Duchenne Muscular Dystrophy (DMD) is a fatal multi-system neuromuscular disease caused by loss of dystrophin. The loss of dystrophin from membranes of contractile muscle cells and the dysregulation of the DAPC, induces chronic inflammation due to tissue necrosis and eventual replacement with collagen which weakens muscular force and strength. Dystrophin deficiency may cause under-diagnosed features of DMD include mood disorders such as depression and anxiety and dysfunction of the gastrointestinal tract. The first study in the thesis examined mood in the dystrophin-deficient mdx mouse model of DMD and examined the effects of the tri-cyclic antidepressant, amitriptyline on behaviours. Amitriptyline had anti-depressant and anxiolytic effects in the mdx mice possibly through effects on stress factors such as corticotrophin-releasing factor (CRF). This antidepressant also reduced skeletal muscle inflammation and caused a reduction in circulating interleukin (IL)-6 levels. In the second and third studies, we specifically blocked IL-6 signalling and used Urocortin 2, CRFR2 agonist to investigate their potential as therapeutic targets in mdx mice pathophysiology. Isometric and isotonic contractile properties of the diaphragm, were compared in mdx mice treated with anti IL-6 receptor antibodies (anti IL-6R) and/or Urocortin 2. Deficits in force production, work and power detected in mdx mice were improved with treatment. In study three I investigated contractile properties in gastrointestinal smooth muscle. As compared to wild type mice, mdx mice had slower faecal transit times, shorter colons with thickened muscle layers and increased contractile activity in response to recombinant IL-6. Blocking IL-6 signalling resulted in an increase in colon length, normalised faecal output times and a reduction in IL-6-evoked contractile activity. The findings from these studies indicate that for both diaphragm and gastrointestinal function in a dystrophin-deficient model, targeting of IL-6 and CRFR2 signalling has beneficial therapeutic effects.

Aims. This paper is a report of a study examining the association between ownership type and perceived team climate among older people care staff. In addition, we examined whether work stress factors (time pressure, resident-related stress, role conflicts and role ambiguity) mediated or moderated the above mentioned association. Background. There has been a trend towards contracting out in older people care facilities in Finland and the number of private for-profit firms has increased. Studies suggest that there may be differences in employee well-being and quality of care according to the ownership type of older people care. Methods. Cross-sectional survey data was collected during the autumn of 2007 from 1084 Finnish female older people care staff aged 1869 years were used. Team Climate Inventory was used to measure team climate. Ownership type was divided into four categories: for-profit sheltered homes, not-for-profit sheltered homes, public sheltered homes and not-for-profit nursing homes. Analyses of covariance were used to examine the associations. Results. Team climate dimensions participative safety, vision and support for innovation were higher in not-for-profit organizations (both sheltered homes and nursing homes) compared to for-profit sheltered homes and public sheltered homes. Stress factors did not account for these associations but acted as moderators in a way that in terms of task orientation and participative safety employees working in for-profit organizations seemed to be slightly more sensitive to work-related stress than others. Conclusion. Our results suggest that for-profit organizations and public organizations may have difficulties in maintaining their team climate. In consequence, these organizations should focus more effort on improving their team climate.

The prlmaiy objective of this study was to Identify and describe the physical and psychological stress factors that elementary school teachers experience and how teachers cope with stress. A secondary objective was to offer boards and teachers potential coping strategies counteracting stress and the effects of stress. The sample consisted of 120 elementaiy teachers from southern Ontario. Ten elementaiy schools were randomly chosen. The Teacher Stress Inventory questionnaire (Flmian, 1989) was used. Data were analyzed using a variety of statistics. Test norms and interpretations were performed based on standard results obtained from the author of the questionnaire (Flmian, 1988). Overall, the results indicated that work-related stressors were the main factor for teacher stress. This Included such factors as caseload/class is too big, too much administrative paperwork, and having little time to prepare lessons. Implications for further research and practical suggestions for further reseairch are discussed. Also a variety of recommendations to boards and for individual use are discussed. Some recommendations are having counselling available for teachers, workshops on how to handle stress, and learning how to breathe and using calm visualization.

