Almost everyone sketches. People use sketches day in and day out in many different and heterogeneous fields, to share their thoughts and clarify ambiguous interpretations, for example. The media used to sketch varies from analog tools like flipcharts to digital tools like smartboards. Whereas analog tools are usually affected by insufficient editing capabilities like cut/copy/paste, digital tools greatly support these scenarios. Digital tools can be grouped into informal and formal tools. Informal tools can be understood as simple drawing environments, whereas formal tools offer sophisticated support to create, optimize and validate diagrams of a certain application domain. Most digital formal tools force users to stick to a concrete syntax and editing workflow, limiting the user’s creativity. For that reason, a lot of people first sketch their ideas using the flexibility of analog or digital informal tools. Subsequently, the sketch is "portrayed" in an appropriate digital formal tool. This work presents Scribble, a highly configurable and extensible sketching framework which allows to dynamically inject sketching features into existing graphical diagram editors, based on Eclipse GEF. This allows to combine the flexibility of informal tools with the power of formal tools without any effort. No additional code is required to augment a GEF editor with sophisticated sketching features. Scribble recognizes drawn elements as well as handwritten text and automatically generates the corresponding domain elements. A local training data library is created dynamically by incrementally learning shapes, drawn by the user. Training data can be shared with others using the WebScribble web application which has been created as part of this work.

Adherens junctions (AJs) and cell polarity complexes are key players in the establishment and maintenance of apical-basal cell polarity. Loss of AJs or basolateral polarity components promotes tumor formation and metastasis. Recent studies in vertebrate models show that loss of AJs or loss of the basolateral component Scribble (Scrib) cause deregulation of the Hippo tumor suppressor pathway and hyperactivation of its downstream effectors Yes-associated protein (YAP) and Transcriptional coactivator with PDZ-binding motif (TAZ). However, whether AJs and Scrib act through the same or independent mechanisms to regulate Hippo pathway activity is not known. Here, we dissect how disruption of AJs or loss of basolateral components affect the activity of the Drosophila YAP homolog Yorkie (Yki) during imaginal disc development. Surprisingly, disruption of AJs and loss of basolateral proteins produced very different effects on Yki activity. Yki activity was cell-autonomously decreased but non-cell-autonomously elevated in tissues where the AJ components E-cadherin (E-cad) or α-catenin (α-cat) were knocked down. In contrast, scrib knockdown caused a predominantly cell-autonomous activation of Yki. Moreover, disruption of AJs or basolateral proteins had different effects on cell polarity and tissue size. Simultaneous knockdown of α-cat and scrib induced both cell-autonomous and non-cell-autonomous Yki activity. In mammalian cells, knockdown of E-cad or α-cat caused nuclear accumulation and activation of YAP without overt effects on Scrib localization and vice versa. Therefore, our results indicate the existence of multiple, genetically separable inputs from AJs and cell polarity complexes into Yki/YAP regulation.

Les anomalies du tube neural (ATN), incluant l'anencéphalie et le spina-bifida, représentent un groupe de malformations congénitales très fréquentes chez l'homme. Ces anomalies sont causées par un défaut partiel ou complet de la fermeture du tube neurale au cours de l'embryogenèse. Les ATN ont une étiologie complexe et multifactorielle impliquant des facteurs environnementaux et génétiques. La voie de signalisation non-canonique du Frizzled (Fz)/Dishevelled (Dvl) contrôle la polarité cellulaire planaire (PCP) et le processus morphogénétique appelé l’extension convergente qui est essentiel pour la gastrulation et la fermeture du tube neural. Très important, des mutations des gènes de cette voie étaient fortement associées aux ATN chez la souris et l’humain. Scribble est un gène de la voie PCP qui cause une sévère ATN chez la souris Circletail. Notre étude vise à analyser le rôle de SCRIBBLE1 dans les ATN humains par des analyses de séquence de son cadre de lecture et ses jonctions exon-introns. Notre étude comporte 396 patients recrutés au Centre Spina Bifida de l’hôpital Gaslini en Gènes, Italie et 83 patients recrutés au Centre Spina Bifida de l’hôpital Sainte Justine. Les patients sont affectés par plusieurs formes d’ATN. Nous avons identifié neuf mutations rares et non synonymes chez 10 patients, p.Asp93Ala (c. 435G>A), p.Gly145Arg (c. 278A>C), p.Gly263Ser (c. 786C>A), p.Gly469Ser (c. 1405G>A), p.Pro649His (c. 1946C>A), p.Gln808His (c. 2424G>T), p.Val1066Met (c. 3196G>A), p.Arg1150Gln (c. 3480G>A) et p.Thr1422Met (c. 4266C>T). Cinque mutations, p.Gly263Ser, p.Pro649His, p.Gln808His, p.Arg1150Gln, p.Thr1422Met, étaient absentes dans les contrôles analysés et prédites d’être pathogéniques in silico. Cette étude montre que des mutations rares dans SCRIB1 pourraient augmenter le risque des ATN dans une fraction des patients. L’identification des gènes prédisposant aux ATN nous aidera à mieux comprendre les mécanismes pathogéniques impliqués dans ces maladies.

