Hemiorchidectomy (HO) in the adult male bonnet monkey results in a selective increase in circulating concentrations of FSH and testosterone, and this is accompanied by compensatory increase in sperm production by the remaining testis. We investigated the possible role of increased FSH concentration that occurs after HO in the compensatory increase in the activity of the remaining testis. Of eight adult male bonnet monkeys that underwent HO, four received i.v. injections every other day for 30 days of a well-characterized ovine FSH antiserum (a/s) that cross-reacts with monkey FSH. The remaining four males received normal monkey serum (NMS) as control treatment in a protocol similar to that employed for ais-treated males. Blood samples were collected between 2100 and 2200 h before and 1/2, 1, 3, 5, 7, 14, 22, and 29 days after HO. Testicular weight, number of 3 beta-hydroxy steroid dehydrogenase-positive (3 beta-HSD+) cells, and DNA flow cytometric analysis of germ cell populations were obtained for testes collected before and at the termination of NMS or ais treatment. In NMS-treated males, circulating serum FSH concentrations progressively increased to reach a maximal level by Day 7 after HO (1.95 +/- 0.3 vs. 5.6 +/- 0.7 ng/ml on Days -1 and 7, respectively). Within 30 min of ais injection, FSH antibodies were detected in circulation, and the antibody level was maintained at a constant level between Day 7 and end of treatment (exhibiting 50-60% binding to I-125-hFSH). Although circulating mean nocturnal serum testosterone concentration showed an initial decrease, it rose gradually to pre-HO concentrations by Day 7 in NMS-treated males. In contrast, nocturnal mat serum testosterone concentrations in a/s-treated males remained lower than in NMS-treated controls (p < 0.05) up to Day 22 and thereafter only marginally increased. Testicular weights increased (p < 0.05) over the pre-HO weight in NMS- but not in ais-treated males. After HO, the number of 3 beta-HSD+ cells (Leydig cells) was markedly increased but was significantly (p < 0.05) higher in NMS-treated males compared to a/s-treated males. A significant (p < 0.05) reduction in the primary spermatocyte population of germ cells was observed in ais-treated compared to NMS-treated males. These results suggest that the increased FSH occurring after HO could be intimately involved in increasing the compensatory functional activity of the remaining testis in the male bonnet monkey.

A study of 140 days duration was performed to examine if human male volunteers (n = 5) respond to ovine follicle stimulating hormone (oFSH) immunization (administered adsorbed on Alugel on days 1, 20, 40 and 70) by producing antibodies capable of both binding and neutralizing bioactivity of human FSH. The kinetics of antibody production for both the immunogen (oFSH) and the cross-reactive antigen (hFSH) were essentially similar, The volunteers responded only to the first two immunizations, The boosters given on days 40 and 70 were ineffective, probably because of the presence of substantial amounts of circulating antibody to oFSH. Of the antibodies generated to oFSH, 25-45% bound hFSH with a mean binding affinity of 0.65 x 10(9) +/- 0.53 M(-1). The binding capacities at the time of high (30-80 days of immunization) and low (>110 days) titres were 346 +/- 185 and 10.5 +/- 5.8 ng hFSH/ml respectively, During the period of high titre, free serum FSH (value in normal males 1-5 ng/ml) was not monitorable, A 50 mu l aliquot of the antiserum obtained from different volunteers between days 30 and 80 and on day 140 blocked binding of I-125-labelled hFSH to its receptor by 82 +/- 9.7 and 53 +/- 12.2% respectively, The antibody produced was specific for FSH, and no significant change in the values of related glycoprotein hormones (luteinizing hormone/testosterone and thyroid stimulating hormone/thyroxine) were recorded, Seminal plasma transferrin, a marker of Sertoli cell as well as of seminiferous tubular function, showed marked reduction (30-90%) following immunization with oFSH. Considering that endogenous FSH remained neutralized for approximately one sperm cycle only (65 days), the reduction in sperm counts (30-74%) exhibited by some volunteers is encouraging, Immunization with oFSH did not result in any significant changes in haematology, serum biochemistry or hormonal profiles, There was no production of antibodies capable of interacting with non-specific tissues, It is concluded that it should be possible to obtain a sustained long-term blockade of endogenous FSH action in men by using oFSH as an immunogen, This is a prerequisite for obtaining significant reduction in the quality and quantity of spermatozoa produced, thus leading to infertility.

Results of Western blot analysis carried out with an interstitial cell extract from male guinea pig and ovarian extract from immature female rats administered equine chorionic gonadotropin (eCG) provide supportive evidence to our earlier suggestion that an 8-kDa peptide is involved in acquisition of steroidogenic capacity by the rat Leydig cells. It was found that though the signal was observed in other tissues such as liver, kidney and lung which do not produce gonadal hormones, the peptide was modulated only by lutenizing hormone (LH) in the rat Leydig cells.

