The present work evaluated the performance of two treatment systems in reducing indicators of biological contamination in swine production wastewater. System I consisted of two upflow anaerobic sludge blanket (UASB) reactors, with 510 and 209 L in volume, being serially arranged. System II consisted of a UASB reactor, anaerobic filter, trickling filter, and decanter, being also organized in series, with volumes of 300, 190, 250, and 150 L, respectively. Hydraulic retention times (HRT) applied in the first UASB reactors were 40, 30, 20, and 11 h in systems I and II. The average removal efficiencies of total and thermotolerant coliforms in system I were 92.92% to 99.50% and 94.29% to 99.56%, respectively, and increased in system II to 99.45% to 99.91% and 99.52% to 99.93%, respectively. Average removal rates of helminth eggs in system I were 96.44% to 99.11%, reaching 100% as in system II. In reactor sludge, the counts of total and thermotolerant coliforms ranged between 10(5) and 10(9) MPN (100 mL)(-1), while helminth eggs ranged from 0.86 to 9.27 eggs g(-1) TS.

Water remains a predominant vector for human enteric pathogens not just for developing countries but also developed nations, where numerous infectious disease outbreaks, linked to the contamination of drinking water have been documented. Private drinking water wells are a source of drinking water that is largely unstudied even though a significant percentage of the population in Ontario relies on wells as their primary water source. As there exists little to no systematic surveillance for enteric infections or outbreaks related to well water sources, these individuals may be at higher risk of waterborne infectious diseases. The relationships between various fecal indicators in the water of private drinking water wells, including E. coli, Total Coliforms (TC) and Bacteroides, and enteric pathogens, including Campylobacter jejuni, Salmonella spp., and Shiga toxin producing E. coli, were studied. Convenience private well water samples collected from various regions of interest during the summer of 2014 underwent membrane filtration and culture to determine quantities of E. coli and TC colony forming units. 289 E. coli positive and 230 TC-only waters were successfully analyzed by individual qPCR assays for the aforementioned enteric pathogens. Microbial source tracking methods targeted to specific Bacteroides were used to determine the source of fecal contamination as either human or bovine. The source of fecal contamination varied by geographic region and is thought to be due to such things as differences in septic tank density and underlying geology, among others. Fecal indicators, E. coli and Bacteroides, were significantly correlated. E. coli as measured by qPCR was more strongly correlated to both total and human-specific Bacteroides genetic markers than culturable E. coli. Lastly, 1.9% of samples showed molecular evidence of contamination with enteric pathogens. Although low, this finding is significant given the limited volume of water available for testing, and suggests a potential health risk to consumers. Knowing the extent of contamination, as well as the biologic source, can better inform risk assessment and the development of potential intervention strategies for private well water in specific regions of Ontario.

Forty-six bottled water samples representing 16 brands from Dhaka, Bangladesh were tested for the numbers of total coliforms, fecal indicator bacteria (i.e., thermotolerant Escherichia coli and Enterococcus spp.) and potential bacterial pathogens (i.e., Aeromonas hydrophil, Pseudomonas aeruginos, Salmonella spp., and Shigella spp.). Among the 16 brands tested, 14 (86%), ten (63%) and seven (44%) were positive for total coliforms, E. coil and Enterococcus spp., respectively. Additionally, a further nine (56%), eight (50%), six (37%), and four (25%) brands were PCR positive for A. hydrophila lip, P. aeruginosa ETA, Salmonella spp. invA, and Shigella spp. ipaH genes, respectively. The numbers of bacterial pathogens in bottled water samples ranged from 28 ± 12 to 600 ± 45 (A. hydrophila lip gene), 180 ± 40 to 900 ± 200 (Salmonella spp. invA gene), 180 ± 40 to 1,300 ± 400 (P. aeruginosa ETA gene) genomic units per L of water. Shigella spp. ipaH gene was not quantifiable. Discrepancies were observed in terms of the occurrence of fecal indicators and bacterial pathogens. No correlations were observed between fecal indicators numbers and presence/absence of A. hydrophila lip (p = 0.245), Salmonella spp. invA (p = 0.433), Shigella spp. ipaH gene (p = 0.078), and P. aeruginosa ETA (p = 0.059) genes. Our results suggest that microbiological quality of bottled waters sold in Dhaka, Bangladesh is highly variable. To protect public health, stringent quality control is recommended for the bottled water industry in Bangladesh. This project developed and assessed a standard operating procedure for monitoring microbiological aerosol levels and dispersal from Australian industrial composting facilities. Development occurred via seasonal monitoring of such operations with evaluation of optimal microbial indicator organisms, sampling and analysis logistics. The resultant procedure allows practical end-user assessment of compost-associated bioaerosol levels, and potential health risks to proximal residential populations encroaching on such composting facilities and on-site industrial operations personnel.

