The knockdown and toxic effects of insecticides of different chemical groups and modes of action registered for citrus in Brazil were investigated for effective control of Bucephalogonia xanthophis, a sharpshooter vector of Xylella fastidiosa in citrus. The active ingredients dimethoate (1.2 mL/1.2L), imidacloprid (0.24 mL/1.2L) and lambda-cyhalothrin (0.24 mL/1.2L), as well as a control (water), were sprayed onto branches of potted-citrus nursery trees to evaluate the effect of residual contact. The insects were confined on sprayed branches by using sleeve cages, in groups of 10 per branch (5 branches/treatment). Lambdacyhalothrin showed a knockdown effect on B. xanthophis (>70% mortality within 2 h of exposure), and the residues were effective for approximately one wk. Imidacloprid, lambdacyhalothrin and dimethoate suppressed the vector populations for up to 3 wk after application, when the insects were exposed to sprayed plants for at least 24 h. In another experiment, 2 neonicotinoid insecticides (thiamethoxam and imidacloprid) were applied by soil drench to potted nursery trees, in order to study their systemic effect, i.e., mortality by ingestion on sharpshooter adults. Thiamethoxam and imidacloprid effectively controlled the vectors at all concentrations tested, when the insects were exposed to treated plants for 24 h (>80% mortality) or 48 h (near 100% mortality). The knockdown effect of thiamethoxam and lambda-cyhalothrin might be particularly important to prevent vector transmission of X. fastidiosa in citrus groves.

The purpose of this study was to evaluate the efficacy of a Coulomb Controlled Iontophoresis system (CCI) in the local delivery of corticosteroids for the treatment of uveitis. The therapeutic efficacy of Dexamethasone (Dex) administered by CCI was compared to systemic injection and to topical application with the iontophoresis apparatus in the absence of electrical current. The evaluation was done in the treatment of the endotoxin-induced uveitis (EIU) model, and in the effect on TNF gene expression in the iris/ciliary body as well as in the retina and on TNF levels in aqueous humor and vitreous. Dex was administered either at the time of LPS injection or 5 hours later. For iontophoresis, we used a 1 ml reservoir-electrode covering the cornea, the limbus, and the first millimeter of the sclera. The applied electrical current was of 400 microA during four minutes with a total surface charge of 0.4 C cm-2. EIU was evaluated by clinical examination, by counts of intraocular inflammatory cells on histological sections, and by measuring the protein levels in the aqueous humor and in the vitreous. The TNF-alpha gene expression in the iris and ciliary body, and in the retina was evaluated by RT-PCR. The systemic effect of Dex delivered by CCI was evaluated on the level of serum TNF-alpha in EIU. Our results demonstrated that local administration of Dex by CCI inhibited anterior and posterior signs of intraocular inflammation as effectively as systemic administration, with no effect on systemic level of TNF. In the anterior and posterior segments of the eye, the protein exudation. TNF levels and the cellular infiltration were inhibited. The TNF-alpha gene expression was inhibited in the anterior as well as the posterior segment of the eye. No clinical nor histological damage were caused by the CCI apparatus. In conclusion, CCI administration of Dex allows for a therapeutic effect on the posterior as well as the anterior segment of the eye, and may present a viable alternative to systemic administration of glucocorticoids in severe ocular inflammations.

