A variety of type reduction (TR) algorithms have been proposed for interval type-2 fuzzy logic systems (IT2 FLSs). The focus of existing literature is mainly on computational requirements of TR algorithm. Often researchers give more rewards to computationally less expensive TR algorithms. This paper evaluates and compares five frequently used TR algorithms from a forecasting performance perspective. Algorithms are judged based on the generalization power of IT2 FLS models developed using them. Four synthetic and real world case studies with different levels of uncertainty are considered to examine effects of TR algorithms on forecasts accuracies. It is found that Coupland-Jonh TR algorithm leads to models with a better forecasting performance. However, there is no clear relationship between the width of the type reduced set and TR algorithm.

This paper introduces a new type reduction (TR) algorithm for interval type-2 fuzzy logic systems (IT2 FLSs). Flexibility and adaptiveness are the key features of the proposed non-parametric algorithm. Lower and upper firing strengths of rules as well as their consequent coefficients are fed into a neural network (NN). NN output is a crisp value that corresponds to the defuzzified output of IT2 FLSs. The NN type reducer is trained through minimization of an error-based cost function with the purpose of improving modelling and forecasting performance of IT2 FLS models. Simulation results indicate that application of the proposed TR algorithm greatly enhances modelling and forecasting performance of IT2 FLS models. This benefit is achieved in no cost, as the computational requirement of the proposed algorithm is less than or at most equivalent to traditional TR algorithms.

Despite several years of research, type reduction (TR) operation in interval type-2 fuzzy logic system (IT2FLS) cannot perform as fast as a type-1 defuzzifier. In particular, widely used Karnik-Mendel (KM) TR algorithm is computationally much more demanding than alternative TR approaches. In this work, a data driven framework is proposed to quickly, yet accurately, estimate the output of the KM TR algorithm using simple regression models. Comprehensive simulation performed in this study shows that the centroid end-points of KM algorithm can be approximated with a mean absolute percentage error as low as 0.4%. Also, switch point prediction accuracy can be as high as 100%. In conjunction with the fact that simple regression model can be trained with data generated using exhaustive defuzzification method, this work shows the potential of proposed method to provide highly accurate, yet extremely fast, TR approximation method. Speed of the proposed method should theoretically outperform all available TR methods while keeping the uncertainty information intact in the process.

Karnik-Mendel (KM) algorithm is the most used and researched type reduction (TR) algorithm in literature. This algorithm is iterative in nature and despite consistent long term effort, no general closed form formula has been found to replace this computationally expensive algorithm. In this research work, we demonstrate that the outcome of KM algorithm can be approximated by simple linear regression techniques. Since most of the applications will have a fixed range of inputs with small scale variations, it is possible to handle those complexities in design phase and build a fuzzy logic system (FLS) with low run time computational burden. This objective can be well served by the application of regression techniques. This work presents an overview of feasibility of regression techniques for design of data-driven type reducers while keeping the uncertainty bound in FLS intact. Simulation results demonstrates the approximation error is less than 2%. Thus our work preserve the essence of Karnik-Mendel algorithm and serves the requirement of low
computational complexities.

Karnik-Mendel (KM) algorithm is the most widely used type reduction (TR) method in literature for the design of interval type-2 fuzzy logic systems (IT2FLS). Its iterative nature for finding left and right switch points is its Achilles heel. Despite a decade of research, none of the alternative TR methods offer uncertainty measures equivalent to KM algorithm. This paper takes a data-driven approach to tackle the computational burden of this algorithm while keeping its key features. We propose a regression method to approximate left and right switch points found by KM algorithm. Approximator only uses the firing intervals, rnles centroids, and FLS strnctural features as inputs. Once training is done, it can precisely approximate the left and right switch points through basic vector multiplications. Comprehensive simulation results demonstrate that the approximation accuracy for a wide variety of FLSs is 100%. Flexibility, ease of implementation, and speed are other features of the proposed method.

As an integral part of interval type-2 fuzzy logic system (IT2FLS), type reduction (TR) plays a vital role in determining the performance of IT2FLS. Out of many type reduction algorithms, only Karnik-Mendel type TR algorithms capture the essence of interval type-2 fuzzy sets in type reduction. Enhanced Karnik-Mendel (EKM) algorithm is the most commonly used TR algorithm. In this work, we propose three new initializations for EKM algorithm. It is shown they are performing better than EKM and one of the proposed initializations significantly outperforms others. The performance gain can be upto 40% as per comprehensive simulation results demonstrated in this paper. Our findings are justified by computational time savings and iteration requirement for switch point search.

