The basic reproduction number of a pathogen, R 0, determines whether a pathogen will spread (R0>1R 0>1), when introduced into a fully susceptible population or fade out (R0<1R 0<1), because infected hosts do not, on average, replace themselves. In this paper we develop a simple mechanistic model for the basic reproduction number for a group of tick-borne pathogens that wholly, or almost wholly, depend on horizontal transmission to and from vertebrate hosts. This group includes the causative agent of Lyme disease, Borrelia burgdorferi, and the causative agent of human babesiosis, Babesia microti, for which transmission between co-feeding ticks and vertical transmission from adult female ticks are both negligible. The model has only 19 parameters, all of which have a clear biological interpretation and can be estimated from laboratory or field data. The model takes into account the transmission efficiency from the vertebrate host as a function of the days since infection, in part because of the potential for this dynamic to interact with tick phenology, which is also included in the model. This sets the model apart from previous, similar models for R0 for tick-borne pathogens. We then define parameter ranges for the 19 parameters using estimates from the literature, as well as laboratory and field data, and perform a global sensitivity analysis of the model. This enables us to rank the importance of the parameters in terms of their contribution to the observed variation in R0. We conclude that the transmission efficiency from the vertebrate host to Ixodes scapularis ticks, the survival rate of Ixodes scapularis from fed larva to feeding nymph, and the fraction of nymphs finding a competent host, are the most influential factors for R0. This contrasts with other vector borne pathogens where it is usually the abundance of the vector or host, or the vector-to-host ratio, that determine conditions for emergence. These results are a step towards a better understanding of the geographical expansion of currently emerging horizontally transmitted tick-borne pathogens such as Babesia microti, as well as providing a firmer scientific basis for targeted use of acaricide or the application of wildlife vaccines that are currently in development.

These included 84 random amplified polymorphic DNA (RAPD) markers, 32 Sequence-Tagged RAPD (STAR) markers, 5 cDNAs, and 5 microsatellites in 232 F1 intercross progeny from a single, field-collected P1 female. A preliminary linkage map of 616 cM was generated across 14 linkage groups with one marker every 10.8 cM. Assuming a genome size of ~ 10 9 bp, the relationship of physical to genetic distance was found to be ~ 300 kb/cM in the I. scapularis genome.

These included 84 random amplified polymorphic DNA (RAPD) markers, 32 Sequence-Tagged RAPD (STAR) markers, 5 cDNAs, and 5 microsatellites in 232 F1 intercross progeny from a single, field-collected P1 female. A preliminary linkage map of 616 cM was generated across 14 linkage groups with one marker every 10.8 cM. Assuming a genome size of ∼109 bp, the relationship of physical to genetic distance is ∼300 kb/cM in the I. scapularis genome.

La chita Anisotremus scapularis es un pez marino que habita las costas de Perú, es muy valorado para el consumo humano directo, y, es considerada una especie con potencial acuícola. El cultivo larval en muchas especies de peces marinos se considera como uno de los cuellos de botella para el desarrollo de la tecnología de cultivo a escala comercial, este es el caso de la chita. Como primer alimento y durante los primeros días de vida de las larvas se les suministra presas vivas, y posteriormente, cuando poseen un sistema digestivo más desarrollado, se les suministra alimento balanceado. Sin embargo, los alimentos vivos más empleados no satisfacen los requerimientos nutricionales de las larvas convirtiéndose en un punto crítico. Es por ello, que en los últimos años diversas investigaciones han concentrado sus esfuerzos en la sustitución del alimento vivo por alimento balanceado; no obstante, esto exige un buen conocimiento de la organización y funcionalidad del sistema digestivo. En este sentido, en el presente trabajo se investigan algunos aspectos de la fisiología nutricional de las larvas de chita durante los primeros 30 días de vida. Las larvas fueron cultivadas a partir de huevos obtenidos de desoves naturales de un lote de reproductores mantenidos en cautiverio en el Laboratorio de Cultivo de Peces del Instituto del Mar del Perú (IMARPE). El estudio del crecimiento en longitud, los