The rice blast fungus, Magnaporthe grisea, generates enormous turgor pressure within a specialized cell called the appressorium to breach the surface of host plant cells. Here, we show that a mitogen-activated protein kinase, Mps1, is essential for appressorium penetration. Mps1 is 85% similar to yeast Slt2 mitogen-activated protein kinase and can rescue the thermosensitive growth of slt2 null mutants. The mps1–1Δ mutants of M. grisea have some phenotypes in common with slt2 mutants of yeast, including sensitivity to cell-wall-digesting enzymes, but display additional phenotypes, including reduced sporulation and fertility. Interestingly, mps1–1Δ mutants are completely nonpathogenic because of the inability of appressoria to penetrate plant cell surfaces, suggesting that penetration requires remodeling of the appressorium wall through an Mps1-dependent signaling pathway. Although mps1–1Δ mutants are unable to cause disease, they are able to trigger early plant-cell defense responses, including the accumulation of autofluorescent compounds and the rearrangement of the actin cytoskeleton. We conclude that MPS1 is essential for pathogen penetration; however, penetration is not required for induction of some plant defense responses.

A short interspersed nuclear element, Mg-SINE, was isolated and characterized from the genome of the rice blast fungus, Magnaporthe grisea. Mg-SINE was isolated as an insertion element within Pot2, an inverted-repeat transposon from M. grisea and shows typical features of a mammalian SINE. Mg-SINE is present as a 0.47-kb interspersed sequence at approximately 100 copies per haploid genome in both rice and non-rice isolates of M. grisea, indicating a common evolutionary origin. Secondary structure analysis of Mg-SINE revealed a tRNA-related region at the 5' end which folds into a cloverleaf structure. Genomic fusions resulting in chimeric Mg-SINEs (Ch-SINEs) composed of a sequence homologous to Mg-SINE at the 3' end and an unrelated sequence at its 5' end were also isolated, indicating that this and other DNA rearrangements mediated by these elements may have a major effect on the genomic architecture of this fungus.

The objective of this study was to report single season effects of wood biochar (char) application coupled with N fertilization on soil chemical properties, aerobic rice growth and grain yield in a clayey Rhodic Ferralsol in the Brazilian Savannah. Char application effected an increase in soil pH, K, Ca, Mg, CEC, Mn and nitrate while decreasing Al content and potential acidity of soils. No distinct effect of char application on grain yield of aerobic rice was observed. We believe that soil properties impacted by char application were inconsequential for rice yields because neither water, low pH, nor the availability of K or P were limiting factors for rice production. Rate of char above 16 Mg ha^(−1) reduced leaf area index and total shoot dry matter by 72 days after sowing. The number of panicles infected by rice blast decreased with increasing char rate. Increased dry matter beyond the remobilization capacity of the crop, and high number of panicles infected by rice blast were the likely cause of the lower grain yield observed when more than 60 kg N ha^(−1) was applied. The optimal rate of N was 46 kg ha^(−1) and resulted in a rice grain yield above 3 Mg ha^(−1).

We describe in this study punchless, a nonpathogenic mutant from the rice blast fungus M. grisea, obtained by plasmid-mediated insertional mutagenesis. As do most fungal plant pathogens, M. grisea differentiates an infection structure specialized for host penetration called the appressorium. We show that punchless differentiates appressoria that fail to breach either the leaf epidermis or artificial membranes such as cellophane. Cytological analysis of punchless appressoria shows that they have a cellular structure, turgor, and glycogen content similar to those of wild type before penetration, but that they are unable to differentiate penetration pegs. The inactivated gene, PLS1, encodes a putative integral membrane protein of 225 aa (Pls1p). A functional Pls1p-green fluorescent protein fusion protein was detected only in appressoria and was localized in plasma membranes and vacuoles. Pls1p is structurally related to the tetraspanin family. In animals, these proteins are components of membrane signaling complexes controlling cell differentiation, motility, and adhesion. We conclude that PLS1 controls an appressorial function essential for the penetration of the fungus into host leaves.

During surveys of wild and cultivated rice in northern Queensland in 2014 and 2015, 92 fungal isolates were obtained from plants that were afflicted by foliar diseases, including the rice blast pathogen, Pyricularia oryzae, and the brown spot pathogen, Bipolaris oryzae. Seven species of Curvularia were found, viz. Curvularia aeria, C. alcornii, C. asianensis, C. clavata, C. lunata, C. muehlenbeckiae and an undescribed species. To remove uncertainty about the identity of the host plants from which the fungi were isolated, a DNA barcoding strategy was developed using regions of the chloroplast genome. Pathogenicity tests using wild rice isolates of P. oryzae indicated that many local rice varieties are susceptible to infection.

