still pending. Methods: Four distant tumor areas of 40 organ-confined clear cell renal cell carcinomas were selected for histopathological and immunohistochemical evaluation. Tumor size, cell type (clear/granular), Fuhrman's grade, Staging, as well as immunostaining with Snail, ZEB1, Twist, Vimentin, E-cadherin, beta-catenin, PTEN, p-Akt, p110 alpha, and SETD2, were analyzed for intratumor heterogeneity using a classification and regression tree algorithm. Results: Cell type and Fuhrman's grade were heterogeneous in 12.5 and 60 % of the tumors, respectively. If cell type was homogeneous (clear cell) then the tumors were low-grade in 88.57 % of cases. Immunostaining heterogeneity was significant in the series and oscillated between 15 % for p110a and 80 % for Snail. When Snail immunostaining was homogeneous the tumor was histologically homogeneous in 100 % of cases. If Snail was heterogeneous, the tumor was heterogeneous in 75 % of the cases. Average tumor diameter was 4.3 cm. Tumors larger than 3.7 cm were heterogeneous for Vimentin immunostaining in 72.5 % of cases. Tumors displaying negative immunostaining for both ZEB1 and Twist were low grade in 100 % of the cases. Conclusions: Intratumor heterogeneity is a common event in clear cell renal cell carcinoma, which can be monitored by immunohistochemistry in routine practice. Snail seems to be particularly useful in the identification of intratumor heterogeneity. The suitability of current sampling protocols in renal cancer is discussed.
