Description of a simple method for counting bacteria with active electron transport systems in water and sediment samples. Sodium succinate, NADH and NADPH served as electron donors. It is possible to see several sites of electron transport in the larger cells. Especially impressive are the plankton-algae, protozoa, and small metazoa. Increased boating activities and new waterfront developments have contributed an estimated 3,000 dismantled, abandoned, junked, wrecked, derelict vessels to Florida coastal waters. This report outlines a method of siting and prioritizing derelict vessel removal using the Florida Keys as a test area. The data base was information on 240 vessels, obtained from Florida Marine Patrol files. Vessel location was plotted on 1:250,000 regional and 1:5,000 and 1:12,000 site maps. Type of vessel, length, hull material, engine, fuel tanks, overall condition, afloat and submerged characteristics, and accessibility, were used to derive parametric site indices of removal priority and removal difficulty. Results indicate 59 top priority cases which should be the focus of immediate clean up efforts in the Florida Keys. Half of these cases are rated low to moderate in removal difficulty; the remainder are difficult to remove. Removal difficulty is a surrogate for removal cost: low difficulty -low cost, high difficulty - high cost. The rating scheme offers coastal planners options of focusing removal operations either on (1) specific areas with clusters of high priority derelict vessels or on (2) selected targeted derelicts at various, specific locations. There is a clear need to develop fisheries independent methods to quantify individual sizes, density, and three dimensional characteristics of reef fish spawning aggregations for use in population assessments and to provide critical baseline data on reproductive life history of exploited populations. We designed, constructed, calibrated, and applied an underwater stereo-video system to estimate individual sizes and three dimensional (3D) positions of Nassau grouper (Epinephelus striatus) at a spawning aggregation site located on a reef promontory on the western edge of Little Cayman Island, Cayman Islands, BWI, on 23 January 2003. The system consists of two free-running camcorders mounted on a meter-long bar and supported by a SCUBA diver. Paired video “stills” were captured, and nose and tail of individual fish observed in the field of view of both cameras were digitized using image analysis software. Conversion of these two dimensional screen coordinates to 3D coordinates was achieved through a matrix inversion algorithm and calibration data. Our estimate of mean total length (58.5 cm, n = 29) was in close agreement with estimated lengths from a hydroacoustic survey and from direct measures of fish size using visual census techniques. We discovered a possible bias in length measures using the video method, most likely arising from some fish orientations that were not perpendicular with respect to the optical axis of the camera system. We observed 40 individuals occupying a volume of 33.3 m3, resulting in a concentration of 1.2 individuals m–3 with a mean (SD) nearest neighbor distance of 70.0 (29.7) cm. We promote the use of roving diver stereo-videography as a method to assess the size distribution, density, and 3D spatial structure of fish spawning aggregations.

The procedure to conduct horizontal starch gel electrophoresis on enzymes is described in detail. Areas covered are (I) collection and storage of specimens, (2) preparation of tissues, (3) preparation of a starch gel, (4) application of enzyme extracts to a gel, (5) setting up a gel for electrophoresis, (6) slicing a gel, and (7) staining a gel. After having described in our first paper (Haase et al. 2005) the main features of an easy and fast method for determination of carbon monoxide in fish and the equipment and chemicals necessary as well as first results measured on fish samples, this part deals with the influence of sample preparation, variation of the size of samples, type of solvent, duration of extraction and further conditions on the result of analyses. Both variants of the method are evaluated with regard to measuring expected errors. The single components of the equipment, including prises, are listed to allow a reliable assessment of costs. Additional instrumental colour and DSC measurements on both untreated and CO-treated tuna illustrate the effects.

The present work appertains to a series of investigations in the field of trophodynamics of water bodies conducted by the department of hydrobiology ”Mosrybvtuza” under the leadership of Prof. Director N.S. Gaevska. Usually with quantitative collections of benthos ostracoda are hardly taken account of. But with the employment of special methods it is possible to be convinced that they are encountered in massive numbers. The role of ostracods in the nutrition of fish is recognised as important by many authors. On the question of the feeding of ostracods, in the literature there are only the fragmentary indications about the occurrence in the gut of ostracods of algae (diatoms, desmids) and detritus. The study of the feeding of ostracods was conducted both by the method of dissecting the guts of ostracods taken from natural waters, and by the method of feeding ostracods under laboratory conditions with a variety of food objects. Moreover, with a view to revision of obtained quantitative data, for determination of food requirements of ostracods the respiration method was applied.

Changes in management practices and agricultural productivity over the past twenty years have lead to nitrate pollution and eutrophication of lakes and rivers. Information on nitrate concentrations and discharge has been collected on the River Frome at East Stoke since 1965, using the same analytical nitrate method so that the results are comparable. These records of weekly spot values of nitrate concentration and daily mean discharges have been analysed for trends and seasonal patterns in both concentration and nitrate loadings. In this extension of our nitrate contract, a new automated method of intensive sampling has been used to monitor short-term variability and to assess how well similar routine (weekly) sampling schemes can represent the true nitrate record.

