This thesis proposes the development of a narrative methodology in the British Methodist Church. Such a methodology embraces and communicates both felt experience and critical theological thinking, thus producing and presenting a theology that might have a constructive transformative impact on wider society. In chapter one I explore the ways in which the Church speaks in public, identify some of the challenges it faces, and consider four models of engagement. If the Church is to engage in public discourses then I argue that its words need to be relevant and connect with people’s experiences. To ground the thinking I focus on the context of the British Methodist Church and explore how the Church engages in theological reflection through the lens of its thinking on issues of human sexuality. Chapter two reviews how theological reflection is undertaken in the British Methodist Church. I describe how the Methodist Quadrilateral of Scripture, tradition, reason and experience remains a foundational framework for theological reflection within the Methodist Church and consider the impact of institutional processes and the ways in which the Methodist people actually engage with theological thinking. The third and fourth chapters focus on how the British Methodist Church has produced its theology of human sexuality, giving particular attention to the use of personal and sexual stories in this process. I find that whilst there has been a desire to listen to the stories of the Methodist people, there has not been a corresponding interrogation or analysis of their stories so as to enable robust and constructive theological reflection on these experiences. Using resources from Foucauldian approaches to discourse analysis, I critique key statements and the processes involved in their production, offering an analysis of this body of theological thinking and indicating where possibilities for alternative ways of thinking and acting arise. The proposed methodology draws upon resources from social science methodologies, and in chapter five I look at the use of personal experience and relevant strategies of inquiry that prompt reflection on the hermeneutical process and employ narrative approaches in undertaking, analysing and presenting research. The exploration shows that qualitative research methodologies offer resources and methods of inquiry that could help the Church to engage with personal stories in its theological thinking in a robust, interrogative and imaginative way. In chapter six an examination of story and narrative is undertaken, to show how they have been understood as ways of knowing and how they relate to theological inquiry. Whilst acknowledging some of the limitations of narrative, I indicate how it offers constructive possibilities for theological reflection and could be a means for the British Methodist Church to engage in public discourse. This is explored further in chapter seven, which looks in more detail at how the British Methodist Church has used narrative in its theological thinking, and outlines areas requiring further attention in order for a narrative theological methodology to be developed, namely: attention to the question ‘whose experience?’; investigation of issues of power and the dynamics involved in the process of the production of theological thought; how personal stories and experiences are interrogated and how narrative is constructed; and how narrative might be employed within the Methodist Quadrilateral. The final chapter considers the advantages and limitations of such an approach, whether the development of such a method is possible in the Methodist Church today and its potential for helping the Church to engage in public discourse more effectively. Niniejsza praca powstała z rozpoznanej potrzeby projektowania nowych ujęć poznawczych w teologii obejmujących, w tym samym stopniu, odniesienia do dziedzin teologicznych, jak i innych nauk. Podjęto próbę skonstruowania procedur badawczych, które mogą stać się punktem wyjścia ku stworzeniu pełnej metody integralnej w teologii fundamentalnej. Stąd tytuł pracy Ku metodzie integralnej w teologii fundamentalnej. Budowanie nowych zasad aktywności badawczej związane jest zawsze z etapem konstrukcyjnym i etapem weryfikacyjnym przygotowanych narzędzi, co zostało wyartykułowane w podtytułach: Sposoby konstruowania interdyscyplinarnych działań w poszukiwaniu »locorum theologicorum«. Zastosowanie i weryfikacja na przykładzie życia i twórczości Simone Weil. Tym samym praca jest dwuczęściowa. Celem pierwszej części jest przedstawienie, w jaki sposób konstruować poszukiwania nowych locorum theologicorum. Druga część pracy ma charakter weryfikacyjny. Opracowane narzędzia badawcze zostały zastosowane do odpowiedzi