This study explores whether the high variability of vascular plant diversity among alpine plant communities can be explained by stress and/or disturbance intensities. Species numbers of 14 alpine plant communities were sampled in the Swiss Alps. To quantify the intensity of 13 stress and 6 disturbance factors potentially controlling plant life in these communities, a survey was conducted by asking numerous specialists in alpine vegetation to assess the importance of the different factors for each community. The estimated values were combined in stress- and disturbance-indices which were compared with diversity according to the Intermediate Stress Hypothesis, the Intermediate Disturbance Hypothesis, and the Dynamic Equilibrium Model, respectively. Each of these theories explained a part of the variability in the species richness, but only the Dynamic Equilibrium Model provided a complete and consistent explanation. The last model suggests that community species richness within the alpine life zone is generally controlled by stress intensity. Disturbance and competition seem to play a secondary role by fine-tuning diversity in specific communities. As diversity is primarily limited by stress, a moderation of temperature-related stress factors, as a result of global warming, may cause a shift of the equilibrium between stress, disturbance, and competition in alpine ecosystems.

Adolescence is a time of physical, social and emotional development, and this development can be accompanied by feelings of stress. The Adolescent Stress Questionnaire is a 56-item scale measuring stress in 10 domains. Developed in Australia, the scale has been translated, and its reliability and validity have been tested in a number of countries across Europe, where the 10-factor, 56-item version of the scale has received little support. The present study tested the factor structure, construct validity and reliability in a sample (n=610) of adolescents in the United Kingdom. Support was found for the 10-factor, 56-item version of the scale, and correlations with self-concept measures, sex scores on stress factors and Cronbach's α-values, suggesting that the scale may be a viable assessment tool for adolescent stress. Results for alcohol-specific analyses support the domain-specific nature of the scale. Future work may seek to investigate the stability of age-specific stress domains (e.g. the stress of Emerging Adult Responsibility) in samples that include younger adolescents.

A formação inicial de professores possui caraterísticas muito próprias e a complexidade envolta sobre o período de estágio na prática pedagógica, requer um olhar aprofundado sobre as questões subjacentes ao stress tanto no estagiário, como no supervisor e orientador. A presente investigação inter e multicultural, pretende conhecer junto a estes três atores as principais fontes indutoras de stress e seus sintomas no decorrer do período de estágio pedagógico, as estratégias de coping e a satisfação com o suporte social. Desenvolvemos três estudos de cariz comparativo entre Portugal e Brasil, em oito Instituições de Ensino Superior (4 de Portugal e 4 do Brasil) numa amostra total de 1.194 indivíduos. O primeiro de índole qualitativa envolveu 24 professores supervisores e orientadores de estágio, os quais identificaram as principais causas de stress que permitiram a construção do questionário “Avaliação do stress do professor/orientador de Estágio Pedagógico” (ASPOEP). Contamos no segundo estudo com uma amostra de 204 docentes (122 orientadores e 82 supervisores) no exercício da supervisão pedagógica e no terceiro estudo com 966 estagiários (476 Portugal e 490 Brasil). Nestes dois últimos estudos, para a identificação dos fatores de stress, os supervisores responderam ao questionário ASPOEP e os estagiários ao questionário ASAE (Avaliação do stress do aluno estagiário). Foram ainda utilizados os questionários de “Avaliação dos sintomas de stress”, o de “Estratégias de coping” e para a análise do suporte social fizemos uso da “Escala de satisfação com o suporte social”. Na análise dos dados foi utilizado o SPSS, tendo sido realizadas as estatísticas descritivas e inferenciais. Os resultados globais evidenciam o estágio pedagógico como uma fonte indutora de stress. Nos supervisores de estágio, a sobrecarga de trabalho, as relações interpessoais, o desempenho do estagiário e sobretudo a estrutura e acompanhamento do estágio, marcaram os fatores de maior stress, que foi mais evidente no gênero feminino e na população brasileira. Entre estagiários, a insegurança e a indisciplina são os fatores de maior stress, sobretudo no gênero feminino de Portugal. O stress apesar de moderado para a maioria da amostra, evidencia índices de stress elevado em 10,7% dos docentes e 19,1% dos estagiários portugueses e 23,8% dos docentes e 12,7% dos estagiários brasileiros. Os sintomas cognitivos são os mais evidentes nestas duas populações estudadas, porém nos casados e com filhos existe evidência significativa de sintoma comportamental. A estratégia de coping centrada no indivíduo foi mais prevalente, porém evidencia-se que os brasileiros e também os pais com filhos apresentam significativo coping centrado no outro. O suporte social da amizade e da intimidade foram os que demonstraram maior satisfação, sendo assim eficaz na manutenção de um stress moderado. Os portugueses apresentam maior satisfação com o suporte social que os brasileiros. Ficou evidente que a satisfação e as estratégias de coping contribuem de forma positiva na diminuição dos sintomas de stress, bem como a satisfação com estágio diminui os níveis de stress. Sugere-se que na formação de professores sejam integrados programas de formação sobre gestão e controlo do stress para supervisor, orientador e estagiário, bem como devem ser propostas mudanças a nível curricular, ligado à estrutura e acompanhamento do estágio, para que este possa ser um período de desenvolvimento de competências profissionais, alicerçadas no bem-estar para todos os intervenientes.