análisis histológicos y la caracterización enzimática se realizaron a partir de muestras de larvas colectadas a los 0, 1, 2, 3, 4, 6, 8, 10, 14, 18, 22, 26 y 30 días después de la eclosión (DDE). Para los análisis histológicos, las larvas fueron sometidas a un proceso de deshidratación e inclusión en parafina, y teñidos utilizando la técnica de hematoxilina-eosina. La cuantificación de las enzimas digestivas de la chita, proteasas totales (ácidas y alcalinas), tripsina, leucina aminopeptidasa, lipasas dependientes de sales biliares y alfa-amilasa se llevó a cabo con técnicas espectrofotométricas. Al momento de la eclosión, las larvas poseen un tubo digestivo recto e histológicamente indiferenciado ubicado dorsalmente con respecto al saco vitelino, que se caracteriza por poseer una gota de aceite. Entre los 2 y 4 DDE, se observaron los mayores cambios en el desarrollo del sistema digestivo, entre los más importantes están la diferenciación de la bucofaringe, esófago e intestino, plegamiento de la mucosa intestinal, la formación de las microvellosidades en los enterocitos y el desarrollo de sus glándulas anexas, hígado y páncreas, los cuales permitieron la ingestión, digestión y absorción de los primeros alimentos exógenos ingeridos por las larvas. El mayor cambio que se observó en días posteriores fue la observación de un estómago en desarrollo, con la diferenciación de las glándulas gástricas (26 DDE). Las enzimas digestivas analizadas, fueron detectadas desde el momento de la eclosión de las larvas, antes de la apertura de la boca, y se incrementaron con la edad de los organismos. En general, se observó incrementos en la actividad total de las enzimas digestivas a partir de los 8 y 10 DDE o a los 4 mm de longitud total (LT), particularmente con aumentos más marcados en la actividad de las lipasas y de la leucina aminopeptidasa. Para la actividad de proteasas alcalinas totales, tripsina y alfa-amilasa, los aumentos fueron evidentes a partir de los 22 DDE o entre 6 y 7 mm de LT. La actividad de las proteasas ácidas se evidenció e incrementó a partir del 26 DDE o a los 9 mm de LT. En cuanto los patrones de actividad específica, en general, se observaron incrementos súbitos de la actividad que se relacionan con los periodos de transición de la alimentación endógena a exógena (3-6 DDE), y con el cambio de alimentación de rotíferos a metanauplios de Artemia (26 DDE). También, exceptuando la actividad de la leucina aminopeptidasa, se observaron estos incrementos súbitos antes de la primera alimentación. Con base al análisis histológico y bioquímico de las larvas de chita, se recomienda iniciar el destete a los 26 DDE.

Los estudios taxonómicos realizados en el Perú por ictiólogos nacionales y extranjeros han permitido la determinación de 727 especies marinas, pertenecientes a 388 géneros y 138 familias, estimándose aproximadamente en 900 las que integran la ictiodiversidad del mar peruano, citada en claves, catálogos y listas publicadas. Del total de especies, 73 son de importancia económica, elevándose este número a 112 si se consideran las de consumo local. El área situada al norte de 06°56' S, que comprende las zonas nerítico tropical y de manglares, es mucho más rica en diversidad específica, con integrante de la Provincia Panameña, mientras que el área al sur de dicha latitud, templada o fría, presenta menor diversidad y para algunas familias con equivalencias de 14% a 25% de sus especies. Las especies que ocurren en el ecosistema de la Corriente Peruana corresponden a las que integran la Provincia Peruano Chilena (06°56' S- 42° S), caracterizándose por la apreciable magnitud de sus poblaciones: Engraulis ringens ("anchoveta"), Sardinops sagax sagax ("sardina") y Trachurus picturatus murphyi ("jurel"). Otras especies de interés por su importancia comercial dentro de este ecosistema son principalmente, Odontesthes regia regia ("pejerrey"), Sarda chiliensis chiliensis ("bonito"), Seriolella víolacea ("cojinoba"), Cilus gilberti ("corvina"), Sciaena deliciosa ("lorna"), Anisotremus scapularis ("chita:") e Isacia onceptionis ("cabinza"). Se determina que en el nivel actual del conocimiento existen vacíos en: áreas poco estudiadas, como la parte sur del litoral y las áreas muy costeras; los grupos de peces poco accesibles; la regularidad en las observaciones; el número suficiente de ictiólogos; la dedicación a grupos taxonómicos; así como es necesaria mayor información bioecológica de los diferentes ecosistemas, que de por sí plantean los requerimientos de investigación a corto y largo plazo.

