Background: In the violaxanthin (V) cycle, V is de-epoxidized to zeaxanthin (Z) when strong light or light combined with other stressors lead to an overexcitation of photosystems. However, plants can also suffer stress in darkness and recent reports have shown that dehydration triggers V-de-epoxidation in the absence of light. In this study, we used the highly stress-tolerant brown alga Pelvetia canaliculata as a model organism, due to its lack of lutein and its non-photochemical quenching independent of the transthylakoidal-ΔpH, to study the triggering of the V-cycle in darkness induced by abiotic stressors. Results: We have shown that besides desiccation, other factors such as immersion, anoxia and high temperature also induced V-de-epoxidation in darkness. This process was reversible once the treatments had ceased (with the exception of heat, which caused lethal damage). Irrespective of the stressor applied, the resulting de-epoxidised xanthophylls correlated with a decrease in Fv/Fm, suggesting a common function in the down-regulation of photosynthetical efficiency. The implication of the redox-state of the plastoquinone-pool and of the differential activity of V-cycle enzymes on V-de-epoxidation in darkness was also examined. Current results suggest that both violaxanthin de-epoxidase (VDE) and zeaxanthin-epoxidase (ZE) have a basal constitutive activity even in darkness, being ZE inhibited under stress. This inhibition leads to Z accumulation. Conclusion: This study demonstrates that V-cycle activity is triggered by several abiotic stressors even when they occur in an absolute absence of light, leading to a decrease in Fv/Fm. This finding provides new insights into an understanding of the regulation mechanism of the V-cycle and of its ecophysiological roles.

In this article an index decomposition methodology is used to estimate the effect of intersectorial and intrasectorial changes in explaining the 38% reduction in industrial energy intensity in the Basque Autonomous Community from 1982 to 2001. Period-wise additive decomposition results show that (1) the decline is fully explained by intrasectorial changes and that (2) intersectorial changes have not contributed to reduce but to increase the energy intensity of the Basque industrial sector. However, timeseries decomposition analysis shows that (1) four different phases can be distinguished in the evolution of energy intensity of the Basque industry from 1982 to 2001 and (2) that the evolution of the “Iron and Steel” sector is determinant when explaining those phases. Moreover, the analysis stresses the necessity to disaggregate the “Iron and Steel” sector in order to be able to distinguish purely technological effects from the rest of intrasectorial changes.

This paper sets out an optimum synthesis methodology for wheel profiles of railway vehicles in order to secure good dynamic behaviour with different track configurations. Specifically, the optimisation process has been applied to the case of rail wheelsets mounted on double gauge bogies, that move over two different gauges, which also have different types of rail: the Iberian gauge (1668 mm) and the UIC gauge (1435 mm). Optimisation is performed using Genetic Algorithms and traditional optimisation methods in a complementary way. The objective function used is based on an ideal equivalent conicity curve which ensures good stability on straight sections and also proper negotiation of curves. To this end the curve is constructed in such a way that it is constant with a low value for small lateral wheelset displacements (with regard to stability), and increases as the displacements increase (to facilitate negotiation of curved sections). Using this kind of ideal conicity curve also enables a wheel profile to be secured where the contact points have a larger distribution over the active contact areas, making wear more homogeneous and reducing stresses. The result is a wheel profile with a conicity that is closer to the target conicity for both gauges studied, producing better curve negotiation while maintaining good stability on straight sections of track. The paper shows the resultant wheel profile, the contact curves it produces, and a number of dynamic analyses demonstrating better dynamic behaviour of the synthesised wheel on curved sections with respect to the original wheel.

Nanostructured FeNi-based multilayers are very suitable for use as magnetic sensors using the giant magneto-impedance effect. New fields of application can be opened with these materials deposited onto flexible substrates. In this work, we compare the performance of samples prepared onto a rigid glass substrate and onto a cyclo olefin copolymer flexible one. Although a significant reduction of the field sensitivity is found due to the increased effect of the stresses generated during preparation, the results are still satisfactory for use as magnetic field sensors in special applications. Moreover, we take advantage of the flexible nature of the substrate to evaluate the pressure dependence of the giant magneto-impedance effect. Sensitivities up to 1 Omega/Pa are found for pressures in the range of 0 to 1 Pa, demostrating the suitability of these nanostructured materials deposited onto flexible substrates to build sensitive pressure sensors.