According to climate change predictions, water availability might change dramatically in Europe and adjacent regions. This change will undoubtedly have an adverse effect on existing tree species and affect their ability to cope with a lack or an excess of water, changes in annual precipitation patterns, soil salinity and fire disturbance. The following chapter will describe tree species and proven-ances used in European forestry practice which are the most suitable to deal with water stress, salinity and fire. Each subchapter starts with a brief description of each of the stress factors and discusses the predictions of the likelihood of their occurrence in the near future according to the climate change scenarios. Tree spe-cies and their genotypes able to cope with particular stress factor, together with indication of their use by forest managers are then introduced in greater detail.

Data on fertilisation and embryo quality in dairy cattle are presented and the main factors responsible for the low fertility of single-ovulating lactating cows and embryo yield in superovulated dairy cattle are highlighted. During the past 50 years, the fertility in high-producing lactating dairy cattle has decreased as milk production increased. Recent data show conception rates to first service to be approximately 32% in lactating cows, whereas in heifers it has remained above 50%. Fertilisation does not seem to be the principal factor responsible for the low fertility in single-ovulating cows, because it has remained above 80%. Conversely, early embryonic development is impaired in high-producing dairy cows, as observed by most embryonic losses occurring during the first week after fertilisation. However, in superovulated dairy cattle, although fertilisation failure is more pronounced, averaging approximately 45%, the percentage of fertilised embryos viable at 1 week is quite high (>70%). Among the multifactorial causes of low fertility in lactating dairy cows, high feed intake associated with low concentrations of circulating steroids may contribute substantially to reduced embryo quality. Fertilisation failure in superovulated cattle may be a consequence of inappropriate gamete transport due to hormonal imbalances.

