The original method, proposed by Yentsch (1957), of determination of chlorophyll directly in the cells, attracts attention by its simplicity. In order to measure the content of chlorophyll by this method, a determined volume of suspension of algae is filtered through a membrane filter. The latter is dried a little, clarified by immersion oil, clamped between two glasses, and spectrophotometrized. Extinction is read off at , wavelengths equal to 670 millimicrons (around the maximum absorption of chlorophyll a in the cell) and 750 millimicrons (correction for non- specific absorption and dispersion of light by particles of the preparation). The method of Yentsch was employed by the authors for determination of chlorophyll-a in samples of phytoplankton. They conclude that in spite of the simplicity and convenience of determination the method must be applied sufficiently carefully. It is more suitable for analysis of cultures of algae, where, non-specific absorption of light is insignificant.

ENGLISH: In this paper, a method of analysis described by Gulland (1963) has been used to estimate the fishing mortality rates of tagged yellowfin and skipjack tuna for specific areas and years. Fishing mortality rates obtained for tagged tunas will also represent those for the entire population from which the tagged fishes were drawn, provided the assumptions used and corrections made for these analyses are valid. Total mortality rates of tagged fishes have also been computed. These are not assumed to be directly equivalent to the total mortality rates of the untagged populations, since tagged fishes are subject to additional types of attrition. These additional sources of mortality are also examined in this study.

ENGLISH: Age composition of catch, and growth rate, of yellowfin tuna have been estimated by Hennemuth (1961a) and Davidoff (1963). The relative abundance and instantaneous total mortality rate of yellowfin tuna during 1954-1959 have been estimated by Hennenmuth (1961b). It is now possible to extend this work, because more data are available; these include data for 1951-1954, which were previously not available, and data for 1960-1962, which were collected subsequent to Hennemuth's (1961b) publication. In that publication, Hennemuth estimated the total instantaneous mortality rate (Z) during the entire time period a year class is present in the fishery following full recruitment. However, this method may lead to biased estimates of abundance, and hence mortality rates, because of both seasonal migrations into or out of specific fishing areas and possible seasonal differences in availability or vulnerability of the fish to the fishing gear. Schaefer, Chatwin and Broadhead (1961) and Joseph etl al. (1964) have indicated that seasonal migrations of yellowfin occur. A method of estimating mortality rates which is not biased by seasonal movements would be of value in computations of population dynamics. The method of analysis outlined and used in the present paper may obviate this bias by comparing the abundance of an individual yellowfin year class, following its period of maximum abundance, in an individual area during a specific quarter of the year with its abundance in the same area one year later. The method was suggested by Gulland (1955) and used by Chapman, Holt and Allen (1963) in assessing Antarctic whale stocks. This method, and the results of its use with data for yellowfin caught in the eastern tropical Pacific from 1951-1962 are described in this paper.

The radioautographic method of determination of the number of autotrophic microorganisms was initially suggested for counting methane-oxidizing bacteria. With the help of this method colonies of hydrogen-oxidizing bacteria are differentiated even more clearly from heterotrophic. Under laboratory conditions it was shown that colonies grown on membrane filters from a pure culture of thionic bacteria on a nutrient medium with radio- active carbonate, give better prints on film. This method was tested by the authors for determining the number of these bacteria in the meromictic Lake Vae de San Juan during the expedition to Cuba in the summer of 1973. The study showed that that the thionic bacteria are found throughout the pelagial. It proved that the thionic bacteria can be well considered in water-bodies by the radioautographic method.

The proper matching of the pull exerted by a trawler and the size of trawl is important for maximizing the catching efficiency. The available pull is more dependent on the propeller and its working conditions than the installed engine power. The normal practice is to directly connect net size to the installed power in the boat by formulae without reference to the prope1ler dimensions or the available trawling pull and this is not adequate to find out the optimum combination. By the method outlined in this paper, the accurate calculation of trawling pull is possible by taking into account only the propeller diameter, pitch and r. p. m. The predictions by the method are compared for trawlers with powers between 30 and 60 hp and agreement is found to be within + 5%. The power absorbed by the propeller in trawling condition can also be calculated by this method for checking whether the engine is being overloaded.

The effects of temperature and relative humidity on the rate of drying of split open fish and salted fish in a tunnel dryer have been studied at a constant air velocity. By a judicious combination of these two, the rate of drying could be considerably accelerated, 10 to 12 hours only being required for drying to moisture levels below 30% in the case of mackerel, lactarius, otolithes and kilimeen (Nemipterus japonicus)

An experimental procedure along with a method of analysis to judge the suitability of an individual to be included in a taste panel is described. The procedure is based on comparison of the organoleptic scores assigned by the individual to pairs of fish samples whose qualities are known and a set of physical measurements of the same samples. Fisher's Exact Probability Test provides a criterion for the judgement.

The recognition of individual animals is crucial to many aspects of research. Prawns (Penaeus monodon) present unique difficulties in this respect since they molt regularly. Thus, almost all tagging and marking methods developed for prawns so far have proven inadequate. Some tags or marks are lost during molting; others cause injury to the prawns. A new and efficient method has been developed at the Igang Seafarming Station of the Aquaculture Department. Rectangular brass tags measuring 5 mm by 20 mm and numbered consecutively are used. The prawn is held gently but firmly at the base of the carapace with the left hand while the right hand slips the brass tag around the stalk of the unablated eye and presses the tag gently. All tagging must be made under water to avoid stress. From May 29 to September 7 1977 to a total of 348 unilaterally-ablated adult female prawns were tagged on the unablated eyestalk in 5 batches to enable individual observations on gonadal maturation, molting, and growth. Periodic examinations were made four times a month to coincide with the different phases of the lunar cycle. On each examination, survival and recovery rates were recorded. The data included death due to immediate mortality during ablation and loss to cannibalism for the duration of the experiments. In all five tagging experiments, most of the prawns recovered had their tags intact. These included even dead and molting animals. The eyestalk tagging method is suitable for prawns because the tags can be attached without causing injury and has no effect on the rate of growth, maturity, molting and behavior of the animal. The tags are identifiable and permanent; they remain attached to the animal even after death.

The importance of studies on the biology of marine foulers and borers needs no emphasis since they cause damage to submerged wooden structures resulting in considerable economic loss. In order to study their occurrence, settlement and to assess the rate of destruction to timber, several immersion techniques using iron racks, cages and rafts have been adopted. On the east and west coasts of India, at the Marine Centres of Forest Research Institute (Waltair, Madras, Cohin and Bombay) investigations have so far been carried out employing the first two methods.
