Amostras de músculo de carpa comum (Cyprinus carpio) e de água de três viveiros fertilizados com dejetos de suínos e de um viveiro em que os peixes foram alimentados com ração comercial para peixes foram analisadas durante seis meses. O objetivo foi observar a presença de bactérias patogênicas, como Salmonella, Staphylococcus aureus e coliformes totais e fecais, em peixes comercializados in natura no município de Chapecó, Santa Catarina. Foram aferidos quinzenalmente, na água dos quatro viveiros, oxigênio dissolvido, pH, turbidez, alcalinidade total, dureza total, amônia, nitrato e ortofosfato, bem como a quantidade de coliformes totais e fecais. A análise microbiológica músculo dos peixes demonstrou ausência de Salmonella em todas as amostras. A contagem dos demais microrganismos manteve-se inferior ao limite máximo permitido pela legislação vigente, em todas as amostras. Portanto, os peixes analisados estavam aptos ao consumo humano, pois atenderam aos padrões recomendados pela Divisão Nacional de Vigilância Sanitária de Alimentos (DINAL). A comparação entre os viveiros fertilizados com dejetos de suínos e o viveiro fertilizado com ração comercial para peixes não apresentou diferença significativa, porém apresentou efeito significativo sobre oxigênio dissolvido, turbidez, alcalinidade total e amônia. A matéria orgânica foi responsável pelos maiores valores, com exceção da turbidez, que foi menor no viveiro fertilizado com ração comercial para peixes. Entretanto, após análise de todos os parâmetros físico-químicos e microbiológicos da água dos viveiros, pode-se constatar que todos os viveiros se enquadraram nas classes II ou III da classificação de águas interiores estabelecida pelo CONAMA.

As condições ideais dos frangos de corte no momento do abate devem ser conhecidas a fim de possibilitar a produção de carne de excelente qualidade, uma vez que diversos fatores pré e pós-abate estão envolvidos na qualidade final. em condições normais de abate e processamento, a retirada de ração é feita de 6 a 8 horas antes da apanha das aves, resultando em um período total de jejum de 8 a 12 horas antes do abate, para esvaziar o intestino e com isso minimizar a contaminação no abatedouro. A escalda, depena e evisceração são pontos importantes de contaminação cruzada no abatedouro devido à grande quantidade de microorganismos aderidos às penas, pele e patas das aves e ao rompimento das vísceras durante a evisceração. Entretanto, a desidratação da carcaça começa imediatamente após o início do jejum. Períodos prolongados de jejum podem afetar o pH das diversas partes do intestino, aumentando a presença de Salmonella e outros microorganismos patogênicos. Além disso, determinam uma maior contaminação pela bile, e são, subjetivamente, associados à fragilidade dos intestinos durante a evisceração mecânica. Portanto, os esquemas de processamento devem ser estabelecidos levando-se em conta a integridade e o esvaziamento do intestino e da vesícula biliar, bem como a desidratação e os seus efeitos sobre o bem estar das aves, contaminação da carcaça e qualidade da carne. Como alguns efeitos do jejum ainda não são bem conhecidos, sugerem-se pesquisas nas seguintes áreas: definir o tempo ótimo de jejum para atender o bem estar das aves, minimizar a contaminação e otimizar os parâmetros de qualidade de carcaça; estudar os efeitos de períodos prolongados de jejum sobre o pH e a colonização do papo, pró-ventrículo, moela, intestino delgado, intestino grosso e cecos por enterobactérias, como Salmonella, por exemplo; efeito do jejum sobre o tamanho e cor do fígado. O resultado esperado é um aumento na qualidade final dos produtos aliado a uma redução nas perdas e no custo de produção.

The microbiological flora of 108 water samples was explored to evaluate the role of recreational waters as a possible source of human diseases in Araraquara, S.P., Brazil. These waters included six swimming pools and three lakes with beaches. The number of total and fecal coliforms, Escherichia coli, fecal streptococci, Pseudomonas aeruginosa, Staphylococcus aureus, Candida albicans, and heterotrophic organisms was determined. As was the occurrence of Salmonella, Shigella, Yersinia, enteropathogenic E. coli (EPEC), enteroinvasive E. coli (EIEC), Mycobacteria, yeasts in general and dermatophytes. Shigella, Yersinia, EIEC and dermatophytes were not isolated. Other organisms or groups of microorganisms were found in variable proportions. From this study it is concluded that recreational waters used by the population of Araraquara, may be contaminated with potentially pathogenic microorganism and this may serve as a source of human diseases.

The objectives of the present study were: to evaluate the variation in total number of microorganisms in laying hen fecal samples and to identify the pathogenic microorganisms possibly present such as Staphylococcus aureus, Salmonella sp., Clostridium botulinum, C chauvoei, Campylobacter sp., Escherichia coli, and Corynebacterium sp. The experiment was conducted at the School of Agrarian and Veterinarian Sciences of Jaboticabal, - UNESP. Fecal samples were collected from the aviary. A fully randomized experimental design was used with 7 treatments and 4 replications, for a total of 28 samples. A decrease in total number of microorganisms was detected when treatment T0 (zero days of storage) was compared with treatment T6 (42 days of storage), with oscillations in numbers being observed during this period of time. Pathogenic bacteria such as Clostridium noyvi, C perfringens, C sordelli, C septicum and C noyvi type B were detected, as well as non-pathogenic bacteria such as Bacillus sp. and Staphylococcus epidermidis.

