Within this thesis, new approaches for the concepts of peptide-polymer conjugates and peptide-based hybrid nanomaterials are investigated. In the first part, the synthesis of a triblock polymer-peptide-polymer is carried out following a typical peptide coupling reaction, both in solution and on solid-phase. The peptide sequence is chosen, so that it is cleaved by an enzyme preparation of trypsin. End-functionalized polystyrene is used as a model hydrophobic polymer and coupled to the peptide sequence. The results show successful coupling reactions in both methods, while the solid phase method produced a more defined product. Suspensions, consisting of peptide-polymer conjugates particles, are prepared in water by ultrasonication. In contact with the enzyme, the peptide constituting the conjugated particles is cleaved. This demonstrates the enzymatic cleavage in heterophase of enzymatic sequence bond to hydrophobic polymers, and is of great interest for the encapsulation and delivery of hydrophobic molecules.rnA second approach is the preparation of peptide-based hybrid nanocapsules. This is achieved by interfacial polyaddition in inverse miniemulsion with the peptide sequence functionalized with additional amino acids. A method suitable to the use of a peptide sequence for interfacial polyaddition was developed. It is shown that, the polarity of the dispersed phase influences the structures prepared, from particle-like to polymeric shell with a liquid core.rnThe peptide sequence is equipped with a FRET pair (more exactly, an internally-quenched fluorescent system) which allows the real-time monitoring of the enzymatic cleavage of the recognition site. This system shows the successful cleavage of the peptide-based nanocapsules when trypsin preparation is added to the suspensions. A water-soluble fluorescent polymer is efficiently entrapped and its possible use as marker for the capsules is highlighted. Furthermore, a small water-soluble fluorescent dye (SR-101) is successfully encapsulated and the encapsulation efficiency as a function of the functionality of the peptide and the amount of comonomer equivalent (toluene diisocyanate) is studied. The dye is encapsulated at such a high concentration, that self-quenching occurs. Thus, the release of the encapsulated dye triggered by the enzymatic cleavage of the peptide results in a fluorescence recovery of the dye. The fluorescence recovery of the FRET pair in the peptide and of the encapsulated dye correlate well.rnFinally, nanocapsules based on a hepsin-cleavable peptide sequence are prepared. Hepsin is an enzyme, which is highly upregulated in prostate cancer cells. The cleavage of the nanocapsules is investigated with healthy and “cancerous” (hepsin-expressing) cell cultures. The degradation, followed via fluorescence recovery of the FRET system, is faster for the suspensions introduced in the hepsin expressing cell cultures.rnIn summary, this work tackles the domain of responsive nanomaterials for drug delivery from a new perspective. It presents the adaptation of the miniemulsion process for hybrid peptide-based materials, and their successful use in preparing specific enzyme-responsive nanoparticles, with hydrophilic payload release properties.rn