过氧化氢(Hydrogen peroxide,H2O2)是植物和病原微生物互作中快速合成的一种早期活性氧类(reactive oxygen species, ROS ),它在植物受到病原微生物侵染后引发的一系列防御反应中起着非常重要的作用,因此通过外源基因导入提高植物体内过氧化氢的含量,可以增强植物的广谱抗病性。葡萄糖氧化酶(glucose oxidase, GO)可以催化β-D-葡萄糖氧化生成过氧化氢和葡萄糖酸,此酶已在数种细菌和真菌中检测到,但在植物和动物中仍未发现。为了尝试将此酶应用于水稻广谱抗病基因工程,本研究将葡萄糖氧化酶基因插入具有潮霉素抗性选择标记的双元载体pCAMBIA1301,新构建为水稻高效表达载体pCAG1301。将此质粒导入根癌农杆菌(Agrobacterium tumefaciens )菌株LBA4404后,转化粳稻(Oryza sativa )品种日本晴(Nipponbare)成熟胚来源的愈伤组织和幼胚,并由筛选出的潮霉素抗性愈伤组织分化再生植株。对所得到的潮霉素抗性植株的Southern杂交分析表明GO基因已整合到受体基因组,为单拷贝或双拷贝插入。利用过氧化氢与淀粉-碘化钾反应显蓝色的特性检测到了转基因植株产生的过氧化氢,证实GO基因表达产生的葡萄糖氧化酶已经在水稻中发挥功能,这是将GO基因转入单子叶植物的首例报道。 基于过氧化氢诱导的植物防御反应没有种属专一性的优点,可以预期所得转基因水稻植株很可能对水稻的多种病原菌具有良好的抗性。已完成的抗病性鉴定表明,所得转基因水稻植株对稻瘟病具有良好的抗性。

Das aus wissenschaftlicher und ökonomischer Sicht wichtigste Pflanzenpathogen M. oryzae entwickelte im Laufe der Evolution konservierte aber auch einzigartige Mechanismen zur Signaltransduktion. Das Erforschen dieser Mechanismen und Prozesse ist essenziell für das Verständnis von Differenzierungsprozessen bei der Pathogen-Wirt-Interaktion.rnIm ersten Teil der vorliegenden Arbeit wurde der Signalweg zur Osmoregulation, der „High Osmolarity Glycerol“ (HOG)-Signalweg, erstmals anhand physiologischer Experimente in entsprechenden Mutantenstämmen in M. oryzae untersucht. Dabei konnten klare Unter-schiede zum HOG-Signalweg von S. cerevisiae aufgezeigt werden. rnDas in M. oryzae bisher noch nicht beschriebene Gen MoYPD1, welches das Phosphotransferprotein MoYpd1p kodiert, wurde erfolgreich inaktiviert. Diese Inaktivierung ist in S. cerevisiae und vielen anderen Pilzen letal und resultierte bei M. oryzae in einer apathoge¬nen Albinomutante, deren Konidiogenese gestört ist. Insbesondere die Funktion des Phosphotransferproteins MoYpd1p, sowohl im Phosphorelaysystem des HOG-Signal¬wegs als auch im Wirkmechanismus des Fungizids Fludioxonil, konnte eindeutig mittels Y2H- und Western Blot-Analysen nachgewiesen werden.rnEs wurden entscheidende Fortschritte für das Verständnis des Aufbaus und der Funktion des HOG-Signalwegs sowohl als physiologisches Regulationssystem für Umweltreize als auch als Fungizidtarget im Pflanzenschutz erzielt. Dabei konnte gezeigt werden, dass die Zweikompo-nenten-Hybrid-Histidinkinase (HIK) MoSln1p als Signalsensor für Salzstress und MoHik1p als Signalsensor für Zuckerstress fungiert. Die Beteiligung der Histidinkinasen MoHik5p und MoHik9p als Sensorproteine für Hypoxie im HOG-Signalweg ist durchaus denk¬bar und wurde durch erste Ergebnisse bekräftigt. rnSo konnte der HOG-Signalweg in mehreren Modellen dargestellt werden. Die Modelle der Signalerkennung und –transduktion von osmotischem Stress, von Hypoxie und der Wirkmecha¬nismus von Fludioxonil wurden erstmals in diesem Umfang für M. oryzae ausgearbei¬tet.rnDer zweite Teil dieser Arbeit repräsentiert die erste umfassende Untersuchung aller zehn HIK-codierender Gensequenzen, die im Genom von M. oryzae identifiziert werden konnten. Diese Signalproteine waren bisher noch nicht Gegenstand wissenschaftlicher Studien. Die Untersuchung beginnt mit einer phylogenetischen Einordnung aller untersuchten Proteinsequen¬zen in die verschiedenen Gruppen von Histidinkinasen in Pilzen. Eine ausführli-che phänotypische Charakterisierung aller HIK-codierender Gene folgt und wurde anhand von Mutanten durchgeführt, in denen diese Gene einzeln inaktiviert wurden.rnDie Beteiligung von MoHik5p und MoHik9p als mögliche Sauerstoffsensoren im HOG-Signal-weg konnte dokumentiert werden und die anschließenden Western Blot-Analysen bestätig¬ten erstmals die Aktivierung des HOG-Signalwegs bei hypoxieähnlichen Zuständen.rnDes Weiteren wurden mit MoHik5p und MoHik8p zwei neue Pathogenitätsfaktoren in M. oryzae identifiziert. Die apathogenen Mutantenstämme ΔMohik5 und ΔMohik8 sind in der Konidiogenese gestört und nicht in der Lage Appressorien zu differenzieren. Der Einsatz dieser Proteine als Fungizidtarget im protektiven Pflanzenschutz in der Zukunft ist somit denk-bar.rn

