Das Ziel dieser Arbeit besteht darin, die Möglichkeiten der Sprühtrocknung für die Generierung von Inhalationspulvern zur Therapie von Lungenkrankheiten zu nutzen. Die Erzeugung von physikalisch stabilen und leicht dispergierbaren Partikeln steht hierbei im Vordergrund. Aufgrund von physiko-chemischen Untersuchungen (Glasübergangstemperatur, Fragilität, Relaxationsverhalten, Hygroskopizität) unterschiedlicher amorpher Hilfsstoffe (Lactose, Raffinose, Dextrane, Cyclodextrine) ist für Hydroxypropyl-β-Cyclodextrin das größte Potential für die Stabilisierung eines Wirkstoffes innerhalb einer amorphen Matrix erkennbar. Sprühgetrocknete Partikel weisen im Vergleich zu strahlgemahlenen Partikeln günstigere Dispergier- und Depositionseigenschaften auf. Dies ist vorrangig auf größere Berührungsflächen zwischen strahlgemahlenen Partikeln zurückzuführen. Kugelförmige sprühgetrocknete Partikel besitzen dagegen aufgrund einer punktförmigen Berührung geringere Haftkräfte. Versuche mit unterschiedlich stark gefalteten Partikeloberflächen weisen auf geringere Haftkräfte hin, wenn sich die Partikel an Stellen geringerer Krümmungsradien berühren. Dispergierversuche in einer definierten Rohrströmung (Deagglomerator) lassen auf einen kaskadenartigen Agglomeratzerfall schließen. Durch Sprüheinbettung unterschiedlicher Modellwirkstoffe (Salbutamolsulfat, Ipratropiumbromid, Budesonid) in Hydroxypropyl-β-Cyclodextrin konnten sowohl Einzelformulierungen als auch eine Kombinationsformulierung mit allen drei Wirkstoffen erzeugt werden. Diese weisen bei einem Wirkstoffgehalt bis max. 14% selbst nach vierwöchiger Offenlagerung bei 40°C und 75% r.F. keine bzw. nur geringfügige Veränderungen in der „Fine Particle Dose“ (FPD) auf. Die „Fine Particle Fraction“ (FPF) liegt bei diesen Formulierungen im Bereich von 40% bis 75%. In Verbindung mit einem geeigneten Pack- bzw. Trockenmittel, ist hierbei mit einer physikalischen Stabilität zu rechnen, die eine sinnvolle Produktlaufzeit eines Inhalationspulvers ermöglicht. Formulierungen mit höheren Wirkstoffkonzentrationen zeigen dagegen stärkere Veränderungen nach Stresslagerung. Als Beispiel einer kristallinen Sprühtrocknungsformulierung konnte ein Pulver bestehend aus Mannitol und Budesonid erzeugt werden.

Il siero di latte e la scotta sono effluenti provenienti rispettivamente dal processo di trasformazione del latte in formaggio e ricotta. Il siero di latte contiene minerali, lipidi, lattosio e proteine; la scotta contiene principalmente lattosio. Il siero può essere riutilizzato in diversi modi, come l'estrazione di proteine o per l’alimentazione animale, mentre la scotta è considerata solamente un rifiuto. Inoltre, a causa degli ingenti volumi di siero prodotti nel mondo, vengono a crearsi seri problemi ambientali e di smaltimento. Destinazioni alternative di questi effluenti, come le trasformazioni biotecnologiche, possono essere un modo per raggiungere il duplice obiettivo di migliorare il valore aggiunto dei processi agroindustriali e di ridurre il loro impatto ambientale. In questo lavoro sono state studiate le condizioni migliori per produrre bioetanolo dal lattosio del siero e della scotta. Kluyveromyces marxianus è stato scelto come lievito lattosio-fermentante. Sono state effettuate fermentazioni su scala di laboratorio aerobiche e anaerobiche in batch, fermentazioni semicontinue in fase dispersa e con cellule immobilizzate in alginato di calcio,. Diverse temperature sono state testate per migliorare la produzione di etanolo. Le migliori prestazioni, per entrambe le matrici, sono state raggiunte a basse temperature (28°C). Anche le alte temperature sono compatibili con buone rese di etanolo nelle fermentazioni con siero. Ottimi risultati si sono ottenuti anche con la scotta a 37°C e a 28°C. Le fermentazioni semicontinue in fase dispersa danno le migliori produzioni di etanolo, in particolare con la scotta. Invece, l'uso di cellule di lievito intrappolate in alginato di calcio non ha migliorato i risultati di processo. In conclusione, entrambi gli effluenti possono essere considerati adatti per la produzione di etanolo. Le buone rese ottenute dalla scotta permettono di trasformare questo rifiuto in una risorsa.

