In this study, the influence of tool rotation speed and feed rate on the forming limit of friction stir welded Al 6061-T651 sheets has been investigated. The forming limit curve was evaluated by limit dome height test performed on all the friction stir welded sheets. The welding trials were conducted at a tool rotation speed of 1300 and 1400 r/min and feed rate of 90 and 100 mm/min. A third trial of welding was performed at a rotational speed of 1500 r/min and feed rate 120 mm/min. It is found that with increase in the tool rotation speed, from 1300 to 1400 r/min, for a constant feed rate, the forming limit of friction stir welded blank has improved and with increase in feed rate, from 90 to 100 mm/min, for a constant tool rotation speed, it has decreased. The forming limit of friction stir welded sheets is better than unwelded sheets. The thickness gradient after forming is severe in the cases of friction stir welded blanks made at higher feed rate and lower rotation speed. The strain hardening exponent of weld (n) increases with increase in tool rotation speed and it decreases with increase in feed rate. It has been demonstrated that the change in the forming limit of friction stir welded sheets with respect to welding parameters is due to the thickness distribution severity and strain hardening exponent of the weld region during forming. There is not much variation in the dome height among the friction stir welded sheets tested. When compared with unwelded sheets, dome height of friction stir welded sheets is higher in near-plane-strain condition, but it is lesser in stretching strain paths.

The main aim of the present work is to analyze the influence of shoulder diameter and plunge depth on the formability of friction stir welded sheets. The base material used for welding and forming was AA6061-T6. Formability evaluation was performed through limiting dome height tests. The forming limit curve, FLC (only in the stretching region), thickness distribution, and strain hardening exponent of the weld region were monitored during formability studies. It is found from the work that the forming limit of friction stir welded sheets is better than unwelded sheets. In general, with an increase in shoulder diameter and plunge depth, the forming limit is found to improve considerably. With a decrease in thickness gradient severity and an increase in strain hardening exponent (n) of the weld region, the forming limit is found to increase. The increase in n value of the weld region is believed to occur because of the reduction in dislocation density. The maximum thickness difference is higher in the retreating side, rather than in the advancing side, of the weld. This is due to the differential straining and hardness levels attained by both sides during friction stir welding.

In this work, a combined forming and fracture limit diagram, fractured void coalescence and texture analysis have been experimentally evaluated for the commercially available aluminum alloy Al 8011 sheet annealed at different temperatures viz. 200 degrees C, 250 degrees C, 300 degrees C and 350 degrees C. The sheets were examined at different annealing temperatures on microstructure, tensile properties, formability and void coalescence. The fractured surfaces of the formed samples were examined using scanning electron microscope (SEM) and these images were correlated with fracture behavior and formability of sheet metals. Formability of Al 8011 was studied and examined at various annealing temperatures using their bulk X-ray crystallographic textures and ODF plots. Forming limit diagrams, void coalescence parameters and crystallographic textures were correlated with normal anisotropy of the sheet metals annealed at different temperatures. Histone deacetylase inhibitors (HDACi) are anti-cancer drugs that primarily act upon acetylation of histones, however they also increase levels of intracellular reactive oxygen species (ROS). We hypothesized that agents that cause oxidative stress might enhance the efficacy of HDACi. To test this hypothesis, we treated acute lymphocytic leukemia cells (ALL) with HDACi and adaphostin (ROS generating agent). The combination of two different HDACi (vorinostat or entinostat) with adaphostin synergistically induced apoptosis in ALL. This synergistic effect was blocked when cells were pre-treated with the caspase-9 inhibitor, LEHD. In addition, we showed that loss of the mitochondrial membrane potential is the earliest event observed starting at 12 h. Following this event, we observed increased levels of superoxide at 16 h, and ultimately caspase-3 activation. Pre-treatment with the antioxidant N-acetylcysteine (NAC) blocked ROS generation and reversed the loss of mitochondrial membrane potential for both combinations. Interestingly, DNA fragmentation and caspase-3 activity was only blocked by NAC in cells treated with vorinostat-adaphostin; but not with entinostat-adaphostin. These results suggest that different redox mechanisms are involved in the induction of ROS-mediated apoptosis. To further understand these events, we studied the role of the antioxidants glutathione (GSH) and thioredoxin (Trx). We found that the combination of entinostat-adaphostin induced acetylation of the antioxidant thioredoxin (Trx) and decreased intracellular levels of GSH. However, no effect on Trx activity was observed in either combination. In addition, pre-treatment with GSH ethyl ester, a soluble form of GSH, did not block DNA fragmentation. Together these results suggested that GSH and Trx are not major players in the induction of oxidative stress. Array data examining the expression of genes involved in oxidative stress demonstrated a differential regulation between cells treated with vorinostat-adaphostin and entinostat-adaphostin. Some of the genes differentially expressed between the combinations include aldehyde oxidase 1, glutathione peroxidase-5, -6, peroxiredoxin 6 and myeloperoxidase. Taken together, these experimental results indicate that the synergistic activity of two different HDACi with adaphostin is mediated by distinct redox mechanisms in ALL cells. Understanding the mechanism involved in these combinations will advance scientific knowledge of how the action of HDACi could be augmented in leukemia models. Moreover, this information could be used for the development of effective clinical trials combining HDACi with other anticancer agents.