We studied the effect of magnetoelastic anisotropy on domain wall (DW) dynamics and remagnetization process of magnetically bistable Fe-Co-rich microwires with metallic nucleus diameters (from 1.4 to 22 mu m). We manipulated the magnetoelastic anisotropy applying the tensile stresses and changing the magnetostriction constant and strength of the internal stresses. Microwires of the same composition of metallic nucleus but with different geometries exhibit different magnetic field dependence of DW velocity with different slopes. Application of stresses resulted in decrease of the DW velocity, v, and DW mobility, S. Quite fast DW propagation (v until 2,500 m/s at H about 30 A/m) has been observed in low magnetostrictive magnetically bistable Co56Fe8Ni10Si10B16 microwires. Consequently, we observed certain correlation between the magnetoelastic energy and DW dynamics in microwires: decreasing the magnetoelastic energy, K (me), DW velocity increases.

This paper studies the feasibility of calculating strains in aged F114 steel specimens with Fiber Bragg Grating (FBG) sensors and infrared thermography (IT) techniques. Two specimens have been conditioned under extreme temperature and relative humidity conditions making comparative tests of stress before and after aging using different adhesives. Moreover, a comparison has been made with IT tecniques and conventional methods for calculating stresses in F114 steel. Implementation of Structural Health Monitoring techniques on real aircraft during their life cycle requires a study of the behaviour of FBG sensors and their wiring under real conditions, before using them for a long time. To simulate aging, specimens were stored in a climate chamber at 70 degrees C and 90% RH for 60 days. This study is framed within the Structural Health Monitoring (SHM) and Non Destructuve Evaluation (NDE) research lines, integrated into the avionics area maintained by the Aeronautical Technologies Centre (CTA) and the University of the Basque Country (UPV/EHU).

[EU]Hurrengo orrialdeetan aurkezten den proiektu honetan, lehiaketa motor baten direkzio piparen diseinua proposatzen da zeinak Moto Engineering Foundation fundazioak sustatutako nazioarteko Motostudent txapelketan parte hartzeko motorrera egokituta egon beharko den. Honako helburu honekin beraz, direkzio piparen diseinu egoki bat egin ahal izateko piezak sufrituko dituen karga egoerak aurreikusi beharko ditugu hauen arabera jasan beharreko tentsio egoerak aztertu ahal izateko. Piezaren tentsioen analisia aurrera eraman beharko da beraz, diseinaturiko piezaren funtzionamendu egokia ziurtatu ahal izateko. Historian zehar existitutako diseinu modelo ezberdinak eta baita gaur egunean gehien erabilitakoak ere aurkeztu eta gero, gure kasura hobekien moldatuko zaion adibidetik ekingo zaio diseinuari, aldez aurretik eskuratutako datu eta errodamenduetatik abiatuz. Horrenbestez, oinarritzat hartutako direkzio sistema modeloa eta bestelako elementuen neurriak datutzat hartuta, tija zein norabide ardatzarena esate baterako, direkzio piparen hasierako geometria definituko da. Eskuragarri ditugun analisi metodo ezberdinak aztertu eta gure kasura hobekien moldatzen dena aukeratu eta eraikitako piezari aplikatuko zaio. Lorturiko emaitzen arabera hasiera batean planteaturiko diseinua onartu edo baztertzea erabaki beharko dugu. Beraz, gure diseinua optimizatuz joateko aukera izango dugu, baldintzak eta betebeharrak egiztatuko dituen amaierako pieza diseinua lortu arte. Bukatzeko, proiektuari itxiera emango dion atala aurkeztuko da, hala nola, proiektuaren garapenean zehar lorturiko emaitzak eta ondorioak biltzen dituen atala non lorturiko direkzio piparen funtzionamendu egokia bermatzen dituzten parametroak aurkeztuko diren. Gainera, direkzio sistemaren funtsezko atala izango diren errodamenduen kalkulu eta aukeraketa bideratuko da baita ere.

Background: Staphyloccocal nuclease domain-containing protein 1 (SND1) is involved in the regulation of gene expression and RNA protection. While numerous studies have established that SND1 protein expression is modulated by cellular stresses associated with tumor growth, hypoxia, inflammation, heat- shock and oxidative conditions, little is known about the factors responsible for SND1 expression. Here, we have approached this question by analyzing the transcriptional response of human SND1 gene to pharmacological endoplasmic reticulum (ER) stress in liver cancer cells. Results: We provide first evidence that SND1 promoter activity is increased in human liver cancer cells upon exposure to thapsigargin or tunicamycin or by ectopic expression of ATF6, a crucial transcription factor in the unfolded protein response triggered by ER stress. Deletion analysis of the 5'-flanking region of SND1 promoter identified maximal activation in fragment (-934, +221), which contains most of the predicted ER stress response elements in proximal promoter. Quantitative real- time PCR revealed a near 3 fold increase in SND1 mRNA expression by either of the stress- inducers; whereas SND1 protein was maximally upregulated (3.4-fold) in cells exposed to tunicamycin, a protein glycosylation inhibitor. Conclusion: Promoter activity of the cell growth- and RNA-protection associated SND1 gene is up-regulated by ER stress in human hepatoma cells.

