La neurogenèse persiste à l’âge adulte dans deux régions du système nerveux central (SNC) des mammifères : la zone sous-ventriculaire (SVZ) du cerveau antérieur et la zone sous-granulaire (SGZ) de l’hippocampe. Cette neurogenèse est possible grâce à la capacité de prolifération des cellules souches présentes dans les niches de la SVZ et la SGZ, mais en vieillissant, le cerveau subit une diminution dramatique du nombre de cellules souches neurales adultes (CSNa), une diminution de la prolifération cellulaire et une altération des niches de neurogenèse. Cependant, une importante question reste sans réponse : comment la perte tardive des CSNa est temporellement reliée aux changements de l’activité de prolifération et de la structure de la principale niche de neurogenèse (la SVZ)? Afin d’avoir un aperçu sur les événements initiaux, nous avons examiné les changements des CSNa et de leur niche dans la SVZ entre le jeune âge et l’âge moyen. La niche de la SVZ des souris d’âge moyen (12 mois) subit une réduction de l’expression des marqueurs de plusieurs sous-populations de précurseurs neuraux en comparaison avec les souris jeunes adultes (2 mois). Anatomiquement, cela est associé avec des anomalies cytologiques, incluant une atrophie générale de la SVZ, une perte de la couche de cellules sousépendymaires par endroit et l’accumulation de gouttelettes lipidiques de grande taille dans l’épendyme. Fonctionnellement, ces changements sont corrélés avec une diminution de l’activité de la SVZ et une réduction du nombre de nouveaux neurones arrivant aux bulbes olfactifs. Pour déterminer si les CSNa de la SVZ ont subi des changements visibles, nous avons évalué les paramètres clés des CSNa in vivo et in vitro. La culture cellulaire montre qu’un nombre équivalent de CSNa ayant la capacité de former des neurosphères peut être isolé du cerveau du jeune adulte et d’âge moyen. Cependant, à l’âge moyen, les précurseurs neuraux semblent moins sensibles aux facteurs de croissance durant leur différenciation in vitro. Les CSNa donnent des signes de latence in vivo puisque leur capacité d’incorporation et de rétention du BrdU diminue. Ensemble, ces données démontrent que, tôt dans le processus du vieillissement, les CSNa et leur niche dans la SVZ subissent des changements significatifs, et suggèrent que la perte de CSNa liée au vieillissement est secondaire à ces événements.

The intracellular mechanisms that determine the response of neural progenitor cells to growth factors and regulate their differentiation into either neurons or astrocytes remain unclear. We found that expression of SOCS2, an intracellular regulator of cytokine signaling, was restricted to mouse progenitor cells and neurons in response to leukemia inhibitory factor (LIF)-like cytokines. Progenitors lacking SOCS2 produced fewer neurons and more astrocytes in vitro, and Socs2(-/-) mice had fewer neurons and neurogenin-1 (Ngn1)-expressing cells in the developing cortex, whereas overexpression of SOCS2 increased neuronal differentiation. We also report that growth hormone inhibited Ngn1 expression and neuronal production, and this action was blocked by SOCS2 overexpression. These findings indicate that SOCS2 promotes neuronal differentiation by blocking growth hormone-mediated downregulation of Ngn1.