Ethmalosa growth curves (calculated by the least squares method) were determined from weekly samplings in Ebrié Lagoon. In order to obtain more accurate results than with a modal decomposition, the author used directly the modal values of the samples. One-year-old ethmalosa is about 15 cm long (fork length). For older fish, growth data seem to be disturbed by migrations: fish measuring >25 cm do not appear in the lagoon. Ethmalosa would spend the first year of its life in the lagoon, where it hatches and reproduces, and would migrate to the sea during its second year.

The long-snouted seahorse (Hippocampus guttulatus) (Cuvier, 1829), was used to validate the pre-dictive accuracy of three progressively realistic models for estimating the realized annual fecundity of asyn-chronous, indeterminate, multiple spawners. Underwater surveys and catch data were used to estimate the duration of the reproductive season, female spawning frequency, male brooding frequency, and batch fecun-dity. The most realistic model, a generalization of the spawning fraction method, produced unbiased estimates of male brooding frequency (mean ±standard deviation [SD]=4.2 ±1.6 broods/year). Mean batch fecundity and realized annual fecundity were 213.9 (±110.9) and 903.6 (±522.4), respectively. However, females prepared significantly more clutches than the number of broods produced by males. Thus, methods that infer spawning frequency from patterns in female egg production may lead to significant overestimates of realized annual fecundity. The spawning fraction method is broadly applicable to many taxa that exhibit parental care and can be applied nondestructively to species for which conservation is a concern.

Length frequency data of six sciaenids (Johnius macrorhynus, J. vogleri, Otolithes cuvieri, J. sina, Pennahia macrophthalamus, J. dussumieri) were collected from shrimp trawlers at New Ferry Wharf and Sasson Docks landing centers off Greater Mumbai (India). Growth parameters of these species were analyzed via modal progression analysis using Bhattacharya's method. Natural mortality (m) was estimated using Cushing's formula. Comparison of growth parameters was done using the 0' index. The growth parameters obtained were compared with the results of earlier growth studies which used other techniques. This study concludes that the growth parameters obtained are consistent with earlier estimates.

In many developing countries, availability of suitable tags for use in genetics and aquaculture research has been a problem as they are often expensive and have to be imported from other countries. A simple and inexpensive method used by scientists in Ghana that can be tried and improved by others is described in this article.

The snakehead (Channa striata ) is a common freshwater fish species in Malaysia. Details are given of a simple technique for breeding this species, suitable for small-scale farmers practising backyard aquaculture. Two techniques were used to induce spawning - the first used water level manipulation to simulate rain and the second used injected with human chorionic gonadotropic hormone. The former, more natural, spawning technique was found to provide a viable alternative for the small-scale farmer, being much simpler than hormone injection.

The lengths of otoliths and other skeletal structures recovered from the scats of pinnipeds, such as Steller sea lions (Eumetopias jubatus), correlate with body size and can be used to estimate the length of prey consumed. Unfortunately, otoliths are often found in too few scats or are too digested to usefully estimate prey size. Alternative diagnostic bones are frequently recovered, but few bone-size to prey-size correlations exist and bones are also reduced in size by various degrees owing to digestion. To prevent underestimates in prey sizes consumed techniques are required to account for the degree of digestion of alternative bones prior to estimating prey size. We developed a method (using defined criteria and photo-reference material) to assign the degree of digestion for key cranial structures of two prey species: walleye pollock (Theragra chalcogramma) and Atka mackerel (Pleurogrammus monopterygius). The method grades each structure into one of three condition categories; good, fair or poor. We also conducted feeding trials with captive Steller sea lions, feeding both fish species to determine the extent of erosion of each structure and to derive condition-specific digestion correction factors to reconstruct the original sizes of the structures consumed. In general, larger structures were relatively more digested than smaller ones. Mean size reduction varied between different types of structures (3.3−26.3%), but was not influenced by the size of the prey consumed. Results from the observations and experiments were combined to be able to reconstruct the size of prey consumed by sea lions and other pinnipeds. The proposed method has four steps: 1) measure the recovered structures and grade the extent of digestion by using defined criteria and photo-reference collection; 2) exclude structures graded in poor condition; 3) multiply measurements of structures in good and fair condition by their appropriate digestion correction factors to derive their original size; and 4) calculate the size of prey from allometric regressions relating corrected structure measurements to body lengths. This technique can be readily applied to piscivore dietary studies that use hard remains of fish.

The proper matching of the pull exerted by a trawler and the size of trawl is important for maximizing the catching efficiency. The available pull is more dependent on the propeller and its working conditions than the installed engine power. The normal practice is to directly connect net size to the installed power in the boat by formulae without reference to the prope1ler dimensions or the available trawling pull and this is not adequate to find out the optimum combination. By the method outlined in this paper, the accurate calculation of trawling pull is possible by taking into account only the propeller diameter, pitch and r. p. m. The predictions by the method are compared for trawlers with powers between 30 and 60 hp and agreement is found to be within + 5%. The power absorbed by the propeller in trawling condition can also be calculated by this method for checking whether the engine is being overloaded.

The effects of temperature and relative humidity on the rate of drying of split open fish and salted fish in a tunnel dryer have been studied at a constant air velocity. By a judicious combination of these two, the rate of drying could be considerably accelerated, 10 to 12 hours only being required for drying to moisture levels below 30% in the case of mackerel, lactarius, otolithes and kilimeen (Nemipterus japonicus)