Recent efforts to develop large-scale neural architectures have paid relatively little attention to the use of self-organizing maps (SOMs). Part of the reason is that most conventional SOMs use a static encoding representation: Each input is typically represented by the fixed activation of a single node in the map layer. This not only carries information in an inefficient and unreliable way that impedes building robust multi-SOM neural architectures, but it is also inconsistent with rhythmic oscillations in biological neural networks. Here I develop and study an alternative encoding scheme that instead uses limit cycle attractors of multi-focal activity patterns to represent input patterns/sequences. Such a fundamental change in representation raises several questions: Can this be done effectively and reliably? If so, will map formation still occur? What properties would limit cycle SOMs exhibit? Could multiple such SOMs interact effectively? Could robust architectures based on such SOMs be built for practical applications? The principal results of examining these questions are as follows. First, conditions are established for limit cycle attractors to emerge in a SOM through self-organization when encoding both static and temporal sequence inputs. It is found that under appropriate conditions a set of learned limit cycles are stable, unique, and preserve input relationships. In spite of the continually changing activity in a limit cycle SOM, map formation continues to occur reliably. Next, associations between limit cycles in different SOMs are learned. It is shown that limit cycles in one SOM can be successfully retrieved by another SOM’s limit cycle activity. Control timings can be set quite arbitrarily during both training and activation. Importantly, the learned associations generalize to new inputs that have never been seen during training. Finally, a complete neural architecture based on multiple limit cycle SOMs is presented for robotic arm control. This architecture combines open-loop and closed-loop methods to achieve high accuracy and fast movements through smooth trajectories. The architecture is robust in that disrupting or damaging the system in a variety of ways does not completely destroy the system. I conclude that limit cycle SOMs have great potentials for use in constructing robust neural architectures.

Esta tesis propone un sistema biométrico de geometría de mano orientado a entornos sin contacto junto con un sistema de detección de estrés capaz de decir qué grado de estrés tiene una determinada persona en base a señales fisiológicas Con respecto al sistema biométrico, esta tesis contribuye con el diseño y la implementación de un sistema biométrico de geometría de mano, donde la adquisición se realiza sin ningún tipo de contacto, y el patrón del usuario se crea considerando únicamente datos del propio individuo. Además, esta tesis propone un algoritmo de segmentación multiescala para solucionar los problemas que conlleva la adquisición de manos en entornos reales. Por otro lado, respecto a la extracción de características y su posterior comparación esta tesis tiene una contribución específica, proponiendo esquemas adecuados para llevar a cabo tales tareas con un coste computacional bajo pero con una alta precisión en el reconocimiento de personas. Por último, este sistema es evaluado acorde a la norma estándar ISO/IEC 19795 considerando seis bases de datos públicas. En relación al método de detección de estrés, esta tesis propone un sistema basado en dos señales fisiológicas, concretamente la tasa cardiaca y la conductancia de la piel, así como la creación de un innovador patrón de estrés que recoge el comportamiento de ambas señales bajo las situaciones de estrés y no-estrés. Además, este sistema está basado en lógica difusa para decidir el grado de estrés de un individuo. En general, este sistema es capaz de detectar estrés de forma precisa y en tiempo real, proporcionando una solución adecuada para sistemas biométricos actuales, donde la aplicación del sistema de detección de estrés es directa para evitar situaciónes donde los individuos sean forzados a proporcionar sus datos biométricos. Finalmente, esta tesis incluye un estudio de aceptabilidad del usuario, donde se evalúa cuál es la aceptación del usuario con respecto a la técnica biométrica propuesta por un total de 250 usuarios. Además se incluye un prototipo implementado en un dispositivo móvil y su evaluación. ABSTRACT: This thesis proposes a hand biometric system oriented to unconstrained and contactless scenarios together with a stress detection method able to elucidate to what extent an individual is under stress based on physiological signals. Concerning the biometric system, this thesis contributes with the design and implementation of a hand-based biometric system, where the acquisition is carried out without contact and the template is created only requiring information from a single individual. In addition, this thesis proposes an algorithm based on multiscale aggregation in order to tackle with the problem of segmentation in real unconstrained environments. Furthermore, feature extraction and matching are also a specific contributions of this thesis, providing adequate schemes to carry out both actions with low computational cost but with certain recognition accuracy. Finally, this system is evaluated according to international standard ISO/IEC 19795 considering six public databases. In relation to the stress detection method, this thesis proposes a system based on two physiological signals, namely heart rate and galvanic skin response, with the creation of an innovative stress detection template which gathers the behaviour of both physiological signals under both stressing and non-stressing situations. Besides, this system is based on fuzzy logic to elucidate the level of stress of an individual. As an overview, this system is able to detect stress accurately and in real-time, providing an adequate solution for current biometric systems, where the application of a stress detection system is direct to avoid situations where individuals are forced to provide the biometric data. Finally, this thesis includes a user acceptability evaluation, where the acceptance of the proposed biometric technique is assessed by a total of 250 individuals. In addition, this thesis includes a mobile implementation prototype and its evaluation.

