The nature of the spatial representations that underlie simple visually guided actions early in life was investigated in toddlers with Williams syndrome (WS), Down syndrome (DS), and healthy chronological age- and mental age-matched controls, through the use of a "double-step" saccade paradigm. The experiment tested the hypothesis that, compared to typically developing infants and toddlers, and toddlers with DS, those with WS display a deficit in using spatial representations to guide actions. Levels of sustained attention were also measured within these groups, to establish whether differences in levels of engagement influenced performance on the double-step saccade task. The results showed that toddlers with WS were unable to combine extra-retinal information with retinal information to the same extent as the other groups, and displayed evidence of other deficits in saccade planning, suggesting a greater reliance on sub-cortical mechanisms than the other populations. Results also indicated that their exploration of the visual environment is less developed. The sustained attention task revealed shorter and fewer periods of sustained attention in toddlers with DS, but not those with WS, suggesting that WS performance on the double-step saccade task is not explained by poorer engagement. The findings are also discussed in relation to a possible attention disengagement deficit in WS toddlers. Our study highlights the importance of studying genetic disorders early in development. The aim of the present study is to investigate the developmental profile of three aspects of prosody function, i.e. affect, focus and turn-endings in children with Williams and in those with Down’s syndrome compared to typically developing English speaking children. The tasks used were part of the computer-based battery, Profiling Elements of Prosody for Speech Communication (Peppe, McCann & Gibon, 2003). Cross-sectional developmental trajectories linking chronological and non-verbal mental age and affects and turn-ending functions of prosody were constructed. The results showed an atypical profile in both clinical populations. More interestingly, the profiles were atypical for different reasons, suggesting multiple and possibly different developmental pathways to the acquisition of prosody in these two populations.

Plasma transferrin binding in Down syndrome and Alzheimer's disease is significantly reduced compared with age matched controls and it was thought this may help elucidate a pathological time sequence for the onset of dementia in Down syndrome. In Down syndrome, there was a reduction in gallium and aluminium transferrin binding both with age and the onset of dementia. Non-transferrin bound gallium species were identified as non-transportable phosphate or silicate. Thus, the route of entry of metals into the brain must be via a transferrin mediated complex only. A clear sequence of pathological events has been demonstrated in Down syndrome which shows the pathway to development of plaques and dementia and this is believed to have an immunological origin.

Esta tese se propõe a explorar as possibilidades de lidar com o trabalho enquanto atividade, compreendendo-o, a partir da perspectiva ergológica (SCHWARTZ, 2000), portando um paradoxo ou um tipo de dialética entre o Registro 1 das normas antecedentes que marcam fortemente a antecipação desse trabalho; e o Registro 2 das dimensões singulares do encontro de encontros, que se produz em todo trabalhar. A partir da permanente dialética entre esses dois registros, a perspectiva ergológica vai buscar métodos que sejam capazes de dar visibilidade à dimensão gestionária que atravessa todo trabalhar, para que os protagonistas da atividade possam se apropriar de sua atividade, individual e coletivamente, ampliando seus níveis de autonomia frente ao trabalho e à vida. O termo ergo(trans)formação afirma uma prática de pesquisa-intervenção que se propõe transformadora da realidade, apostando numa intervenção micropolítica no seio da experiência social, conforme Rocha e Aguiar (2003). Este modo de compreensão de pesquisa e de afirmação da produção coletiva do conhecimento pauta-se numa abordagem construtivista onde conhecer não é representar uma realidade pré-existente, mas é um processo de invenção de si e do mundo (KASTRUP, 1999), em um acoplamento direto com a matéria. Enfrentamos o seguinte problema: o trabalho pode ser pensado como invenção de si e do mundo, entrando em curto-circuitagem com as formas de produzir em conformidade com organização formal do trabalho, afirmando o primado do agir próprio do trabalhador como atividade, frente ao acontecimental do trabalho? Para dar conta deste problema, propomos desenvolver uma cartografia, enquanto método de análise da atividade de trabalho, que possibilite acompanhar a curto-circuitagem entre as formas de produzir em conformidade com organização do trabalho e o agir próprio do trabalhador. São apresentadas duas experimentações de ergo(trans)formação desenvolvidas em dois territórios distintos dos mundos do trabalho: o desenvolvimento de uma Oficina de Vendas e a formação de educadores internos, em empresa financeira pública e a possibilidade de formação educacional e profissional de jovens com transtorno mental grave em uma rede de restaurantes que se desdobra na cartografia do trabalho coletivo e do coletivo de trabalho, tendo um jovem portador de síndrome de Down como protagonista da atividade. As escolhas metodológicas se fizeram em duas perspectivas distintas e complementares: (1) o método cartográfico elaborado a partir de Deleuze e Guattari (1995), Guattari e Rolnik (1986), assim como Passos, Kastrup e Escóssia (2009); e (2) o que propõe Schwartz (1999), o Dispositivo Dinâmico de Três Polos (DD3P), incorporando a reformulação conceitual proposta por Athayde e Brito (2003) denominada Comunidade Ampliada de Pesquisa. Como conclusão, refletimos se é possível pensar a atividade inventiva de trabalho, a partir das experimentações de ergo(trans)formação que apresentamos, considerando suas condições e efeitos e, mesmo, se podemos nomeá-las assim, levando-se em conta as diferenças de atividades envolvidas. Estas experimentações nos impuseram pensar o trabalho (enquanto atividade) em sua indissociabilidade com a formação humana e com a produção de subjetividade