Erosão dos solos em Cabo Verde: estudo dos processos e quantificação à escala de três bacias hidrográficas O arquipélago de Cabo Verde é constituído por 10 ilhas vulcânicas pertence à zona do Sahel que se estende do Atlântico ao Mar Vermelho. Desde então, várias décadas, Cabo Verde é afectado pela desertificação causada principalmente pela recessão climatica e a erosão do solo. Esses fatores, aliados à alta pressão humana sobre os recursos, a topografia acidentada e chuvas tropicais por vezes torrenciais, causam sérios danos aos solos. No entanto, desde sua independência em 1975, o Governo realizou um amplo programa de arborização, recuperação de áreas degradadas e a correcção dos leitos das ribeiras. No entanto, a investigação, muito pouco foi realizada para avaliar as acções de protecção e conservação do solo e da água. Portanto, não há dados sobre o problema da degradação das terras nem balanços. Como parte deste trabalho, foram estudados vários factores que controlam a erosão do solo pela água. Especificamente, buscou-se diferenciar os efeitos das actividades humanas, incluindo a agricultura, os factores climáticos, como chuva e geração de escoamento. Também estabeleceu os primeiros balanços das exportações de matérias em suspensão e em solução no contexto do arquipélago de Cabo Verde. O estudo foi realizado em três bacias hidrográficas da ilha de Santiago, Cabo Verde. Estas três bacias hidrográficas (Longueira, Grande e Godim) estão localizadas na parte central da ilha de Santiago e representam os diversos tipos de uso da terra e as diferentes zonas bioclimaticas da ilha. Existe um gradiente climático entre as três bacias hidrográficas. Na verdade, Longueira que abrange uma área de 4,18 km2, tem um declive médio de 47 %, uma zona florestada de 69% e uma área agrícola de 17 %. Grande com uma área de 1,87 km2, é localizada numa zona sub humida com um declive médio de 50%, é essencialmente agrícola. Godim, com uma área de 2,0 km2, é localizado numa zona semi-árida com um declive médio de 32%, é particularmente uma zona agricola. Para estes três bacias hidrográficas, as cheias foram medidas e amostradas de 2004 a 2009. A bacia de Longueira teve um maior acompanhamento, nomeadamente em termos de amostragem e monitoramento dos escoamentos. Em cada amostra foram feitas a determinação da concentração de matérias em suspensão e a análise dos principais elementos quimicos. Os resultados mostram que a erosão mecânica nas três bacias hidrográficas é caracterizada por uma forte variabilidade espacial e temporal. Durante o período de 2005-2009, o balanço anual média para as bacias hidrográficas de Longueira, Grande e Godim é: 4266, 157 e 10,1 t.km2.an-1, respectivamente. A estação das chuvas de 2006 foi a mais erosiva para as três bacias, particularmente em Longueira, com 2 cheias excepcionais, que têm gerado uma concentração média de sólidos em suspensão superior a 100 g / l. Porém, as estações do ano de 2005 e 2008 foram de uma forma geral menos erosivas porque as concentrações médias não inferiores a 20 g / l. Além disso, não houve cheias para as temporadas 2005 e 2007 para a bacia do Godim. Na bacia de Longueira, o estudo dos fenómenos de histerese na caracterização das cheias mostrou que a evolução temporal das exportações de sólidos em suspensão durante a temporada é fortemente influenciada pelas atividades agrícolas. Na verdade, a primeira cheia causou uma exportação maciça de sedimento disponível e localizado no leito da ribeira. Assim, a segunda cheia exportou menos sedimentos. Um mês após as primeiras chuvas, a prática da monda que reduz a densidade da cobertura vegetal e destructura a camada superficial do solo, gerou uma grande quantidade de sedimento que novamente permitiu uma exportação muito forte de sedimentos durante a terceira forte cheia. Os resultados da erosão química na bacia de Longueira indicam que a taxa de erosão é de 45 t.km2.an-1 com uma forte variabilidade temporal. Na verdade, as temporadas de 2006 e 2007 são as mais erosivas, enquanto 2005 teve uma exportação de matérias disolvidas baixa. A utilização do modelo EMMA (End- Members Mixing Analysis) mostra que os escoamentos hipodermico e profundo, alimentandos os fluxos de elementos dissolvidos são os principais factores da erosão química. É mostrado que esses fluxos causam mais de 90% dos fluxos de erosão química. O escoamento superficial, que contribui com cerca de 70% na formação das cheias, é o maior factor da erosão mecânica do solo.

