This work studies the effect of NTMP (nitrilotris(methylenephosphonic acid)) on the adsorption of Cu(II), Zn(II), and Cd(II) onto boehmite in the pH range 5-9.5. The data were analyzed using the 2-pK constant capacitance model (CCM) assuming ternary surface complex formation. Under stoichiometric conditions, NTMP is more effective for removing Cu(II) than Zn(II) from solution and the contribution of ternary surface complexes are important to model the adsorption of both metals. Under nonstoichiometric conditions and high surface loading with a Me(II)/NTMP ratio of 1:5, Cu(II) and Zn(II) adsorption is significantly suppressed. In the case of Cd(II) the free metal adsorption is the most dominant species.

The aim of this study was to determine whether inclusion complexes between 2-hydroxypropyl-beta-cyclodextrin (HP beta CD) and finasteride (FIN) are formed, and to characterize these. Equimolar FIN/HP beta CD solid systems in the presence or absence of 0.1% (w/v) of polyvinylpyrrolidone K30 (PVP K30) or 0.3% of chitosan were prepared by coevaporation and freeze-drying methods. The systems were characterized by phase solubility, NMR, DSC, and XRD analysis. The results suggest that true binary and ternary inclusion complexes were formed. (c) 2009 Elsevier Ltd. Stable ternary transcription complexes assembled in vitro, using a HeLa whole-cell extract, have been isolated and visualized by electron microscopy. The formation of these stable complexes on the DNA fragment used as template, the 5' end region of the Xenopus laevis vitellogenin gene B2, depends on factors present in the whole-cell extract, RNA polymerase II and at least two nucleotides. Interestingly, bending in the DNA fragment was frequently observed at the binding site of RNA polymerase II. Dinucleotides that can prime initiation within a short sequence of approximately 10 contiguous nucleotides centered around the initiation site used in vivo, also favour the formation of stable complexes. In addition, pre-initiation complexes were isolated and it was shown that factors in the extract involved in their formation are more abundant than the RNA polymerase II molecules available for binding. The possible implication of this observation relative to the in vivo situation is discussed.

The binding selectivity of the M(phen)(edda) (M = Cu, Co, Ni, Zn; phen = 1,10-phenanthroline, edda = ethylenediaminediacetic acid) complexes towards ds(CG)(6), ds(AT)(6) and ds(CGCGAATTCGCG) B-form oligonucleotide duplexes were studied by CD spectroscopy and molecular modeling. The binding mode is intercalation and there is selectivity towards AT-sequence and stacking preference for A/A parallel or diagonal adjacent base steps in their intercalation. The nucleolytic properties of these complexes were investigated and the factors affecting the extent of cleavage were determined to be: concentration of complex, the nature of metal(11) ion, type of buffer, pH of buffer, incubation time, incubation temperature, and the presence of hydrogen peroxide or ascorbic acid as exogenous reagents. The fluorescence property of these complexes and its origin were also investigated. The crystal structure of the Zn(phen)(edda) complex is reported in which the zinc atom displays a distorted trans-N4O2 octahedral geometry; the crystal packing features double layers of complex molecules held together by extensive hydrogen bonding that inter-digitate with adjacent double layers via pi...pi interactions between 1,10-phenanthroline residues. The structure is compared with that of the recently described copper(II) analogue and, with the latter, included in molecular modeling. (C) 2008 Elsevier B.V. All rights reserved.

Dapsone (DAP) is a synthetic sulfone drug with bacteriostatic activity, mainly against Mycobacterium leprae. In this study we have investigated the interactions of DAP with cyclodextrins, 2-hydroxypropyl-beta-cyclodextrin (HP beta CD) and beta-cyclodextrin (beta CD), in the presence and absence of water-soluble polymers, in order to improve its solubility and bioavailability. Solid systems DAP/HP beta CD and DAP/beta CD, in the presence or absence of polyvinylpyrrolidone (PVP K30) or hydroxypropyl methylcellulose (HPMC), were prepared. The binary and ternary systems were evaluated and characterized by SEM, DSC, XRD and NMR analysis as well as phase solubility assays, in order to investigate the interactions between DAP and the excipients in aqueous solution. This study revealed that inclusion complexes of DAP and cyclodextrins (HP beta CD and beta CD) can be produced in order to improve DAP solubility and bioavailability in the presence or absence of polymers (PVP K30 and HPMC). The more stable inclusion complex was obtained with HP beta CD, and consequently HP beta CD was more efficient in improving DAP solubility than beta CD, and the addition of polymers had no influence on DAP solubility or on the stability of the DAP/CDs complexes.