Der Suche nach neuen Wirkstoffen für den chemischen Pflanzenschutz kommt insbesondere vor dem Hintergrund der steigenden Weltbevölkerung und weniger zur Verfügung stehenden kulturfähigen Ackerflächen eine stetig wachsende Bedeutung zu. Ziel dieser Arbeit war die Synthese von cyclischen Peptiden und Depsipeptiden, die aufgrund ihrer biologischen Aktivität als potentielle Insektizide für den chemischen Pflanzenschutz in Frage kommen. Darüber hinaus sollten von Kohlenhydraten abgeleitete Katalysatoren zur enantioselektiven Cyanhydrinsynthese entwickelt werden, um einen leichten Zugang zu den Bausteinen der Depsipeptide zu ermöglichen. Als vielversprechender Naturstoff mit insektiziden Eigenschaften gilt das cyclische Pentapeptid Cycloaspeptid E, dessen Totalsynthese in 10 Stufen mit einer Gesamtausbeute von 25% erreicht wurde, sodass die Verbindung für biologische Tests bereitgestellt werden konnte. Zusätzlich gelang die Kristallisation der Verbindung, was eine Röntgenstrukturanalyse ermöglichte. Ein Derivat von Cycloaspeptid E sollte 2-Aminonicotinsäure anstelle von Anthranilsäure enthalten. Die Synthese dieser Verbindung wurde auf drei Wegen versucht. Dabei zeigte sich, dass es bei einer zur Totalsynthese des Naturstoffs analogen Strategie zur quantitativen Bildung eines Diketopiperazins kommt. Auf den anderen Routen ließ sich entweder ein Kupplungsschritt nicht realisieren, oder die Verbindung erwies sich unter den gewählten Bedingungen als instabil. Die Darstellung eines 2-Aminonicotinsäure-Derivats von Cycloaspeptid E bleibt daher weiterhin ein ungelöstes Problem, das weiterer Forschung bedarf. Verticilid A1 ist ein cyclisches Depsipeptid, das aufgrund seiner Bindungsfähigkeit an den Ryanodinrezeptor von Insekten, als Leitstruktur für die Suche nach neuen Insektiziden von Interesse ist. Um zu untersuchen, wie wichtig die Esterbindungen im Molekül für die biologische Aktivität sind, sollte das entsprechende Amid-Derivat und das Cyclodepsipeptid mit nur zwei statt vier Esterbindungen hergestellt werden. Hierbei zeigte sich, dass eine zur Darstellung von Verticilid A1 analoge Syntheseroute zu einer ausgeprägten Epimerisierung führt. Eine lineare Synthese der Derivate endet in der Bildung des Diketopiperazins. Weiterhin wurden zwei neue, zueinander pseudoenantiomere Vanadium(IV)-Katalysatoren auf Basis von D-Glucose einerseits und L-Xylose andererseits dargestellt. Diese lassen sich in fünf bzw. 14 Stufen synthetisieren und liefern in der enantioselektiven Katalyse von Mandelsäurenitril Enantiomerenüberschüsse von 89% bzw. 91% bei hohen Ausbeuten. Zusammenfassend lässt sich feststellen, dass im Rahmen dieser Arbeit die Totalsynthese von Cycloaspeptid E erfolgreich durchgeführt wurde, und die Syntheseversuche von weiteren cyclischen Peptiden wichtige Erkenntnisse für weitere Synthesen lieferten. Mit den beiden hergestellten Vanadium(IV)-Komplexen wurden zwei potente, auf Kohlenhydraten basierende Katalysatoren für die enantioselektive Synthese von Cyanhydrinen entwickelt.

Il peptide correlato al gene della calcitonina (CGRP) è una molecola presente nei neuroni del midollo spinale di diverse specie di Mammiferi, inclusi topi, ratti, conigli, cani, gatti, pecore, scimmie e uomo. Nonostante la distribuzione dei neuroni contenenti questo neuropeptide sia stata studiata in maniera dettagliata nel midollo spinale delle suddette specie, non sono disponibili, in letteratura, informazioni relative alla presenza di queste cellule nel midollo spinale dei Cetacei. Di conseguenza, è stata condotta la presente ricerca che ha avuto lo scopo di determinare, mediante metodiche di immunoistochimica, la distribuzione e la morfologia dei neuroni esprimenti il CGRP nel midollo spinale di tursiope (Tursiops truncatus). In questa specie, la distribuzione laminare (secondo Rexed) dei neuroni CGRP-immunoreattivi è assai simile a quella che si osserva nei Roditori, nei Carnivori e nei Primati; infatti, i corpi cellulari immunopositivi sono localizzati soprattutto in corrispondenza dell’apice del corno dorsale (lamine I e II) e nel corno ventrale (lamine VIII e IX). La distribuzione e la morfologia dei neuroni esprimenti CGRP nel midollo spinale di tursiope suggeriscono come tale neuropeptide possa essere coinvolto nella trasmissione delle informazioni sia sensitive (somatiche e viscerali) che motorie. I neuroni CGRP-immunoreattivi localizzati nelle lamine I e II del midollo spinale di tursiope, come dimostrato in altre specie, potrebbero agire da interneuroni modulando le informazioni nocicettive che dai gangli spinali vengono trasmesse al midollo spinale. Nelle lamine I e II sono presenti anche numerosi processi immunopositivi che, oltre ad appartenere a neuroni locali, derivano, molto probabilmente, dai ai neuroni pseudounipolari dei gangli spinali. In accordo con quanto appena affermato, è opportuno sottolineare come le fibre afferenti primarie provenienti dai gangli spinali utilizzino il CGRP per la trasmissione delle informazioni dolorifiche. La presenza di CGRP nei neuroni della lamina VIII, invece, indica come questo neuropeptide possa essere implicato nella trasmissione di segnali di natura motoria, utilizzando meccanismi presinaptici. Infine, la presenza di numerosi motoneuroni immunoreattivi per il CGRP nella lamina IX indicherebbe un’azione diretta svolta da questo neuropeptide nell’interazione tra motoneurone inferiore e muscolo scheletrico.