Die Untersuchung der Adhäsionskräfte mit Colloid Probe Technik, einer Weiterentwicklung der Rasterkraftmikroskopie (Atomic Force Microscopy=AFM), an erzeugten Carrier- und Wirkstoffkristallen bei Laborbedingungen und unter Einfluss der Luftfeuchte zeigte, dass die Adhäsion von Tiotropiumbromid Monohydrat an Mannitol deutlich höher ist als an Lactose Monohydrat. Die Kohäsionskräfte des Wirkstoffes sind stärker als die Adhäsionskräfte an Carriermaterialien. Auf dieser Grundlage wurde die Hypothese aufgestellt, dass eine Mischung mit Mannitol als Carrier eine kleinere Feinpartikeldosis liefert als eine Mischung mit Lactose. Diese Theorie wurde an interaktiven Pulvermischungen unter Variation von verschiedenen Einflussfaktoren überprüft. Die binare und ternäre Lactose-basierte Mischung lieferte unabhängig vom Kapselmaterial (Gelatine- und Polyethylenkapsel) eine höhere Feinpartikeldosis als die entsprechenden Mannitol-basierten Formulierungen. Die ternäre Komponente bewirkte nur bei Mannitol-basierten Mischungen eine Verbesserung der Feinpartikeldosis. Die detaillierte Untersuchung der aerodynamischen Verteilung ternärer Mischungen zeigte, dass das Kapselmaterial nur unter dem Einfluss der Luftfeuchte und Permeabilität der Blisterverpackung die interpartikulären Wechselwirkungen beeinflusst. Mischungen mit Mannitol als Carrier lieferten unabhängig vom Kapselmaterial, von Luftfeuchte/Lagerungsbedingungen und Permeabilität der Blisterverpackung eine kleinere Feinpartikeldosis als Mischungen mit Lactose als Carrier. Die Carrierart, die Permeabilität der Blisterverpackung und die Luftfeuchte wurden als Haupteinflussfaktoren auf die aerodynamischen Eigenschaften identifiziert. Es konnte gezeigt werden, dass AFM einen wertvollen Beitrag zum Verständnis der interpartikulären Wechselwirkungen leistet und aufgrund prädiktiver Eigenschaften hilfreich in der Entwicklung inhalativer Darreichungs-formen sein kann.

Although ability to digest lactose generally declines after weaning in all mammals, in some human populations it persists also in adult individuals, a condition named lactase persistence (LP). Studies on the prevalence of the LP phenotype in worldwide human populations have shown that the frequency of this trait is highly variable in different ethnic groups, appearing to be positively correlated with the importance of milk in the diet. In particular, several single-nucleotide polymorphisms (SNPs) in the proximity of the LCT gene have been proved to be associated with LP. Nevertheless, few studies have till now analyzed genetic variation underlying LP in a wide set of Eurasian populations and, especially, in the Italian one. In the present study, we thus typed 40 SNPs surrounding the LCT gene in more than 1,000 samples from Italian and Arabic peninsulas to investigate patterns of LP-related genetic diversity in two regions which have played a pivotal role in the recent human evolutionary history according to their geographical position and historical/archaeological records. Our results underline a high and complex variability of the explored genomic region in both studied populations. In particular, a clear diversification of Northern Italian groups from the rest of the peninsula, was observed, with the formers being genetically more similar to Northern European populations than to Southern Italians. These observation are consistent with known decreasing pattern of LP from Northern to Southern Italy and suggest the possibility of an independent evolution of LP-associated genotypes in Northern Italy. A similar scenario was observed in the Arabian peninsula, with Dhofari Arabs from Southern Oman and Yemeni clustering together with respect to Arabs from Northern Oman and the subgroup of Omanis of Asian origin which appeared instead to be genetically closer to Europeans than to the rest of Arabic groups.