Mon étude vise à évaluer la propagation d’une zoonose en émergence au Québec, la maladie de Lyme, en conséquence du réchauffement climatique. Le pathogène responsable de cette infection, Borrelia burgdorferi, est transmis par l’intermédiaire d’une tique parasite, Ixodes scapularis, de plus en plus commune au Québec en raison de l’augmentation de la température moyenne du climat depuis les dernières décennies. Puisque la tique a une capacité de déplacement très restreinte, on s'attend à ce que sa dispersion soit liée à celle de son hôte primaire, soit la souris à pattes blanches (Peromyscus leucopus). Je décrirai donc d’abord les espèces impliquées, leur écologie et leur rôle dans ce système à trois niveaux (hôte/pathogène/vecteur). Puis, à l’aide de séquences d’ADN mitochondrial, je comparerai la phylogéographie des deux principales espèces de souris au Québec, la souris à pattes blanches et la souris sylvestre (P. maniculatus). Des analyses d’arbres et de réseaux d’haplotypes ont révélé des différences significatives dans la structure génétique et ainsi montré que les populations de P. leucopus seraient en expansion dans le sud du Québec. Cette étude nous a finalement permis d’émettre des hypothèses sur le patron d’établissement de la maladie de Lyme au Québec.

Depuis les années 90, les études réalisées au Canada ont permis d’identifier de nouvelles zones endémiques de l’agent de la maladie de Lyme, Borrelia burgdorferi, ou de sa tique vectrice, Ixodes scapularis. Ces régions représentent des zones privilégiées pour étudier le cycle de transmission dans son contexte environnemental. L’objectif principal de ce projet est d’étudier les relations spirochètes – tiques - hôtes et les facteurs environnementaux impliqués dans le cycle de transmission à partir des données du vecteur et de l’agent pathogène recueilli dans le Sud-Ouest du Québec de 2007 à 2008. Tout d’abord sera décrite la saisonnalité des tiques et des associations entre le vecteur et les hôtes rongeurs capturés. En effet, l’identification de la saisonnalité spécifique à chaque stade de la tique I. scapularis permet d’établir quels seront les mois propices pour effectuer les futures collectes de tiques. La saisonnalité synchrone des tiques immatures en quête peut également être un indicateur de la nature ou des souches de B. burgdorferi retrouvées. L’association des tiques immatures à différentes espèces ou à différentes classes d’hôtes (c.-à-d. âge, sexe, statut reproductif) a également été explorée. Nos résultats montrent que les souris du genre Peromyscus, principalement les mâles adultes, contribuent significativement à la survie et au développement du complexe I. scapularis - B. burgdorferi. Les tamias et les écureuils contribuent aussi à la survie et au développement des populations de la tique I. scapularis. Ensuite les associations entre le vecteur et les hôtes cervidés ont été examinées en tenant compte des facteurs environnementaux associés à leur niveau d’infestation. Ceci a permis d’évaluer l’utilisation des cerfs à titre de sentinelles pour le vecteur et les agents pathogènes. D’après nos résultats, bien qu’ils soient des sentinelles efficaces pour détecter Anaplasma phagocytophilum, les cerfs semblent des sentinelles inefficaces pour détecter les zones d’établissement du complexe I. scapularis-B. burgdorferi. Enfin, une analyse de l’impact de la diversité des hôtes et de l’habitat sur l’abondance de la tique I. scapularis et la prévalence de B. burgdorferi a été effectuée et ce, en tenant compte d’autres facteurs environnementaux. Ces analyses ont permis de déterminer les facteurs critiques pour l’établissement du complexe I. scapularis – B. burgdorferi et d’explorer la contribution relative de diverses espèces d’hôtes. D’après nos études, la diversité de la communauté d’hôte et la diversité de l’habitat influencent le complexe I. scapularis - B. burgdorferi. De plus, le climat (la température et les précipitations) joue un rôle significatif dans l’établissement, la survie et le développement des populations d’I. scapularis. Ce projet de recherche a permis d’explorer et d’identifier divers facteurs environnementaux biotiques et abiotiques influençant l’établissement du complexe I. scapularis - B. burgdorferi dans le Sud-Ouest du Québec. Ceux-ci pourraient être utilisés à titre d’indicateurs environnementaux du risque de la maladie de Lyme au Québec et possiblement ailleurs au Canada.

