Techniques are described for preparing acetate peels of sectioned valves of ocean quahogs, Arctica islandica, for age determinations. The respective sequence of preparation begins by sectioning left valves oriented to include a single hinge tooth, bleaching to remove the heavy periostracum, embedding the valves in an epoxy resin, grinding and polishing the embedments to a high luster, etching the exposed cut valve surfaces, and applying sheet acetate with acetone. Annuli are clearly defined relative to growth increments in the peel preparations for all sizes and ages of ocean quahogs. (PDF file contains12 pages.)

Three types of prototype (Lege) traps with different numbers of entrance valves were evaluated in River Rima, north western Nigeria. The traps contained 4,6 and 8 valves, tagged 4-V, 6-V and 8-V respectively. The experiment was carried out in a randomized complete block design with one factor each replicated three times. Data collected on fish diversity, number biomass and sizes were subjected to descriptive statistics and analysis of variance. The results of the catch composition showed close diversity index of 0.86 for 6-V, 0.80 for 8-V and 0.60 for 4-V Lege traps. However, the number (41%) and biomass 48%) of fish caught in the 6-V Lege trap were significantly (P0.05) higher than those caught in the other traps. There was no definite trend in the sizes (length and girth) of fish caught in the traps. On the basis of species diversity, and the number and biomass of fish caught, the 6-V Lege showed preference for adoption than the other two traps. However, further studies are recommended on the appropriate mesh size net for the trap in line with the provisions of fisheries edicts

Taxonomic descriptions, line drawings, and references are given for the 30 named and 5 unnamed species of North American fish Eimeriidae. In addition, a key was developed based on available morphologic data to distinguish between similar species. Taxa are divided into two genera: Eimeria (27 species) which are tetr&sporocystic with dizoic, nonbivalved sporocysts, and Goussia (3 species) which are tetrasporocystic with dizoic, bivalved sporocysts that lack Stleda bodies and have sporocyst walls composed of two longitudinal valves. Based on the findings and diagnostic survey, a new fishing trap, christened Lege trap was designed and fabricated, and the performance evaluated. The 8-valve Lege trap was assessed concurrently with Malian and Ndurutu traps by the fishermen in the study area. The experiment was conducted in a randomized complete block design with one factor each replicated three times. Data collected on fish diversity number, biomass and size were subjected to descriptive statistics and analysis of variance. The results showed that 22 fish species belonging to thirteen families were caught. The prototype (Lege) trap recorded higher species diversity index (0.90) than the Malian (0.50) and Ndurutu (0.50) traps. The Lege trap also accounted for the largest number (55%) and biomass (63%) of fish caught which was significantly (P<0.05) higher than those of the Malian and Nduutu traps were. The mean length (15.03~c5.70cm), weight (60.43~c48.61g) and girth (4.77~c1.65cm) of fishes caught in the Lege trap were also significantly (P0.05) higher than those caught in the other two traps. These results demonstrated better performance of the new trap than the two conventional traps, even though the sizes of some of the fish species caught in all the traps were below those allowed by the Sokoto State Fisheries Edict where the study was conducted. Therefore, since it is desirable to develop conservation-oriented trap at a least cost, it is necessary to research further on the number of valves and mesh size of the new trap

One of the supposed effects of the observed ozone depletion is the increase of solar UV-B irradiation at the seasurface. This will cause an impact on certain compartments of marine ecosystems. Especially, sensitive developmental stages of pelagic fish embryos might be affected. Embryos of dab (Limanda limanda) and plaice (Pleuronectes plalessa) were experimentally exposed 10 different amounts of UVB irradiation in a sunshine simulator. This programmable device allows the dosage of realistic solar irradiation in quality and guantity. Experiments were carried out in March 1995 and February 1996. Either artificially inserninated and reared emhryos of dab and plaice or embryos caught in the German Bight were exposed to simulated solar irradiation. The 1995 experiments served to identify the effective irradiation dosages. For the 1996 experiments irradiation applied was much lower, being dose to realistic valucs expected over the North Sea as a consequence of ozone depletion. The following end points were studied: 1. Mortality, 2. sublethal morphological effects (malformations), 3. DNA damage, 4. changes in buoyancy of embryos measured as changes in osmolarity of the perivitelline fluid. Conditions for the simulation of daylight were a c1oudless sky with a solar zenith distance of 34 % (air mass 1.2). The adopted ozone depletion was 40 % corresponding to 180 DU (Dobson Units) instead of 300 DU. In the 1995 experiments time and dosage dependent influenccs on mortality and buoyancy of embryos of dab and plaice were found. Even in those embryos which were protected from the UV-B spectral range a loss of buoyancy was registered after 12 hours in the simulator. No diffcrences in DNA integrity as determined by DNA unwinding of exposed and control embryos were found. Also with lower amounts of irradiation in the 1996 experiments dosage dependent acute mortality, malformations, and impact on the buoyancy of the emhryos was registered. Sublethal effects occurred as well in embryos protected against UV-B in the exposure chambers, but were not found in the dark controls. The impact of low dosages of UV-B on the buoyancy of pelagic fish embryos might indicate an important ecological threat and deserves further studies.