In den vergangenen Jahren wurden einige bislang unbekannte Phänomene experimentell beobachtet, wie etwa die Existenz unterschiedlicher Prä-Nukleations-Strukturen. Diese haben zu einem neuen Verständnis von Prozessen, die auf molekularer Ebene während der Nukleation und dem Wachstum von Kristallen auftreten, beigetragen. Die Auswirkungen solcher Prä-Nukleations-Strukturen auf den Prozess der Biomineralisation sind noch nicht hinreichend verstanden. Die Mechanismen, mittels derer biomolekulare Modifikatoren, wie Peptide, mit Prä-Nukleations-Strukturen interagieren und somit den Nukleationsprozess von Mineralen beeinflussen könnten, sind vielfältig. Molekulare Simulationen sind zur Analyse der Formation von Prä-Nukleations-Strukturen in Anwesenheit von Modifikatoren gut geeignet. Die vorliegende Arbeit beschreibt einen Ansatz zur Analyse der Interaktion von Peptiden mit den in Lösung befindlichen Bestandteilen der entstehenden Kristalle mit Hilfe von Molekular-Dynamik Simulationen.rnUm informative Simulationen zu ermöglichen, wurde in einem ersten Schritt die Qualität bestehender Kraftfelder im Hinblick auf die Beschreibung von mit Calciumionen interagierenden Oligoglutamaten in wässrigen Lösungen untersucht. Es zeigte sich, dass große Unstimmigkeiten zwischen etablierten Kraftfeldern bestehen, und dass keines der untersuchten Kraftfelder eine realistische Beschreibung der Ionen-Paarung dieser komplexen Ionen widerspiegelte. Daher wurde eine Strategie zur Optimierung bestehender biomolekularer Kraftfelder in dieser Hinsicht entwickelt. Relativ geringe Veränderungen der auf die Ionen–Peptid van-der-Waals-Wechselwirkungen bezogenen Parameter reichten aus, um ein verlässliches Modell für das untersuchte System zu erzielen. rnDas umfassende Sampling des Phasenraumes der Systeme stellt aufgrund der zahlreichen Freiheitsgrade und der starken Interaktionen zwischen Calciumionen und Glutamat in Lösung eine besondere Herausforderung dar. Daher wurde die Methode der Biasing Potential Replica Exchange Molekular-Dynamik Simulationen im Hinblick auf das Sampling von Oligoglutamaten justiert und es erfolgte die Simulation von Peptiden verschiedener Kettenlängen in Anwesenheit von Calciumionen. Mit Hilfe der Sketch-Map Analyse konnten im Rahmen der Simulationen zahlreiche stabile Ionen-Peptid-Komplexe identifiziert werden, welche die Formation von Prä-Nukleations-Strukturen beeinflussen könnten. Abhängig von der Kettenlänge des Peptids weisen diese Komplexe charakteristische Abstände zwischen den Calciumionen auf. Diese ähneln einigen Abständen zwischen den Calciumionen in jenen Phasen von Calcium-Oxalat Kristallen, die in Anwesenheit von Oligoglutamaten gewachsen sind. Die Analogie der Abstände zwischen Calciumionen in gelösten Ionen-Peptid-Komplexen und in Calcium-Oxalat Kristallen könnte auf die Bedeutung von Ionen-Peptid-Komplexen im Prozess der Nukleation und des Wachstums von Biomineralen hindeuten und stellt einen möglichen Erklärungsansatz für die Fähigkeit von Oligoglutamaten zur Beeinflussung der Phase des sich formierenden Kristalls dar, die experimentell beobachtet wurde.

We recently reported that brief, remotely controlled intrameal hepatic-portal vein infusions of glucagon-like peptide-1 (GLP-1) reduced spontaneous meal size in rats. To investigate the neurobehavioural correlates of this effect, we equipped male Sprague-Dawley rats with hepatic-portal vein catheters and assessed (i) the effect on eating of remotely triggered infusions of GLP-1 (1 nmol/kg, 5 min) or vehicle during the first nocturnal meal after 3 h of food deprivation and (ii) the effect of identical infusions performed at dark onset on c-Fos expression in several brain areas involved in the control of eating. GLP-1 reduced (P < 0.05) the size of the first nocturnal meal and increased its satiety ratio. Also, GLP-1 increased (P < 0.05) the number of c-Fos-expressing cells in the nucleus tractus solitarii, the area postrema and the central nucleus of the amygdala, but not in the arcuate or paraventricular hypothalamic nuclei. These data suggest that the nucleus tractus solitarii, the area postrema and the central nucleus of the amygdala play a role in the eating-inhibitory actions of GLP-1 infused into the hepatic-portal vein; it remains to be established whether activation of these brain nuclei reflect satiation, aversion, or both.