The isotherms of adsorption of CuX2 (X=Cl-, Br-, ClO4-) by silica gel chemically modified with 2-amino-1,3,4-thiadiazole were studied in acetone and ethanol solutions: at 298 K. The following equilibria constants (in 1 mol(-1)) were determined: (a) CuCl2: 3.5 x 10(3) (ac), 2.0 x 10(3) (eth); (b) CuBr2: 2.8 x 10(3) (ac), 2.0 x 10(3) (eth); (c) Cu(ClO4)(2): 1.8 x 10(3) (ac), 1.0 x 10(3) (eth); ac = acetone, eth = ethanol. The electron spin resonance spectra of the surface complexes indicated a tetragonal distorted structure in the case of lower degrees of metal loading on the chemically modified surface. The d-d electronic transition spectra showed that for the ClO4-, complex, the peak of absorption did not change for any degree of metal loading, and for Cl- and Br- complexes, the peak maxima shifted to a higher energy region with a lower metal loading. This study comprised batch experiments, direct speciation studies via EXAFS, and modelling with the 2SPNE SC/CE model to elucidate the mechanisms of Np(V) sorption on montmorillonite and, for reference, on γ-Al2O3. The sorption of pM 239Np(V) and µM 237Np(V) on montmorillonite (STx-1, 4 g/L) and γ-Al2O3 (0.5 g/L) was studied at room temperature in the presence and absence of ambient CO2 covering a pH-range from 2.5 (STx-1) or 5 (γ-Al2O3) to 10.5 with 0.01 or 0.1M NaClO4 as background electrolyte. The Np(V) uptake was determined by γ spectroscopy of the supernatants and calculated as percentage as well as distribution coefficient Kd. Sorption starts from pH ~6 and, under exclusion of CO2, increases continuously, while, in the presence of ambient air, it reaches a maximum at pH ~8.5 (γ-Al2O3: log Kd max ≈ 4 mL/g; STx-1: log Kd max ≈ 2.7 mL/g). Beyond that it decreases again due to the formation of queous neptunium carbonate complexes. Furthermore, neptunium sorption on montmorillonite is influenced by ionic strength at pH <6 through ion exchange processes pointing towards the formation of outer-sphere surface complexes there. Isotherms measured at the sorption maximum showed the precipitation of resumably neptunium carbonate complexes above 3∙10^-5 M under ambient air conditions. Additionally, they indicated progressive saturation of the sorption sites of γ-Al2O3. At selected pH (STx-1: 5.0, 7.0, 8.0, 8.5, 9.0, 9.5; γ-Al2O3: 8.5, 9.5) EXAFS samples were prepared as wet pastes with µM 237Np and measured at room temperature in fluorescence mode at ANKA and ESRF. Several spectra were averaged and analysed with EXAFSPAK and FEFF 8.20 employing models of NaNpO2(CO3) or soddyite, (UO2)2SiO4∙2(H2O). The shorter atomic distances of the neptunyl ion at pH 5 compared to the others hinted at the retention of the hydration shell and, thus, at outer-sphere sorption. On average the bond lengths for Np(V) sorbed on STx-1 at high pH were Oax ≈ 1.84 Å and Oeq ≈ 2.53 Å. At high pH, ternary neptunyl carbonate surface complexes could be identified for montmorillonite (C ≈ 3.00 Å), but not for γ-Al2O3, where an interaction of neptunium with the aluminium surface atoms according to the soddyite model gave better agreement with the experimental data. However, neither structure as suggested by the two models could be excluded for both systems rendering a combination most likely. Modelling of the sorption data provided further evidence for the existence of ternary neptunium carbonate surface complexes in both cases. The results of this study can aid environmental risk assessment for clay-based nuclear waste repositories by providing valuable input data for simulations of radionuclide migration from a final disposal site.