L’archipel du Cap Vert constitué de 10 îles volcaniques appartient à la zone sahélienne qui s’étend de l’atlantique jusqu’à la mer rouge. Depuis, plusieurs décennies le Cap Vert est affecté par la désertification causée en grande partie par la récession climatique et l’érosion des sols. Ces facteurs, associés à la forte pression anthropique sur les ressources, à l’orographie accidentée et à des pluies tropicales parfois diluviennes, provoquent de sérieuses pertes du patrimoine foncier. Cependant, depuis son Indépendance en 1975, le Gouvernement a mené un vaste programme d’arborisation, de restauration des terres et d’aménagement des cours d’eau. Pourtant, très peu de recherches ont été menées pour évaluer les actions de protection et de conservation des sols et des eaux. Par conséquent, il n’existe quasiment pas de données sur la problématique de la dégradation des terres ni de bilans. Dans le cadre de ce travail, nous avons étudié les différents facteurs qui contrôlent l’érosion hydrique des sols. Nous avons plus particulièrement cherché à différencier les effets des activités humaines, notamment agricoles, de ceux des facteurs climatiques comme les précipitations et la génération des écoulements. Nous avons également établi les premiers bilans d’exportations de matières en suspension et en solution dans le contexte de l’archipel du Cap Vert. L’étude a été menée à l’échelle de trois bassins versants de l’ile de Santiago Cap-Vert. Ces trois bassins versant (Longueira, Grande et Godim) sont localisés dans la partie centrale de l’île de Santiago et représentatifs des divers modes d’occupation du sol et des différents climats de l’île. Il existe un gradient climatique entre les trois bassins versants. En effet, Longueira qui présente une superficie de 4,18 km2, une pente moyenne de 47 %, se localise dans une zone humide couverte à 69 % par une forêt et une surface agricole de 17 %. Grande avec une superficie de 1,87 km2, se localise en zone sub humide pour une pente moyenne de 50 %, il est essentiellement agricole. Godim, avec une superficie de 2,0 km2, se localise en zone semi aride, il est particulièrement agricole et sa pente moyenne est de 32 %. Pour ces trois bassins versants, les écoulements de crue à l’exutoire ont été mesurés et échantillonnés de 2004 à 2009. Le bassin versant de Longueira a fait l’objet d’un suivi plus poussé, notamment en termes de fréquence d’échantillonnage et de suivi des écoulements hors crue. Sur chaque échantillon nous avons procédé à la détermination de la concentration des matières en suspension ainsi qu’à l’analyse des éléments majeurs. Les résultats obtenus montrent que l’érosion mécanique dans les 3 bassins versants est caractérisée par une forte variabilité spatiale et temporelle. Sur la période 2005-2009, le bilan moyen annuel pour les bassins versants de Longueira, Grande et Godim est de : 4266, 157 et 10,1 t.km2.an-1 respectivement. La saison humide 2006 a été la plus érosive pour l’ensemble des trois bassins versants et particulièrement dans Longueira avec 2 crues exceptionnelles qui ont généré une concentration moyenne de matières en suspension supérieure à 100 g/l. En revanche, les saisons 2005 et 2008 ont été dans l’ensemble peu érosives car les concentrations moyennes ne dépassèrent pas 20 g/l. Par ailleurs, il n’y a pas eu de lames d’eau écoulées pour les saisons 2005 et 2007 pour le bassin de Godim. Sur le bassin de Longueira, l’étude des phénomènes d’hystérésis permet de caractériser chaque crue et de montrer que l’évolution temporelle des exportations de matières en suspension au cours de la saison est fortement influencée par les activités agricoles. En effet, la première crue provoque l’exportation massive des sédiments disponibles et localisés dans le lit du cours d’eau. En conséquence, la seconde est moins exportatrice de sédiments. Un mois après les premières pluies, les activités de sarclage diminuent la densité du couvert végétal et destructurent la partie superficielle des sols, ce qui provoque à nouveau une très forte exportation de sédiments lors de la troisième crue. Les résultats de l’érosion chimique sur le bassin de Longueira indiquent que le taux d’érosion chimique moyen s’élève à 45 t.km2.an-1 avec une forte variabilité temporelle. En effet, les saisons les plus humides de 2006 et 2007 sont les plus exportatrices de matières en solution, alors que 2005 a eu une faible exportation. L’utilisation du modèle de mélanges EMMA (End-Members Mixing Analysis) montre que les écoulements hypodermique et profond, qui alimentent le cours d’eau en éléments dissous, sont les principaux facteurs de l’érosion chimique. On montre ainsi que les écoulements hors crue sont à l’origine de plus de 90% des flux d’érosion chimique. L’écoulement superficiel, qui contribue à environ 70 % du débit du cours d’eau en crue, constitue un facteur de premier plan de l’érosion mécanique des sols.

