The diversity and beneficial characteristics of endophytic microorganisms have been studied in several host plants. However, information regal-ding naturally, occurring seed-associated endophytes and vertical transmission among different life-history stages of hosts is limited. Endophytic bacteria were isolated from seeds and seedlings of 10 Eucalyptus species and two hybrids. The results showed that endophytic bacteria, Such as Bacillus, Enterococcus, Paenibacillus and Methylobacterium, are vertically transferred from seeds to seedlings. In addition, the endophytic bacterium Pantoea agglomerans was tagged with the gfp gene, inoculated into seeds and further reisolated from seedlings. These results suggested it novel approach to change the profile of the plants, where the bacterium is a delivery vehicle for desired traits. This is the first report of an endophytic bacterial community residing in Eucalyptus seeds and the transmission of these bacteria from seeds to seedlings. The bacterial species reported ill this work have been described as providing benefits to host plants. Therefore, we Suggest that endophytic bacteria can be transmitted vertically from seeds to seedlings, assuring the support of the bacterial community in the host plant.

(Morpho-anatomical studies of seeds and seedlings of wild indigo, ""anileira"", Indigofera-Leguminosae). The common name ""wild indigo"" specifies Indigofera will L., I. suffruticosa Mill. (legitimate name) and I. truxillensis Kunth (legitimate name) that are very similar due to their external morphology. This work analyzed diagnostic characteristics of seeds and seedlings of these species since such features are widely used in taxonomic approaches within Leguminosae. We studied surface features and morpho-anatomy of seeds and cotyledons with scanning electronic microscopy and light microscopy, and described seedling phases. Although seedlings are similar, seed characteristics (size, shape, surface ornamentation, shape of the hilum and embryo size) and cotyledon characteristics (shape, trichome ornamentation, organization of spongy parenchyma along central veins and metabolite distribution in the tissues) have diagnostic features for the species. I. anil differs from I. suffruticosa in its larger seeds and acuminate-margined cotyledons. I. truxillensis is recognized by its cylindrical seeds and kidney-shaped cotyledons with large oil drops. We assume that the characteristics examined, plus fruit morphology and foliar anatomy suggest that I. anil, I. suffruticosa and I. truxillensis should not be synonymized.

SFTI-1 is a recently discovered cyclic peptide trypsin inhibitor from sunflower seeds comprising 14 amino acid residues. It is the most potent known Bowman-Birk inhibitor and the only naturally occurring cyclic one. The solution structure of SFTI-1 has been determined by H-1-NMR spectroscopy and compared with a synthetic acyclic permutant. The solution structures of both are remarkably similar. The lowest energy structures from each family of 20 structures of cyclic and acyclic SFTI-1 have an rmsd over the backbone and heavy atoms of 0.29 Angstrom and 0.66 Angstrom, respectively. The structures consist of two short antiparallel beta -strands joined by an extended loop containing the active site at one end. Cyclic SFTI-1 also has a hairpin turn completing the cycle. Both molecules contain particularly stable arrangements of cross-linking hydrogen bonds between the beta -strands and a single disulfide bridge, making them rigid and well defined in solution. These stable arrangements allow both the cyclic and acyclic variants of SFTI-1 to inhibit trypsin with very high potencies (0.5 nM and 12.1 nM, respectively). The cyclic nature of SFTI-1 appears to have evolved to provide higher trypsin inhibition as well as higher stability. The solution structures are similar to the crystal structure of the cyclic inhibitor in complex with trypsin. The lack of a major conformational change upon binding suggests that the structure of SFTI-1 is rigid and already pre-organized for maximal binding due to minimization of entropic losses compared to a more flexible ligand. These properties make SFTI-1 an ideal platform for the design of small peptidic pharmaceuticals or pesticides. (C) 2001 Academic Press.