na pytanie: Na ile życie i twórczość francuskiej myślicielki, Simone Weil, może stać się źródłem dla tworzonych współcześnie opinii teologicznych? W przedstawianej pracy powiązano, dość ściśle, konstruowanie integralnego ujęcia badawczego z poszukiwaniem nowych czy reinterpretacją już rozeznanych miejsc teologicznych. Główny cel pracy związany jest z opisywaniem działań, które uprawdopodobniają otrzymanie nowych treści. Przyjęty sposób działania to wyjaśnianie, nie mające statusu pełnej prawdziwości logicznej; jednak jest ono procedurą, która pozwala w pewnym stopniu obiektywizować subiektywne dane płynące z otaczającej nas rzeczywistości. Pozwoliło to na kompetentne poruszanie się w przestrzeni zdań uznanych za prawdziwe zarówno w ramach teologii, jak i innych nauk. Dlatego w tej pracy skupiono się na przedstawieniu procedur poszukiwań, weryfikacji i możliwego zastosowania w badaniach naukowych współczesnych miejsc teologicznych. Ważniejszy jest sposób otrzymywania określonych wyników, a wynik tychże działań doceniony jest dopiero na etapie weryfikacyjnym skonstruowanych narzędzi badawczych. W części pierwszej pracy skupiono się na tworzeniu ogólnych zasad myślenia w ramach teologii fundamentalnej. Konstruowanie narzędzi badawczych rozpoczęto od rozeznania i przedstawienia najgłębszych fundamentów, korzeni zastosowanych procedur poprzez opis zasad opracowywania i przygotowywania działań operacyjnych. Określenie tych zasad związane jest z wprowadzeniem nowego sposobu ujmowania problematyki badawczej poprzez tak zwane myślenie modelowe. Pozwala ono na włączenie w proces uzasadniania otrzymanych wyników, oprócz dowodzenia i tłumaczenia, również tak zwanego wyjaśniania, poprzez zataczanie coraz wyższych kręgów po metodycznej spirali, której osią jest odniesienie do Bożego Objawienia odczytanego we wspólnocie Kościoła. Dzięki temu otrzymano śmiałą hipotezę, w której uwzględniono w tym samym stopniu Objawienie Boga, jak i adresata tegoż, z jego pełnym osadzeniem w rzeczywistościach ziemskich. Holistyczne, w ramach tworzonego modelu, ujmowanie problematyki pozwoliło również na ukazywanie ciągle nowych aspektów tworzonej metody integralnej. Opracowane zostały zasady konstruowania modelu ujmowania rzeczywistości Objawienia w aspekcie przedmiotowym. Jako podstawę działań przyjęto istniejącą już w teologii fundamentalnej metodę historiozbawczą. Przechodząc do systematycznej prezentacji, jako odniesienia graniczne tworzonego modelu «przestrzeni eklezjalnej» wprowadzono ujęcia teologiczne św. Ireneusza z Lyonu i św. Justyna Męczennika. Konstruując narzędzia pozwalające na ujęcia podmiotowe treści przeżyć odbiorcy Objawienia jako odniesienia graniczne wprowadzono koncepcje Umberto Eco, Geralda O’Collinsa i Paula Ricoeura. Pozwalają one na modelowe ujęcie ludzkich przeżyć jako «dzieła otwartego». W etapie czwartym podjęto próbę ustalenia zasad wzajemnego przenikania się ujęć przedmiotowych i podmiotowych problematyki badawczej, by stworzyć ujęcie integralne w opracowywaniu treści teologicznych. Powstały model «wzajemnych oddziaływań» okazał się przydatnym narzędziem w ocenie teologicznego potencjału interdyscyplinarnych oddziaływań pomiędzy teologią fundamentalną a treściami innych nauk. Pozwolił on doprecyzować warunki możliwości przyjęcia jakiejś ludzkiej aktywności jako locus theologicus z zachowaniem odpowiedniej relacji do Objawienia odczytanego we wspólnocie Kościoła z możliwym do osiągnięcia stopniem obiektywizacji. W cześć drugiej pracy realizuje się konieczną w naukach weryfikację opracowanych narzędzi badawczych i uwiarygodnienia działań operacyjnych poprzez zastosowanie do analizy konkretnego przypadku, którym jest życie i twórczość Simone Weil. Praca w tej części również została podzielona na etapy według aspektowego ujęcia umożliwiającego różne punkty widzenia tego samego przedmiotu badawczego. Ponieważ opracowane modele teoretyczne tworzą ramy dla nowej twórczości badacza, kolejne etapy weryfikacyjne są dalszym ciągiem budowania sposobów ujęcia ukonkretnionej tematyki. Życie i twórczość Simone Weil zostały przedstawione w czterech perspektywach badawczych. Pierwsza z nich jest ujęciem z punktu widzenia relacji «dzieło – autor dzieła», kolejna wprowadza dodatkowe odniesienia związane z relacją «dzieło – odbiorca dzieła». Trzeci aspekt skupia się na możliwych do odczytania z twórczości Simone Weil, treściach teologicznych na bazie relacji «dzieło – badacz dzieła». Najpłodniejszym badawczo okazało się odniesienie «dzieła», któremu na imię Simone Weil ku wymiarowi eschatologicznemu chrześcijaństwa. Opracowana metoda i przedstawione warunki weryfikacyjne ustalające kryteria w uznaniu jej przemyśleń i koncepcji jako locus theologicus ukazały się jako niezwykle przydatne.