The molecular mechanism of factor Xa (FXa) inhibition by Alboserpin, the major salivary gland anticoagulant from the mosquito and yellow fever vector Aedes albopictus, has been characterized. cDNA of Alboserpin predicts a 45-kDa protein that belongs to the serpin family of protease inhibitors. Recombinant Alboserpin displays stoichiometric, competitive, reversible and tight binding to FXa (picomolar range). Binding is highly specific and is not detectable for FX, catalytic site-blocked FXa, thrombin, and 12 other enzymes. Alboserpin displays high affinity binding to heparin (K(D) similar to 20 nM), but no change in FXa inhibition was observed in the presence of the cofactor, implying that bridging mechanisms did not take place. Notably, Alboserpin was also found to interact with phosphatidylcholine and phosphatidylethanolamine but not with phosphatidylserine. Further, annexin V (in the absence of Ca(2+)) or heparin outcompetes Alboserpin for binding to phospholipid vesicles, suggesting a common binding site. Consistent with its activity, Alboserpin blocks prothrombinase activity and increases both prothrombin time and activated partial thromboplastin time in vitro or ex vivo. Furthermore, Alboserpin prevents thrombus formation provoked by ferric chloride injury of the carotid artery and increases bleeding in a dose-dependent manner. Alboserpin emerges as an atypical serpin that targets FXa and displays unique phospholipid specificity. It conceivably uses heparin and phosphatidylcholine/phosphatidylethanolamine as anchors to increase protein localization and effective concentration at sites of injury, cell activation, or inflammation.

Sialostatin L (SialoL) is a secreted cysteine protease inhibitor identified in the salivary glands of the Lyme disease vector Ixodes scapularis. In this study, we reveal the mechanisms of SialoL immunomodulatory actions on the vertebrate host. LPS-induced maturation of dendritic cells from C57BL/6 mice was significantly reduced in the presence of SialoL. Although OVA degradation was not affected by the presence of SialoL in dendritic cell cultures, cathepsin S activity was partially inhibited, leading to an accumulation of a 10-kDa invariant chain intermediate in these cells. As a consequence, in vitro Ag-specific CD4(+) T cell proliferation was inhibited in a time-dependent manner by SialoL, and further studies engaging cathepsin S(-/-) or cathepsin L(-/-) dendritic cells confirmed that the immunomodulatory actions of SialoL are mediated by inhibition of cathepsin S. Moreover, mice treated with SialoL displayed decreased early T cell expansion and recall response upon antigenic stimulation. Finally, SialoL administration during the immunization phase of experimental autoimmune encephalomyelitis in mice significantly prevented disease symptoms, which was associated with impaired IFN-gamma and IL-17 production and specific T cell proliferation. These results illuminate the dual mechanism by which a human disease vector protein modulates vertebrate host immunity and reveals its potential in prevention of an autoimmune disease. Ticks are blood feeding parasites transmitting a wide variety of pathogens to their vertebrate hosts. The vector competence of ticks is tightly linked with their immune system. Despite its importance, our knowledge of tick innate immunity is still inadequate and the limited number of sufficiently characterized immune molecules and cellular reactions are dispersed across numerous tick species. The phagocytosis of microbes by tick hemocytes seems to be coupled with a primitive complement-like system, which possibly involves self/nonself recognition by fibrinogen-related lectins and the action of thioester-containing proteins. Ticks do not seem to possess a pro-phenoloxidase system leading to melanization and also coagulation of tick hemolymph has not been experimentally proven. They are capable of defending themselves against microbial infection with a variety of antimicrobial peptides comprising lysozymes, defensins and molecules not found in other invertebrates. Virtually nothing is known about the signaling cascades involved in the regulation of tick antimicrobial immune responses. Midgut immunity is apparently the decisive factor of tick vector competence. The gut content is a hostile environment for ingested microbes, which is mainly due to the antimicrobial activity of hemoglobin fragments generated by the digestion of the host blood as well as other antimicrobial peptides. Reactive oxygen species possibly also play an important role in the tick-pathogen interaction. The recent release of the Ixodes scapularis genome and the feasibility of RNA interference in ticks promise imminent and substantial progress in tick innate immunity research.