This dissertation: 1) determines the factor(s) responsible for spawning induction in NematosteJla vectensis; 2) isolates, describes, and documents the source of jelly from egg masses of N. vectensis; and 3) describes N. vectensis' early development. Namatostella vectensis were maintained on a 7-day mussel feeding/water change regime over 159 days. Within 36 hours of mussel feeding/water change. 69.1% of females and 78.5% of males spawned reliably. Through manipulation of feeding, water change, oxygen and nitrogenous waste concentrations, spawning induction was found to be triggered by the oxygen concentration associated with water change, and not by feeding. Ammonia, anemones' major waste product, inhibited this induction in a concentration-dependent manner. Female N. vectensis release eggs in a persistent jellied egg mass which is unique among the Actiniaria. The major component of this egg mass jelly was a positive periodic acid-Schiffs staining, 39.5-40.5 kD glycoprotein. Antibodies developed in rabbits against this glycoprotein bound to jelly of intact egg masses and to granules (~ 2.8 IJm in diameter) present in female anemone mesenteries and their associated filaments. Antibodies did not label male tissues. Nematostella vecfensis embryos underwent first karyokinesis -60 minutes following the addition of sperm to eggs. Second nuclear division took place, followed by first cleavage, 90-120 minutes later. Each of the 4 blastomeres that resulted from first cleavage contained a single nucleus. Arrangement of these blastomeres ranged from radial to pseudospiral. Embryonic development was both asynchronous and holoblastic. Following formation of the 4-cell stage, 71% of embryos proceeded to cleave again to form an 8-cell stage. In each of the remaining 29% of embryos, a fusion of from 2-4 blastomeres resulted in 4 possible patterns which had no affect on either cleavage interval timing or subsequent development. The fusion event was not due to ooplasmic segregation. Blastomeres isolated from 4-celled embryos were regulative and developed into normal planula larvae and juvenile anemones that were 1/4 the size of those that developed from intact 4-celled embryos. Embryos exhibiting the fusion phenomenon were examined at the fine structural level. The fusion phenomenon resulted in formation of a secondary syncytium and was not a mere compaction of blastomeres.

Menlicirrhus americanus in the northwestern Gulf of Mexico mature at 150-220 mm TL and 12-14 months of age, with males maturing when 10-40 mm smaller than females. Spawning occurs within a broad period from February through November with two discrete peaks which coincide with the periodicity of downcoast alongshore currents (towards Mexico) in spring and fall. This species occurs at depths of less than 5 to 27 m, being most abundant at 5 m or shallower. Young-of-the-year recruit primarily at 5-9 m or shallower and gradually expand their bathymetric range. Age determination by length frequency is feasible in M. americanus but not as simple as in species that spawn in one major period of the year. Only one or two spawned groups normally predominated at anyone time and no more than three co-occurred with few possible exceptions. Observed mean sizes were 138 mm TL at 6 months, and 192 and 272 mm at ages I and II, respectively. Typical maximum size was 296-308 mm and typical maximum age is probably 2-3 years. The largest fISh captured were 392 and 455 mm. Observed sex ratio was 1.2 females to 1 male. Weight, girth, and length-length regressions are presented.(PDF file contains 27 pages.)

Ethmalosa fimbriata (Bowdich) is the most abundant clupeid in the Ivorian artisanal fisheries. It represents over 60% of lagoon catches. In Aby lagoon the size at first sexual maturity is situated at 9.6 cm for male and 9.9 cm for female. The reproduction takes place twice a year during the high salinity periods from February to May and July to August. This reproduction does not affect significantly the sex-ratio, the mean of which varies from 35% to 36%, depending the period. The fecondity is very fluctuating and varies from 11000 eggs to 148000 eggs per female.

The aim of this study was to evaluate the normalized response speed (Vrn) of the knee musculature (flexor and extensor) in high competitive level volleyball players using tensiomyography (TMG) and to analyze the muscular response of the vastus medialis (VM), rectus femoris (RF), vastus lateralis (VL), and biceps femoris (BF) in accordance with the specific position they play in their teams. One hundred and sixty-six players (83 women and 83 men) were evaluated. They belonged to eight teams in the Spanish women's superleague and eight in the Spanish men's superleague. The use of Vrn allows avoiding possible sample imbalances due to anatomical and functional differences and demands. We found differences between Vrn in each of the muscles responsible for extension (VM, RF, and VL) and flexion (BF) regardless of the sex. Normalized response speed differences seem to be larger in setters, liberos and outside players compared to middle blockers and larger in males when compared to females. These results of Vrn might respond to the differences in the physical and technical demands of each specific position, showing an improved balance response of the knee extensor and flexor musculature in male professional volleyball players.