The localization of the stretch sensitivity and myogenic automatism in the systemic heart of Octopus vulgaris has been studied on an isolated preparation in which the ventricle was zoned by ligatures. Each region has been submitted to two different levels of internal hydrostatic pressure (1 and 2 kPa). Only the two atrio-ventricular regions were able to contract regularly when submitted to internal pressure, with a frequency dependent from the pressure value, while the ventricle-aortic region was insensitive to the stretching by internal pressure. This result supports the hypothesis that the automatism in this heart is localized. Electrocardiogram recordings from different areas of an isolated and perfused preparation of the systemic heart ventricle are also reported, which suggest that the electrical activity of the ventricle originates in two narrow areas near the atrio-ventricular valves.

Cryogenic preservation trials of spermatozoa of Labeo rohita were carried out. Twenty four cryodiluents (extender + cryoprotectant), with the combination of six extenders such as egg-yolk citrate, urea-egg-yolk, 0.9% NaCl, Kurokura-2, Ma and Mb and four cryoprotectants viz. DMSO, glycerol, methanol and ethanol, were used to screen out the suitable cryodiluents. Sperm was preserved in 0.25ml plastic straw in programmable freezer. Two step freezing method was followed. Sperm preserved with egg-yolk citrate and urea-egg-yolk containing 10% DMSO showed best post-thaw motility (80%) followed by 0.9% NaCl (60%) and Kurokura-2(30%) solutions. Sperm with the extenders M" and Mb clotted at the time of equilibration and also after few days of preservation. Egg-yolk citrate mixed with ethanol and methanol also showed good percentage of motility (80%) but egg-yolk citrate with glycerol showed less sperm motility (>60%). To determine suitable dilution ratio of milt and cryodiluent two best extender eggyolk citrate and urea-egg-yolk with four cryoprotectants such as DMSO, glycerol, methanol and ethanol at different ratio viz 1:2,1:4,1:7,1:10,1:15 and 1:20 were used. Highest post-thaw motility (>80%) was observed when milt was preserved with egg-yolk citrate containing 10% DMSO at 1:2, 1:4, 1:7 and 1:10 dilutions. Meanwhile using glycerol as cryoprotectants provided less post thaw motility at lower dilution ratio but with the increase of its dilution showed good sperm motility compared with other cryoprotectants. Finally, evaluation on the effect of cryoprotectant concentration on post-thaw sperm motility was conducted. Egg-yolk citrate and four cryoprotectant i.e. DMSO, glycerol, methanol and ethanol with six different concentrations namely 5%,7%, 10%, 15%, 20% and 30%.were evaluated. Among the cryoprotectants DMSO, methanol and ethanol showed highest post-thaw motility (about 80%) at 7% and 10% concentrations. Although glycerol was not suitable at low concentration but its 20% and 30% concentration levels provided best post-thaw motility. No post-thaw motility was obtained with DMSO at 30% concentration. The overall analysis on cryoprotectant concentration indicated that below 5% and above 20% cryoprotectant concentrations could not be suitable for effective cryopreservation of spermatozoa.

96h acute toxicity tests were performed using commercial grade metasystox on the marine wedge clam, Donax cuneatus during summer 1985. The behaviour and mortality rates were recorded periodically. Most of the dams responded in opening the shell valves and extending the siphons quicker in low test concentrations (0.004-0.0052 p.p.m) but this was slow and late in high concentrations (0.0056-0.008 p.p.m). Mortality began to occur in 0.008 p.p.m. from 12 h, whereas, in 0.0052 p.p.m. from 60 h onwards. The observed LC sub(0) value was 0.004 p.p.m. and LC sub(50) 0.0064 p.p.m. The regression equation established was Y = 79.0891 + 33.4523 X. The rate of oxygen concentration increased at LC sub(0) and LC sub(50) values compared to control indicating the disturbed physiological adjustment. The results are correlated with physico-chemical parameters of seawater and discussed in the light of pesticide toxicity to the dam.