Response surface methodology based on Box-Behnken (BBD) design was successfully applied to the optimization in the operating conditions of the electrochemical oxidation of sanitary landfill leachate aimed for making this method feasible for scale up. Landfill leachate was treated in continuous batch-recirculation system, where a dimensional stable anode (DSA(©)) coated with Ti/TiO2 and RuO2 film oxide were used. The effects of three variables, current density (milliampere per square centimeter), time of treatment (minutes), and supporting electrolyte dosage (moles per liter) upon the total organic carbon removal were evaluated. Optimized conditions were obtained for the highest desirability at 244.11 mA/cm(2), 41.78 min, and 0.07 mol/L of NaCl and 242.84 mA/cm(2), 37.07 min, and 0.07 mol/L of Na2SO4. Under the optimal conditions, 54.99 % of chemical oxygen demand (COD) and 71.07 ammonia nitrogen (NH3-N) removal was achieved with NaCl and 45.50 of COD and 62.13 NH3-N with Na2SO4. A new kinetic model predicted obtained from the relation between BBD and the kinetic model was suggested.

Didanosine-loaded chitosan microspheres were developed applying a surface-response methodology and using a modified Maximum Likelihood Classification. The operational conditions were optimized with the aim of maintaining the active form of didanosine (ddI), which is sensitive to acid pH, and to develop a modified and mucoadhesive formulation. The loading of the drug within the chitosan microspheres was carried out by ionotropic gelation technique with sodium tripolyphosphate (TPP) as cross-linking agent and magnesium hydroxide (Mg(OH)2) to assure the stability of ddI. The optimization conditions were set using a surface-response methodology and applying the Maximum Likelihood Classification, where the initial chitosan concentration, TPP and ddI concentration were set as the independent variables. The maximum ddI-loaded in microspheres (i.e. 1433mg of ddI/g chitosan), was obtained with 2% (w/v) chitosan and 10% TPP. The microspheres depicted an average diameter of 11.42μm and ddI was gradually released during 2h in simulated enteric fluid.

Um inquérito de base populacional foi conduzido na população urbana de todas as capitais e do Distrito Federal no Brasil para fornecer informações sobre a prevalência de hepatites virais e fatores de risco, entre 2005 e 2009. Este artigo descreve o delineamento e a metodologia do estudo que envolveu a população com idade entre 5 e 19 anos para hepatite A e 10 a 69 anos para hepatite B e C. As entrevistas e amostras de sangue foram obtidas através de visitas domiciliares e a amostra selecionada a partir de uma amostragem estratificada em múltiplos estágios (por conglomerado) com igual probabilidade para cada domínio de estudo (região e faixa etária). Nacionalmente, 19.280 residências e ~31.000 indivíduos foram selecionados. O tamanho da amostra foi suficiente para detectar uma prevalência em torno de 0,1% e para avaliar os fatores de risco por região. A metodologia apresentou-se viável para distinguir entre diferentes padrões epidemiológicos da hepatite A, B e C. Estes dados serão de valia para a avaliação das políticas de vacinação e para o desenho de estratégias de controle.