Age, growth, and reproductive data were obtained from dolphinfish (Coryphaena hippurus, size range: 89 to 1451 mm fork length [FL]) collected between May 2002 and May 2004 off North Carolina. Annual increments from scales (n=541) and daily increments from sagittal otoliths (n=107) were examined; estimated von Bertalanffy parameters were L∞ (asymptotic length)=1299 mm FL and k (growth coefficient)=1.08/yr. Daily growth increments reduced much of the residual error in length-at-age estimates for age-0 dolphinfish; the estimated average growth rate was 3.78 mm/day during the first six months. Size at 50% maturity was slightly smaller for female (460 mm FL) than male (475 mm FL) dolphinfish. Based on monthly length-adjusted gonad weights, peak spawning occurs from April through July off North Carolina; back-calculated hatching dates from age-0 dolphinfish and prior reproductive studies on the east coast of Florida indicate that dolphinfish spawning occurs year round off the U.S. east coast and highest levels range from January through June. No major changes in length-at-age or size-at-maturity have occurred since the early 1960s, even after substantial increases in fishery landings.

We studied a small artisanal fishery for the spotted eagle ray (Aetobatus narinari) off Margarita Island in northeastern Venezuela. We analyzed data from 413 fishing trips directed at A. narinari over a 29-month sampling period (August 2005–December 2007). These trips yielded 55.9 metric tons and 1352 individuals from which a subsample of 846 females and 321 males was used for biological data. Maximum fishing effort and landings occurred between February and May, and catch per unit of effort was highest between December and February and between July and October with an overall average of 3 individuals and 133 kg per trip. The overall sex ratio was significantly different from 1:1 with a predominance of females. Females ranged in size with disc widths (DW) from 64 to 226 cm. Males ranged in size between 97 and 190 cm DW. There was no statistically significant difference between male and female length-weight relationships. Mean fecundity was estimated at 3.09 embryos per female, and the largest embryo measured 44.5 cm DW. Females in different maturity stages were found in all months, except November 2007, the month when all females were immature. Postgravid females occurred mainly during the periods of January–May and July–October. Mean length (L50) at maturity was estimated at 129.2 cm DW for males and 134.9 cm DW for females. This study provides much needed information on the biology and life history of A. narinari for the management of an intensive, directed, small-scale fishery for this little known species in northeastern Venezuela.

The presence of endogenous opioid peptides in different testicular cell types has been extensively characterized and provides evidence for the participation of the opioid system in the regulation of testicular function. However, the exact role of the opioid system during the spermatogenesis has remained controversial since the presence of the mu-, delta-and kappa-opioid receptors in spermatogenic cells was yet to be demonstrated. Through a combination of quantitative real-time PCR, immunofluorescence, immunohistochemistry and flow cytometry approaches, we report for the first time the presence of active mu-, deltaand kappa-opioid receptors in mouse male germ cells. They show an exposition time-dependent response to opioid agonist, hence suggesting their active involvement in spermatogenesis. Our results contribute to understanding the role of the opioid receptors in the spermatogenesis and could help to develop new strategies to employ the opioid system as a biochemical tool for the diagnosis and treatment of male infertility.

We examined 536 permit (Trachinotus falcatus, 65–916 mm FL) collected from the waters of Florida Keys and from the Tampa Bay area on Florida’s Gulf coast to describe their growth and reproduction.Among permit that we sexed, females ranged from 266 to 916 mm in length (mean=617) and males ranged from 274 to 855 mm (mean=601). Ages of 297 permit ranging from 102 to 900 mm FL were estimated from thin-sectioned otoliths (sagittae). The large proportion of otoliths with an annulus on the margin and an otolith from an OTC-injected fish suggested that a single annulus was formed each year during late spring or early summer.Permit reach a maximum age of at least 23 years.Permit grew rapidly until an age of about five years, and then growth slowed considerably. Male and female von Bertalanffy growth models were not significantly different, and the sexes-combined growth model was FL=753.1(1–e –0.348(Age+0.585)). Gonad development was seasonal, and spawning occurred during late spring and summer over artificial and natural reefs at depths of 10–30 m. Ovaries that contained oocytes in the final stages of oocyte maturation or postovulatory follicles were found during May–July. We estimated that 50% of the females in the population had reached sexual maturity by 547 mm and an age of 3.1 years and that 50% of the males in the population had reached sexual maturity by 486 mm and an age of 2.3 years. Because Florida regulations restrict the maximum size of permit caught in recreational and commercial fisheries to 20-inch (508-mm), most fish harvested are sexually immature. With the current size selectivity of the fishery, the spawning stock biomass of permit could decrease quickly in response to moderate levels of fishing mortality; thus, the regulations in place in Florida to restrict harvest levels appear to be justified.

To characterise central neurons in the pedal ganglia of both male and female green lipped mussel, Perna canaliculus immunohistochemical techniques were used. Mollusc antibodies were used against neuropeptides and neurotansmitters known to control reproduction and spawning. Anti-ELH and anti-APGWamide showed very strong immunoreactivity in small type of neurons. Anti-5-HT and anti-DA immunoreactivity was mostly in large type of neurons. The labelled neurons are consistent with descriptions of neurosecretory cells implicated in the control of reproduction and spawning on the basis of earlier histological staining techniques used in this species. The use of selective immunological markers for peptides and amines appears to be a, promising tool for further characterisation of neurosecretory cells, and to isolate an'tl characterise neuropeptides and other biologically active materials involved in the control of reproduction in Perna canaliculus.