The aim of this study is to quantify the prevalence and types of rare chromosome abnormalities (RCAs) in Europe for 2000-2006 inclusive, and to describe prenatal diagnosis rates and pregnancy outcome. Data held by the European Surveillance of Congenital Anomalies database were analysed on all the cases from 16 population-based registries in 11 European countries diagnosed prenatally or before 1 year of age, and delivered between 2000 and 2006. Cases were all unbalanced chromosome abnormalities and included live births, fetal deaths from 20 weeks gestation and terminations of pregnancy for fetal anomaly. There were 10,323 cases with a chromosome abnormality, giving a total birth prevalence rate of 43.8/10,000 births. Of these, 7335 cases had trisomy 21,18 or 13, giving individual prevalence rates of 23.0, 5.9 and 2.3/10,000 births, respectively (53, 13 and 5% of all reported chromosome errors, respectively). In all, 473 cases (5%) had a sex chromosome trisomy, and 778 (8%) had 45,X, giving prevalence rates of 2.0 and 3.3/10,000 births, respectively. There were 1,737 RCA cases (17%), giving a prevalence of 7.4/10,000 births. These included triploidy, other trisomies, marker chromosomes, unbalanced translocations, deletions and duplications. There was a wide variation between the registers in both the overall prenatal diagnosis rate of RCA, an average of 65% (range 5-92%) and the prevalence of RCA (range 2.4-12.9/10,000 births). In all, 49% were liveborn. The data provide the prevalence of families currently requiring specialised genetic counselling services in the perinatal period for these conditions and, for some, long-term care.

La présente thèse par articles aborde différentes facettes du dépistage prénatal de certaines aneuploïdies au premier trimestre de la grossesse. L’introduction retrace l’historique du dépistage prénatal et énonce les différents marqueurs biochimiques et échographiques associés aux aneuploïdies. La première publication démontre que le tabagisme maternel abaisse significativement les niveaux sanguins maternels de PAPP-A et de la fraction libre de la β-hCG et augmente significativement la clarté nucale, confirmant la nécessité de contrôler cette co-variable dans le calcul de risque final, du moins pour la trisomie 18. Le deuxième article identifie des seuils de clarté nucale au-dedesquels la biochimie génétique n’apporte aucune valeur additionnelle au dépistage prénatal de la trisomie 21 et de la trisomie 18. Pour les fœtus avec clarté nucale supérieure aux seuils établis, un diagnostic prénatal intrusif devrait être offert sans délai. Le troisième et dernier article porte sur la première détermination des niveaux plasmatiques maternels de la protéine FLRG (follistatin-related gene) au premier trimestre de grossesse et sur son rôle potentiel à titre de marqueur biochimique dans le dépistage prénatal de la trisomie 21. Bien que détectables, les niveaux plasmatiques maternels de FLRG ne sont pas significativement altérés en présence de fœtus avec syndrome de Down. Dans la discussion générale, les trois articles sont abordés sous un angle plus spécifique au Québec. Des données complémentaires et originales y sont présentées. Une discussion sur l’évolution future du dépistage prénatal est entamée et des axes de recherche sont proposés.

