Amino functionalised mesoporous silica nanoparticles (AM-41) have been identified as a promising vaccine delivery material. The capacity of AM-41 to stabilise vaccine components at ambient temperature (23–27 °C) was determined by adsorbing the model antigen ovalbumin (OVA) to AM-41 particles (OVA-41). The OVA-41 was successfully freeze-dried using the excipients 5% trehalose and 1% PEG8000. Both the immunological activity of OVA and the nanoparticle structure were maintained following two months storage at ambient temperature. The results of immunisation studies in mice with reconstituted OVA-41 demonstrated the induction of humoral and cell-meditated immune responses. The capacity of AM-41 particles to facilitate ambient storage of vaccine components without loss of immunological potency will underpin the further development of this promising vaccine delivery platform.

The changes occurring in water and salt extractable protein and non-protein fractions in prawn muscle of different species during freezing, freeze drying and subsequent prolonged storage have been studied. There is no denaturation of water extractable proteins, whereas salt extractable proteins were rendered insoluble to the extent of 21% due to freeze drying. The freeze dried products remained in good edible condition for 32 months of storage up to which storage characteristics were followed.

Behaviour of freeze dried ready-to-serve fish based food preparations during prolonged storage at room temp has been studied and reported in this paper. Storage life of such products under the conditions employed in this study can be reckoned in years at our ambient temps.

MTDSC is a software modification of the traditional DSC thermal analysis technique that allows more accurate determination of the glass transition as well as measurement of the endothermic relaxation that often accompanies the transition. The glass transition is an essential parameterboth of the original frozen solution and of the end product. Measurement of endothermic relaxation allows the determination of molecularrelaxation times in the freeze-dried product that may be useful in predicting the effect of formulation variables and storage conditions on physical stability.

To formulate therapeutic proteins into polymeric devices the protein is typically in the solid state, which can be achieved by the process of freeze-drying. However, freeze-drying not only risks denaturing the protein but it can adversely affect the cure characteristics of protein-loaded silicone elastomers. This study demonstrates that a variation in the parameters of the freeze-dryer can significantly affect the residual moisture content of freeze-dried BSA, which in turn has an effect on the bulk density and flow properties of the BSA. The bulk density and flow properties of the BSA subsequently affect the cure characteristics of BSA-loaded silicone elastomers. An increase in the residual moisture content results in the freeze-dried BSA having a decreased bulk density and poor flow properties which can have a detrimental effect on the cure characteristics of a freeze-dried BSA-loaded silicone elastomer. © 2012 Wiley Periodicals, Inc.