In the present study, we compared 2 methods for collecting ixodid ticks on the verges of animal trails in a primary Amazon forest area in northern Brazil. (i) Dragging: This method was based on passing a 1-m(2) white flannel over the vegetation and checking the flannel for the presence of caught ticks every 5-10 m. (ii) Visual search: This method consisted of looking for guesting ticks on the tips of leaves of the vegetation bordering animal trails in the forest. A total of 103 adult ticks belonging to 4 Amblyomma species were collected by the visual search method on 5 collecting dates, while only 44 adult ticks belonging to 3 Amblyomma species were collected by dragging on 5 other collecting dates. These values were statistically different (Mann-Whitney Test, P = 0.0472). On the other hand, dragging was more efficient for subadult ticks, since no larva or nymph was collected by visual search, whereas 18 nymphs and 7 larvae were collected by dragging. The visual search method proved to be suitable for collecting adult ticks in the Amazon forest: however, field studies should include a second method, such as dragging in order to maximize the collection of subadult ticks. The cellular and molecular characteristics of a cell line (BME26) derived from embryos of the cattle tick Rhipicephalus (Boophilus) microplus were studied. The cells contained glycogen inclusions, numerous mitochondria, and vesicles with heterogeneous electron densities dispersed throughout the cytoplasm. Vesicles contained lipids and sequestered palladium meso-porphyrin (Pd-mP) and rhodamine-hemoglobin, suggesting their involvement in the autophagic and endocytic pathways. The cells phagocytosed yeast and expressed genes encoding the antimicrobial peptides (microplusin and defensin). A cDNA library was made and 898 unique mRNA sequences were obtained. Among them, 556 sequences were not significantly similar to any sequence found in public databases. Annotation using Gene Ontology revealed transcripts related to several different functional classes. We identified transcripts involved in immune response such as ferritin, serine proteases, protease inhibitors,. antimicrobial peptides, heat shock protein, glutathione S-transferase, peroxidase, and NADPH oxidase. BME26 cells transfected with a plasmid carrying a red fluorescent protein reporter gene (DsRed2) transiently expressed DsRed2 for up to 5 weeks. We conclude that BME26 can be used to experimentally analyze diverse biological processes that occur in R. (B.) microplus such as the innate immune response to tick-borne pathogens. Desenvolvido no Parque das Dunas, segunda reserva ambiental urbana do Brasil, ocupando uma área de 1.172,80 hectares, com característica de mata atlântica de dunas, situada numa faixa litorânea na região urbana do município de Natal (05° 46 S, 35º 12 W), o presente estudo, realizado durante os anos de 2004 a 2006, teve como objetivo identificar as espécies de culicídeos existentes no Parque das Dunas, capazes de transmitir arbovírus, tendo em vista que em 2004 houve uma epizootia de saguis (Callitrix jacchus), que causou grande mortandade, sem definição do agente etiológico. No ano de 2004, foram pesquisados sete pontos no interior da mata, com instalação de 20 armadilhas de ovitrampas e 20 de bambu para coleta dos imaturos. Para os adultos, durante quatro vezes por semana, foram usadas as armadilhas de Sannhon. Foram coletados 5.691 imaturos, sendo 839 Ae. aegypti, 3.184 Ae. albopictus e 1.668 Hg. leucocelaenus. A coleta dos adultos foi realizada de 2004 a 2006, etapa em que se recolheu 17.506 culicídeos adultos, sendo 17.244 Wy. bourrouli, 255 Ae. aegypti, 593 Ae. albopictus, 1.275 Hg. leucocelaenus, 294 Oc. scapularis, 05 Oc. taeniorynchus, 02 Oc. serratus e 3 Li. durhami. Para os imaturos houve correlação significativa entre