Esta dissertação é o resultado de uma pesquisa que teve como objetivo analisar como um grupo de ribeirinhos, moradores da ilha João Pilatos (Ananindeua - PA), relaciona os conhecimentos ministrados no curso de formação para empreendedores rurais com os conhecimentos que possui da tradição ribeirinha e, em especial, os conhecimentos matemáticos. A parte empírica dessa pesquisa aconteceu em dois lugares e em momentos diferentes: o primeiro deles ocorreu na ilha João Pilatos, onde foram coletadas informações sobre o histórico das comunidades existentes na ilha, sobre os afazeres do cotidiano e sobre as práticas de pesca, de plantio, de coleta na floresta, de preparação do carvão e sobre a comercialização desses produtos. Nesse momento, os únicos recursos utilizados foram caderno e caneta para anotações e a técnica da escuta e do diálogo. O segundo momento ocorreu no curso de formação para empreendedores rurais ministrado pelo SENAR (Serviço Nacional de Aprendizagem Rural). Nesse local, minha participação no grupo estava restrita a escutar e olhar os gestos e expressões faciais de todos os presentes. Em função disso, a observação seguida de anotações foram as técnicas utilizadas durante esse período. Após o curso, foram realizadas entrevistas semi-estruturadas com os sujeitos dessa pesquisa com o objetivo de complementar as anotações feitas durante o curso. A Sociologia e a Etnomatemática foram as bases teóricas utilizadas na organização e análise das informações de campo. No campo da sociologia, busquei Antony Giddens, Renato Ortiz e Georges Balandier para analisar o fenômeno da globalização econômica e social sobre a cultura ribeirinha e Berger & Luckmann com a sua teoria "A construção social da realidade" para analisar o cotidiano e a produção de conhecimento na vida cotidiana. No campo da Etnomatemática, utilizei a Etnomatemática numa abordagem D'Ambrosiana e as pesquisas da área. Ao final das análises, no que diz respeito à matemática, percebi que ter domínio sobre os cálculos e fórmulas dessa área do conhecimento não era condição suficiente para que os sujeitos da pesquisa alcançassem o objetivo do curso, uma vez que tanto para os empreendedores quanto para os ribeirinhos a matemática era uma forma de ver e explicar o mundo que tinha significados específicos de cada contexto. A partir desse conflito cultural, é possível indicar que os sujeitos da pesquisa atingiriam o objetivo do curso e estabeleceriam o maior número de relações entre a matemática escolar e os saberes da tradição se a matemática estudada, no curso, fosse ressignificada no contexto ribeirinho. Como consequência da primeira análise, também é possível concluir que, mediante a concepção atualmente existente no ambiente escolar que separa e não apenas distingue os valores culturais, que considera a produção de conhecimentos proveniente apenas de ambientes acadêmicos científicos e que esses tipos de conhecimentos devem ser socializados de forma disciplinar, as possibilidades de um maior número de relações entre os saberes da tradição cultural e da tradição científica têm mais chances de ocorrer em ambientes não escolares.

Defects in apical-basal cell polarity and abnormal expression of cell polarity determinants are linked to human cancer. Loss of polarity is highly correlated with malignancy. In Drosophila, perturbation of apical-basal polarity, including overexpressing the apical determinant Crumbs, can lead to uncontrolled tissue growth. Cells mutant for the basolateral determinant scribble overproliferate and can form neoplastic tumors. Interestingly, scribble mutant clones that arise in wild-type tissues are eliminated and therefore do not manifest their tumorigenic potential. However, the mechanisms by which cell polarity coordinates with growth control pathways in developing organs to achieve appropriate organ size remain obscure. To investigate the function of apical determinants in growth regulation, I investigated the mechanism by which the apical determinant Crumbs affects growth in Drosophila imaginal discs. I found that crumbs gain and loss of function cause overgrowth and induction of Hippo target genes. In addition, Crumbs is required for the proper localization of Expanded, an upstream component of the Hippo pathway. Furthermore, we uncoupled the cell polarity and growth control function of Crb through structure-functional analysis. Taken together, our data identify a role of Crb in growth regulation specifically through modulation of the Hippo pathway. To further explore the role of polarity in growth control, I investigated how cells mutant for basolateral determinants are eliminated by using patches of cells mutant for scribble (scribble mutant clones) as a model system. We found that competitive cell-cell interactions eliminate tumorigenic scribble cells by modulation of the Hippo pathway. The regulation of Hippo signaling is required and sufficient to restrain the tumorous growth of scribble mutant cells. Artificially increasing the relative fitness of scribble mutant cells unleashes their tumorigenic potential. Therefore, we have identified a novel tumor-suppression mechanism that depends on signaling between normal and tumorigenic cells. These data identify evasion of cell competition as a critical step toward malignancy and illustrate a role for wild-type tissue in eliminating abnormal cells and preventing the formation of tumors.
