Torção testicular (TT) é uma síndrome urológica comumente encontrada em recém nascidos, crianças e adolescentes. Neste trabalho foi estudada as lesões morfológicas e a função reprodutiva em ratos adultos que sofreram TT, em diferentes idades de maturidade sexual e o efeito protetor da L-arginina, contra os danos causados pela isquemia/reperfusão na torção testicular. Dezoito ratos pré-púberes (4 semanas de vida) dezessete púberes (6 semanas de vida) e dezessete adultos (9 semanas de vida) foram submetidos à TT. Sob anestesia o testículo direito foi rotacionado em 720 e fixado, sendo então destorcido após 4 horas. Vinte e quatro ratos (ARG4, n=8, ARG6, n=8 e ARG9 n=8) foram submetidos ao tratamento com 650mg/kg de L-arginina, por via oral, durante 7 dias. Outros trinta ratos de mesma idade sofreram cirurgia simulada (SH4, n=10, SH6, n=10 e SH9, n=10). Com 12 semanas de idade, foram submetidos ao acasalamento controlado com 3 fêmeas e ao vigésimo dia de gestação o número de fetos, corpos lúteos, absorções e implatações foram contados. Na 14 semana, os ratos foram mortos e os espermatozóides coletados da cauda dos epidídimos, foi anotado o peso corporal, o peso e volume testicular. O soro foi usado para dosagem de testosterona. Foram avaliadas a concentração, motilidade e a viabilidade espermática. Os testículos coletados foram fixados em Bouin, pós-fixados em formalina e processados em parafina. Utilizando o programa de imagem Image J, lâminas coradas com HE, mensuramos a altura do epitélio, densidade volumétrica e diâmetro do túbulo seminífero. Para avaliar a integridade do epitélio seminífero foi utilizado a frequência dos estágios do ciclo do epitélio e o escorre de Johnsen. Também foi avaliado a proliferação do compartimento tubular e intertubular, através da imunomarcação com PCNA. Os dados foram tabulados e as médias dos grupos comparadas pelo teste de ANOVA com pós-teste de Bonferroni ou teste de Kruskal-Wallis com pós teste de Dunns (programa Graphpad Prism, com p < 0,05). Os resultados revelaram grandes danos produzidos pela injuria testicular, no testículo ipsilateral, com diminuição da capacidade reprodutiva, da concentração, viabilidade e mobilidade, do peso e volume testicular. Animais TT9 não apresentaram espermatozoides nas amostras coletas. Houve diminuição do diâmetro dos túbulos seminíferos e da altura do epitélio, aumento do compartimento intertubular, com ênfase dos vasos sanguíneos, aumento da proliferação celular estromal e diminuição da proliferação epitelial. Houve diminuição da concentração sérica de testosterona no grupo TT4, quando comparado como grupo TT9. Em geral, os animais submetidos à torção na fase adulta foram os mais acometidos. Não foram encontradas alterações dignas de nota, nos testículo contralaterais. Animais tratados com L-arginina obtiveram melhora dos índices reprodutivos, com aumento da potência em todas as idades. Houve aumento da concentração espermática no testículo contralateral de ARG4 e ARG6, mostrando que a L-arginina atuou como antioxidante. Não houve proteção para as lesões causadas pela torção na maioria dos grupos, mas os animais tratados ARG9 apresentaram concentração espermática mensuravel, quando comparados aos ratos TT9, que tinham azospermia.

The objective of this study was to evaluate the possible use of biometric testicular traits as selection criteria for young Nellore bulls using Bayesian inference to estimate heritability coefficients and genetic correlations. Multitrait analysis was performed including 17,211 records of scrotal circumference obtained during andrological assessment (SCAND) and 15,313 records of testicular volume and shape. In addition, 50,809 records of scrotal circumference at 18 mo (SC18), used as an anchor trait, were analyzed. The (co) variance components and breeding values were estimated by Gibbs sampling using the Gibbs2F90 program under an animal model that included contemporary groups as fixed effects, age of the animal as a linear covariate, and direct additive genetic effects as random effects. Heritabilities of 0.42, 0.43, 0.31, 0.20, 0.04, 0.16, 0.15, and 0.10 were obtained for SC18, SCAND, testicular volume, testicular shape, minor defects, major defects, total defects, and satisfactory andrological evaluation, respectively. The genetic correlations between SC18 and the other traits were 0.84 (SCAND), 0.75 (testicular shape), 0.44 (testicular volume), -0.23 (minor defects), -0.16 (major defects), -0.24 (total defects), and 0.56 (satisfactory andrological evaluation). Genetic correlations of 0.94 and 0.52 were obtained between SCAND and testicular volume and shape, respectively, and of 0.52 between testicular volume and testicular shape. In addition to favorable genetic parameter estimates, SC18 was found to be the most advantageous testicular trait due to its easy measurement before andrological assessment of the animals, even though the utilization of biometric testicular traits as selection criteria was also found to be possible. In conclusion, SC18 and biometric testicular traits can be adopted as a selection criterion to improve the fertility of young Nellore bulls.