Mesembryanthemum crystallinum L. (Aizoaceae) is a facultative annual halophyte and a C-3-photosynthesis/crassulacean acid metabolism intermediate species currently used as a model plant in stress physiology. Both salinity and high light irradiance stress are known to induce CAM in this species. The present study was performed to provide a diagnosis of alterations at the photosystem 11 level during salinity and irradiance stress. Plants were subjected for up to 13 days to either 0.4M NaCl salinity or high irradiance of 1000 mu mol m(-2) s(-1), as well as to both stress factors combined (LLSA = low light plus salt; HLCO = high light of 1000 mu mol m(-2)s(-1), no salt; HLSA = high light plus salt). A control of LLCO = low light of 200 mu mol m(-2) s(-1), no salt was used. Parameters of chlorophyll a fluorescence of photosystem 11 (PSII) were measured with a pulse amplitude modulated fluorometer. HLCO and LLSA conditions induced a weak degree of CAM with day/night changes of malate levels (Delta malate) of similar to 12 mM in the course of the experiment, while HLSA induced stronger CAM of Delta malate similar to 20mM. Effective quantum yield of PSII, Delta F/F'(m), was only slightly affected by LLSA, somewhat reduced during the course of the experiment by HLCO and clearly reduced by HLSA. Potential quantum efficiency of PSII, F-v/F-m, at predawn times was not affected by any of the conditions, always remaining at >= 0.8, showing that there was no acute photoinhibition. During the course of the days HL alone (HLCO) also did not elicit photoinhibition; salt alone (LLSA) caused acute photoinhibition which was amplified by the combination of the two stresses (HLSA). Non-photochemical, NPQ, quenching remained low (< 0.5) under LLCO, LLSA and HLCO and increased during the course of the experiment under HLSA to 1-2. Maximum apparent photosynthetic electron transport rates, ETRmax, declined during the daily courses and were reduced by LLSA and to a similar extent by HLSA. It is concluded that A crystallinum expresses effective stress tolerance mechanisms but photosynthetic capacity is reduced by the synergistic effects of salinity and tight irradiance stress combined. The impact of simultaneous environmental stresses on plants and how they respond to combined stresses compared with single stresses is largely unclear. By using a transgene (RD29A-LUC) consisting of the firefly luciferase coding sequence (LUC) driven by the stress-responsive RD29A promoter, we investigated the interactive effects of temperature, osmotic stress, and the phytohormone abscisic acid (ABA) in the regulation of gene expression in Arabidopsis seedlings. Results indicated that both positive and negative interactions exist among the studied stress factors in regulating gene expression. At a normal growth temperature (22°C), osmotic stress and ABA act synergistically to induce the transgene expression. Low temperature inhibits the response to osmotic stress or to combined treatment of osmotic stress and ABA, whereas low temperature and ABA treatments are additive in inducing transgene expression. Although high temperature alone does not activate the transgene, it significantly amplifies the effects of ABA and osmotic stress. The effect of multiple stresses in the regulation of RD29A-LUC expression in signal transduction mutants was also studied. The results are discussed in the context of cold and osmotic stress signal transduction pathways.

Sjuksköterskor som arbetar på akutmottagningar utsätts regelbundet för stress. Det medför psykiska och fysiska besvär för sjuksköterskor och försämrad omvårdnad för patienten. Studien avser att utifrån vetenskaplig litteratur sammanställa de faktorer som leder till stress för sjuksköterskor som arbetar på akutmottagningar och hur vården påverkas av stressade sjuksköteskor. Syfte Syftet med litteraturstudien var att beskriva vilka faktorer på akutmottagningen som upplevdes bidra till stress för sjuksköterskor som arbetar på en akutmottagning och hur stressen påverkar sjuksköterskors möjligheter att erbjuda en god vård för patienter på akutmottagningen. Metod Studien har genomförts som en litteraturstudie. I studien användes 15 artiklar som bestod av både kvalitativ och kvantitativ ansats. Materialet hämtades i databaserna CINAHL och PubMed. Resultat Resultatet visar att det finns flera olika faktorer som bidrar till stress för sjuksköterskor på akutmottagningen. Stressande faktorer visade sig utifrån studierna i resultatet vara; hög arbetsbelastning och låg bemanning, avsaknad av tid för reflektion för sjuksköterskan, att vårda barn i stressade situationer, hot och våld på akutmottagningen, kommunikation samt smärta och lidande. Hur vården påverkas av sjuksköterskors stress på akutmottagningen var utifrån studierna att patientsäkerheten blev försämrad och att vårdrelationen påverkades. Slutsats Författarna drar slutsatsen att stress påverkar sjuksköteskors arbetsuppgifter på akutmottagningen. Patienters möjlighet till patientsäker och god vård på akutmottagningen påverkas negativt av stressade sjuksköterskor. För att komma till rätta med sjuksköterskebristen på akutmottagningar så är det av betydelse för sjukhusledningen att arbeta preventivt mot stressade sjuksköterskor.