The adsorption of As(V) onto alpha -Al2O3 was investigated at 25, 50 and 70 degreesC using batch adsorption experiments. Results indicate that As is strongly adsorbed at low pH and gets progressively released to the fluid with increasing pH above 7. At any pH, increasing temperature favors aqueous species of As over surface species. Surface complexation constants were determined at the experimental temperatures by fitting the adsorption data. Adsorption reactions were then converted to semi-isocolumbic reactions, i.e, reactions with balanced like-charged aqueous species. Intrinsic adsorption constants of semi-isocolumbic reactions change linearly when plotted against inverse temperature, suggesting that the heat capacity of these reactions remains constant over the temperature range considered. This permitted thermodynamic parameters of intrinsic surface complexation constants to be determined. Changes in surface complexation constants result in a change in the surface speciation with increasing temperature. This change is similar to the one observed for aqueous species, i.e. increasing temperature favors less negatively charged species below a pH of 9 and more negatively charged species above a pH of 10. Comparison with the stability of As surface complexes with Fe suggests that surface complexes with Al are more stable. (C) 2001 Elsevier Science Ltd. All rights reserved.

The structural saturation and stability, the energy gap, and the density of states of a series of small, silicon-based clusters have been studied by means of the PM3 and some ab initio (HF/6-31G* and 6-311++G**, CIS/6-31G* and MP2/6-31G*) calculations. It is shown that in order to maintain a stable nanometric and tetrahedral silicon crystallite and remove the gap states, the saturation atom or species such as H, F, Cl, OH, O, or N is necessary, and that both the cluster size and the surface species affect the energetic distribution of the density of states. This research suggests that the visible luminescence in the silicon-based nanostructured material essentially arises from the nanometric and crystalline silicon domains but is affected and protected by the surface species, and we have thus linked most of the proposed mechanisms of luminescence for the porous silicon, e.g., the quantum confinement effect due to the cluster size and the effect of Si-based surface complexes.

Zur Modellierung von Vergasungs- und Verbrennungsprozessen zur energetischen Nutzung von Biomasse ist die Kenntnis von reaktionskinetischen Daten für die Sauerstoff-Oxidation von Biomassepyrolysaten erforderlich. Eine ausführliche Literaturübersicht zeigt den Stand der Forschung bezüglich der experimentellen Ermittlung von reaktionskinetischen Parametern für die Oxidation von Pyrolysaten aus Lignin, Cellulose und pflanzlicher Biomasse sowie der Suche nach einem plausiblen Reaktionsmechanismus für die Reaktion von Sauerstoff mit festen Kohlenstoffmaterialien. Es wird eine Versuchsanlage mit einem quasistationär betriebenen Differentialreaktor konstruiert, die eine Messung der Reaktionskinetik und der reaktiven inneren Oberfläche (RSA) für die Reaktion eines Pyrolysats aus Maispflanzen mit Sauerstoff ermöglicht. Die getrockneten und zerkleinerten Maispflanzen werden 7 Minuten lang bei 1073 K in einem Drehrohrofen pyrolysiert. Das Pyrolysat zeichnet sich vor allem durch seine hohe Porosität von über 0,9 und seinen hohen Aschegehalt von 0,24 aus. Die RSA wird nach der Methode der Messung von Übergangskinetiken (TK) bestimmt. Die Bestimmung der RSA erfolgt für die Reaktionsprodukte CO und CO2 getrennt, für die entsprechend ermittelten Werte werden die Bezeichnungen CO-RSA und CO2-RSA eingeführt. Die Abhängigkeit dieser Größen von der Sauerstoffkonzentration läßt sich durch eine Langmuir-Isotherme beschreiben, ebenso das leichte Absinken der CO-RSA mit der Kohlendioxidkonzentration. Über dem Abbrand zeigen sich unterschiedliche Verläufe für die CO-RSA, CO2-RSA und die innere Oberfläche nach der BET-Methode. Zur Charakterisierung der Oberflächenzwischenprodukte werden temperaturprogrammierte Desorptionsversuche (TPD) durchgeführt. Die Ergebnisse zeigen, daß eine Unterscheidung in zwei Kohlenstoff-Sauerstoff-Oberflächenkomplexe ausreichend ist. Die experimentellen Untersuchungen zum Oxidationsverlauf werden im kinetisch bestimmten Bereich durchgeführt. Dabei werden die Parameter Temperatur, Sauerstoff-, CO- und CO2-Konzentration variiert. Anhand der Ergebnisse der reaktionskinetischen Untersuchungen wird ein Reaktionsmechanismus für die Kohlenstoff-Sauerstoff-Reaktion entwickelt. Dieser Reaktionsmechanismus umfaßt 7 Elementarreaktionen, für welche die reaktionskinetischen Parameter numerisch ermittelt werden. Darüber hinaus werden reaktionskinetische Parameter für einfachere massenbezogene Reaktionsgeschwindigkeitsansätze berechnet und summarische Reaktionsgeschwindigkeitsansätze für die Bildung von CO und CO2 aus dem Reaktionsmechanismus hergeleitet.