Ae. aegypti e umidade relativa do ar p = 0, 049 e pluviometria p = 0,00, Ae. albopictus apresentou correlação significativa positiva com a pluviometria, enquanto Hg. leucocelaenus não apresentou nenhuma das variáveis climáticas. Para os adultos, a análise de série temporal aponta flutuação sazonal significativa para Ae. aegypti (p = 0,003); Ae. albopictus (p = 0,04); Oc. scapularis (p = 0,008 ) e Hg. leucocelaenus (p = 0,003). Uma correlação significativa negativa foi observada entre o número de Ae. albopictus coletado e a temperatura (Corr= - 0,50, p = 0,01); isto é, para cada 1°C a mais há diminuição de 7 espécimes. Este estudo teve a participação de uma equipe multidisciplinar: biólogos, entomologistas, para confirmação das espécies; técnicos de laboratório, para acompanhamento diário das larvas eclodidas das armadilhas de ovitrampas. Teve a importante colaboração de profissionais da Fundação Oswaldo Cruz FIOCRUZ/Rio de Janeiro, da Universidade de São Paulo USP para identificação do grupo Wyeomyia.

The genus Herpsilochmus is composed mainly of cryptic species, among them is Herpsilochmus rufimarginatus, which is currently represented by four subspecies: H. r. rufimarginatus, H. r. frater, H. r. scapularis and H. r. exiguus. Differences in plumage and vocalization suggest that there are more than one species involved in this complex. Thus this and other subspecific taxa need urgent revision, the disjunct distribution of this species also allows us to infer the relationship between birds that occur in this biome and / or different centers of endemism. This study aims to make a taxonomic revision of the taxa included in the complex time Herpsilochmus rufimarginatus based on morphological, morphometric, vocals and geographical distribution of this bird. Besides creating distribution models current potential and make the reconstruction of the distribution bygone using ecological niche modeling, and testing the niche conservatism and divergence between different subspecies. Consultations for examination of the skins of specimens of the museums: Museum of Zoology, University of São Paulo (MZUSP), National Museum of Rio de Janeiro (MN) and Emilio Goeldi Museum of Pará (MPEG), and the skins deposited at the collection of Ornithological Federal University of Rio Grande do Norte (COUFRN). We studied the following measures length of specimens: exposed culmen, culmen and total culmen nostril, tarsus, wing and tail flattened. The voice analysis was performed with vocalizations banks and / or digital banks people where 17 voice parameters were measured. This information and more available in the literature were used to assemble a bunch of data under the limit distribution of taxa and generate ecological niche models. This analyzes carried out in the program Maxent, having as model selection criterion the AUC, and the models were greater than 0.80 are considered good models. Environmental data for the realization of the modeling were downloaded on the website of Worldclim. The morphometric information, vocals and geographic distribution point for the separation of these taxa to be considering various uni and multivariate analyzes. The potential distribution models performed well (AUC> 0.80), and its distribution associated with environmental characteristics of the Amazon forest and Atlantic forest (forests of south and southeast, northeast and forest). The reconstruction of the distribution indicates a possible contact between the southern part of the Atlantic forest in the northern part of the Amazon. The analysis of niche overlap showed a low overlap between taxa and comparisons between the null model and the generated overlay link probably occurring niche conservatism. The data suggest that the taxa that occur in the Amazon and Atlantic forest represent three distinct species