Mechanisms of testicular thermoregulation, the relationship of scrotal, testicular vascular cone (TVC), and testicular morphology with thermoregulatory capability, and their effects on semen quality and sperm production were studied in 20 Bos indicus, 28 crossbred, and 26 Bos taurus bulls. The ratio of testicular artery length and volume to testicular volume were larger (P < 0.05) in B. indicus and crossbred bulls than in B. taurus bulls (1.03 and 0.94 cm/cm(2). versus 0.48 cm/cm(3); 0.034 and 0.047 ml/cm(3) versus 0.017 ml/cm(3), respectively). Testicular artery wall thickness (average 192.5, 229.0, and 290.0 mum, respectively) and arterial-venous blood distance in the TVC (average 330.5, 373.7, and 609.4 pm, respectively) were smallest in B. indicus, intermediary in crossbred, and greatest in B. taurus bulls (P < 0.05); the proximity between arterial and venous blood was consistent with the estimated decrease in arterial blood temperature after passage through the TVC (5.9, 5.0, and 2.9 degreesC, in B. indicus, crossbred, and B. taurus bulls, respectively). In crossbred and B. taurus bulls, there was a positive top-to-bottom scrotal temperature gradient and a negative testicular subtunic temperature gradient. However, in B. indicus bulls, both scrotal and testicular subtunic temperatures gradients were positive. Differences in the vascular arrangement, characteristics of the artery (e.g. wall thickness) or thickness of the tunica albuginea may have affected the testicular arterial blood and subtunic temperatures in B. indicus bulls. Better testicular thermoregulatory capability was associated with increased scrotal shape (pendulosity), testicular artery length and volume, and top-to-bottom gradient of the distance between the artery wall and the veins in the TVC. Increased semen quality was associated with increased testicular volume and scrotal subcutaneous (SQT) temperature gradient, and with decreased scrotal surface and testicular temperatures. Increased sperm production was associated with increased testicular artery volume, testicular volume, and SQT temperature gradient, and with decreased testicular artery wall thickness, scrotal circumference (SC), and scrotal surface, testicular subtunic, and epididymal temperatures. In conclusion, morphology of the TVC may contribute to the greater resistance of B. indicus bulls to high ambient temperatures by conferring a better testicular blood supply and by facilitating heat transfer between the testicular artery and veins. Testicular thermoregulation was associated with opposing scrotal and testicular subtunic temperatures gradients only in crossbred and B. taurus bulls. Scrotal, TVC, and testicular morphology influence testicular thermoregulatory capability and were associated with differences in semen quality and sperm production. (C) 2003 Elsevier B.V. The objectives were to determine the effects of age and genetic group on characteristics of the scrotum, testes and testicular vascular cones (TVC), and on sperm production and semen quality in 107 Bos indicus, B, taurus and cross-bred bulls at three artificial insemination (AI) centers in Brazil. In addition, predictors of sperm production and semen quality were identified. In general, scrotal circumference (SC), scrotal shape score, scrotal neck perimeter, and testicular size (length, width and volume) increased (P < 0.05) with age. Although there were no significant differences among genetic groups for SC or testicular size, B. indicus bulls had the least pendulous scrotal shape, the shortest scrotal neck length, and the greatest scrotal neck perimeter (P < 0.05). Fat covering the TVC was thinner (P < 0.05) in bulls <= 36 months of age and in B. taunts bulls than in older bulls and B. indicus bulls, respectively. Age and genetic group did not affect testicular ultrasonic echotexture. B. indicus bulls tended (P < 0.1) to have the lowest average scrotal surface temperature (SST). In general, ejaculate volume, total number of spermatozoa and number of viable spermatozoa increased (P < 0.05) with age. However, there was no significant effect of age on sperm concentration, motility, major and total defects. The proportion of spermatozoa with minor defects was highest (P < 0.05) in bulls 37-60 months of age. B. indicus bulls had higher (P < 0.01) sperm concentration, total number of spermatozoa and number of viable spermatozoa than B. taunts bulls, with intermediate values for cross-bred bulls. Increased sperm production was associated with increased testicular volume, SC, TVC fat cover, and SST top-to-bottom gradient. Decreased semen quality was associated with increased SC and bottom SST, and decreased scrotal shape, scrotal neck perimeter and vascular cone diameter. In summary, age and genetic group affected the characteristics of the scrotum, testes, and TVC, sperm production and semen quality. In addition, characteristics of the scrotum, testes and TVC were associated with sperm production and semen quality in bulls and could be assessed for breeding soundness evaluation. Aiming to determine the relationship between the frequency of testicular shape and the andrological aspects in young Nellore bulls, 18,676 animals were assessed. All andrological examinations were performed between the years 2000 and 2008. Animals were classified as able for breeding, able for breeding in natural mating system, unable for breeding and discarded. The testicular shape was classified as long, fairly long, oval-long, spherical-oval, and spherical. The analysis of Pearson correlations was performed for testicular shape with scrotal circumference, testicular volume, progressive motility, sperm vigor, major defects, minor defects and total defects. Testicles with oval shape prevailed (99.61%). It was obseved that 76.34; 66.34; 64.34; 58.33 and 50.00% of the animals were classified as sound for breeding for shapes long, fairly long, oval-long, spherical-oval, and spherical, respectively. Correlations between testicular shape with scrotal circumference, testicular volume, progressive motility, sperm vigor, major, minor and total defects were 0.26; 0.08; 0.00; 0.11; -0.02; 0.02 and -0.01, respectively. Testicular shape had no influence upon the andrological examination results. Testicles of long shape were prevalent within the population.

