Um total de 281 espécimes de peixes ornamentais de água doce - das espécies Leporacanthicus galaxias, Lasiancistrus saetiger, Cochliodon sp., Hypostomus sp., Pseudacanthicus spinosus, Ancistrus sp. e Rineloricaria cf. lanceolata - foram capturados na bacia hidrográfica do rio Guamá, Pará, Brasil. A infecção por Trypanosoma spp. foi inspecionada. A caracterização morfológica e morfométrica dos parasitas e os parâmetros hematológicos foram determinados. Todas as espécies foram infectadas e todos os espécimes de Leporacanthicus galaxias e Pseudacanthicus spinosus estavam parasitados. As outras espécies mostraram uma prevalência variável da infecção. Os parasitas mostraram claramente morfotipos e dimensões diferentes, e provavelmente, pertencem a espécies diferentes. A resposta hematológica à infecção variou de acordo com o hospedeiro. Em Cochliodon sp. não houve diferença entre peixes infectados e não infectados. Em outras espécies diversas modificações em alguns parâmetros hematológicos foram encontrados, mas aparentemente sem causar doença. Ressalta-se a possibilidade de introdução de parasitas em novos ambientes devido aos movimentos artificiais destes peixes ornamentais.

Trypanosomiasis has been identified as a neglected tropical disease in both humans and animals in many regions of sub-Saharan Africa. Whilst assessments of the biology of trypanosomes, vectors, vertebrate hosts and the environment have provided useful information about life cycles, transmission, and pathogenesis of the parasites that could be used for treatment and control, less information is available about the effects of interactions among multiple intrinsic factors on trypanosome presence in tsetse flies from different sites. It is known that multiple species of tsetse flies can transmit trypanosomes but differences in their vector competence has normally been studied in relation to individual factors in isolation, such as: intrinsic factors of the flies (e.g. age, sex); habitat characteristics; presence of endosymbionts (e.g. Wigglesworthia glossinidia, Sodalis glossinidius); feeding pattern; host communities that the flies feed on; and which species of trypanosomes are transmitted. The purpose of this study was to take a more integrated approach to investigate trypanosome prevalence in tsetse flies. In chapter 2, techniques were optimised for using the Polymerase Chain Reaction (PCR) to identify species of trypanosomes (Trypanosoma vivax, T. congolense, T. brucei, T. simiae, and T. godfreyi) present in four species of tsetse flies (Glossina austeni, G. brevipalpis, G. longipennis and G. pallidipes) from two regions of eastern Kenya (the Shimba Hills and Nguruman). Based on universal primers targeting the internal transcribed spacer 1 region (ITS-1), T. vivax was the predominant pathogenic species detected in flies, both singly and in combination with other species of trypanosomes. Using Generalised Linear Models (GLMs) and likelihood ratio tests to choose the best-fitting models, presence of T. vivax was significantly associated with an interaction between subpopulation (a combination between collection sites and species of Glossina) and sex of the flies (X2 = 7.52, df = 21, P-value = 0.0061); prevalence in females overall was higher than in males but this was not consistent across subpopulations. Similarly, T. congolense was significantly associated only with subpopulation (X2 = 18.77, df = 1, P-value = 0.0046); prevalence was higher overall in the Shimba Hills than in Nguruman but this pattern varied by species of tsetse fly. When associations were analysed in individual species of tsetse flies, there were no consistent associations between trypanosome prevalence and any single factor (site, sex, age) and different combinations of interactions were found to be significant for each. The results thus demonstrated complex interactions between vectors and trypanosome prevalence related to both the distribution and intrinsic factors of tsetse flies. The potential influence of the presence of S. glossinidius on trypanosome presence in tsetse flies was studied in chapter 3. A high number of Sodalis positive flies was found in the Shimba Hills, while there were only two positive flies from Nguruman. Presence or absence of Sodalis was significantly associated with subpopulation while trypanosome presence showed a significant association with age (X2 = 4.65, df = 14, P-value = 0.0310) and an interaction between subpopulation and sex (X2 = 18.94, df = 10, P-value = 0.0043). However, the specific associations that were significant varied across species of trypanosomes, with T. congolense and T. brucei but not T. vivax showing significant interactions involving Sodalis. Although it has previously been concluded that presence of Sodalis increases susceptibility to trypanosomes, the results presented here suggest a more complicated relationship, which may be biased by differences in the distribution and intrinsic factors of tsetse flies, as well as which trypanosome species are considered. In chapter 4 trypanosome status was studied in relation to blood meal sources, feeding status and feeding patterns of G. pallidipes (which was the predominant fly species collected for this study) as determined by sequencing the mitochondrial cytochrome B gene using DNA extracted from abdomen samples. African buffalo and African elephants were the main sources of blood meals but antelopes, warthogs, humans, giraffes and hyenas were also identified. Feeding on multiple hosts was common in flies sampled from the Shimba Hills but most flies from Nguruman had fed on single host species. Based on Multiple Correspondence Analysis (MCA), host-feeding patterns showed a correlation with site of sample collection and Sodalis status, while trypanosome status was correlated with sex and age of the flies, suggesting that recent host-feeding patterns from blood meal analysis cannot predict trypanosome status. In conclusion, the complexity of interactions found suggests that strategies of tsetse fly control should be specific to particular epidemic areas. Future studies should include laboratory experiments that use local colonies of tsetse flies, local strains of trypanosomes and local S. glossinidius under controlled environmental conditions to tease out the factors that affect vector competence and the relative influence of external environmental factors on the dynamics of these interactions.