The cytochromes P450 (P450s) are a remarkable class of heme enzymes that catalyze the metabolism of xenobiotics and the biosynthesis of signaling molecules. Controlled electron flow into the thiolate-ligated heme active site allows P450s to activate molecular oxygen and hydroxylate aliphatic C–H bonds via the formation of high-valent metal-oxo intermediates (compounds I and II). Due to the reactive nature and short lifetimes of these intermediates, many of the fundamental steps in catalysis have not been observed directly. The Gray group and others have developed photochemical methods, known as “flash-quench,” for triggering electron transfer (ET) and generating redox intermediates in proteins in the absence of native ET partners. Photo-triggering affords a high degree of temporal precision for the gating of an ET event; the initial ET and subsequent reactions can be monitored on the nanosecond-to-second timescale using transient absorption (TA) spectroscopies. Chapter 1 catalogues critical aspects of P450 structure and mechanism, including the native pathway for formation of compound I, and outlines the development of photochemical processes that can be used to artificially trigger ET in proteins. Chapters 2 and 3 describe the development of these photochemical methods to establish electronic communication between a photosensitizer and the buried P450 heme. Chapter 2 describes the design and characterization of a Ru-P450-BM3 conjugate containing a ruthenium photosensitizer covalently tethered to the P450 surface, and nanosecond-to-second kinetics of the photo-triggered ET event are presented. By analyzing data at multiple wavelengths, we have identified the formation of multiple ET intermediates, including the catalytically relevant compound II; this intermediate is generated by oxidation of a bound water molecule in the ferric resting state enzyme. The work in Chapter 3 probes the role of a tryptophan residue situated between the photosensitizer and heme in the aforementioned Ru-P450 BM3 conjugate. Replacement of this tryptophan with histidine does not perturb the P450 structure, yet it completely eliminates the ET reactivity described in Chapter 2. The presence of an analogous tryptophan in Ru-P450 CYP119 conjugates also is necessary for observing oxidative ET, but the yield of heme oxidation is lower. Chapter 4 offers a basic description of the theoretical underpinnings required to analyze ET. Single-step ET theory is first presented, followed by extensions to multistep ET: electron “hopping.” The generation of “hopping maps” and use of a hopping map program to analyze the rate advantage of hopping over single-step ET is described, beginning with an established rhenium-tryptophan-azurin hopping system. This ET analysis is then applied to the Ru-tryptophan-P450 systems described in Chapter 2; this strongly supports the presence of hopping in Ru-P450 conjugates. Chapter 5 explores the implementation of flash-quench and other phototriggered methods to examine the native reductive ET and gas binding events that activate molecular oxygen. In particular, TA kinetics that demonstrate heme reduction on the microsecond timescale for four Ru-P450 conjugates are presented. In addition, we implement laser flash-photolysis of P450 ferrous–CO to study the rates of CO rebinding in the thermophilic P450 CYP119 at variable temperature. Chapter 6 describes the development and implementation of air-sensitive potentiometric redox titrations to determine the solution reduction potentials of a series of P450 BM3 mutants, which were designed for non-native cyclopropanation of styrene in vivo. An important conclusion from this work is that substitution of the axial cysteine for serine shifts the wild type reduction potential positive by 130 mV, facilitating reduction by biological redox cofactors in the presence of poorly-bound substrates. While this mutation abolishes oxygenation activity, these mutants are capable of catalyzing the cyclopropanation of styrene, even within the confines of an E. coli cell. Four appendices are also provided, including photochemical heme oxidation in ruthenium-modified nitric oxide synthase (Appendix A), general protocols (Appendix B), Chapter-specific notes (Appendix C) and Matlab scripts used for data analysis (Appendix D).

Neste trabalho avaliou-se a perda de vigor para produção de frutos imaturos com sucessivas gerações de autofecundação (S0, S1, S2, S3, S4 e S5), em uma população de pepino caipira, obtida pelo cruzamento dos híbridos Safira x Hatem. O delineamento experimental utilizado foi em blocos ao acaso, sete tratamentos (seis gerações de autofecundação com diferentes níveis de endogamia e o híbrido Safira como testemunha), seis repetições e cinco plantas por parcela conduzidas em ambiente protegido. Foram avaliados a produção de frutos imaturos total e comercial (número e peso), porcentagem de frutos comerciais e peso médio de frutos. As médias foram comparadas pelo teste de Tukey (5%) e a avaliação da depressão por endogamia foi feita com análise de regressão sem o híbrido Safira. As populações S0 e S1 foram iguais ou superiores ao híbrido Safira para as características número de frutos totais, peso total por planta e peso de frutos comercial por planta, demonstrando o potencial desta população para obtenção de uma nova cultivar ou híbrido do tipo caipira. Houve menor produção de frutos (total e comercial) a partir da população S2, demonstrando possível perda de vigor nesta população com as autofecundações sucessivas.