Selen kann in verschiedenen Oxidationszuständen (+6, +4, ±0, -2) in unterschiedlichen Umweltkompartimenten auftreten. Verbundenen damit sind verschiedene Eigenschaften, wie z. B. die Wasserlöslichkeit, die in direktem Zusammenhang mit der Migrationsfähigkeit sowie der Bioverfügbarkeit steht. Im Rahmen der vorliegenden Arbeit wurden die Verfügbarkeit anorganischer Selenspezies und damit die Mobilisierbarkeit dieser in verschiedenen Laborexperimenten untersucht. Hierbei wurde an Goethit adsorbiertes Selenit sowohl mit einer Reinkultur des aktiv methylierenden Pilzes Alternaria alternata als auch mit einer angereicherten Umweltmischkultur inkubiert und die mikrobiologische Zugänglichkeit anhand der Bildung leichtflüchtiger, alkylierter Selenmetabolite wie z. B. Dimethylselenid und Dimethyldiselenid beobachtet. Zur Analyse dieser wurde eine cryotrapping-cryofocussing-GC-ICP-MS-Kopplung etabliert. Die Anteile der methylierten Selenverbindungen stiegen bei Verwendung von A. alternata mit der Inkubationszeit auf 10 % des gelösten Selens und 1 % des Gesamtselens an. Dieser Trend konnte während der Inkubation der Umweltmischkultur nicht beobachtet werden. Hier lagen die Anteile über den gesamten Untersuchungszeitraum bei ca. 0,5 % des gelösten bzw. 0,1 % des Gesamtselens, inklusive eines leichten Abwärtstrends, welcher wahrscheinlich durch die Nutzung der Alkylselenide als Kohlenstoffquelle hervorgerufen wurde. Weiterhin wurde das reduzierte Eisenselenidmineral Ferroselit eingesetzt, um dessen Stabilität gegenüber der Aktivität des sulfatreduzierenden Bakteriums Desulfovibrio gigas zu untersuchen. Mit zunehmender Inkubationszeit und damit verbundener, zunehmender Reduktion des im Nährmedium vorhandenen Sulfates konnte ein Anstieg leichtflüchtiger Organoselenverbindungen in der Gasphase der Kulturansätze festgestellt werden, die im unteren Nanogrammbereich lagen. Einhergehend damit wurde auch die Zunahme der Gehalte an gelöstem Selen und somit die biologisch bedingte Rücklösung aus der Mineral- in die Wasserphase beobachtet. Es konnte gezeigt werden, dass die Aktivität von Mikroorganismen einen deutlichen Einfluss auf die Stabilität von Oberflächenkomplexen des Selenits als auch von mineralischen Selenidspezies hat.