La grossesse s’accompagne d’importantes modifications hormonales et hémodynamiques. Parmi celles-ci, le système rénine-angiotensine-aldostérone (SRAA) est activé très tôt durant la grossesse. De plus, cette augmentation du SRAA est accompagnée d’élévations du débit cardiaque et du volume plasmatique ainsi que des baisses paradoxales de la pression artérielle et de la résistance vasculaire périphérique. Ceci suggère que la grossesse induit un remaniement des réponses physiologiques normales au SRAA. Une résistance vasculaire à l’action des vasopresseurs est également observée durant la gestation. Ce phénomène serait causé par la modification de la fonction des canaux calciques et potassiques. De plus, il serait possiblement dû à la participation de la Na+/K+-ATPase, par son influence sur le potentiel membranaire des cellules des muscles lisses vasculaires (VSMC). La présence des récepteurs minéralocorticoïdes (MR) dans les VSMC laisse croire que l’aldostérone peut influencer le tonus vasculaire par des effets génomiques et non-génomiques. Compte tenu des connaissances actuelles, nous avons émis l’hypothèse que l’augmentation des taux sériques d’aldostérone durant la grossesse est responsable des changements hémodynamiques observés et que ces effets sont causés par l’activation des MR. Des rates gestantes ont été traitées avec du canrénoate de potassium (20 mg/kg•jr), un antagoniste des MR, durant la dernière semaine de gestation (sur 3). Sur des anneaux aortiques dénudés de leur endothélium, nous avons mesuré les réponses contractiles à la phényléphrine (PhE) et au KCl en présence d’un bloqueur des canaux calciques dépendants du voltage (VDCC), la nifédipine, et d’activateurs des canaux potassiques à large conductance (BKCa) et ceux dépendants de l’ATP (KATP), respectivement le NS-1619 et la cromakalim. Les réponses à la PhE et au KCl sont réduites à partir du 17e jour de gestation et le traitement au canrénoate augmente ces réponses dans tous les groupes. Les modulateurs de canaux ioniques atténuent les réponses à la PhE et au KCl. Cependant, le canrénoate modifie aussi les effets des modulateurs sur les aortes. Aucun effet ou une baisse des réponses est observable chez les rates non gestantes, tandis qu’une hausse de leur effet inhibiteur est notée chez les rates gestantes. Ces effets du canrénoate font croire que l’aldostérone participe à l’adaptation de la réactivité vasculaire durant la grossesse. Par ailleurs, le potentiel membranaire des VSMC pourrait être affecté dans la gestation. Pour vérifier cette hypothèse, nous avons évalué l’activité de la Na+/K+-ATPase, impliquée dans le contrôle du potentiel membranaire. Nos résultats démontrent que l’activité de la pompe est inhibée à partir du 19e jour de gestation. Cet effet est renversé par le canrénoate. Toutefois, comme le renversement de l’inhibition de la pompe est également présent chez les rates gestantes traitées avec du PST 2238, un antagoniste de l’ouabaïne sur la Na+/K+-ATPase, et que le canrénoate agit également comme agoniste partiel de la pompe, nous croyons que la diminution d’activité associée à la gestation est liée à une inhibition de la Na+/K+-ATPase par des stéroïdes cardiotoniques plutôt qu’à un effet des minéralocorticoïdes. L’augmention d’activité de la pompe liée au canrénoate s’accompagne d’une diminution de l’expression de la sous-unité α1, suggérant que la sous-unité α2 est responsable des variations de contractilité de l’aorte, puisque son expression n’est pas modifiée par le canrénoate. Les effets de la diminution de l’expression de la sous-unité α1, influencée par la signalisation du MR, restent à être déterminés. Néanmoins, nos résultats montrent que les modifications d’activité de la Na+/K+-ATPase influencent l’activité des canaux potassiques et que la pompe pourraient être un des éléments primordiaux dans le contrôle de la réactivité vasculaire durant la grossesse. Comme le canrénoate modifie la réactivité vasculaire, nous voulions déterminer ses impacts sur la pression artérielle. Des rates gestantes ont été traitées avec du canrénoate (20 ou 60 mg/kg•jr) et les paramètres hémodynamiques ont été évalués par radiotélémétrie. Aucune modification de la pression artérielle, du rythme cardiaque et de la pression pulsée ne sont mesurées chez les rates recevant le traitement. Toutefois, des augmentations de l’osmolalité, des taux sériques d’aldostérone et de corticostérone ainsi que de l’activité rénine plasmatique sont observées chez les animaux recevant 60 mg/kg•jr. Le canrénoate bloque donc le rétrocontrôle du SRAA. Par contre, les MR ne sont pas les principaux responsables du contrôle de la pression artérielle durant la grossesse. En conclusion, nous avons démontré que le traitement des rates au canrénoate influence la réactivité vasculaire de l’aorte durant la gestation. Cet effet est causé par la modification de l’activité de certains canaux ioniques (VDCC, BKCa et KATP). De plus, le canrénoate renverse l’inhibition de la Na+/K+-ATPase observée durant la gestation. Finalement, les actions locales de cet antagoniste des MR sur les vaisseaux sanguins ne se répercutent pas sur l’effet systémique global et aucune modification de la pression artérielle n’est observée. D’autres études seront toutefois nécessaires pour déterminer les voies de signalisation par lesquelles l’aldostérone module les réponses des canaux ioniques dans les VSMC.

