The herbal medicine Sanativo® is produced by the Pernambucano Laboratory since 1888 with indications of healing and hemostasis. It is composed of a fluid extract about Piptadenia colubrina, Schinus terebinthifolius, Cereus peruvianus and Physalis angulata. Among the plants in their composition, S. terebinthifolius and P. colubrina have in common phenolic compounds which are assigned most of its pharmacological effects. The tannins, gallic acid and catechin were selected as markers for quality control. The aim of this study was the development and validation of analytical method by HPLC/UV/DAD for the separation and simultaneous quantification of gallic acid (GAC) and catechin (CTQ) in Sanativo®. The chromatographic system was to stationary phase, C-18 RP column, 4,6 x 150 mm (5 mm) under a temperature of 35 ° C, detection at 270 and 210 nm. The mobile phase consisted of 0.05% trifluoroacetic acid and methanol in the proportions 88:12 (v/v), a flow rate of 1 ml/min. The analytical method presented a retention factor of 0.30 and 1.36, tail factor of 1.8 and 1.63 for gallic acid and catechin, respectively, resolution of 18.2, and theoretical plates above 2000. The method validation parameters met the requirements of Resolution n º 899 of May 29, 2003, ANVISA. The correlation coefficient of linear regression analysis for GAC and CTQ from the standard solution was 0.9958 and 0.9973 and when performed from the Sanativo® 0.9973 and 0.9936, the matrix does not interfere in the range 70 to 110 %. The limits of detection and quantification for GAC and CQT were 3.25 and 0.863, and 9.57 and 2.55 mg/mL, respectively. The markers, GAC and CQT, showed repetibility (coefficient of variation of 0.94 % and 2.36 %) and satisfactory recovery (100.02 ± 1.11 % and 101.32 ± 1.36 %). Schinus terebinthifolius Raddi (Anacardiaceae) é uma espécie nativa da América do Sul com grande distribuição geográfica e muito comum em áreas de restinga. Sua grande plasticidade ecológica e boa interação biótica a torna espécie-chave na restauração de ambientes, porém, sua dominância pode estar relacionada à liberação de compostos alelopáticos. O objetivo do estudo foi avaliar o potencial alelopático dos extratos aquosos e óleos essenciais de folhas de Schinus terebinthifolius nas diferentes estações do ano sobre espécies-alvo nativas da restinga (Cereus fernambucensis, Erythroxylum ovalofolium, Pilosocereus arrabidae), uma espécie agrícola (Lactuca sativa) e nela própria (autoalelopatia). Para isso, suas folhas foram coletadas sazonalmente na restinga de Massambaba para o preparo dos extratos aquosos e extração dos óleos essenciais. Os extratos aquosos foram obtidos através da secagem das folhas a 60 C, trituração em liquidificador, diluição em água destilada e filtração, obtendo as concentrações de 5, 10, 15 e 20%. As extrações dos óleos foram realizadas através da hidrodestilação de 300 g de folhas frescas. A análise da composição dos óleos essenciais foi realizada através de cromatografia gasosa utilizando fibra SPME. O efeito fitotóxico foi avaliado nas espécies-alvo através da porcentagem de germinação, velocidade de germinação, crescimento aéreo e radicular, condutividade elétrica e massa seca. O efeito alelopático dos extratos aquosos foi observado em todas as espécies-alvo, principalmente sobre as espécies nativas. Esse efeito variou nas estações do ano de forma dose-dependente e espécie-específica. Os óleos essenciais de S. terebinthifolius também foram capazes de inibir a germinação e crescimento das espécies-alvo, e, da mesma forma que os extratos aquosos, esses efeitos variaram nas estações do ano e para cada espécie. Sugere-se que essas diferenças estejam relacionadas às fenofases de S. terebinthifolius e condições ambientais da restinga. Esse efeito fitotóxico, se comprovado em campo, pode restringir a utilização de S. terebinthifolius na restauração de ambientes.

Avaliou-se a segurança de um fitoterápico, constituído de extratos fluidos de Aristolochia cymbifera (“cassaú”), Plantago major L.(“transagem”), Luehea grandiflora Mart.(“açoita-cavalo”), Myrocarpus frondosus Allemão (“cabreúva”), Piptadenia colubrina Benth (“angico”) (Cassaú Composto® ), através de estudos de toxicidade aguda e subcrônica, tendo como base a resolução Nº 90, de 16 de março de 2004 da ANVISA (Agência Nacional de Vigilância Sanitária). Para o teste de toxicidade aguda, ratos Wistar de ambos os sexos foram tratados por via oral com uma única dose de 26 ml/kg, correspondendo a 20 vezes a dose terapêutica indicada pelo fabricante para seres humanos adultos. Os resultados revelaram haver sinais de toxicidade sistêmica com o aparecimento de ataxia, porém de forma transitória e reversível, não causando interferência no desenvolvimento ponderal dos animais, nos consumos de água e ração, nas produções de urina e fezes, bem como alterações macroscópicas nos órgãos dos animais. Avaliou-se também a exposição a doses repetidas do fitoterápico (toxicidade subcrônica). Constituiram-se 4 grupos experimentais (10 animais/sexo/dose), onde administrou-se por via oral a ratos Wistar, durante 30 dias, doses diárias de 1,3 ml/kg, 6,5 ml/kg e 13 ml/kg, respectivamente a dose terapêutica indicada pelo fabricante para seres humanos adultos, 5 vezes, e 10 vezes a dose terapêutica, além de um grupo controle, onde administrou-se o veículo do fitoterápico. Os resultados revelaram ausência de toxicidade sistêmica, fundamentados na ausência de alterações hematológicas e bioquímicas sangüíneas, bem como peso e análises histopatológicas dos órgãos, nos diferentes grupos. As flutuações nos consumos de água e ração, bem como produções de urina e fezes, não influenciaram de maneira negativa o desenvolvimento ponderal dos animais. Concluiu-se portanto, que a utilização do fitoterápico nas doses e períodos referidos pode ser considerado segura.