Fifty Bursa of Fabricius (BF) were examined by conventional optical microscopy and digital images were acquired and processed using Matlab® 6.5 software. The Artificial Neuronal Network (ANN) was generated using Neuroshell® Classifier software and the optical and digital data were compared. The ANN was able to make a comparable classification of digital and optical scores. The use of ANN was able to classify correctly the majority of the follicles, reaching sensibility and specificity of 89% and 96%, respectively. When the follicles were scored and grouped in a binary fashion the sensibility increased to 90% and obtained the maximum value for the specificity of 92%. These results demonstrate that the use of digital image analysis and ANN is a useful tool for the pathological classification of the BF lymphoid depletion. In addition it provides objective results that allow measuring the dimension of the error in the diagnosis and classification therefore making comparison between databases feasible.

Blends of milk fat and canola oil (MF:CNO) were enzymatically interesterified (EIE) by Rhizopus oryzne lipase immobilized on polysiloxane-polyvinyl alcohol (SiO(2)-PVA) composite, in a solvent-free system. A central composite design (CCD) was used to optimize the reaction, considering the effects of different mass fractions of binary blends of MF:CNO (50:50, 65:35 and 80:20) and temperatures (45, 55 and 65 degrees C) on the composition and texture properties of the interesterified products, taking the interesterification degree (ID) and consistency (at 10 degrees C) as response variables. For the ID variable both mass fraction of milk fat in the blend and temperature were found to be significant, while for the consistency only mass fraction of milk fat was significant. Empiric models for ID and consistency were obtained that allowed establishing the best interesterification conditions: blend with 65 % of milk fat and 35 %, of canola oil, and temperature of 45 degrees C. Under these conditions, the ID was 19.77 %) and the consistency at 10 degrees C was 56 290 Pa. The potential of this eco-friendly process demonstrated that a product could be obtained with the desirable milk fat flavour and better spreadability under refrigerated conditions.

This paper proposes a new design methodology for discrete multi-pumped Raman amplifier. In a multi-objective optimization scenario, in a first step the whole solution-space is inspected by a CW analytical formulation. Then, the most promising solutions are fully investigated by a rigorous numerical treatment and the Raman amplification performance is thus determined by the combination of analytical and numerical approaches. As an application of our methodology we designed an photonic crystal fiber Raman amplifier configuration which provides low ripple, high gain, clear eye opening and a low power penalty. The amplifier configuration also enables to fully compensate the dispersion introduced by a 70-km singlemode fiber in a 10 Gbit/s system. We have successfully obtained a configuration with 8.5 dB average gain over the C-band and 0.71 dB ripple with almost zero eye-penalty using only two pump lasers with relatively low pump power. Introduction. This method is used to forecast the harvest date of banana bunches from as early as the plant shooting stage. It facilitates the harvest of bunches with the same physiological age. The principle, key advantages, time required and expected results are presented. Materials and methods. Details of the four steps of the method ( installation of the temperature sensor, tagging bunches at the flowering stage, temperature sum calculation and estimation of bunch harvest date) are described. Possible problems are discussed. Results. The application of the method allows drawing a curve of the temperature sum accumulated by the bunches which have to be harvested at exactly 900 degree-days physiological age.

An in vivo study was conducted to verify the ability of laser fluorescence (LF) to assess the activity status of occlusal caries in primary teeth, using different air-drying times. Occlusal sites (707) were examined using LF (DIAGNOdent) after air-drying for 3 s and 15 s, and the difference between readings (DIF15 s-3 s) was calculated. For concurrent validation of LF, visual criteria-Nyvad (NY) and Lesion Activity Assessment associated with the International Caries Detection and Assessment System (LAA-ICDAS)-were the reference standards for lesion activity. Histological exam using a pH-indicator dye (0.1% methyl red) was performed in 46 exfoliated/extracted teeth for criterion validation. LF readings and DIF15 s-3 s were compared using Kruskall-Wallis and Mann-Whitney tests. Receiver operating characteristic analyses were performed and validity parameters calculated, considering the caries activity assessment. Using NY, active lesions (3 s: 30.0 +/- 29.3; 15 s: 34.2 +/- 30.6) presented higher LF readings than inactive lesions (3 s: 17.0 +/- 16.3; 15 s: 19.2 +/- 17.3; p <0.05), different from LAA-ICDAS. Active cavitated caries resulted in higher LF readings (3 s: 50.3 +/- 3.5; 15 s: 54.7 +/- 30.2) than inactive cavitated caries (3 s: 19.9 +/- 16.3; 15 s: 22.8 +/- 16.8). Therefore, LF can distinguish cavitated active and inactive lesions classified by NY, but not by LAA-ICDAS; however, this difference might be related to the visual system rather than to LF. The air-drying time could be an alternative to improve the caries activity assessment; however, longer air-drying time is suggested to be tested subsequently. The conditions for maximization of the enzymatic activity of lipase entrapped in sol-gel matrix were determined for different vegetable oils using an experimental design. The effects of pH, temperature, and biocatalyst loading on lipase activity were verified using a central composite experimental design leading to a set of 13 assays and the surface response analysis. For canola oil and entrapped lipase, statistical analyses showed significant effects for pH and temperature and also the interactions between pH and temperature and temperature and biocatalyst loading. For the olive oil and entrapped lipase, it was verified that the pH was the only variable statistically significant. This study demonstrated that response surface analysis is a methodology appropriate for the maximization of the percentage of hydrolysis, as a function of pH, temperature, and lipase loading.