Dentre os Kinetoplastida, Trypanosoma é o gênero com maior ocorrência, infectando populações de peixes marinhos e de água doce em todo o mundo. Apresenta elevados níveis de prevalência, ocasiona impactos na saúde dos peixes e consequente perdas econômicas, principalmente para populações de peixes em situação de estresse. Este estudo investigou a infecção por Trypanosoma spp. e sanguessugas em Hypostomus spp. e parâmetros sanguíneos desse hospedeiro do sistema de tributários do Rio Tapajós, no Estado do Pará, Amazônia Oriental, Brasil. De 47 hospedeiros examinados, 89,4% estavam parasitados por Trypanosoma spp., e 55,4% tinham também sanguessugas na região da boca. A intensidade de Trypanosoma spp. aumentou com o tamanho dos hospedeiros, mas as condições corporais não foram influenciadas pelo parasitismo. O número de eritrócitos, hematócrito, hemoglobina, VCM, HCM, CHCM, número de leucócitos e trombócitos totais apresentaram variações e correlação negativa com a intensidade de Trypanosoma spp. no sangue dos hospedeiros. Os resultados sugerem que sanguessugas foram os vetores de Trypanosoma spp. in Hypostomus spp.

Os animais silvestres estão expostos a inúmeros patógenos,dentre eles estão os hemoparasitas. Podem-se destacar espécies do gênero Trypanosoma e do grupo das hemogregarinas,que ocorrem com freqüência parasitando anuros (rãs, pererecas e sapos). Normalmente, a descrição destes hemoparasitas é feita através da morfologia dos estágios observados nosangue periférico do hospedeiro e as pesquisas sobre o ciclobiológico desses hemoparasitas são escassas. Os objetivos dopresente estudo foram avaliar a presença de hemogregarinas eTrypanosoma spp. em anuros capturados nos Estados de São Paulo e Mato Grosso do Sul e fazer a caracterização morfológica e morfométrica dos seus hemoparasitas. As espécies deanuros examinadas foram: Dendropsophus nanus, D. minutus, Leptodactylus chaquensis, L. podicipinus, L. labyrinthicus, L. fuscus, Bufo granulosus, B. schneideri, Phyllomedusahypocondrialis, Trachicephalus venulosus, Scinax fuscovarius e Hypsiboas albopunctatus. Dos 40 animais estudados, foramencontrados quatro (10%) positivos para hemogregarinas e oito(20%) positivos para Trypanosoma spp. Foram observadosgamontes de hemogregarinas com morfologia variável e, alémdas formas intraeritrocíticas, também foram observados gamontes fora das hemácias. As formas de Trypanosoma encontradas eram muito polimórficas, conforme é descrito na literatura, sendo na sua maioria, larga e oval.