Fungal pathogens perceive and respond to molecules from the plant, triggering pathogenic development. Transduction of these signals may use heterotrimeric G proteins, and it is thought that protein phosphorylation cascades are also important. We have isolated a mitogen-activated protein kinase homolog from the corn pathogen Cochliobolus heterostrophus to test its role as a component of the transduction pathways. The new gene, CHK1, has a deduced amino acid sequence 90% identical to Pmk1 of the rice blast fungus Magnaporthe grisea and 59% identical to Fus3 of Saccharomyces cerevisiae. A series of chk1 deletion mutants has poorly developed aerial hyphae, autolysis, and no conidia. No pseudothecia are formed when a cross between two Δchk1 mutants is attempted. The ability of Δchk1 mutants to infect corn plants is reduced severely. The growth pattern of hyphae on a glass surface is strikingly altered from that of the wild type, forming coils or loops, but no appressoria. This set of phenotypes overlaps only partially with that of pmk1 mutants, the homologous gene of the rice blast fungus. In particular, sexual and asexual sporulation both require Chk1 function in Cochliobolus heterostrophus, in contrast to Pmk1, but perhaps more similar to yeast, where Fus3 transmits the mating signal. Chk1 is required for efficient colonization of leaf tissue, which can be compared with filamentous invasive growth of yeast, modulated through another closely related mitogen-activated protein kinase, Kss1. Ubiquitous signaling elements thus are used in diverse ways in different plant pathogens, perhaps the result of coevolution of the transducers and their targets.

Blast is a major disease of rice in Brazil, the largest rice-producing country outside Asia. This study aimed to assess the genetic structure and mating-type frequency in a contemporary Pyricularia oryzae population, which caused widespread epidemics during the 2012/13 season in the Brazilian lowland subtropical region. Symptomatic leaves and panicles were sampled at flooded rice fields in the states of Rio Grande do Sul (RS, 34 fields) and Santa Catarina (SC, 21 fields). The polymorphism at ten simple sequence repeats (SSR or microsatellite) loci and the presence of MAT1-1 or MAT1-2 idiomorphs were assessed in a population comprised of 187 isolates. Only the MAT1-2 idiomorph was found and 162 genotypes were identified by the SSR analysis. A discriminant analysis of principal components (DAPC) of SSR data resolved four genetic groups, which were strongly associated with the cultivar of origin of the isolates. There was high level of genotypic diversity and moderate level of gene diversity regardless whether isolates were grouped in subpopulations based on geographic region, cultivar host or cultivar within region. While regional subpopulations were weakly differentiated, high genetic differentiation was found among subpopulations comprised of isolates from different cultivars. The data suggest that the rice blast pathogen population in southern Brazil is comprised of clonal lineages that are adapting to specific cultivar hosts. Farmers should avoid the use of susceptible cultivars over large areas and breeders should focus at enlarging the genetic basis of new cultivars.