Strain-based failure criteria have several advantages over stress-based failure criteria: they can account for elastic and inelastic strains, they utilise direct, observables effects instead of inferred effects (strain gauges vs. stress estimates), and model complete stress-strain curves including pre-peak, non-linear elasticity and post-peak strain weakening. In this study, a strain-based failure criterion derived from thermodynamic first principles utilising the concepts of continuum damage mechanics is presented. Furthermore, implementation of this failure criterion into a finite-element simulation is demonstrated and applied to the stability of underground mining coal pillars. In numerical studies, pillar strength is usually expressed in terms of critical stresses or stress-based failure criteria where scaling with pillar width and height is common. Previous publications have employed the finite-element method for pillar stability analysis using stress-based failure criterion such as Mohr-Coulomb and Hoek-Brown or stress-based scalar damage models. A novel constitutive material model, which takes into consideration anisotropy as well as elastic strain and damage as state variables has been developed and is presented in this paper. The damage threshold and its evolution are strain-controlled, and coupling of the state variables is achieved through the damage-induced degradation of the elasticity tensor. This material model is implemented into the finite-element software ABAQUS and can be applied to 3D problems. Initial results show that this new material model is capable of describing the non-linear behaviour of geomaterials commonly observed before peak strength is reached as well as post-peak strain softening. Furthermore, it is demonstrated that the model can account for directional dependency of failure behaviour (i.e. anisotropy) and has the potential to be expanded to environmental controls like temperature or moisture.

This work deals with the formulation and implementation of finite deformation viscoplasticity within the framework of stress-based hybrid finite element methods. Hybrid elements, which are based on a two-field variational formulation, are much less susceptible to locking than conventional displacement-based elements. The conventional return-mapping scheme cannot be used in the context of hybrid stress methods since the stress is known, and the strain and the internal plastic variables have to be recovered using this known stress field.We discuss the formulation and implementation of the consistent tangent tensor, and the return-mapping algorithm within the context of the hybrid method. We demonstrate the efficacy of the algorithm on a wide range of problems.

Existing soil nailing design methodologies are essentially based on limit equilibrium principles that together with a lumped factor of safety or a set of partial factors on the material parameters and loads account for uncertainties in design input parameter values. Recent trends in the development of design procedures for earth retaining structures are towards load and resistance factor design (LRFD). In the present study, a methodology for the use of LRFD in the context of soil-nail walls is proposed and a procedure to determine reliability-based load and resistance factors is illustrated for important strength limit states with reference to a 10 m high soil-nail wall. The need for separate partial factors for each limit state is highlighted, and the proposed factors are compared with those existing in the literature.