O stress é a doença da actualidade, devido à exigência e à pressão emocional que o meio pode exercer sobre todas as pessoas, quer no seu ambiente pessoal quer no ambiente profissional. O termo stress serve cada vez mais como justificação ou forma de expressão de irritações, mal-estar físico e mental, cansaço, ansiedade e depressões. Todavia torna-se pertinente percebermos a evolução do stress enquanto conceito e enquanto doença. Neste trabalho contextualizamos o stress no âmbito da Emergência Pré – Hospitalar, explorando-o, avaliando-o e analisando-o, de modo a podermos perceber os níveis de stress e de que forma os Profissionais de Emergência a ele estão sujeitos. Considerando a base teórica deste estudo definimos, como objectivo geral do trabalho, a avaliação de níveis e factores de stress percepcionados pelos profissionais de emergência médica e como objectivos específicos do estudo a descrever e caracterizar, a nível da região centro, os indivíduos que desempenham funções na emergência préhospitalar, verificar a percepção dos mesmos no que respeita ao stress na sua vida profissional, conhecer como percepcionam os níveis de stress em situações profissionais de emergência e identificar a sua percepção no que concerne aos níveis de stress diário no seu quotidiano. Com estes objectivos, utilizamos uma amostra de 184 profissionais de Emergência Médica divididos por quatro profissões [Médicos, Enfermeiros, Técnicos de Ambulância de Emergência (TAE) e Operadores de Central de Emergência Médica(OPCEM)], com idades compreendidas entre os 20 e os 56 anos. Realizámos ainda um estudo descritivo explorando os questionários de stress no INEM (Instituto Nacional de Emergência Médica) e o Questionário de Vulnerabilidade do Stress com variáveis do estudo demográfico. Podemos concluir que o stress está presente nas actividades de emergência. A sua presença relaciona-se, por um lado, com uma maior ou menor discriminação dos factores stressantes e, por outro, numa maior ou menor centração no sujeito, nomeadamente, um maior perfeccionismo e intolerância à frustração que se apresentam como distressantes para a actividade profissional de emergência. Os aspectos estudados permitem-nos clarificar o sentido e a utilidade do conceito, a sua operacionalidade e a sua importância nos profissionais de emergência. / Stress is the disease of our time, due to the requirement and emotional pressure that the media can have on all people, both in their personal environment and in the professional environment. The word stress increasingly serves as a justification or form of expression of irritation, physical discomfort and mental fatigue, anxiety and depression. However it is pertinent to realize the evolution of stress as a concept and as a disease. In this work, we contextualize under the stress of the Pre - Hospital Emergency, exploring it, assessing it and analyzing it, so you can see the stress levels and how the Emergency Professionals are subject to it. Whereas the theoretical basis of this study as a strategic objective of the work, the assessment of levels and stress factors perceived by medical emergency professionals and specific objectives of the study description and characterization, the level of the center, individuals who perform functions in pre-hospital emergency, check their perception with regard to the stress in your life, know how they experience the stress levels of professionals in emergency situations and identify the perceptions of individuals regarding the levels of daily stress in their daily lives. For this we used a sample of 184 professionals of Medical Emergency divided by four professions [(Doctors, Nurses, Technicians Ambulance Emergency (TAE), Operators of Emergency Medical Center (OPCEM)], aged 20 to 56 years. We conducted a descriptive study also exploring the questionnaires of stress in INEM(National Institute of Medical Emergency) and Vulnerability Questionnaire Stress with variables of the study population. We can conclude that stress is present in emergency activities. Its presence is related to, first, with more or less stressful factors of discrimination and, secondly, a greater or lesser concentration on the subject, in particular, greater perfectionism and intolerance to frustration that stand as unstressed for the activity of emergency professionals. The areas studied will clarify the meaning and usefulness of the concept, its operation and its importance in day-to-day occupation of emergency.