It is contended that competitiveness is better understood as a discourse rather than as a characteristic that is supposedly possessed. The discourse of competitiveness derives its legitimacy from the enterprise culture that came to dominance during the 1980s. Current popularized theories of competitiveness are constituent parts of this broader discourse, which has had significant material implications for the UK construction sector. The dominant discourse of competitiveness amongst contracting firms is shaped by the need to achieve structural flexibility to cope with fluctuations in demand. Fashionable espoused improvement recipes such as total quality management, business process re-engineering, and lean construction legitimize and reinforce the material manifestations of the enterprise culture. In consequence, the UK industry is characterized by a plethora of hollowed-out firms that have failed to invest in their human capital. While the adopted model may be rational for individual firms, the systemic effect across the sector as a whole equates to a form of anorexia. However, the discourse of competitiveness is by no means monolithic and continues to be contested locally. There have also been numerous counter-discourses that have been mobilized in response to the undesirable externalities of unbridled enterprise. Currently, important counter-discourses promote the ideas of sustainability and corporate social responsibility.

Visando buscar métodos alternativos no controle da mosca-branca Bemisia tabaci (Gennadius) biótipo B em tomateiro, foram realizados testes de atratividade e preferência para oviposição em casa-de-vegetação, utilizando-se quatorze extratos aquosos a 3% (peso/volume). Os extratos foram preparados com partes de Azadirachta indica, Trichilia pallida,Chenopodium ambrosioides,Piper nigrum,Melia azedarach,Ruta graveolens,Ricinus communis,Mentha pulegium,Tagetes erecta,Eucalyptus citriodora,Cymbopogon nardus e Coriandrum sativum. Numa segunda etapa, os extratos mais eficientes em casa-de-vegetação foram observados em laboratório, a fim de avaliar o possível efeito sistêmico dos mesmos sobre ninfas da mosca-branca. Constatou-se que as plantas de tomateiro pulverizadas com extratos à base de folhas de M. pulegium e folhas e sementes de A. indica foram menos atrativas aos adultos do inseto. Plantas pulverizadas com extratos de folhas de A. indica e folhas + ramos de R. communis mostraram efeitos deterrentes à oviposição do inseto, reduzindo o número de ovos; em contrapartida, o extrato à base de folhas de C. nardus estimulou a oviposição da mosca-branca sobre as plantas. O uso dos extratos por via sistêmica não afetou o período de desenvolvimento (ovo-adulto) da mosca-branca; entretanto, a presença de extratos de sementes e folhas de A. indica e de folhas de M. pulegium provocou aumento significativo na mortalidade de ninfas de B. tabaci biótipo B.