The Jurassic (approximately 145 Ma) Nambija oxidized gold skarns are hosted by the Triassic volcanosedimentary Piuntza unit in the sub-Andean zone of southeastern Ecuador. The skarns consist dominantly of granditic garnet (Ad(20-98)) with subordinate pyroxene (Di(46-92)Hd(17-42)Jo(0-19)) and epidote and are spatially associated with porphyritic quartz-diorite to granodiorite intrusions. Endoskarn is developed at the intrusion margins and grades inwards into a potassic alteration zone. Exoskarn has an outer K- and Na-enriched zone in the volcanosedimentary unit. Gold mineralization is associated with the weakly developed retrograde alteration of the exoskarn and occurs mainly in sulfide-poor vugs and milky quartz veins and veinlets in association with hematite. Fluid inclusion data for the main part of the prograde stage indicate the coexistence of high-temperature (500A degrees C to > 600A degrees C), high-salinity (up to 65 wt.% eq. NaCl), and moderate- to low-salinity aqueous-carbonic fluids interpreted to have been trapped at pressures around 100-120 MPa, corresponding to about 4-km depth. Lower-temperature (510-300A degrees C) and moderate- to low-salinity (23-2 wt.% eq. NaCl) aqueous fluids are recorded in garnet and epidote of the end of the prograde stage. The microthermometric data (Th from 513A degrees C to 318A degrees C and salinity from 1.0 to 23 wt.% eq. NaCl) and delta(18)O values between 6.2aEuro degrees and 11.5aEuro degrees for gold-bearing milky quartz from the retrograde stage suggest that the ore-forming fluid was dominantly magmatic. Pressures during the early retrograde stage were in the range of 50-100 MPa, in line with the evidence for CO(2) effervescence and probable local boiling. The dominance of magmatic low-saline to moderately saline oxidizing fluids during the retrograde stage is consistent with the depth of the skarn system, which could have delayed the ingression of external fluids until relatively low temperatures were reached. The resulting low water-to-rock ratios explain the weak retrograde alteration and the compositional variability of chlorite, essentially controlled by host rock compositions. Gold was precipitated at this stage as a result of cooling and pH increase related to CO(2) effervescence, which both result in destabilization of gold-bearing chloride complexes. Significant ingression of external fluids took place after gold deposition only, as recorded by delta(18)O values of 0.4aEuro degrees to 6.2aEuro degrees for fluids depositing quartz (below 350A degrees C) in sulfide-rich barren veins. Low-temperature (< 300A degrees C) meteoric fluids (delta(18)O(water) between -10.0aEuro degrees and -2.0aEuro degrees) are responsible for the precipitation of late comb quartz and calcite in cavities and veins and indicate mixing with cooler fluids of higher salinities (about 100A degrees C and 25 wt.% eq. NaCl). The latter are similar to low-temperature fluids (202-74.5A degrees C) with delta(18)O values of -0.5aEuro degrees to 3.1aEuro degrees and salinities in the range of 21.1 to 17.3 wt.% eq. CaCl(2), trapped in calcite of late veins and interpreted as basinal brines. Nambija represents a deep equivalent of the oxidized gold skarn class, the presence of CO(2) in the fluids being partly a consequence of the relatively deep setting at about 4-km depth. As in other Au-bearing skarn deposits, not only the prograde stage but also the gold-precipitating retrograde stage is dominated by fluids of magmatic origin.

Mutations of the huntingtin protein (HTT) gene underlie both adult-onset and juvenile forms of Huntington's disease (HD). HTT modulates mitotic spindle orientation and cell fate in mouse cortical progenitors from the ventricular zone. Using human embryonic stem cells (hESC) characterized as carrying mutations associated with adult-onset disease during pre-implantation genetic diagnosis, we investigated the influence of human HTT and of an adult-onset HD mutation on mitotic spindle orientation in human neural stem cells (NSCs) derived from hESCs. The RNAi-mediated silencing of both HTT alleles in neural stem cells derived from hESCs disrupted spindle orientation and led to the mislocalization of dynein, the p150Glued subunit of dynactin and the large nuclear mitotic apparatus (NuMA) protein. We also investigated the effect of the adult-onset HD mutation on the role of HTT during spindle orientation in NSCs derived from HD-hESCs. By combining SNP-targeting allele-specific silencing and gain-of-function approaches, we showed that a 46-glutamine expansion in human HTT was sufficient for a dominant-negative effect on spindle orientation and changes in the distribution within the spindle pole and the cell cortex of dynein, p150Glued and NuMA in neural cells. Thus, neural derivatives of disease-specific human pluripotent stem cells constitute a relevant biological resource for exploring the impact of adult-onset HD mutations of the HTT gene on the division of neural progenitors, with potential applications in HD drug discovery targeting HTT-dynein-p150Glued complex interactions.