The water produced by the Cristais River Drinking Water Treatment Plant (CR-DWTP) repeatedly produced mutagenic responses that could not be explained by the presence of disinfection byproducts (DBPs) generated by the reaction of humic acids and chlorine. In order to determine the possible role of chlorinated dye products in this mutagenic activity, solutions of a black dye commercial product (BDCP) composed of C. I. Disperse Blue 373, C. I. Disperse Orange 37, C. I. Disperse Violet 93, and chemically reduced BDCP (R-BDCP) were chlorinated in a manner similar to that used by the CR-DWTP. The resulting solutions were extracted with XAD-4 along with one drinking water sample collected from the CR-DWTP. All extracts showed mutagenic activity in the Salmonella/microsome assay. Dye components of the BDCP as well as its reduced chlorinated (Cl-R-BDCP) derivative were detected in the drinking water sample by analysis with a high performance liquid chromatography/diode array detector (HPLC/DAD). The mutagenicity results of these products suggest that they are, at least in part, accounting for the mutagenic activity detected in the drinking water samples from the Cristais River. The data obtained in this study have environmental and health implications because the chlorination of the BDCP and the R-BDCP leads to the formation of mutagenic compounds (Cl-BDCP and Cl-R-BDCP), which are potentially important disinfection byproducts that can contaminate the drinking water as well as the environment.

An indirect ELISA using soluble whole cell antigen was used to screen serum samples obtained from breeder and layer flocks some of which had shown clinical or bacteriological evidence of infection with Salmonella Gallinarum or S. Pullorum. There was good correlation between Salmonella infection and the presence of serum samples showing high optical density (OD) values. Sera from seven flocks showing high values were retested using group D (0-1, 9, 12) lipopolysaccharide (LPS) and g,m-H flagella as detecting antigens. Sera from six flocks produced high OD values with LPS and low values with flagella confirming infection with a non-flagellate, group D Salmonella while one produced high values with both antigens indicating mixed infection with another group D serotype.

The aims of this study were to estimate the changes in total bacterial counts (TBC) in poultry litter samples, consisting of rice hulls, after storage, and to identify pathogenic bacteria. For the countings Plate Count agar (Difco) was used. Enrichment and selective media such as blood agar, MacConkey, Baird Parker, brain and heart agar, and egg yolk solid media, and cooked meat and brain and heart infusion, incubated under aerobic or anaerobic conditions were used to isolate Staphylococcus aureus, Salmonella sp, Clostridium perfringens, C. botulinum, C. chauvoei, Campylobacter sp, Escherichia coli, and Corynebacterium sp. Litter samples were collected from the houses of the Veterinary School experimental aviary. A fully randomized experimental design was used with four treatments and four replications, for a total of 16 samples. A decrease in TBC was detected when treatment T1 (zero days of storage) was compared with treatments T2 (14 days of storage). on the other hand the treatments T3 (28 days of storage) and T4 (42 days of storage) presented significantly superior counting in relation to treatment T1. Some pathogenic bacteria of Enterobacteriaceae such as Escherichia coil, Proteus, Arizona, Providencia, Edwardsiella, as well as Staphylococcus aureus, S. epidermidis, different species of genus Clostridium as C. perfringens, C. sordelli, C. chauvoei, C. tetani and C. novyi as well as some strains of Corynebacterium pyogenes were isolated.

This work shows the chemical characterization of a dye processing plant effluent that was contributing to the mutagenicity previously detected in the Cristais river, São Paulo, Brazil, that had an impact on the quality of the related drinking water. The mutagenic dyes Disperse Blue 373, Disperse Orange 37 and Disperse Violet 93, components of a Black Dye Commercial Product (BDCP) frequently used by the facility, were detected by thin layer chromatography (TLC). The blue and orange dyes were quantified by high performance liquid chromatography (HPLC/DAD) in a raw and treated effluent samples and their contribution to the mutagenicity was calculated based on the potency of each dye for the Salmonella YG1041. In the presence of S9 the Disperse Blue 373 accounted for 2.3% of the mutagenic activity of the raw and 71.5% of the treated effluent. In the absence of S9 the Disperse Blue 373 accounted for 1.3% of the mutagenic activity of the raw and 1.5% of the treated effluent. For the Disperse Orange 37, in the presence of S9, it contributed for 0.5% of the mutagenicity of the raw and 6% of the treated effluent. In the absence of S9; 11.5% and 4.4% of the raw and treated effluent mutagenicity, respectively. The contribution of the Disperse Violet 93 was not evaluated because this compound could not be quantified by HPLC/DAD. Mutagenic and/or carcinogenic aromatic amines were also preliminary detected using gas chromatograph/mass spectrometry in both raw and treated and are probably accounting for part of the observed mutagenicity. The effluent treatment applied by the industry does not seem to remove completely the multagenic compounds. The Salmomella/microsome assay coupled with TLC analysis seems to be an important tool to monitor the efficiency of azo dye processing plant effluent treatments. (c) 2006 Elsevier B.V. To determine the inhibitory capacity of lactic acid bacteria due to the action of antagonistic substances, we tested 474 isolates of Lactobacillus from the crop and cecum of chickens against gram-positive and gram-negative indicator microorganisms by the spot-on-the-lawn and well-diffusion antagonism methods. of the 474 isolates, 265 demonstrated antimicrobial activity against the indicator microorganisms. Isolates identified as L. reuteri, L. salivarius, or Lactobacillus spp. inhibited Enterococcus faecalis, E. faecium, Listeria monocytogenes, and Salmonella spp. but not L. casei, L. delbrueckii, L. fermentum, or L. helveticus by the well-diffusion simultaneous antagonism method under anaerobic incubation conditions. The antagonistic substances produced by some of the Lactobacillus isolates were inactivated after treatment by proteolytic enzymes, which suggested that the substances could be antimicrobial peptides or bacteriocins.