1. With the global increase in CO2 emissions, there is a pressing need for studies aimed at understanding the effects of ocean acidification on marine ecosystems. Several studies have reported that exposure to CO2 impairs chemosensory responses of juvenile coral reef fishes to predators. Moreover, one recent study pointed to impaired responses of reef fish to auditory cues that indicate risky locations. These studies suggest that altered behaviour following exposure to elevated CO2 is caused by a systemic effect at the neural level. 2. The goal of our experiment was to test whether juvenile damselfish Pomacentrus amboinensis exposed to different levels of CO2 would respond differently to a potential threat, the sight of a large novel coral reef fish, a spiny chromis, Acanthochromis polyancanthus, placed in a watertight bag. 3. Juvenile damselfish exposed to 440 (current day control), 550 or 700 µatm CO2 did not differ in their response to the chromis. However, fish exposed to 850 µatm showed reduced antipredator responses; they failed to show the same reduction in foraging, activity and area use in response to the chromis. Moreover, they moved closer to the chromis and lacked any bobbing behaviour typically displayed by juvenile damselfishes in threatening situations. 4. Our results are the first to suggest that response to visual cues of risk may be impaired by CO2 and provide strong evidence that the multi-sensory effects of CO2 may stem from systematic effects at the neural level.

Aggregation chimeras were formed between C57BL/6 mice heterozygous for the Apcmin (Min) mutation and wild-type SWR mice, that differ in their Pla2g2a status, a modifier of Apcmin, and also in their resistance to intestinal polyp formation. Variation in the dolichos biflorus agglutinin-staining patterns of the intestines of these mouse strains was used to determine the chimeric composition of the intestine in individual mice and to examine the clonal composition of adenomas. Macroscopic adenoma numbers in chimeric mice were compared with the expected adenoma numbers based on the percentage of C57BL/6J-Apcmin/+ epithelium in individual mice. These results unexpectedly show that there was no apparent inhibitory effect of the SWR-derived (Pla2g2a wild-type) tissue on adenoma formation in the C57BL/6J-Apcmin/+ epithelium. This suggests that the main genetic modifiers of the Min phenotype act at a cellular or crypt-restricted level with no discernable systemic effect. All adenomas were seen to contain C57BL/6J-Apcmin/+-derived epithelium, confirming that the germ-line mutation of the mApc gene is necessary to initiate tumorigenesis in this model system, and that the mApc gene acts in a cell autonomous fashion.

Le récepteur éboueur CD36 facilite l’internalisation des acides gras libres non estérifiés (AGNE) au niveau des tissus cardiaque et périphériques. Lors d’une ischémie-reperfusion du myocarde (MI/R), les dommages produits sont en partie liés à l’internalisation des AGNE et à la production d’espèces réactives de l’oxygène, contrairement à ce qui est observé chez des souris déficientes en CD36 (CD36-/-). Nous avons émis l’hypothèse selon laquelle le CP-3(iv), un ligand synthétique du récepteur CD36, exercerait un effet cardioprotecteur en réduisant la taille de la zone myocardique infarcie lors d’une ischémie transitoire du myocarde. Nos objectifs étaient 1) de déterminer l’effet cardioprotecteur du CP-3(iv) et 2) de définir son mécanisme. Pour cela, des études in vivo et ex vivo ont été faites. Des souris de type sauvage ont été traitées avec le CP-3(iv) (289 nmol/kg) par voie sous-cutanée pendant 14 jours avant d’être soumises à 30 minutes d’ischémie suivant la ligature de l’artère coronaire gauche descendante et de sa reperfusion pendant une période de 6 ou 48 heures. De plus, des coeurs isolés de souris ont été perfusés 30 minutes, suivi de 40 minutes à faible débit (10%) et de 30 minutes de reperfusion pendant laquelle le coeur est perfusé avec le CP-3(iv) à une concentration de 10-6 M. Nos travaux ont montré que l’effet cardioprotecteur d’un traitement préventif par le CP-3(iv) permet de diminuer la taille de l’infarctus et préserve l’hémodynamie cardiaque de façon dépendante du CD36 puisque cet effet est non visible chez les souris CD36-/-. De plus, le CP-3(iv) exerce non seulement un effet systémique, mais aussi un effet cardioprotecteur direct sur le coeur isolé.