The decoction of Brazilian pepper tree barks (Schinus terebinthifolius, Raddi), is used in medicine as wound healing and antiinflamatory. Once extracts from this plant are used for acceleration of scar s process, it is important to study their mutagenic and genotoxic potential. In previous works in our laboratory, it was observed mutagenicity caused by the decoction when in high concentrations. Among the chemical compounds of this plant that could be able to induce mutation, the flavonoids were the only group that was referred to have either an oxidant or antioxidant potential. The flavonoids were isolated, purified and quantified by adsorptive column chromatography under silica gel, bacterial and in vitro genotoxic tests were realized to determine if the flavonoids were the responsible agents for this mutagenicity found. The tests realized with plasmidial DNA were indicative that the flavonoids are probably genotoxic, due to the presence of correlation between increase of the flavonoid concentration and in plasmidial DNA double strand breakage visualized in agarose gel, as well as they were capable to generated abasic sites shown by the in vitro treatment with exonuclease III. The same tests with plasmidial DNA in the presence of copper [10 µM] and of a Tris-HCl pH 7.5 [10 µM] buffer were realized with the isolated flavonoids to determine if there would be or not participation of reactive oxygen species (ROS). The transformation of plasmidial DNA in different bacterial strains proficient and deficient in DNA repair enzymes in the presence or not of a Tris-HCl buffer, suggests that the enzymes that repair oxidative lesions are necessary to repair the lesions generated by the flavonoids and that ROS are generated and are necessary to promote the lesions. Bacterial tests with Escherichia coli strains of the CC collection (deficient or not for DNA repair enzymes), showed that the flavonoids are able to increase the frequency of mutations, mainly in strains mutated in repair enzymes (MutM, MutY-glicosylases and double mutant), suggesting that these agents are responsible for the enhancement in the mutation rate. In order to determine the mutation spectrum caused by the flavonoids of the Brazilian pepper tree stem bark, plasmidial DNA previously treated with the flavonoids were transformed in bacterial strains deficient and proficient in the DNA repair enzymes, followed by a blue-white selection with X-gal, DNA amplification by PCR and sequencing the positive mutant clones. Analysis of the mutants obtained from strains CC104, CC104mutM, CC104mutY, CC104mutMmutY, BW9101, BW9109 indicated a predominance of some mutations like G:C to C:G that can be correlated with the origin of 8-oxoG, due to oxidative lesions caused by the flavonoids. So it can concluded that the flavonoid isolated or in fractions enriched on them are genotoxic and mutagenic, and their mutations are predominantly oxidative, mediated by ROS, and the lesions are recognized by the BER system. In this way it is proposed that the flavonoids can act in two different ways to generate the DNA lesion: 1. in a Fenton-like reaction, when the flavonoid are in the presence of metal ions and that together with the water generate ROS that promotes the DNA lesions; 2. in another way the lesions can be generated by the formation of ROS due to the internal chemical structure of the flavonoid molecule due to the quantity and location of hydroxyl groups, and so producing the DNA lesions, those lesions can be directly (suggested by the in vitro experiments) or indirectly done (supported by the experiments using the CC bacterial strains)

Schinus terebinthifolius Raddi is used in the treatment of skin and mucosal injuries, infections of respiratory, digestive and genitourinary systems. Currently one of the biggest problems faced for the industry of phytopharmaceuticals with regard to the quality of raw materials is the microbial contamination. The aim this study was to evaluate the antimicrobial action of the hidroalcoholic extract of aroeira, beyond testing the effectiveness of the preservative system in hidrogel to the base of this extract. The extracts were prepared by maceration in the ratio of 1:10 of solvent plant/with alcohol 40%. The methods for microbial count were pour plate and test for specific microorganisms, analyzing in third copy each one of the samples. The antimicrobial activity of aroeira extracts was performed using an agar diffusion method, using strains of S. aureus, P. aeruginosa, E. coli, B. subtilis, C. albicans, C. tropicalis, C. krusei, C. guilliermondii, T. rubrum, M. gypseum, A. flavus and A. niger. The formula with aroeira was evaluated by the challenge test. This method consisted of artificial contamination the sample with separate inóculos of A. niger, C. albicans, E. coli e S. aureus aeruginosa and determinations of survivors for the method of counting for pour plate , during times 0, 24h, 48h, 7 days, 14 days, 21 days and 28 days. How much to the results, one verified that the extract of aroeira in the 13,5 concentration mg/mL presented antimicrobial activity for cepas of E. coli, B. subtilis, P. aeruginosa e S. aureus, producing inhibition zone, on average with 13 mm of diameter. However it did not present no fungi activity. The formula with aroeira containig both methylparaben and propylparaben showed a good efficacy in challenge test front to strains of A. niger, C. albicans, E. coli, S. aureus. The A criteria of European Pharmacopoeia, adopted in this work, was verified that this product revealed the good preservative efficacy for the challenge test, time interval of the 28 days. However, it is interesting to extend this study, in order to carry through the sped up stability and the test of shelf, to establish the validity of this formularization