This study proposes a wastewater electrolysis cell (WEC) for on-site treatment of human waste coupled with decentralized molecular H2 production. The core of the WEC includes mixed metal oxides anodes functionalized with bismuth doped TiO2 (BiOx/TiO2). The BiOx/TiO2 anode shows reliable electro-catalytic activity to oxidize Cl- to reactive chlorine species (RCS), which degrades environmental pollutants including chemical oxygen demand (COD), protein, NH4+, urea, and total coliforms. The WEC experiments for treatment of various kinds of synthetic and real wastewater demonstrate sufficient water quality of effluent for reuse for toilet flushing and environmental purposes. Cathodic reduction of water and proton on stainless steel cathodes produced molecular H2 with moderate levels of current and energy efficiency. This thesis presents a comprehensive environmental analysis together with kinetic models to provide an in-depth understanding of reaction pathways mediated by the RCS and the effects of key operating parameters. The latter part of this thesis is dedicated to bilayer hetero-junction anodes which show enhanced generation efficiency of RCS and long-term stability.
Chapter 2 describes the reaction pathway and kinetics of urea degradation mediated by electrochemically generated RCS. The urea oxidation involves chloramines and chlorinated urea as reaction intermediates, for which the mass/charge balance analysis reveals that N2 and CO2 are the primary products. Chapter 3 investigates direct-current and photovoltaic powered WEC for domestic wastewater treatment, while Chapter 4 demonstrates the feasibility of the WEC to treat model septic tank effluents. The results in Chapter 2 and 3 corroborate the active roles of chlorine radicals (Cl•/Cl2-•) based on iR-compensated anodic potential (thermodynamic basis) and enhanced pseudo-first-order rate constants (kinetic basis). The effects of operating parameters (anodic potential and [Cl-] in Chapter 3; influent dilution and anaerobic pretreatment in Chapter 4) on the rate and current/energy efficiency of pollutants degradation and H2 production are thoroughly discussed based on robust kinetic models. Chapter 5 reports the generation of RCS on Ir0.7Ta0.3Oy/BixTi1-xOz hetero-junction anodes with enhanced rate, current efficiency, and long-term stability compared to the Ir0.7Ta0.3Oy anode. The effects of surficial Bi concentration are interrogated, focusing on relative distributions between surface-bound hydroxyl radical and higher oxide.

The total coliforms, fecal coliforms and fecal streptococci were counted over 1 year at 33 stations in the Ebrié Lagoon near Abidjan (Ivory Coast). Most of the waters show high numbers of bacteria, (D category of the American standards). For the sea-beaches, high numbers of fecal germs are found where the human population is abundant.

This is the Kent estuary survey 18th July 1981 produced by the North West Water Authority in 1981. The report focuses on a tidal cycle survey carried out on the Kent estuary on the 18th August, 1981. The chemical water quality at Arnside Pier and at New Barns was, on the whole, acceptable. However, samples taken downstream of the outfall at low water for total coliforms and E. coli were all above the EEC mandatory level whilst those for faecal streptococci were all above the guideline value. This document shows chemical and bacteriological data analysed at Kent estuary such chloride, dissolved oxygen, BOD, ammonia, nitrate, phosphate, total coliforms, E. coli and Faecal streptococci.