Il microbiota intestinale riveste un ruolo importantissimo nell’influenzare la salute dell’ospite. È stato dimostrato come la composizione della dieta possa condizionare lo stato di benessere dell’animale, inducendo importanti cambiamenti tra le popolazioni batteriche che coabitano l’intestino; l’uso di prebiotici rappresenta una delle strategie maggiormente impiegate per modulare positivamente la composizione ed il metabolismo dell’ecosistema gastroenterico. Il presente progetto di dottorato si è proposto di indagare gli effetti sul microbiota intestinale del cane e del gatto di diete a diverso tenore proteico e contenenti proteine di diversa digeribilità in presenza o meno di sostanze prebiotiche. Inoltre, sono stati valutati gli effetti della presenza di un estratto di Yucca schidigera e di tannini sulla microflora intestinale del gatto. In ultima istanza, sono state valutate le conseguenze di dosi crescenti di lattosio sul benessere intestinale del cane. I risultati del presente studio hanno rilevato come le sostanze prebiotiche influiscono sulla composizione e sul metabolismo della microflora del cane e del gatto, e come l’impiego di diete ricche di proteine possa avere conseguenze negative sull’ambiente intestinale. Tuttavia, la presenza di oligosaccaridi non sembra contrastare gli effetti negativi che diete ad alto tenore proteico potrebbero avere sull’ecosistema intestinale dell’animale. Nella successiva prova è stato evidenziato come l’inclusione nella dieta di estratti di Yucca e tannini possa contribuire a mitigare l’emanazione di sostanze maleodoranti dalle deiezioni degli animali da compagnia. Nel corso dell’ultima prova, nonostante non siano state osservate differenze tra le popolazioni microbiche intestinali, la somministrazione di dosi crescenti di lattosio ha indotto una certa riduzione delle fermentazioni proteolitiche microbiche. Ulteriori studi sono necessari per stabilire in che misura la dieta e gli alimenti “funzionali” possano influire sul microbiota intestinale del cane e del gatto e come queste informazioni possono essere utilizzate per migliorare miratamente l’alimentazione e lo stato di salute degli animali da compagnia.