Este trabalho foi realizado com o objetivo de avaliar os melhores métodos e épocas de realização da enxertia, sobre o pegamento de enxertos de aroeira-pimenteira (Schinus terebinthifolius Raddi). Foram conduzidos dois experimentos, instalados na Faculdade de Ciências Agronômicas, da Universidade Estadual Paulista - Câmpus de Botucatu-SP, nos períodos de 16 de dezembro de 2005 a 30 de janeiro 2006 e de 03 de junho a 17 de julho de 2006. O delineamento utilizado foi de blocos casualizados, com cinco tratamentos, três repetições cada e dez plantas por parcela. Os tratamentos foram: garfagem no topo em fenda cheia, inglês simples, inglês complicado; borbulhia em T normal e T invertido. As diferentes épocas do ano alteraram o índice de pegamento, o que restringe o período de coleta de garfos. A maior porcentagem de pegamento ocorreu no mês de dezembro, com a enxertia em fenda cheia (40%). No mês de junho, não houve pegamento significativo, mostrando então que o mês de dezembro é o mês ideal, entre os estudados, para a realização da enxertia em aroeira. As garfagens em geral foram superiores à borbulhia. Houve baixo índice de pegamento, quando não foi nulo, e, além disso, suas borbulhas não apresentaram brotos dentro de 45 dias. A maior porcentagem de plantas com brotos, aos 45 dias, foi pelo método inglês simples, com 92%.

Objective: To evaluate the clinical efficacy of Schinus terebinthifolius Raddi (aroeira) tincture in the treatment of denture stomatitis. Method: Eighteen removable denture wearers with clinical diagnosis of type II denture stomatts and presence of candidosis associated to the denture use, as confirmed by clinical and mycological examinations, were selected for the study. The patients were allocated to two groups: TG (test group) - treatment with Schinus terebinthifolius Raddi (aroeira) tincture; CG (control group), treatment with nystatin. All patients were instructed to clean the dentures with toothbrush and dentifrice, and then apply the product on the palatal mucosa and on denture surface 3 times a day, during 15 consecutive days, removing the denture at bedtime and keeping it in a receptacle with water. At the 15th day of use, the clinical and mycological examinations were redone to evaluate treatment efficacy. Data were analyzed statistically by Wilcoxon and Mann-Whitney tests at 5% significance level. Results: The inflammatory process and Candida spp. infect on were eliminated in 66.7% and 77.8% of the cases, respectively, in TG. In CG, elimination of the inflammatory process and fungal infect on occurred in 77.8% and 88.9% of the cases, respectively. These results were statistically significant (p=0.01). There was no statistically significant difference between the treatments (p>0.05). In all cases, fungal infect on was detected only on the denture, and C. albicans was the most prevalent microorganism, being present in 94.4% of the cases. Conclusion: The treatment with Schinus terebinthifolius Raddi (aroeira) tincture was effective in the treatment of denture stomatitis, promoting remission of the inflammatory process and Candida spp infection.

Forest species with economic potential, such as aroeira (Schinus terebinthifolius Raddi.), require selection of individuals with superior characteristics for use in forest restoration projects and for establishment of commercial plantations. These plantations can contribute to the sustainability of natural populations of species in the remaining forest fragments, in areas of permanent preservation, legal reserves or other areas of ecological significance. It was evaluated the fruits' yield, morphometry and viability of seeds from 15 individuals of aroeira, collected in different fragments in the region of Lower São Francisco river, Sergipe State. The fruit yield was estimated by Fornier's intensity index, and the morphometric characteristics were obtained with a digital paquimeter and analytical balance. The viability and vigor were evaluated by germination percentage and germination speed index (GSI), under controlled conditions. The results of fruit yield were subjected to variance analysis, and the means were compared by Tukey (p <0.05). The other variables means were compared by Scott-Knott test (p<0.05). The individuals differed in Fournier's indices (indices 1, 2, 3 and 4) and in the size of fruits and seeds. The germination varied from 0 to 83% and the GSI from 0.00 to 0.98. The phenotypic differences observed among individuals for fruit yield, and morphophysiological characteristics can be used in forest restoration and establishment of provenances/progenies tests, aiming discrimination of superior material for future commercial plantations.