The by-catch from the shrimp trawl fishery in Kalpitiya is mainly used for the production of dried fish, which provides an additional source of income for fishermen in the area. It has been observed that current handling practices along the value addition chain are responsible for the poor quality and low price of the end product. This study was aimed at identifying the shortcomings in such handling practices by fishermen and dried fish producers and assessing the quality of shrimp fishery by-catch along the processing chain in order to recommend more efficient utilization methods that will improve the quality of the end product. Fresh fish, dried fish and harbour water samples were tested for total coli forms, faecal coliforms, E. coli and Salmonella in order to assess their microbial quality: In addition, standard plate counts (SPC) of fish samples were also carried out. A survey was carried out from July-October 2006 at Kalpitiya, using a pre-tested questionnaire to collect information from individuals who have been engaged in dried fish processing. Average values obtained for freshly landed and dried fish respectively, were, SPC 9.88x10 super(5) CFU/g and 30.43x10 super(5) CFU/g, total coliforms 23.05 and 24.23 MPN/g and fecal coliforms 8.28 and 9.00 MPN/g. These values exceed the recommendations in the SL standards. A quarter of the landed fresh fish and 38% of dried fish from the producers were positive for E. coli and thus failed to show required end product quality. SPC of harbour water was 14.35x10 super(6) CFU/ml and all samples were found to be contaminated with E. coli. None of the fishermen and dried fish producers were satisfied with the quality of the end product. The reasons for poor quality as indicated by them were: limited availability of ice (75%), lack of infrastructure facilities (65%), uncertainty of markets (52%), lack of emphasis on quality (47%) and poor access to available technologies (41%). Respondents to the questionnaire also identified: unavailability of potable water, insulated boxes, good landing jetty, racks for drying fish, poor cold storage facilities and limitations in dried fish storage facilities, as further factors leading to the loss of quality in their products. Results demonstrate that improvements to the infrastructure facilities and conducting of proper awareness programmes on handling practices could lead for improvements in the quality of value added products prepared from the shrimp fishery by-catch at Kalpitiya.

Em Portugal, nomeadamente nos Açores (Centro Termal das Furnas), durante vários anos foram utilizados pelóides naturais (lamas/argilas obtidos após conveniente maturação com água mineral) como agentes terapêuticos. Apesar das evidências empíricas da aplicação das argilas para fins terapêuticos, o suporte científico é ainda escasso. Além disso, contrariamente ao que se verifica para águas minerais utilizadas para fins termais, no campo da peloterapia, não existe regulamentação que estabeleça critérios quanto à qualidade padrão de um pelóide. Este trabalho visou a avaliação das características biológicas, físico-químicas e mineralógicas de lamas vulcânicas das ilhas de São Miguel e da Terceira para aplicação em peloterapia antes e após maturação. A área de amostragem foi caracterizada sob o ponto de vista geomorfológico, geológico, hidrológico e climático. A análise dos materiais colhidos contemplou estudos de granulometria e de mineralogia da amostra total e da fracção argilosa; caracterização química; e determinação de outras propriedades tais como índice de abrasividade, superfície específica, pH, capacidade de troca catiónica e catiões de troca, limites de consistência (plasticidade e liquidez e índice de plasticidade) e taxa de arrefecimento. A caracterização biológica das lamas incluiu a identificação e quantificação relativa de diatomáceas e a determinação do teor em coliformes totais e microrganismos cultiváveis a 37 ºC e a 22 ºC. Estudos efectuados em amostras comerciais argilosas e em recursos naturais açorianos precederam os ensaios de maturação. Estes estudos visaram, fundamentalmente, aferir as características apropriadas dos materiais para aplicação tópica e detectar e caracterizar a presença de comunidades diatomológicas em locais próximos dos materiais a serem sujeitos a maturação. O estudo de maturação foi desenvolvido em duas etapas sequenciais e complementares com vista à optimização do processo de maturação de lamas vulcânicas açorianas. Foi testada a influência de algumas condições abióticas na maturação e fez-se variar a fracção granulométrica, proporção fase sólida:fase líquida e o tempo de maturação entre os dois ensaios. O uso da granulometria <63 μm, proporção de 2:1 (fase sólida:fase líquida) e condições de luminosidade, sem agitação parecem ser as mais adequadas à maturação dos materiais analisados. A maturação revela-se um processo que promove a melhoria de características importantes dos materiais para aplicação em peloterapia, tais como aumento dos tempos de arrefecimento e da quantidade de catiões de troca. Existe uma comunidade de microalgas e bacteriana capaz de se desenvolver em materiais geológicos vulcânicos durante a maturação e cujo contributo para a acção terapêutica do pelóide deverá ser aferida. Após comparação com materiais de referência, a amostra das Furnas revela as características mais adequadas para aplicação em peloterapia.