DSCAM est exprimé dans le cortex lors du développement et sa mutation altère l’arborisation dendritique des neurones pyramidaux du cortex moteur. Considérant que les souris DSCAM2J possèdent des problèmes posturaux et locomoteurs, nous émettons l’hypothèse que DSCAM est impliqué dans le fonctionnement normal du cortex moteur et de la voie corticospinale. Comparées aux souris contrôles, les souris DSCAM2J vont présenter des problèmes moteurs à basse vitesse et enjamber un obstacle presque normalement à vitesse intermédiaire. Le traçage antérograde de la voie corticospinale révèle un patron d’innervation normal dans le tronc cérébrale et la moelle épinière. Des microstimulations intracorticale du cortex moteur évoque des réponses électromyographiques dans les membres à un seuil et une latence plus élevé. Par contre, une stimulation de la voie corticospinale dans la médulla évoque des réponses électromyographies à un seuil et une latence similaire entre les deux groupes, suggérant une réduction de l’excitabilité du cortex moteur.

Background Motivation has been identified as an area of difficulty for children with Down syndrome. Although individual differences in mastery motivation are presumed to have implications for subsequent competence, few longitudinal studies have addressed the stability of motivation and the predictive validity of early measures for later academic achievement, especially in atypical populations. Method The participants were 25 children with Down syndrome. Mastery motivation, operationalised as persistence, was measured in early childhood and adolescence using tasks and parent report. At the older age, preference for challenge, another aspect of mastery motivation, was also measured and the children completed assessments of academic competence. Results There were significant concurrent correlations among measures of persistence at both ages, and early task persistence was associated with later persistence. Persistence in early childhood was related to academic competence in adolescence, even when the effects of cognitive ability at the younger age were controlled. Conclusions For children with Down syndrome, persistence appears to be an individual characteristic that is relatively stable from early childhood to early adolescence. The finding that early mastery motivation is significant for later achievement has important implications for the focus of early interventions.

Maternal behaviors and child mastery behaviors were examined in 25 children with Down syndrome and 43 typically developing children matched for mental age (24–36 months). During a shared problem-solving task, there were no group differences in maternal directiveness or support for autonomy, and mothers in the two groups used similar verbal strategies when helping their child. There were also no group differences in child mastery behaviors, measured as persistence with two optimally challenging tasks. However, the two groups differed in the relationships of maternal style with child persistence. Children with Down syndrome whose mothers were more supportive of their autonomy in the shared task displayed greater persistence when working independently on a challenging puzzle, while children of highly directive mothers displayed lower levels of persistence. For typically developing children, persistence was unrelated to maternal style, suggesting that mother behaviors may have different causes or consequences in the two groups.

Despite a lack of consistent empirical evidence, there has been an ongoing assumption that intellectual disability is associated with reduced levels of motivation. The participants in this study were 33 children with Down syndrome ages 10–15 years and 33 typically developing 3–8-year-old children. Motivation was measured through observational assessments of curiosity, preference for challenge, and persistence, as well as maternal reports. There were no significant group differences on motivation tasks, but mothers of children with Down syndrome rated their children significantly lower on motivation than did parents of typically developing children. There were some intriguing group differences in the pattern of correlations among observations and parent reports. The findings challenge long-held views that individuals with intellectual disability are invariably deficient in motivation.

Motivational deficits are generally accepted to be part of the behavioural phenotype associated with Down syndrome (DS). A motivational profile comprising low or inconsistent levels of task persistence, avoidance of challenging activities and over-dependence on adult direction has been described. However, comparisons are usually made between children with DS and those who are developing typically, without the inclusion of samples with intellectual disability (ID) from aetiologies other than DS. Such comparisons are needed to determine the extent to which motivational deficits are specific to DS, as opposed to being a feature of ID generally. Methods: The current study collected data about the personality-motivation profiles of children in three groups matched for mental age. They consisted of 80 typically developing (TD)3–7 year old children, 62 children with DS aged 7–15 years, and 54 children with moderate ID aged 7–15 years. Parents completed the 37-item EZ-Personality Questionnaire (EZPQ; Zigler et al., 2002), a measure of personality-motivational functioning. Results: There were significant differences between TD children and those with ID on all EZPQ scales. In most respects children with DS did not differ significantly from others with moderate ID, although they were rated as having greater expectancy of success and fewer negative reactions. Conclusion: The finding that children with DS are less motivated than TD children of the same mental age is consistent with previous studies in which parents have rated motivation. It seems, however, that motivation difficulties are associated with ID more generally, rather than being specific to those with DS. The study raises questions about phenotypic versus experiential effects on motivation for children with ID.