This work presents a thermoeconomic optimization methodology for the analysis and design of energy systems. This methodology involves economic aspects related to the exergy conception, in order to develop a tool to assist the equipment selection, operation mode choice as well as to optimize the thermal plants design. It also presents the concepts related to exergy in a general scope and in thermoeconomics which combines the thermal sciences principles (thermodynamics, heat transfer, and fluid mechanics) and the economic engineering in order to rationalize energy systems investment decisions, development and operation. Even in this paper, it develops a thermoeconomic methodology through the use of a simple mathematical model, involving thermodynamics parameters and costs evaluation, also defining the objective function as the exergetic production cost. The optimization problem evaluation is developed for two energy systems. BACKGROUND: The combined effects of vanillin and syringaldehyde on xylitol production by Candida guilliermondii using response surface methodology (RSM) have been studied. A 2(2) full-factorial central composite design was employed for experimental design and analysis of the results. RESULTS: Maximum xylitol productivities (Q(p) = 0.74 g L(-1) h(-1)) and yields (Y(P/S) = 0.81 g g(-1)) can be attained by adding only vanillin at 2.0 g L(-1) to the fermentation medium. These data were closely correlated with the experimental results obtained (0.69 +/- 0.04 g L(-1) h(-1) and 0.77 +/- 0.01 g g(-1)) indicating a good agreement with the predicted value. C. guilliermondii was able to convert vanillin completely after 24 h of fermentation with 94% yield of vanillyl alcohol. CONCLUSIONS: The bioconversion of xylose into xylitol by C. guilliermondii is strongly dependent on the combination of aldehydes and phenolics in the fermentation medium. Vanillin is a source of phenolic compound able to improve xylitol production by yeast. The conversion of vanillin to alcohol vanilyl reveals the potential of this yeast for medium detoxification. Response surface methodology was used to evaluate optimal time, temperature and oxalic acid concentration for simultaneous saccharification and fermentation (SSF) of corncob particles by Pichia stipitis CBS 6054. Fifteen different conditions for pretreatment were examined in a 2(3) full factorial design with six axial points. Temperatures ranged from 132 to 180 degrees C, time from 10 to 90 min and oxalic acid loadings from 0.01 to 0.038 g/g solids. Separate maxima were found for enzymatic saccharification and hemicellulose fermentation, respectively, with the condition for maximum saccharification being significantly more severe. Ethanol production was affected by reaction temperature more than by oxalic acid and reaction time over the ranges examined. The effect of reaction temperature was significant at a 95% confidence level in its effect on ethanol production. Oxalic acid and reaction time were statistically significant at the 90% level. The highest ethanol concentration (20 g/l) was obtained after 48 h with an ethanol volumetric production rate of 0.42 g ethanol l(-1) h(-1). The ethanol yield after SSF with P. stipitis was significantly higher than predicted by sequential saccharification and fermentation of substrate pretreated under the same condition. This was attributed to the secretion of beta-glucosidase by P. stipitis. During SSF, free extracellular beta-glucosidase activity was 1.30 pNPG U/g with P. stipitis, while saccharification without the yeast was 0.66 pNPG U/g. Published by Elsevier Ltd.