Microbiological studies on the incidence, behaviour, activity and ecological implications of marine micro~organisms, particularly microbial pathogens in coastal waters and estuaries exhibit the increasing concern and awareness of environmental impacts on health and wealth. Marine microbiologists have been active in investigating on the distribution, kinds of organisms and their activity in the environment. However, informations on the effect of environment on the ecology or on the distribution (spatial/temporal) of microbial comunity and competition among groups inhabiting the ecosystem are sparE§L Estuarine environment are complex with respect to diversity of habitats, variation in physicochemical parameters and contamination by terrestrial bacterial species. Being the organisms of‘public health significance, ecological studies on total coliforms, faecal coliforms, faecal streptococci, §. ggli and X. parahaemolyticus have great relevance as studies of these types would provide a wealth of information to environmentalists and to fishery industry. In order to evalé%e the status, role and significance of potentially hazardous bacterial species in natural environment it is necessary to monitor the ecology of such organisms systematically in relation to physico-chemical parameters

Wastewater reuse has become an important alternative to agricultural irrigation; on the other hand, it poses concern with regard to public health. Total coliform and Escherichia coli concentration, presence of helminth eggs and Salmonella, and physical-chemical parameters were evaluated in raw and treated wastewater. Chemical and biochemical oxygen demand removal efficiency was 74.6 and 77.9%, respectively. As for organic nitrogen, total phosphorus, and total suspended solids, total efficiency removal was 17.4, 12.5, and 32.9%, respectively. The average density of total coliforms and E. coli was 3.5 x 10(9) and 1.8 x 10(8) MPN/100 mL and 1.1 x 10(7) MPN/100 mL and 3.9 x 10(5) MPN/100 mL for raw and treated wastewater, respectively. Ascaris eggs were observed in 80.8% of the samples collected, and viable eggs in 42.3% of the samples. Salmonella was detected in 36.4% of the samples. The values observed in treated wastewater did not show the adequate bacteriological quality, as recommended by World Health Organization (Geneva, Switzerland). Therefore, additional measures should be taken to achieve an improved microbiological and parasitological quality.