Die chemische Synthese definierter Glycopeptidstrukturen bildet die Basis einiger vielversprechender Ansätze zur Therapie verschiedener Krankheiten. Die Entwicklung hochaffiner Selektininhibitoren könnte der Behandlung chronischer Entzündungen und zur Unterdrückung der Metastasierung von Tumoren dienen. Vollsynthetische Vakzine auf Basis glycosylierter MUC1-Partialstrukturen sollen das Immunsystem zur Bekämpfung von krankem Gewebe anregen und so perspektivisch eine Impfung gegen Krebs ermöglichen. Da die natürlich vorkommenden O-Glycoside in vivo eine begrenzte Stabilität besitzen, wurde eine Methode entwickelt, welche die modulare Herstellung von stabilen rnC-Glycosylaminosäuren als Mimetika der natürlichen Serin-, Threonin- und Tyrosin-Glycoside ermöglicht. Dazu wurden passend geschützte Kohlenhydrat-Lactone synthetisiert, die in einer mikrowellengestützten Petasis-Olefinierung unter Durchflussbedingungen in die entsprechenden exo-Glycale überführt wurden. Die Reaktionszeit konnte durch diese spezielle Reaktionsführung auf weniger als drei Minuten verringert werden, während konventionell mehrere Stunden benötigt werden. Die C-glycosidische Verknüpfung mit den entsprechenden Aminosäurebausteinen gelang durch eine Hydroborierungs-Suzuki-Kupplungs-Kaskade. Nach umfangreicher Optimierung der Reaktionsparameter ließ sich neben mehreren Monosacchariden auch ein exo-Glycal der Lactose erfolgreich in der Kupplung einsetzen. Nach verschiedenen Schutzgruppenmanipulationen wurden einige der synthetisierten Bausteine zur Synthese C-glycosylierter Partialstrukturen des Mucins MUC1 an der festen Phase herangezogen. In ELISA-Experimenten wurden die C-Glycosylpeptide von an Brustkrebsgewebe bindenden Antikörpern erkannt, die durch Vakzinierung mit ähnlichen Strukturen erhalten worden waren. Zur Synthese zweier Bausteine potenzieller Selektin-Inhibitoren wurde ein Mimetikum des in natürlichen Liganden vorkommenden Tetrasaccharides Sialyl-Lewisx synthetisiert. Bei diesem wurde die terminale Sialinsäure durch (S)-Cyclohexylmilchsäure ersetzt. Die bei der gewählten Syntheseroute notwendige regioselektive Öffnung eines Benzylidenacetals wurde in einem Mikroreaktor durchgeführt, wodurch eine einfache Reaktionsoptimierung mit geringen Substanzmengen möglich war. Die Reaktionszeit liegt mit unter 4 Minuten deutlich unter den üblichen Werten von einer bis mehreren Stunden. In einer Block-Glycosylierung konnte das Pseudotetrasaccharid sowohl an einen C-Lactosyl-Tyrosin-, als auch an einen C-Lactosyl-Serin-Akzeptor angefügt und somit die Synthese der Zielverbindungen abgeschlossen werden. Diese Bausteine können in Zukunft als Bestandteile synthetischer Glycopeptide zum Einsatz kommen, welche Mimetika der natürlichen Selektin-Liganden darstellen sollen.rn

Clinical manifestations of lactase (LCT) deficiency include intestinal and extra-intestinal symptoms. Lactose hydrogen breath test (H2-BT) is considered the gold standard to evaluate LCT deficiency (LD). Recently, the single-nucleotide polymorphism C/T(-13910) has been associated with LD. The objectives of the present study were to evaluate the agreement between genetic testing of LCT C/T(-13910) and lactose H2-BT, and the diagnostic value of extended symptom assessment. Of the 201 patients included in the study, 194 (139 females; mean age 38, range 17-79 years, and 55 males, mean age 38, range 18-68 years) patients with clinical suspicion of LD underwent a 3-4 h H2-BT and genetic testing for LCT C/T(-13910). Patients rated five intestinal and four extra-intestinal symptoms during the H2-BT and then at home for the following 48 h. Declaring H2-BT as the gold standard, the CC(-13910) genotype had a sensitivity of 97% and a specificity of 95% with a of 0.9 in diagnosing LCT deficiency. Patients with LD had more intense intestinal symptoms 4 h following the lactose challenge included in the H2-BT. We found no difference in the intensity of extra-intestinal symptoms between patients with and without LD. Symptom assessment yielded differences for intestinal symptoms abdominal pain, bloating, borborygmi and diarrhoea between 120 min and 4 h after oral lactose challenge. Extra-intestinal symptoms (dizziness, headache and myalgia) and extension of symptom assessment up to 48 h did not consistently show different results. In conclusion, genetic testing has an excellent agreement with the standard lactose H2-BT, and it may replace breath testing for the diagnosis of LD. Extended symptom scores and assessment of extra-intestinal symptoms have limited diagnostic value in the evaluation of LD.