Neurons of the mammalian cerebral cortex comprise two broad classes: pyramidal neurons, which project to distant targets, and the inhibitory nonpyramidal cells, the cortical interneurons. Pyramidal neurons are generated in the germinal ventricular zone, which lines the lateral ventricles, and migrate along the processes of radial glial cells to their positions in the developing cortex in an `inside-out' sequence. The GABA-containing nonpyramidal cells originate for the most part in the ganglionic eminence, the primordium of the basal ganglia in the ventral telencephalon. These cells follow tangential migratory routes to enter the cortex and are in close association with the corticofugal axonal system. Once they enter the cortex, they move towards the ventricular zone, possibly to obtain positional information, before they migrate radially in the direction of the pial surface to take up their positions in the developing cortex. The mechanisms that guide interneurons throughout these long and complex migratory routes are currently under investigation.

We have performed a screen combining subtractive hybridization with PCR to isolate genes that are regulated when neuroepithelial (NE) cells differentiate into neurons. From this screen, we have isolated a number of known genes that have not previously been associated with neurogenesis, together with several novel genes. Here we report that one of these genes, encoding a guanine nucleotide exchange factor (GEF), is regulated during the differentiation of distinct neuronal populations. We have cloned both rat and mouse GEF genes and shown that they are orthologs of the human gene, MR-GEF, which encodes a GEF that specifically activates the small GTPase, Rap1. We have therefore named the rat gene rat mr-gef (rmr-gef) and the mouse gene mouse mr-gef (mmr-gef). Here, we will collectively refer to these two rodent genes as mr-gef. Expression studies show that mr-gef is expressed by young neurons of the developing rodent CNS but not by progenitor cells in the ventricular zone (VZ). The expression pattern of mr-gef during early telencephalic neurogenesis is strikingly similar to that of GABA and the LIM homeobox gene Lhx6, a transcription factor expressed by GABAergic interneurons generated in the ventral telencephalon, some of which migrate into the cortex during development. These observations suggest that mr-gef encodes a protein that is part of a signaling pathway involved in telencephalic neurogenesis; particularly in the development of GABAergic interneurons.

Gap junction (GJ) channels couple adjacent cells, allowing transfer of second messengers, ions, and molecules up to 1 kDa. These channels are composed by a multigene family of integral membrane proteins called connexins (Cx). In the retina, besides being essential circuit element in the visual processing, GJ channels also play important roles during its development. Herein, we analyzed Cx43, Cx45, Cx50, and Cx56 expression during chick retinal histogenesis. Cx exhibited distinct expression profiles during retinal development, except for Cx56, whose expression was not detected. Cx43 immunolabeling was observed at early development, in the transition of ventricular zone and pigmented epithelium. Later, Cx43 was seen in the outer plexiform and ganglion cell layers, and afterwards also in the inner plexiform layer. We observed remarkable changes in the phosphorylation status of this protein, which indicated modifications in functional properties of this Cx during retinal histogenesis. By contrast, Cx45 showed stable gene expression levels throughout development and ubiquitous immunoreactivity in progenitor cells. From later embryonic development, Cx45 was mainly observed in the inner retina, and it was expressed by glial cells and neurons. In turn, Cx50 was virtually absent in the chick retina at initial embryonic phases. Combination of PCR, immunohistochemistry and Western blot indicated that this Cx was present in differentiated cells, arising in parallel with the formation of the visual circuitry. Characterization of Cx expression in the developing chick retina indicated particular roles for these proteins and revealed similarities and differences when compared to other species. A zona subgranular (ZSG) do giro denteado (GD) de mamíferos adultos é conhecida por produzir constantemente novos neurônios. A busca por novas moléculas que possam modular a formação de novas células neurais são bastante atuais. Visto que a Amazônia é conhecida mundialmente pela sua biodiversidade, com um potencial pouco explorado de fármacos naturais derivados de plantas medicinais típicas da região. O trabalho buscou investigar o efeito neurogênico do extrato aquoso (EA) da Physalis angulata e da substância purificada Fisalina D sobre as células-tronco do GD do hipocampo de camundongos adultos. Os camundongos machos (BALB/c), 6 a 8 semanas de idade foram divididos em quatro grupos experimentais: controle e tratados com EA ou substância purificada. Os animais receberam diferentes doses do extrato (0,1; 1 e 5 mg/Kg) e/ou substância purificada (5mg/Kg) ou salina (grupo controle), 5 horas depois uma única dose de 5-Bromodeoxiuridina (BrdU) [50mg/kg]. Em seguida, os animais foram sacrificados 24 horas ou 7 dias após a administração do BrdU. Os cérebros foram coletados e cortes coronais do hipocampo (40 μm) foram realizados para contagem das células BrdU-positivas no GD hipocampal. Para avaliação estatística realizamos análise de variância (ANOVA) das médias amostrais seguida pelo pós-teste t de Student. O EA promoveu um aumento significativo do número de células BrdU positivas no GD dos grupos tratados em relação ao grupo controle [Controle, 92±24 (n=9); 0,1mg/Kg, 160±22 (n=4); 1mg/Kg, 310±5 (n=4); 5mg/Kg, 501±24 (n=3)] nos animais sacrificados 24 horas após administração do BrdU. Quando os animais foram sacrificados 7 dias após administração do BrdU, o número de células BrdU+ no GD também foi maior no grupo tratado em relação ao controle [Controle, 107±7 (n=4); 5mg/Kg, 145±23 (n=4)]. Usando a substância purificada, Fisalina D, também observamos um aumento do número de células BrdU+ no GD do grupo tratado com a droga em relação ao grupo controle [Controle, 92±24 (n=9); Fisalina D, 5mg/Kg, 316±37 (n=3)]. Este resultado sugere que o EA e a sustância purificada, na dose de 5 mg/Kg, estimulam a proliferação de células BrdU-positivas na ZSG do GD do hipocampo de camundongos adultos.