Oceanographic changes in the western equatorial Pacific during the past 6 Ma are inferred from oxygen isotopic analyses of planktic and benthic foraminifera from Ontong Java Plateau (DSDP Site 586). The taxa are Globigerinoides sacculifer, Pulleniatina, Cibicidoides wuellerstorfi, and Oridorsalis umbonatus. Cooling and ice buildup are indicated by an 18O enrichment of 0.3 per mil in the planktic species near 3.4 Ma. This shift apparently is compensated in the benthic data by a warming of the deep waters by between 1° and 2° C. We suggest that the dominant source of upper deep water supply to the Pacific changed from Antarctic to North Atlantic at that time, the North Atlantic-derived water being warmer. Near 2.8 Ma (approximately) the planktic foraminifera again record an enrichment in 18O (Delta delta18O=0.25 per mil). We suggest ice buildup in the northern hemisphere as the cause, because of subsequent sharp increase in fluctuations of the delta18O signal, that is, instability. The enrichment is magnified in the benthic foraminifera (Delta delta18O = 0.5 per mil) by a cooling of the deep water by 1.5° at the time, presumably signalling a glacial-type reduction of North Atlantic Deep Water (NADW) production. Episodic divergence between the signals of G. sacculifer and Pulleniatina in the Pleistocene apparently reflects periods of increased upwelling in the western equatorial Pacific. The amplitude of ice volume fluctuations cannot be reconstructed from delta18O data alone, unless there are constraints on temperature variations. The increase in amplitude of fluctuation of the benthic and planktic signals during the Pleistocene may be attributed either to an increase in maximum ice volume, or to an increase in the fractionation of continental ice, or a combination of both causes.

We describe a dominant-negative approach in vivo to assess the strong, early upregulation of thyroid hormone receptor beta (TR beta) gene in response to thyroid hormone, characteristic of the onset of natural and thyroid hormone-induced amphibian metamorphosis, 3,3',5-Triiodo-thyronine (T3) treatment of organ cultures of premetamorphic Xenopus tadpole tails coinjected in vivo with the wild-type Xenopus TR beta (wt-xTR beta) and three different thyroid responsive element chloramphenicol acetyltransferase (TRE-CAT) reporter constructs, including a direct repeat +4 (DR +4) element in the -200/+87 fragment of the xTR beta promoter, resulted in a 4- to 8-fold enhancement of CAT activity. Two human C-terminal TR beta 1 mutants (delta-hTR beta 1 and Ts-hTR beta 1), an artificial Xenopus C-terminal deletion mutant (mt-xTR beta), and the oncogenic viral homology v-erbA, none of which binds T3, inhibited this T3 response of the endogenous wt-xTR in Xenopus XTC-2 cells cotransfected with the -1600/+87 xTR beta promoter-CAT construct, the potency of the dominant-negative effect of these mutant TRs being a function of the strength of their heterodimerization with Xenopus retinoid X receptor gamma. Coinjection of the dominant-negative Xenopus and human mutant TR beta s into Xenopus tadpole tails totally abolished the T3 responsiveness of the wt-xTR beta with different TREs, including the natural DR +4 TRE of the xTR beta promoter.

It is studied whether there is any regular relationship between the yellow luminescence band and electron mobility of n-type GaN. For a series of GaN samples grown with the same Si doping, it is found that the electron mobility decreases with an increase of relative intensity of yellow luminescence, accompanied by an increase of edge dislocation density. Further research indicates that it is acceptors introduced by edge dislocations which lead to the concomitant changes of yellow luminescence and electron mobility. Similar changes are induced by Si doping in the n-type GaN samples with relatively low edge dislocation density. However, the relationship between the yellow luminescence and electron mobility of n-type GaN is not a simple one. A light Si doping may simultaneously increase yellow luminescence and electron mobility when Si doping plays a dominant role in reducing the carrier scattering. This means that even the intensity of yellow luminescence is often used as an indicator of material quality for GaN, it does not have any monotonous correlation with the electron mobility of GaN. The electrochemical behavior of Dawson-type P2W18O626- adsorbed on a glassy carbon electrode and doped in a polypyrrole film electrode was described. These modified electrodes all display catalytic activity for nitrite reduction, either in acid solutions or in pH > 4.0 solutions.