Coagulation factor XIII (FXIII) stabilizes fibrin fibers and is therefore a major player in the maintenance of hemostasis. FXIII is activated by thrombin resulting in cleavage and release of the FXIII activation peptide (AP-FXIII). The objective of this study was to characterize the released AP-FXIII and determine specific features that may be used for its specific detection. We analyzed the structure of bound AP-FXIII within the FXIII A-subunit and interactions of AP-FXIII by hydrogen bonds with both FXIII A-subunit monomers. We optimized our previously developed AP-FXIII ELISA by using 2 monoclonal antibodies. We determined high binding affinities between the antibodies and free AP-FXIII and demonstrated specific binding by epitope mapping analyses with surface plasmon resonance and enzyme-linked immunosorbent assay. Because the structure of free AP-FXIII had been characterized so far by molecular modeling only, we performed structural analysis by nuclear magnetic resonance. Recombinant AP-FXIII was largely flexible both in plasma and water, differing significantly from the rigid structure in the bound state. We suggest that the recognized epitope is either occluded in the noncleaved form or possesses a structure that does not allow binding to the antibodies. On the basis of our findings, we propose AP-FXIII as a possible new marker for acute thrombotic events.

Glucagon-like peptide-1 (GLP-1) receptors are highly overexpressed in benign insulinomas, permitting in vivo tumour visualisation with GLP-1 receptor scanning. The present study sought to evaluate the GLP-1 receptor status in vitro in other pancreatic disorders leading to hyperinsulinaemic hypoglycaemia, specifically after gastric bypass surgery.

Glucagonlike peptide-1 receptors (GLP-1R) play an increasingly important role in endocrine gastrointestinal tumor management. In particular, virtually all benign insulinomas express GLP-1R in high density. Exendin-4 is a GLP-1 analog that has a longer half-life than GLP-1. Targeting GLP-1R by (111)In-DOTA-exendin-4 or (111)In-DPTA-exendin-4 offers a new approach that permits the successful localization of small benign insulinomas. It is likely that this new noninvasive technique has the potential to replace the invasive localization by selective arterial stimulation and venous sampling.

Gastrinomas, a rare group of neuroendocrine tumors, are responsible for severe peptic disease and diarrhea. Although symptomatic control may be achieved with proton-pump inhibitors (PPIs) and somatostatin analogues (SSAs), data are limited regarding the possible antitumor effect of the peptide receptor radioligand therapy (PRRT) with radiolabeled SSAs in gastrinoma patients. The goal of this study was to assess the effect of PRRT on symptoms, gastrin secretion, and tumor load in patients with progressive malignant gastrinomas.

The G(βγ)-sequestering peptide β-adrenergic receptor kinase (βARK)ct derived from the G-protein-coupled receptor kinase (GRK)2 carboxyl terminus has emerged as a promising target for gene-based heart failure therapy. Enhanced downstream cAMP signaling has been proposed as the underlying mechanism for increased β-adrenergic receptor (βAR) responsiveness. However, molecular targets mediating improved cardiac contractile performance by βARKct and its impact on G(βγ)-mediated signaling have yet to be fully elucidated.

The production of epithelial neutrophil activating peptide-78 (NA-78) and the interleukins IL-8 and IL-6 by endometrial stromal cells is stimulated by pro-inflammatory interleukin-1 (IL-1) and tumour necrosis factor-α (TNF-α). IL-8 is suggested to play a role in the pathogenesis of endometriosis, and in these women the peritoneal fluid concentrations of ENA-78 and IL-8 are increased. TNF-α has been tested together with interferon-γ because of their cooperative stimulation of IL-6. The release of IL-8, however, is inhibited with increasing interferon levels. The aim of the study was the analysis of the production of ENA-78, IL-6 and IL-8 by cultured human endometrial stromal cells in the presence of varying concentrations of IL-1β, TNF-α, and interferon-γ.