Für die Entwicklung des zerebralen Kortex ist die radiale Migration von Neuronen von elementarer Bedeutung. Für diese radiale Migration sind extrazelluläre Signale, die mit den Neuronen interagieren und eine Umgestaltung des Zytoskeletts vermitteln, notwendig. Zu den extrazellulären Signalen gehört auch der Neurotransmitter GABA, der über Depolarisation der Neurone einen Ca2+-Einstrom vermittelt und dadurch die Modulation der Migration über Ca2+-abhängige Signalwege ermöglicht. Auch von Taurin ist bekannt, dass es die neuronale Migration beeinflusst. Frühere Studien zeigten, dass die Depolarisation von GABAA-Rezeptoren durch GABA zu einem Migrationsstop führt, wohingegen Picrotoxin-sensitive Rezeptoren die Migration von der Ventrikulären Zone in die Intermediäre Zone des pränatalen Kortex vermitteln. Obwohl zu den Picrotoxin-sensitiven Rezeptoren GABAA-, GABAC- und bestimmte Glyzinrezeptoren gehören, wurde die Rolle von GABAC- und Glyzinrezeptoren während der radialen Migration nie überprüft. Ziel dieser Dissertation war deshalb, den Einfluss von GABAC- und Glyzinrezeptoren auf die radiale Migration zu untersuchen. Unter Verwendung von Migrationsanalysen, Fluoreszenzmessungen, molekularbiologischen und histologischen Methoden wurde gezeigt, dass GABAC-Rezeptoren im unteren Bereiche des präfrontalen Kortex exprimiert werden, ihre Aktivierung durch GABA in der Intermediären Zone zu einer Depolarisation führt, dass GABAC-Rezeptoren die Migration fördern und dieser Effekt über den migrationsstoppenden Effekt der GABAA-Rezeptoren dominiert. Durch Aktivierung der Glyzinrezeptoren fördert Taurin die Migration.

During the years 1984–1987 Lake Victoria in East Africa experienced what is probably the largest mass extinction of contemporary vertebrates. Within a decade about 200 endemic species of haplochromine cichlids disappeared. The extinctions that occurred in the 1980s have been documented predominantly on species of offshore and sub-littoral waters in the Mwanza Gulf of southern Lake Victoria. Although the littoral fauna of southern Lake Victoria had not been examined in detail, their diversity seemed less affected by the changes in the ecosystem. We give results of the first comprehensive inventory of the littoral cichlid fauna in southern Lake Victoria and discuss its conservation status. We also report on new developments in the sub-littoral fauna after 1990. More than 50 littoral and 15 sub-littoral stations were sampled between the years 1991 to 1995. Of the littoral stations, 34 were sampled for the first time. One hundred sixty three species of haplochromines were collected. Of these, 102 species were previously unknown. About two thirds of them live in rocky areas that were sampled for the first time. Littoral rocky habitats harbored the highest diversity. Since 1990, however, 13 more species disappeared from established sampling stations in littoral habitats. Fishing practices, spreading of exotic fishes, water hyacinth, and eutrophication are considered important threats to the littoral fauna. Also in the upper sub-littoral the number of species declined further. On deeper sub-littoral mud bottoms individual and species numbers increased again, although they are nowhere close to those found before the Nile perch (Lates niloticus) upsurge. This fauna differs from the well studied pre-Nile perch fauna. At well-established sampling stations, the sub-littoral zone is dominated by previously unknown species, and some known species have performed dramatic habitat shifts.