This paper deals with the influence of crystallographic texture on room temperature mechanical behavior of the sheets of the aluminum alloy AA7020 processed to different thicknesses. Three different thicknesses of the alloy sheet, namely 1, 1.85, and 3.6 mm, corresponding to different textures were investigated. Tensile tests were carried out at 0°, 45° and 90° with respect to sheet rolling direction and the resulting in-plane anisotropy in 0.2 proof stress, work hardening and plastic strain ratio (r-value) were determined. Texture derived r-values are also calculated and discussed vis-à -vis the experimentally obtained r-values. Finally the formability of the optimal alloy was studied using forming limit diagrams. Effect of natural aging, with a simulated heat treatment of 70 °C for 2 h on FLD was studied and compared with the as solutionized samples. It was observed that, the strain levels in the bi-axial region of the FLD were not much affected by the heat treatment. © 2012 Elsevier B.V. All rights reserved.

L’insuline est une hormone essentielle qui induit des réponses complexes dans l’organisme pour maintenir l’homéostasie du glucose et des lipides. La résistance à son action est un phénomène pathologique observé dans un large éventail de situations, allant de l’obésité et du syndrome métabolique à la stéatose hépatique et au diabète de type 2, qui aboutissent au développement de l’athérosclérose et de la mortalité. Des avancées remarquables ont été réalisées dans notre compréhension des mécanismes moléculaires responsables du développement de la résistance à l’action de l’insuline. En particulier, l’induction d’un stress cellulaire par des taux élevés d’acides gras libres (AGL) et des cytokines, via l’activation des protéines Ser/Thr kinases, qui augmente la phosphorylation sur des résidus sérine, des molécules critiques impliquées dans la signalisation insulinique (p. ex. IR, IRS et p85) et conduit à la diminution de la réponse cellulaire à l’insuline. Cependant, la plupart des chercheurs ont limité leur travail dans l’investigation du rôle des protéines kinases susceptibles de modifier la réponse cellulaire à l’insuline. Donc, peu de données sont disponibles sur le rôle des Protéines Ser/Thr phosphatases (PS/TPs), même si il est bien établi que la phosphorylation de ces protéines est étroitement régulée par un équilibre entre les activités antagonistes des Ser/Thr kinases et des PS/TPs. Parmi les PS/TPS, PPM1A (également connu sous le nom PP2Cα) est une phosphatase particulièrement intéressante puisqu’il a été suggéré qu’elle pourrait jouer un rôle dans la régulation du métabolisme lipidique et du stress cellulaire. Ainsi, en se basant sur des résultats préliminaires de notre laboratoire et des données de la littérature, nous avons émis l’hypothèse selon laquelle PPM1A pourrait améliorer la sensibilité à l’insuline en diminuant l’activité des protéines kinases qui seraient activées par le stress cellulaire induit par l’augmentation des AGL. Ces effets pourraient finalement améliorer le métabolisme glucidique et lipidique dans l’hépatocyte. Ainsi, pour révéler le rôle physiologique de PPM1A à l’échelle d’un animal entier, nous avons généré un modèle animal qui la surexprime spécifiquement dans le foie. Nous décrivons ici notre travail afin de générer ce modèle animal ainsi que les premières analyses pour caractériser le phénotype de celui-ci. Tout d’abord, nous avons remarqué que la surexpression de PPM1A chez les souris C57BL/6J n’a pas d’effets sur le gain de poids sur une longue période. Deuxièmement, nous avons observé que PPM1A a peu d’effets sur l’homéostasie du glucose. Par contre, nous avons montré que sa surexpression a des effets significatifs sur l’homéostasie du glycogène et des triglycérides. En effet, nous avons observé que le foie des souris transgéniques contient moins de glycogène et de triglycérides que le foie de celles de type sauvage. De plus, nos résultats suggèrent que les effets de la surexpression de PPM1A pourraient refléter son impact sur la synthèse et la sécrétion des lipides hépatiques puisque nous avons observé que sa surexpression conduit à l’augmentation la triglycéridémie chez les souris transgéniques. En conclusion, nos résultats prouvent l’importance de PPM1A comme modulateur de l’homéostasie hépatique du glucose et des lipides. Des analyses supplémentaires restent cependant nécessaires pour confirmer ceux-ci et éclaircir l’impact moléculaire de PPM1A et surtout pour identifier ses substrats.