Co-infections by Leishmania (L.) chagasi, Trypanosoma evansi, Toxoplasma gondii and Neospora caninum in dogs were investigated. Amastigotes forms of Leishmania spp. were detected by cytopathological analysis of lymph nodes in 46,42% (39/84) of dogs. In a male dog, adult, without defined breed, from rural area and positive for Leishmania, were observed flagellated forms of T. evansi in blood smear. By immunofluorescence antibody test, 5,95% (5/84) of dogs were considered reactive to T. gondii, with titer equal to or higher than 1:64, while 3,57% (3/84) were reactive to N. caninum, with titer ≥1:50. Among the animals with visceral leishmaniasis, one showed positive serological response to T. gondii and two for N. caninum. All dogs reactive to N. caninum were from rural area and the predominance of infection by T. gondii was in dogs from urban area. A young male dog from the rural area and seropositive for T. gondii showed Ehrlichia spp. morulae in the cytology and positive reaction for canine distemper virus. Thus, further studies are needed to assess the epidemiology of these infections in canine population, especially with respect to the reservoirs of Trypanosoma spp. in rural areas.

O presente trabalho monitorou a saúde de oito espécies de acaris ornamentais capturados e comercializados no Médio Rio Guamá - Pará, através do estabelecimento do quadro hematológico basal, avaliação de estresse de transporte e de infecção por Trypanosoma spp. São elas: ancistrus (Ancistrus sp. - L338), loricaia (Rineloricaia lanceolata - L10), picoto (Hypostomus sp. - L28), bola (Peckoltia oligospila - L06), pleco (Cochilodon sp. - L145), canoa (Lasiancistrus saetiger - L323), assacu (Pseudacanthicus spinosus - L160) e pinima (Leporacanthicus galaxias - L07). As coletas sanguíneas para a determinação do quadro hematológico basal (Capítulos I e II) foram realizadas ainda no local da captura dos peixes, sob o mínimo de estresse possível. Separarou-se as amostras sanguíneas não infectadas para possibilitar comparações com as do após-estresse de transporte (Capítulos III e IV) e também com as infectados por Trypanosoma spp. (Capítulo V). O estresse de transporte estabelecido durou 3h, com densidade de 1,5 peixe/L, simulando o processo de comercialização dos peixes na região e foi avaliado após 0, 6, 24, 48, 72 e 96h. Nos Capítulos I e II, observou-se que o hemograma basal apresentou diferença significativa (p>0,05) entre os as sete espécies de acaris, apesar de estas pertencerem a mesma família e compartilharem nichos ecológicos semelhantes. O estresse de transporte por 3h (Capítulos III e IV) não comprometeu a saúde dos acaris, pois a maioria dos parâmetros hematológicos retornou aos níveis basais em 24h em bola, em 48h em pleco e em 72h em picoto, sendo estes, respectivamente, os períodos mínimos indicados para a aclimatação destes peixes antes de uma nova comercialização. Todas as oito espécies de acaris estudadas estavam infectadas por Tryopanosoma spp. (Capítulo V). Encontrou-se anemia normocítica-hipocrômica em ancistrus e canoa, e anemia macrocítica-hipocrômica em loricaia. Pinimas infectados apresentaram quadro de estresse com linfocitopenia, neutrofilia e monocitose. Assim, os resultados deste ensaio proporcionaram a avaliação da higidez destas espécies de acaris ornamentais através de exames hematológicos, podendo assim subsidiar o desenvolvimento ou a adequação do manejo menos estressante para estes peixes, de forma a auxiliar a sustentabilidade da cadeia extrativista das espécies ornamentais.