Objectives: The induction of analgesia for many chronic cutaneous lesions requires treatment with an opioid analgesic. In many patients suffering with these wounds such drugs are either contraindicated or shunned because of their association with death. There are now case reports involving over 100 patients with many different types of chronic superficial wounds, which suggest that the topical application of an opioid in a suitable gel leads to a significant reduction in the level of perceived pain. Key findings: Some work has been undertaken to elucidate the mechanisms by which such a reduction is achieved. To date there have been no proven deleterious effects of such an analgesic system upon wound healing. Although morphine is not absorbed through the intact epidermis, an open wound provides no such barrier and for large wounds drug absorption can be problematic. However, for most chronic cutaneous lesions, where data has been gathered, the blood levels of the drug applied ranges from undetectable to below that required for a systemic effect. Summary If proven, the use of opioids in this way would provide adequate analgesia for a collection of wounds, which are difficult to treat in patients who are often vulnerable. Proof of this concept is now urgently required. © 2011 Royal Pharmaceutical Society.

Purpose This study evaluated the effect of severe magnesium (Mg) dietary deficiency on systemic bone density and biomechanical resistance of bone tissue to the removal torque of osseointegrated implants Materials and Methods The sample consisted of 45 rats, each received a titanium implant in their tibial metaphysis After 60 days, the animals were divided into three groups (n = 15) according to their dietary Mg the control group received the recommended content of Mg, group Mg1 received a 75% reduction in dietary Mg content, and group Mg2 was fed a diet with a 90% reduction in Mg con tent Animals were sacrificed 150 days after implant placement Serum concentrations of Mg were measured and the effect of Mg deficiency on systemic bone density was evaluated by densitometry of the lumbar vertebrae and femur Biomechanical characteristics were measured by resistance of the bone tissue to removal of the implants Results Lower Mg serum concentrations were found for the Mg1 and Mg2 groups, however, densitometric analysis and torque evaluations showed a statistically significant difference only in the Mg2 group (P < 05) There was a statistically significant difference in removal torque between the Mg2 group and the control group Conclusions This study showed that a severe deficiency of Mg decreased the systemic bone density and removal torque of osseointegrated implants INT J ORAL MAXILLOFAC IMPLANTS 2010 25 1125-1130

Brain injury is responsible for significant morbidity and mortality in trauma patients, but controversy still exists over therapeutic management for these patients. The objective of this study was to analyze the effect of phototherapy with low intensity lasers on local and systemic immunomodulation following cryogenic brain injury. Laser phototherapy was applied (or not-controls) immediately after cryogenic brain injury performed in 51 adult male Wistar rats. The animals were irradiated twice (3 h interval), with continuous diode laser (gallium-aluminum-arsenide (GaAlAs), 780 nm, or indium-gallium-aluminum-phosphide (InGaAlP), 660 nm) in two points and contact mode, 40 mW, spot size 0.042 cm(2), 3 J/cm(2) and 5 J/cm(2) (3 s and 5 s, respectively). The experimental groups were: Control (non-irradiated), RL3 (visible red laser/ 3 J/cm(2)), RL5 (visible red laser/5 J/cm(2)), IRL3 (infrared laser/ 3 J/cm(2)), IRL5 (infrared laser/5 J/cm(2)). The production of interleukin-1IL-1 beta (IL-1 beta), interleukin6 (IL-6), interleukin-10 (IL-10), and tumor necrosis factor-alpha (TNF-alpha) was analyzed by enzyme immunoassay technique (ELISA) test in brain and blood samples. The IL-1 beta concentration in brain of the control group ;was significantly reduced in 24 h (p < 0.01). This reduction was also observed in the RL5 and IRL3 groups. The TNF-alpha and IL-6 concentrations increased significantly (p < 0.01 and p < 0.05, respectively) in the blood of all groups, except by the IRL3 group. The IL-6 levels in RL3 group were significantly smaller than in control group in both experimental times. IL-10 concentration was maintained stable in all groups in brain and blood. Under the conditions of this study, it is possible to conclude that the laser phototherapy can affect TNF-alpha, IL-1 beta and IL-6 levels in the brain and in circulation in the first 24 h following cryogenic brain injury.