L’adaptation des cellules à leur environnement externe repose sur la transduction adéquate de signaux régulés par une pléthore d'événements moléculaires. Parmi ces événements moléculaires, les modifications post-traductionnelles (MPT) de protéines aident à intégrer, à traduire et à organiser de façon spatiotemporelle ces signaux pour que les cellules puissent réagir aux stimuli externes. Parmi les modifications post-traductionnelles, les petites protéines de la famille de l’Ubiquitine (Ublps, Ubiquitin-like proteins) jouent un rôle majeur dans presque toutes les voies de signalisation. Cette thèse rapporte des études fonctionnelles et structurales des interactions covalentes et non covalentes entre SUMO (Small Ubiquitin related MOdifier), un membre de la famille des Ublps, et trois protéines d'échafaudage, TIF1beta, le corépresseur universel des protéines KRAB-multidoigt de zinc, PIAS1, une ligase E3 pour SUMO et PML, un suppresseur de tumeur. La première étude rapporte l'identification et la caractérisation biochimique des sites de SUMOylation de TIF1beta. Nous avons déterminé que la modification covalente de six résidus lysine par SUMO est essentielle à l’activité de répression de la transcription induit par TIF1beta. En outre, nous présentons des évidences indiquant que la SUMOylation de TIF1 exige non seulement sa capacité à homo-oligomériser, mais est aussi positivement régulée par son interaction avec le domaine KRAB des protéines à doigts de zinc. Partant de ce constat, nous postulons que les protéines KRAB-multidoigt de zinc recrutent leur corépresseur TIF1betaà des gènes cibles, mais aussi accentuent son activité répressive grâce à l'augmentation de sa SUMOylation. Notre seconde étude révèle qu’en plus de réprimer la transcription en tant que MPT covalente, SUMO joue aussi un rôle important dans la répression en tant que partenaire non covalent d’interactions protéine-protéine. Nous avons montré que SUMO interagit simultanément avec deux enzymes de la machinerie de SUMOylation, l’unique enzyme de conjugaison E2, UBC9, et la ligase E3 PIAS1 au sein d’un complexe ternaire répresseur. En outre, nous révélons que la formation du complexe ternaire PIAS1:SUMO:UBC9 est modulée par le niveau de phosphorylation de résidus sérine juxtaposés à un motif d’interaction avec SUMO (SIM) dans PIAS1. Ainsi, SUMO agit comme un adaptateur spécifique qui stabilise les interactions UBC9 E2: E3 PIAS1. Partant de ce constat, nous proposons que les enzymes E2 et E3 des autres systèmes Ublps exploitent des mécanismes similaires dans le cadre de leur fonction Enfin, notre troisième étude explore la régulation des interactions non covalentes de SUMO par la phosphorylation. En utilisant une combinaison d'études in vivo et in vitro, nous démontrons que l'interaction entre SUMO1 et PML est régi par la phosphorylation dépendant de CK2 sur quatre résidus sérine de PML. Les structures cristallographiques des complexes PML-SIM:SUMO1 révèlent que les phospho-sérines de PML contactent des résidus de la région basique de SUMO1. Sachant que la kinase CK2 peut être induite par des kinases activables par le stress, ces résultats suggèrent que les interactions non-covalentes avec SUMO sont modulées par le stress cellulaire. Sur la base de cette constatation, nous postulons que des événements analogues affectent des protéines contenant des séquences SIM ciblées par CK2. En résumé, cette étude révèle qu’en plus de son rôle de MPT, SUMO peut fonctionner comme un adaptateur permettant des interactions spécifiques entre protéines tel que pour les enzymes E3 et E2.

The aims of this work are: (i) to produce new experimental data for fretting fatigue considering the presence of a mean bulk stress and (ii) to assess two design methodologies against failure by fretting fatigue. Tests on a cylinder–flat contact configuration were conducted using a fretting apparatus mounted on a servo-hydraulic machine. The material used for both the pads and fatigue specimen was an aeronautical 7050-T7451 Al alloy. The experimental program was designed with all relevant parameters, apart from the mean bulk load (always applied before the contact loads), kept constant. The mean bulk stress varied from compressive to tensile values while maintaining a high peak pressure in order to encourage crack initiation. Two methodologies against fretting fatigue are proposed and confronted against the experimental data. The non-local stress-based methodology considers the evaluation of a critical plane fatigue criterion at the center of a process zone located beneath the contacting surfaces. The results showed that it correctly predicts crack initiation, but was not capable to provide successful prediction of the integrity of the specimens. Alternatively, we considered a crack arrest criterion which has the potential to provide a more complete description about the integrity of the specimens.