O objetivo foi verificar pela ultrassonografia associada à biometria testicular como ferramenta na seleção de precocidade sexual em machos bubalinos. Foram usados 19 machos mestiços das raças Murrah e Mediterrâneo, com idades entre 11 a 59 meses, no período de 06 de maio a 18 de novembro de 2010. Os animais foram divididos em 12 faixas etárias: 8 animais de 12 a 19 meses; 3 animais de 20 a 29 meses; e 8 animais de 30 a 59 meses. Os animais foram mantidos em sistema extensivo, em área de várzea com pastagem de canarana (Eriochloa SP) sem mineralização. Os animais foram submetidos a exame clínico andrológico, com colheita de sêmen por massagem transretal das ampolas e análises físicas do ejaculado. Os exames ultrassonograficos foram realizados com o aparelho modelo Mindray DP-2200, com transdutor linear transretal 75L50EAV, multi-frequencial de 5,0/7,5/10,0 MHz. A ecodensidade testicular (ECOt) foi expressa em quantidade de pixels/área, utilizando o Programa Image J. Foram coletadas amostras sanguíneas para aferição dos níveis de testosterona. A análise estatística foi feita utilizando-se o programa estatístico SAS 1999. Os resultados da circunferência escrotal mostram médias e desvios padrão mínimo e máximo de 12.88±0.51cm para animais de 12 e 13 meses e de 28.38±0.38cm nos machos com idade maior ou igual a 60 meses, respectivamente. As medidas de volume testicular apresentaram valores mínimo e máximo de 30.28±17.37cm³ em animais de 12 e 13 meses e de 534.25±25.36 cm³ nos animais maiores ou iguais a 60 meses, respectivamente. As médias e desvio padrão da concentração espermática mínima e máxima encontrada foram de 5.5± 3.5(x 10⁶ /mm³) para os animais de 16 e 17 meses e de 51.41± 26.02 (x 10⁶ /mm³) para os maiores ou iguais a 60 meses, respectivamente. Na motilidade espermática as porcentagens mínima e máxima obtidas foram de 10% para animais com 16 e 17 meses e de 80±5.77% para maiores ou iguais a 60 meses, respectivamente. Os defeitos espermáticos totais mínimos e máximos encontrados foram de 18.5 % nos animais de 16 e 17 meses e 3%. Nos maiores ou iguais a 60 meses, respectivamente. Os níveis de testosterona mínimos e máximos encontrados foram de 0.070±0.026 ng/ml para 12 e 13 meses e de 2.762± 0.457 ng/ml para os animais maiores ou iguais a 60 meses, respectivamente. A ecogenicidade para animais na faixa etária de 12 a 13 meses foi de 78.67±6.36 pixel, na faixa de 14 e 15 meses foi de 94.22±3.40 pixel, para os animais com idades de 16 e 17 meses foi de 88.16±3.95 pixel, nos animais entre 18 a 19 meses foi de 96.09±3.40 pixel, para os animais de 20 e 21 meses foi de 103.12±3.86 pixel, para 22 e 23 meses foi de 98.4±5.87 pixel, para a idade de 24 aos 29 meses foi de 114.05±2.42 pixel, na faixa etária de 30 aos 35 meses foi de 109.24±3.13 pixel, para animais de 36 aos 41meses foi de 98.67±3.05 pixel, entre 42 a 47 meses a média foi 99.33±2.01 pixel, entre 48 a 59 meses foi de 96.17±1.90 pixel e nos animais maiores ou iguais a 60 meses de idade foi de 90.13±1.77 pixel. Concluímos, assim, que a ultrassonografia, pela interpretação da ecogenicidade, associada ao exame clinico andrológico, é uma ferramenta que pode ser usada na seleção de precocidade e avaliação de fertilidade de machos bubalinos.