The aim of this study was to investigate the effects of a severe nutrient restriction on mammary tissue morphology and remodeling, mammary epithelial cell (MEC) turnover and activity, and hormonal status in lactating dairy cows. We used 16 Holstein x Normande crossbred dairy cows, divided into 2 groups submitted to different feeding levels (basal and restricted) from 2 wk before calving to wk 11 postpartum. Restricted-diet cows had lower 11-wk average daily milk yield from calving to slaughter than did basal-diet cows (20.5 vs. 33.5 kg/d). Feed restriction decreased milk fat, protein, and lactose yields. Restriction also led to lower plasma insulin-like growth factor 1 and higher growth hormone concentrations. Restricted-diet cows had lighter mammary glands than did basal-diet cows. The total amount of DNA in the mammary gland and the size of the mammary acini were smaller in the restricted-diet group. Feed restriction had no significant effect on MEC proliferation at the time of slaughter but led to a higher level of apoptosis in the mammary gland. Gelatin zymography highlighted remodeling of the mammary extracellular matrix in restricted-diet cows. Udders from restricted-diet cows showed lower transcript expression of alpha-lactalbumin and kappa-casein. In conclusion, nutrient restriction resulted in lower milk yield in lactating dairy cows, partly due to modulation of MEC activity and a lower number of mammary cells. An association was found between feed restriction-induced changes in the growth hormone-insulin-like growth factor-1 axis and mammary epithelial cell dynamics.

It has been suggested that the ratio of lactose to milk oligosaccharides in mammalian milk/colostrum is based on the ratio of expression of a-lactalbumin and glycosyltransferases in the mammary epithelial cells. It has also been suggested that the high secretion of milk in dairy breed cows has been acquired by a high expression of a-lactalbumin expression. As there is a large difference of milk secretion level between dairy and non dairy breed cows, there may be a difference in the ratio of lactose to milk oligosaccharides in milks between dairy and non dairy breed cows. In this study, the concentrations of hexose, sialic acid as well as sialyllactoses, which are representative bovine milk oligosaccharides, were determined in the milks of dairy and non dairy breed cows. The concentration of hexose was significantly higher in the milks of non dairy breed cows than that of dairy breed cows, but there were no significant differences with respect to sialic acid and sialyllactose. The significant difference of the ratio of the concentrations of 3'- and 6'-sialyllactose to total hexose in milk was not observed between dairy and non dairy cows.

The effect of somatic cell count (SCC) and milk fraction on milk composition, distribution of cell populations, and mRNA expression of various inflammatory parameters was studied. Therefore, quarter milk samples were defined as cisternal (C), first 400 g of alveolar (A1), and remaining alveolar milk (A2) during the course of milking. Quarters were assigned to 4 groups according to their total SCC: 1) <12 x 10(3)/mL, 2) 12 to 100 x 10(3)/mL, 3) 100 to 350 x 10(3)/mL, and 4) >350 x 10(3)/mL. Milk constituents of interest were SCC, fat, protein, lactose sodium, and chloride ions as well as electrical conductivity. Cell populations were classified into lymphocytes, macrophages, and neutrophils (PMN). The mRNA expression of the inflammatory factors tumor necrosis factor-alpha, interleukin-1beta, cyclooxygenase-2, lactoferrin, and lysozyme was measured via real-time, quantitative reverse transcription PCR. Somatic cell count decreased from highest levels in C to lowest levels in A1 and increased thereafter to A2 in all groups. Fat content increased from C to A2 and with increasing SCC level. Lactose decreased with increasing SCC level but remained unchanged during milking. Concentrations of sodium and chloride, and electrical conductivity increased with increasing SCC but were higher in C than in A1 and A2. Protein was not affected by milk fraction or SCC level. The distribution of leukocytes was dramatically influenced by milk fraction and SCC. Lymphocytes were the dominating cell population in group 1, but the proportion of lymphocytes was low in groups 2, 3, and 4. Macrophage proportion was highest in group 2 and decreased in groups 3 and 4, whereas that of PMN increased from group 2 to 4. The content of macrophages decreased during milking in all SCC groups whereas that of PMN increased. The proportion of lymphocytes was not affected by milk fraction. The mRNA expression of all inflammatory factors showed an increase with increasing SCC but minor changes occurred during milking. In conclusion, milk fraction and SCC level have a crucial influence on the distribution of leukocyte populations and several milk constituents. The surprisingly high content of lymphocytes and concomitantly low mRNA expression of inflammatory factors in quarters with SCC <12 x 10(3)/mL indicates a different and possibly reduced readiness of the immune system to respond to invading pathogens.