[ES]Este trabajo pretende realizar el diseño de las tijas y del acople de freno para una moto de competición. El objetivo principal es conseguir un diseño óptimo de las tijas y del acople del freno. Para ello se realiza una investigación previa, se estudian las solicitaciones a las que se ven sometidas las piezas y se diseñan con un programa de CAD/MEF (PTC Creo 2.0) con su respectivo análisis de resultados. Con todo esto se pretende la posibilidad de aportar un diseño novedoso y eficaz de las tres piezas para el proyecto MotoStudent o para cualquier otro proyecto.

[EU]Lan honetan, surf oholetan erabiltzen diren gila mota desberdinen analisia egin da. Horretarako, merkatuan garrantzi handiena duten bi enpresek euren katalogoetan eskaintzen dituzten modelo batzuk aztertu dira. Ingeniaritza simulazioak ahalbidetzen dituen ansys softwarea erabiliz egin da aukeratutako modeloen diseinu geometrikoa. Hauen analisiari ekin baino lehenago, gila hauetako bat aztertu da, hau osatzen duen materiala ezagutu eta ezaugarriak zehaztu asmoz. Aztertutakoa modelo guztiek erabiltzen duten ohiko materiala izanik, lortutako emaitzak baliogarriak dira gila guztiak aztertzeko. Materialaren propietateak ezagututa eta aipatutako softwareaz baliatuz, analisi estatikoa egin da. Horretarako, surfean aurkitzen diren ohiko egoera batzuk irudikatu dira, hauetan agertzen diren esfortzuak kalkulatuz. Gila bakoitzaren portaera, jasandako tentsio eta deformazioak aztertuz ondorioztatu da. Horrela, kasu bakoitzean komenigarrienak diren ezaugarriak oinarri hartuz, modelo desberdinen arteko konparaketa egin da.

[EU]Proiektu honen bidez, laser bidezko tenplaketaren ondoriozko piezen gainazalen distortsioaren neurketa kuantitatiboa eta tratamendu honegatik eratorri daitezkeen hondar tentsioen azterketa egin nahi izan da. Hauxe burutzeko hondar tentsioen neurketarako Contour metodoa erabili da, elektro-higadurako ebaketa eginez. IGET Bilboko mekanikako tailerreko ingurugiroan eta bertako langileen inbestigazio lerroetan barneratzeko helburuz, laser tenplaketa tratamenduen azterketan lagungarria izateko xedearekin aurrera eraman da proiektu hau.

Background: The impact of nano-scaled materials on photosynthetic organisms needs to be evaluated. Plants represent the largest interface between the environment and biosphere, so understanding how nanoparticles affect them is especially relevant for environmental assessments. Nanotoxicology studies in plants allude to quantum size effects and other properties specific of the nano-stage to explain increased toxicity respect to bulk compounds. However, gene expression profiles after exposure to nanoparticles and other sources of environmental stress have not been compared and the impact on plant defence has not been analysed. Results: Arabidopsis plants were exposed to TiO2-nanoparticles, Ag-nanoparticles, and multi-walled carbon nanotubes as well as different sources of biotic (microbial pathogens) or abiotic (saline, drought, or wounding) stresses. Changes in gene expression profiles and plant phenotypic responses were evaluated. Transcriptome analysis shows similarity of expression patterns for all plants exposed to nanoparticles and a low impact on gene expression compared to other stress inducers. Nanoparticle exposure repressed transcriptional responses to microbial pathogens, resulting in increased bacterial colonization during an experimental infection. Inhibition of root hair development and transcriptional patterns characteristic of phosphate starvation response were also observed. The exogenous addition of salicylic acid prevented some nano-specific transcriptional and phenotypic effects, including the reduction in root hair formation and the colonization of distal leaves by bacteria. Conclusions: This study integrates the effect of nanoparticles on gene expression with plant responses to major sources of environmental stress and paves the way to remediate the impact of these potentially damaging compounds through hormonal priming.