The gene encoding type II DNA topoisomerase from the kinetoplastid hemoflagellated protozoan parasite Leishmania donovani (LdTOP2) was isolated from a genomic DNA library of this parasite. DNA sequence analysis revealed an ORF of 3711 bp encoding a putative protein of 1236 amino acids with no introns. The deduced amino acid sequence of LdTOP2 showed strong homologies to TOP2 sequences from other kinetoplastids, namely Crithidia and Trypanosoma spp. with estimated identities of 86 and 68%, respectively. LdTOP2 shares a much lower identity of 32% with its human homologue. LdTOP2 is located as a single copy on a chromosome in the 0.7 Mb region in the L.donovani genome and is expressed as a 5 kb transcript. 5′-Mapping studies indicate that the LdTOP2 gene transcript is matured post-transcriptionally with the trans-splicing of the mini-exon occurring at –639 from the predicted initiation site. Antiserum raised in rabbit against glutathione S-transferase fusion protein containing the major catalytic portion of the recombinant L.donovani topoisomerase II protein could detect a band on western blots at ∼132 kDa, the expected size of the entire protein. Use of the same antiserum for immunolocalisation analysis led to the identification of nuclear, as well as kinetoplast, antigens for L.donovani topoisomerase II. The in vitro biochemical properties of the full-length recombinant LdTOP2 when overexpressed in E.coli were similar to the Mg(II) and ATP-dependent activity found in cell extracts of L.donovani.

In this study, we provide phylogenetic and biogeographic evidence that the Trypanosomo cruzi lineages T. cruzi I (TCI) and T. cruzi IIa (TCIIa) circulate amongst non-human primates in Brazilian Amazonia, and are transmitted by Rhodnius species in overlapping arboreal transmission cycles, sporadically infecting humans. TO presented higher prevalence rates, and no lineages other than TCI and TCIIa were found in this study in wild monkeys and Rhodnius from the Amazonian region. We characterised TO and TCIIa from wild primates (16 TO and five TCIIa), Rhodnius spp, (13 TCI and nine TCIIa), and humans with Chagas disease associated with oral transmission (14 TO and five TCIIa) in Brazilian Amazonia. To our knowledge, TCIIa had not been associated with wild monkeys until now. Polymorphisms of ssrDNA, cytochrome b gene sequences and randomly amplified polymorphic DNA (RAPD) patterns clearly separated TCIIa from TCIIb-e and TCI lineages, and disclosed small intra-lineage polymorphisms amongst isolates from Amazonia. These data are important in understanding the complexity of the transmission cycles, genetic structure, and evolutionary history of T cruzi populations circulating in Amazonia, and they contribute to both the unravelling of human infection routes and the pathological peculiarities of Chagas disease in this region. Visceral Leishmaniasis (VL) is a zoonosis caused by Leishmania spp. protozoa. Dog is the main parasite's reservoir, especially in urban areas. Indirect Immunofluorescent Antibody Test (IFAT) is commonly used for canine visceral leishmaniasis (CVL) diagnosis, by detection of antibodies against the parasite. However, Trypanosoma cruzi (T.cruzi) cross reactions can occur in serological diagnosis. In order to evaluate the occurrence of cross reactions between antigens of Leishmania spp. and T.cruzi, 150 blood samples of CVL epidemiological inquiry positive dogs were collected and sera were tested by IFAT. Dogs were culled at Zoonosis Control Center of Bauru (SP), an endemic area for VL. Another 150 dogs' blood samples were collected in Botucatu (SP), a non endemic area for LV, and sera were also tested by IFAT. Serum samples of dogs from Bauru were positive, both for Leishmania spp. and for T. cruzi, showing high cross-reactions incidence, reinforcing the need of diagnosis confirmation by other tests. Serum samples of dogs from Botucatu demonstrated low prevalence of positive results by IFAT, both for Leishmania spp. and T.cruzi. However, despite the low number of positive serological results for these parasites, the epidemiological and serological investigation for LV and Chagas disease in dogs from Botucatu must be kept, considering the importance of dog as domestic reservoir for both parasites.