The National School Nutrition Program is the oldest program in the country directed at food and nutrition safety. Its aims are to offer nutritional food as a supplement to students of public and philanthropic schools. Studying school nutrition transcends the investigation as a purely social program, given that it reaches the fields of public health, nutrition and food, using a wide variety of approaches. Thus, it is characterized by a multidisciplinary study, where the disciplines work side by side in distinct aspects of a single problem. Aim: This study aims to assess hygiene practices during the preparation of meat-based meals in public schools in the city of Natal, Brazil. Methods: A list was applied at 27 schools to identify the procedures of good food preparation practices. In addition, cooking and meal distribution temperature were measured and a microbiological analysis of the final preparation and of the water used in preparing it was performed. For microbiological analyses of the food, we analyzed coliforms at 45°C, coliforms at 35°C and Enterococcus, and for the water, we analyzed thermotolerant coliforms and total coliforms, using the methods recommended by APHA, 1995. Results: Most of the schools did not meet the required standards in all the variables related to good food preparation practices, except for the time spent preparing the meat, in which 89% were within the norm. Cooking temperature of the meals was within the standard; however, the temperature at distribution and the time spent dispensing the meals were inadequate. Of the 27 schools, 22 (81.5%) showed the presence of coliforms at 35° C in at least one meal sample and 18 (66.7%) had values above the recommended limit for coliforms at 45°C. The presence of E. coli was identified in 6.1% of the samples analyzed. The presence of Enterococcus was not found at any of the schools. With respect to the water, the North district of the city was the only one that did not meet the standards for the two indicators evaluated. The contamination found was not associated with the hygiene or food storage problems observed. Conclusions: The results show that the hygiene-sanitary conditions of meat-based public school meals were unsatisfactory, demonstrating the need for improvements in the production process to preserve the health of the student population. The experiment constituted in evaluating the acid silage prepared from Nile tilapia (Oreochromis niloticus) filletage industry residues. This silage was prepared in a filetage industry through the residues milling and 5% of ascetic acid addition and stock piling in a period up to until 201 days. Storaged silages in a period of 7, 12, 22, 27, 41, 48, 61, 75, 84, 91, 96, 110, 140, 151, 181, 187 and 201dias were evaluated. Bromatologics analyses of rude protein(PB), etereal extract (EE), humidity (UM), ashes (CZ) and microbiological analyses of Salmonella, total coliforms, faecals and Escherichia coli of the storaged silages in a period of 7, 91 and 201 days were realized. The pH was evaluated in all the storaged periods. Differences (P > 0.05) in the substance of PB, EE, UM and CZ were not observed. A linear increase in PH was observed, being stabilizing in plateau of 4.74. In relation to the microbiological analysis the presence of Salmonella, total coliforms, faecals and E. Coli were not found. Was concluded that the acid silage gotten from tilapia filetage residues produced with the use of 5% ascetic acid can be stored during 201 days without having Salmonella, total coliforms and faecals proliferation.

O Brasil é o sétimo maior consumidor mundial de águas engarrafadas. Os recipientes mais utilizados, galões plásticos de 20 litros, devem ser submetidos à inspeção individual e posteriormente a sanificação. Recentemente, enfermidades associadas a microrganismos emergentes têm despertado o interesse por novos sanitizantes. Entre estes, o gás ozônio é um dos mais atraentes em virtude da sua segurança e eficácia superiores aos desinfetantes convencionais, não gerando resíduos tóxicos. Neste trabalho, o ozônio foi avaliado como método alternativo na sanificação de galões de água de 20 litros, na cidade de Alfenas, MG. Trinta galões foram avaliados sem tratamento e trinta após a sanificação com água ozonizada (4mg/L/2minutos) quanto à contagem total de microrganismos aeróbios mesófilos heterotróficos, número mais provável (NMP) de coliformes totais e Escherichia coli, Staphyloccocus aureus e Pseudomonas spp. em 100mL de solução enxaguatória. A contagem média de unidades formadoras de colônias (UFC) de microrganismos heterotróficos no estágio de pré-lavagem foi de 5,7/cm² enquanto que o tratamento com a água ozonizada reduziu este valor para 0,003/cm², além de promover a negativação das análises para coliformes Pseudomonas ssp. e somente 13,3% das amostras apresentaram-se positivas para Staphylococcus aureus após a sanificação. Concluiu-se que o tratamento com utilização de ozônio foi eficiente, nas condições testadas.