The capability of goats to maintain milk production during water deprivation is remarkable and not yet fully understood. The aim of the present study was to investigate whether intravenous infusions of hypertonic NaCl cause release of both vasopressin and oxytocin and whether the peptides, in combination with the hyperosmolality, affect milk flow and milk composition. Six Swedish domestic landrace goats in their first to third lactation were milked every 30 min during experiments. Hypertonic NaCl (HNaCl) or isotonic NaCl (IsoNaCl) were infused for 90 min. Goats were not allowed to drink during infusions. Plasma vasopressin concentration increased during HNaCl infusions, and did not change in response to IsoNaCl infusions. Plasma oxytocin concentration did not change during either infusion. Milk flow was maintained during the infusions. Milk fat concentration decreased in the three samples taken before onset of the infusions, but then increased gradually during HNaCl infusions, while it continued to fall during the IsoNaCl infusions. Milk osmolality followed the rise in plasma osmolality during the HNaCl infusions and did not change in IsoNaCl experiments. Milk lactose concentration increased throughout both series of experiments, the concentration being higher during HNaCl infusions. Milk protein concentration did not change during HNaCl infusions, but fell in the IsoNaCl experiments. It is concluded that the hyperosmolality in combination with elevated plasma vasopressin levels did not disturb the secretory activity of the mammary cells, but rather facilitated emptying of the alveolar milk. Such a mechanism may help to explain the sustained milk production in water deprived goats.

BACKGROUND: Contagious bovine pleuropneumonia (CBPP) caused by Mycoplasma mycoides subsp. mycoides small-colony type (SC) is among the most serious threats for livestock producers in Africa. Glycerol metabolism-associated H2O2 production seems to play a crucial role in virulence of this mycoplasma. A wide number of attenuated strains of M. mycoides subsp. mycoides SC are currently used in Africa as live vaccines. Glycerol metabolism is not affected in these vaccine strains and therefore it does not seem to be the determinant of their attenuation. A non-synonymous single nucleotide polymorphism (SNP) in the bgl gene coding for the 6-phospho-beta-glucosidase (Bgl) has been described recently. The SNP differentiates virulent African strains isolated from outbreaks with severe CBPP, which express the Bgl isoform Val204, from strains to be considered less virulent isolated from CBPP outbreaks with low mortality and vaccine strains, which express the Bgl isoform Ala204. RESULTS: Strains of M. mycoides subsp. mycoides SC considered virulent and possessing the Bgl isoform Val204, but not strains with the Bgl isoform Ala204, do trigger elevated levels of damage to embryonic bovine lung (EBL) cells upon incubation with the disaccharides (i.e., beta-D-glucosides) sucrose and lactose. However, strains expressing the Bgl isoform Val204 show a lower hydrolysing activity on the chromogenic substrate p-nitrophenyl-beta-D-glucopyranoside (pNPbG) when compared to strains that possess the Bgl isoform Ala204. Defective activity of Bgl in M. mycoides subsp. mycoides SC does not lead to H2O2 production. Rather, the viability during addition of beta-D-glucosides in medium-free buffers is higher for strains harbouring the Bgl isoform Val204 than for those with the isoform Ala204. CONCLUSION: Our results indicate that the studied SNP in the bgl gene is one possible cause of the difference in bacterial virulence among strains of M. mycoides subsp. mycoides SC. Bgl does not act as a direct virulence factor, but strains possessing the Bgl isoform Val204 with low hydrolysing activity are more prone to survive in environments that contain high levels of beta-D-glucosides, thus contributing in some extent to mycoplasmaemia.