The microbiological quality of various fresh waters in the Araraquara region, state of São Paulo, Brazil was investigated. Ninety-nine water samples were taken from rivers, reservoirs, artesian and non-artesian wells, springs and tap waters, and these waters were analysed using: plate counts of heterotrophic microorganisms (per 1 ml); Most Probable Number (MPN) of fecal coliforms and E. coli (per 100 ml); tests for presence of Salmonella, Shigella, Yersinia, the E. coli pathogens of classes EPEC, ETEC and EIEC and Mycobacterium, Shigella, Yersinia and enteroinvasive E. coli (EIEC) were not isolated. The other types of microorganisms were isolated in varying proportions. We conclude that the waters investigated represent a potential microbiological health risk.

A new isocoumarin with antimicrobial activity was isolated from Paepalanthus vellozioides (a native Brazilian plant) and called paepalantine. This study was carried out to assess the mutagenic activity of this new agent in assays with Salmonella typhimurium TA100, TA98, and TA102 and in Chinese hamster ovary (CHO) cell cultures, as well as cytotoxicity to McCoy cells. Paepalantine caused a significant dose-dependent increase in the frequency of revertants in the three strains used in the assay, both with and without S9 mix, in concentrations varying from 2 to 128 mu g/ plate. The mutagenicity was confirmed in assays with CHO cells treated in the G(1), S, and G(2) phases of the cell cycle. There was an increase in the chromosomal aberration frequency, mainly in the G(2) phase. Furthermore, the mitotic index of the treated cultures (40,80, and 160 mu g/ml) was significantly lower, indicating cytotoxicity. The midpoint cytotoxicity values to McCoy cells by the neutral red (NR) and microculture tetrazolium (MTT) techniques resulted in a NR50, and MTT50 of 30 and 38 mu g/ml, respectively. Alterations to the paepalantine structure are suggested to reduce its mutagenic and cytotoxic activity in investigations for its antineoplasic potential (C) 1997 Wiley-Liss, Inc.

The ability of photoelectrocatalytic oxidation to degrade the commercially important copper-plitalocyanine dye, remazol turquoise blue 15 (RTB) was investigated. The best experimental condition was optimized, evaluating the performance of Ti/TiO2 thin-film electrodes prepared by sol-gel method in the decolourization of 32 mg L-1 RTB dye in 0.5 mol L-1 Na2SO4 pH 8 and applied potential of +1.5 V versus SCE under UV irradiation. Spectrophotometric measurements, high performance liquid chromatography, dissolved organic carbon (TOC) evaluation and stripping analysis of yielding solution obtained after 3 h of photoelectrolysis leads to 100% of absorbance removal from wavelength of 250-800 nm, 79.6% of TOC reduction and the releasing of up to 54.6% dye-bound copper (0.85 mg L-1) into the solution. Both, original and oxidized dye solution did not presented mutagenic activity with the strains TA98 and WOO of Salmonella in the presence and absence of S9 mix at the tested doses. Nevertheless, the yielding photoelectrocatalytic oxidized solution showed an increase in the acute toxicity for Vibrio fischeri bacteria, explained by copper liberation during treatment. The goal of this study was to evaluate the process of salting and dry salting of fillets in pacu (P. mesopotamicus), from cultivation in cages. Fillet with skin were salted at a rate of 40% of salt in relation of the initial weight, for a period of 10 days. After processing, were evaluated for microbiological and centesimal composition of fillets subjected to salting and determining the concentration of sodium chloride (NaCl) and calculation of income. The centesimal composition were evaluated the protein, ether extract, moisture and ash of the raw material fresh and processed products by salting. The microbiological analysis included the determination of coliforms at 45 degrees C, Salmonella and Staphylococcus coagulase positive. The pacus subjected to umided and dry salting were within quality control standards established by the legislation. The content of umided, protein, lipids and chlorides showed differences (p< 0.05) between the processes of salt. However both forms can be used for the development of technological processes of low investment by encouraging new options for fish consumption.