Radial Glia (RG) are a mitotically active population of cells which reside within the ventricular zone at the lateral ventricle and give rise to the pyramidal neurons and astrocytes of the neocortex. Through cellular divisions, RG produce two daughter cells, one which resides in the ventricular zone and becomes another RG while the other is an immature progenitor which migrates away from the ventricle and populates the growing cortex. RG have been found to be a heterogeneous population of cells which express different surface antigens and genetic promoters which may influence the cellular fate of their progeny. In this study we have investigated the progenitor profiles of two promoters, nestin (a neural intermediate filament) and GLAST (astrocyte specific glutamate transporter) within the RG. In-utero electroporation was used to transfect reporter plasmids under the control of promoter driven Cre-Recombinase into the RG lining the lateral ventricle during mid-neurogensesis (E14). It was found that there was a large amount of overlap between the nestin and GLAST expressing populations of RG, however, there was still a small subset of cells which exclusively expressed GLAST. This prompted us to investigate the lineage of these two promoters using the PiggyBac transposon system which uses promoter driven episomal plasmids to incorporate a reporter gene into the genome of the transfected cells, allowing use to trace their full progeny. Our data shows that nestin expressing RG generate mostly neurons and few astrocytes while the GLAST expressing RG generate a greater proportion of astrocytes to neurons.

The sub-Antarctic zone (SAZ) lies between the subtropical convergence (STC) and the sub-Antarctic front (SAF), and is considered one of the strongest oceanic sinks of atmospheric CO2. The strong sink results from high winds and seasonally low sea surface fugacities of CO2 (fCO2), relative to atmospheric fCO2. The region of the SAZ, and immediately south, is also subject to mode and intermediate water formation, yielding a penetration of anthropogenic CO2 below the mixed layer. A detailed analysis of continuous measurements made during the same season and year, February - March 1993, shows a coherent pattern of fCO2 distributions at the eastern (WOCE/SR3 at about 145°E) and western edges (WOCE/I6 at 30°E) of the Indian sector of the Southern Ocean. A strong CO2 sink develops in the Austral summer (delta fCO2 < - 50 µatm) in both the eastern (110°-150°E) and western regions (20°-90°E). The strong CO2 sink in summer is due to the formation of a shallow seasonal mixed-layer (about 100 m). The CO2 drawdown in the surface water is consistent with biologically mediated drawdown of carbon over summer. In austral winter, surface fCO2 is close to equilibrium with the atmosphere (delta fCO2 ± 5 µatm), and the net CO2 exchange is small compared to summer. The near-equilibrium values in winter are associated with the formation of deep winter mixed-layers (up to 700 m). For years 1992-95, the annual CO2 uptake for the Indian Ocean sector of the sub Antarctic Zone (40°-50°S, 20°-150°E) is estimated to be about 0.4 GtC/yr. Extrapolating this estimate to the entire sub-Antarctic zone suggests the uptake in the circumpolar SAZ is approaching 1 GtC/yr.

Extracellular lysophosphatidic acid (LPA) produces diverse cellular responses in many cell types. Recent reports of several molecularly distinct G protein-coupled receptors have raised the possibility that the responses to LPA stimulation could be mediated by the combination of several uni-functional receptors. To address this issue, we analyzed one receptor encoded by ventricular zone gene-1 (vzg-1) (also referred to as lpA1/edg-2) by using heterologous expression in a neuronal and nonneuronal cell line. VZG-1 expression was necessary and sufficient in mediating multiple effects of LPA: [3H]-LPA binding, G protein activation, stress fiber formation, neurite retraction, serum response element activation, and increased DNA synthesis. These results demonstrate that a single receptor, encoded by vzg-1, can activate multiple LPA-dependent responses in cells from distinct tissue lineages.