NcMIC4 is a Neospora caninum microneme protein that has been isolated and purified on the basis of its unique lactose-binding properties. We have shown that this protein binds to galactosyl residues of lactose; antibodies directed against NcMIC4 inhibit host cell interactions in vitro, thus making it a vaccine candidate. Because of this feature, NcMIC4 was first purified on a larger scale in its native, functionally active form using lactose-agarose affinity chromatography. Second, NcMIC4 was expressed in Escherichia coli as a histidine-tagged recombinant protein (recNcMIC4) and purified through Ni-affinity chromatography. Third, NcMIC4 cDNA was cloned into the mammalian pcDNA3.1 DNA vector and expression was confirmed upon transfection of Vero cells in vitro. For vaccination studies, we employed the murine cerebral infection model based on C57Bl/6 mice, employing experimental groups of 10 mice each. Two groups were injected intraperitoneally with purified native NcMIC4 and recNcMIC4, respectively, employing RIBI adjuvant. The third group was vaccinated intramuscularly with pcDNA-NcMIC4. Control groups included an infection control, an adjuvant control, and a pcDNA3.1 control group. Following 3 injections at 4-wk intervals, mice were challenged by i.p. inoculation of 2 x 10(6) N. caninum tachyzoites (Nc-1 isolate). During the course of parasite challenge (3 wk), mice from the 3 different test groups showed varying degrees of symptoms bearing a semblance to neosporosis, i.e., walking disorder, rounded back, apathy, and paralysis of the hind limbs. Control groups showed no symptoms at all. Most notably, vaccination with pcDNA-MIC4 proved antiprotective, with 60% of mice succumbing to infection within 3 wk, and all mice lacking a measurable anti-NcMIC4 IgG response. NcMIC4 in its native form elicited a substantial humoral IgG1 immune response and a reduction in cerebral parasite load compared to the controls, but 20% of mice succumbed to infection. Vaccination with recNcMIC4 also resulted in 20% of mice dying; however, in this group, cerebral parasite load was similar to the controls, and recNcMIC4 vaccination elicited a mixed IgG1/IgG2 response. In conclusion, vaccines based on NcMIC4, especially pcDNA-NcMIC4, render mice more susceptible to cerebral disease upon challenge with N. caninum tachyzoites.

A modified Astra type multistage liquid impinger (MSLI) with integrated bronchial cell monolayers was used to study deposition and subsequent drug absorption on in vitro models of the human airway epithelial barrier. Inverted cell culture of Calu-3 cells on the bottom side of cell culture filter inserts was integrated into a compendial MSLI. Upside down cultivation did not impair the barrier function, morphology and viability of Calu-3 cells. Size selective deposition with subsequent absorption was studied for three different commercially available dry powder formulations of salbutamol sulphate and budesonide. After deposition without size separation the absorption rates from the aerosol formulations differed but correlated with the size of the carrier lactose particles. However, after deposition in the MSLI, simulating relevant impaction and causing the separation of small drug crystals from the carrier lactose, the absorption rates of the three formulations were identical, confirming the bioequivalence of the three formulations.

BACKGROUND: Potentized antimony is traditionally used in anthroposophic medicine to enhance hemostasis in bleeding disorders, but evidence of its effectiveness is scarce. On the other hand, non-toxic and economic additional therapeutic options for hemostatic disorders are desirable. OBJECTIVES: We examined all available literature on the subject and performed a controlled pilot in vitro study to test the procoagulatory potency of antimony D 5. DESIGN: Freshly drawn citrated whole blood of 12 healthy volunteers and 12 patients with bleeding disorders was equally distributed into 344 portions, after which it was mixed with antimony D 5, or its potentized vehicle (lactose D 5) as control solution and tested with thrombelastography. The paired t-test and the Wilcoxon signed rank test were used for statistical analysis. In 5 of the 12 healthy donors, a second blood sample was drawn to assess individual variability and increase the total number of replicates. Thus three separate calculations were performed: for the 12 patients, the 12 healthy donors, and the 5 later samples from the same donors. The analysis was exploratory, and no Bonferroni correction was applied. RESULTS: In the antimony D5 samples of the 12 healthy subjects, but not the patients, there was a tendency toward a shorter clotting time (CT) (p = 0.074) and a trend for an increased clot firmness, expressed as maximal amplitude (MA) (p = 0.058). The increase of MA was significant (p = 0.011) when the later samples were included. No statistical difference was detected for the clot formation time and the clot lysis index. CONCLUSION: The exploratory results of this pilot study are inconclusive as to whether antimony D5 has a procoagulatory effect in vitro, although the results suggest an effect on MA and possibly CT. More research is warranted.