The Euterpe oleracea Mart. (açaí) is a plant from the Amazon region, classified as "super fruit" because of its various functional properties. However, limited investigation has been performed on açaí by-products, such as seeds. Therefore, the aim of this work was to characterized the phenolic compounds of the aqueous extract of açaí seeds and further evaluate its bioactivity (antioxidant and cytotoxic activities. Only proanthocyanidins were detected, being a B-type (epi)catechin tetramer the most abundant; however, procyanidin trimmers were the most predominant form. Açaí seeds extract revealed a high antioxidant (EC50 ranging from 3.6 to 19.4 μg/mL) and cytotoxic activity, being more effective in the cervical carcinoma cell line (HeLa; GI50 = 18 μg/mL); it did not show toxicity for non-tumor cells. Açaí seeds are considered a waste and could have an added economic benefit, through the extraction of natural antioxidants, particularly proanthocyanidins, that could find applications in food and pharmaceutical industries.

Vertebrates show different tendencies in regard to their preference for seeds or fruits infested by insects compared to non-infested ones. Behaviour may include rejection of one type, preferential consumption of one type or no differentiation among them. When comparing infested versus non-infested fruits, most studies have focused on energy content and nutritional components of the food items; but the energy input provided to the consumer is a better measure for the comparison of the value of each type of food. In this study, I calculated the energy assimilated by rodents for the seeds of the palm Attalea butyracea contained in non-infested endocarps and from bruchid beetle larvae contained in infested endocarps. Using the energy assimilation and time of handling by rodents for both types of endocarps, I quantitatively demonstrated that both infested and non-infested endocarps produce a similar energy input. This finding is consistent with the previous hypothesis that there is a trade-off between the energy content and the time required to extract the insect larvae compared with the seeds in endocarps of Attalea butyracea.

The experiment was carried out at the Embrapa Semi-Árido, Petrolina-PE, Brazil, in order to study the physiological responses of umbu plants propagated by seeds and by stem cuttings under water stress conditions, based on leaf water potential and gas exchange measurements. Data were collected in one-year plants established in pots containing 30 kg of a sandy soil and submitted to twenty-day progressive soil water deficit. The evaluations were based on leaf water potential and gas exchange data collection using psychrometric chambers and a portable infra-red gas analyzer, respectively. Plants propagated by seeds maintained a significantly higher water potential, stomatal conductance, transpiration and photosynthesis under decreasing soil water availability. However, plants propagated by stem cuttings were unable to maintain a favorable internal water balance, reflecting negatively on stomatal conductance and leaf gas exchange. This fact is probably because umbu plants propagated by stem cuttings are not prone to formation of root tubers which are reservoirs for water and solutes. Thus, the establishing of umbu plants propagated by stem cuttings must be avoided in areas subjected to soil water deficit.

This study was carried out at the site named Pedregulho in the "Estação Experimental de Itirapina", Itirapina, state of São Paulo, Brazil. Soil samples were collected in the rainy and in the dry season at three depths in the gallery forest, cerrado and cerradão. The total number of seeds was counted after being separated from the soil by wet sieving and the number of viable seeds was determined by emergence of seedlings in soil samples in a greenhouse. The number of seeds and of emerged seedlings decreases as soil samples become deeper. The number of seeds was much larger in the cerrado in the dry season than in the rainy season, but it was lower in the dry than in the rainy season in the gallery forest. The number of seeds in the cerradão was similar in both seasons. There was a heavy discrepancy between the total number of seeds and that of emerged seedlings.

The present work examined the germination and morphology of the fruits and seeds of the cactus Pilosocereus gounellei (xique-xique) and describeb the different stages of seedling growth. Germination tests examined the effects of combinations of two illumination regimes (a 12 hour photoperiod and total darkness) and three temperature regimes (25, 30, and 20-30 ºC) on the seeds of this species. Germination counts were made daily until the 16th day after sowing and the germination percentage and velocity index, and average germination time were evaluated. Descriptions of seed morphology considered both their external and internal aspects. The processes of germination and seedling growth were evaluated during 150 days and eight developmental stages were identified based on morphological changes. The fruits of the xique-xique cactus are 40.67 ± 4.40 mm long and 48.09 ± 3.23 mm in diameter, weigh 53.85 ± 10.03 g, and contain 3713 ± 689.50 seeds. Alternating temperatures of 20-30 ºC together with a photoperiod of 12 hours of light resulted in the highest seed germination rate. The seeds are positively photoblastic and germination is epigeal and fanerocotyledonary. Seedling growth is slow, but at 150 days after sowing the plants had developed epicotyls, large numbers of thorns, and the whole plant averaged 59.56 mm in height.

Iron bioavailability was evaluated in three mixtures of cereals, seeds, and grains ("Human Ration"): light, regular, and homemade provided to rats. The animals received an iron depletion diet for 21 days, followed by a repletion diet containing 12 mg·kg-1 of iron for 14 days. The hemoglobin regeneration efficiency and the relative biological value did not differ between the light mixture and control group. The iron bioavailability of the light mixture of cereals, seeds, and grains and the control group were 99.99±27.62 and 80.02±36.63, respectively, while the regular and homemade mixtures of cereals, seeds, and grains showed lower iron bioavailability, 50.12±35.53 and 66.66±15.44, respectively; the iron content of the diet with light cereal mixture light was statistically similar to that of the control (ferrous sulfate 99.99±27.62). The high content of tannin (202.81±19.53 mg·100-1) in the diet with the regular cereal mixture may have contributed to its low iron bioavailability. The higher intake of soluble fiber by the animals fed the light mixture (21.15±0.92 g) was moderately correlated (r=0.5712, p=0.0018) with the concentration of propionate in the caecal bulk (65.49±11.08 µmol/g). The short chain fatty acids produced by soluble fiber fermentation, associated with the low-content of tannin may have improved iron solubility and absorption in the light cereal mixture diet. The iron bioavailability in the light mixture of cereals, seeds, and grains was similar to that of ferrous sulfate.

Abstract Germinated grains have been known as sources of Gamma-aminobutyric acid (GABA) that provide beneficial effects for human health. This study was aimed to investigate GABA production, dietary fiber, antioxidant activity, and the effect of cooking on GABA loss in germinated legumes and sesame. The highest GABA content was found in germinated mung bean, (0.8068 g kg-1, 24 h incubation) followed by germinated soybean, germinated black bean and soaked sesame. Beside GABA, dietary fiber content also increased in all grains during germination where the insoluble dietary fiber fractions were always found in higher proportions to soluble dietary fiber fractions. Our results also confirmed that germinated mung bean is a rich source of GABA and dietary fibers. Microwave cooking resulted in the smallest loss of GABA in mung bean and sesame, while steaming led to the least GABA content loss in soybean and black bean. Therefore microwave cooking and steaming are the most recommended cooking processes to preserve GABA in germinated legumes and sesame.

A utilização das popularmente chamadas, sementes crioulas de soja ou sementes salvas pelos produtores é uma constante no Centro-Oeste Brasileiro. Este trabalho teve o objetivo de avaliar a qualidade das sementes de soja produzidas, colhidas e armazenadas por uma empresa produtora de sementes, e das sementes utilizadas pelos produtores de grãos, produzidas sem seguir os procedimentos técnicos de produção de sementes. As parcelas conduzidas para produção de sementes comerciais, foram retiradas do campo logo que atingiram a maturação de campo com umidade de 16,3%, receberam quatro aplicações de inseticidas durante o seu ciclo, para o controle de percevejos e, foram colhidas com colhedora de fluxo axial com rotação de cilindro de 500 rpm, enquanto as outras parcelas foram retiradas do campo com umidades de 14,7 e 12,3%, receberam apenas três aplicações de inseticida e, foram colhidas com colhedora de cilindro côncavo com velocidade de rotação do cilindro de 700 rpm. Após a colheita foram utilizados diferentes processos de secagem, beneficiamento e armazenamento para as sementes comerciais e para as sementes salvas. No início e no final do armazenamento, todas as unidades experimentais foram analisadas com os seguintes testes: de germinação, de tetrazólio 1-8 , do pH do exsudato, de envelhecimento acelerado, de desenvolvimento de plântula, de tetrazólio 1-3 e de 1º contagem do teste de germinação. Os resultados permitem concluir que: sementes comerciais, produzidas de acordo com as recomendações técnicas de produção de sementes, apresentam qualidade fisiológica superior; a qualidade fisiológica de materiais conduzidos como grãos é reduzida com o armazenamento, impedindo sua utilização como sementes para formação de lavouras de grãos comerciais.
