It is well known residual gum exists in degummed or retted hemp fibres. Gum removal results in improvement in fibre fineness and the properties of the resultant hemp yarns. However, it is not known what correlation if any exists between the residual gum content in retted hemp fibres and the fibre fineness, described in terms of fibre width in this paper. This study examined the mean width and coefficient of variation (CV) of fibre width of seventeen chemically retted hemp samples with reference to residual gum content. The mean and CV of fibre width were obtained from an Optical fibre diameter analyser (OFDA 100). The linear regression analysis results show that the mean fibre width is directly proportional to the residual gum content. A slightly weaker linear correlation also exists between the coefficient of variation of fibre width and the residual gum content. The strong linear co-relation between the mean of fibre width and the residual gum content is a significant outcome, since testing for fibre width using the OFDA is a much simpler and quicker process than testing the residual gum content. Scanning Electron Microscopy (SEM) reinforces the OFDA findings. SEM micrographs show a flat ribbon like fibre cross-section hence the term “fibre width” is used instead of fibre diameter. Spectral differences in the untreated dry decorticated skin samples and chemically treated and subsequently carded samples indicate delignification. The peaks at 1370 cm−1, 1325 cm−1, 1733 cm−1, and 1600 cm−1 attributed to lignin in the untreated samples are missing from the spectra of the treated samples. The spectra of the treated samples are more amine-dominated with some of the OH character lost.

The demand for eco-friendly apparel and technical textiles has led to a resurgence of interests in bast fibres such as hemp. The lack of fast and objective evaluation of the quality attributes of bast fibres has been a major barrier to the advancement of the bast fibre industry. One of the most important quality attributes of a fibre is its fineness. For bast fibres, the fibre fineness measurement can also reflect the degree of fibre separation during retting or degumming. The traditional method of evaluating the fineness and residual gum content of bast fibres is a very tedious process. In this paper, degummed hemp fibres have been measured for fineness on an Optical Fibre Diameter Analyser (OFDA), and the results have been co-related with the residual gum content in the fibre samples. Since hemp fibres do not have a circular cross section, it is the width of the fibre that gets measured by the OFDA instrument, and this width has been used as an indication of the fibre fineness in this paper. The findings from this study suggest that the optical method can provide a fast and objective way of evaluating the fineness of hemp fibres, and that there is a good correlation between the fibre „width‟ measurement and the residual gum content.

The measurement of fibre quality in bast fibres is related to the amount of gum (lignin, hemicellulose, wax and pectin) left in the fibre after the retting process. Large amounts of gums present represent poor separation of fibre. Efficiency of retting can be monitored by measuring the residual gum content of the retted fibre.

This paper investigated the use of ultrasonic vibration combined with chemical retting as a pre-treatment to improve accuracy of traditional residual gum content test. Work was conducted on chemically retted hemp fibre. Pre-treatment conditions were analysed by determining the best chemical combination, chemical concentration and treatment time. Fibres were examined for successful separation using optical microscopy and optical fibre diameter analysis (OFDA). The work proposed a new method for determining the residual gum content of hemp fibre.

This paper provides an overview of recent research on a range of natural fibres and textiles. The focus is on work carried out at Deakin University’s Centre for Material and Fibre Innovation, which is a multidisciplinary research centre with over 100 researchers. The fibres include hemp, wool, silk, and alpaca fibres. Research on yarns, fabrics, and fine powders made from wool and silk fibres are briefly discussed also.

The within-fibre diameter variation of wool has been examined systematically, which highlights the importance of this hard-to-measure fibre attribute. A relationship between hemp fibre fineness and residual gum content has been established, which provides a rapid means of assessing the residual gum content in the degummed hemp fibres. Silk and wool fibres have been converted into ultrafine powders for advanced applications. The Resistance to Compression (RtC) behaviour of wool and alpaca fibres has been closely examined, which challenges the belief that RtC is a good indicator of fibre softness. Ways of reducing the hairiness of natural fibre yarns, predicting the pilling propensity of wool knits, and functionalising fabrics for superhydrophobicity and photochromic or colour changing effects are discussed.

Solvent-cast films from three polymers, carboxymethylcellulose (CMC), sodium alginate (SA), and xanthan gum, were prepared by drying the polymeric gels in air. Three methods, (a) passive hydration, (b) vortex hydration with heating, and (c) cold hydration, were investigated to determine the most effective means of preparing gels for each of the three polymers. Different drying conditions [relative humidity - RH (6-52%) and temperature (3-45 degrees C)] were investigated to determine the effect of drying rate on the films prepared by drying the polymeric gels. The tensile properties of the CMC films were determined by stretching dumbbell-shaped films to breaking point, using a Texture Analyser. Glycerol was used as a plasticizer, and its effects on the drying rate, physical appearance, and tensile properties of the resulting films were investigated. Vortex hydration with heating was the method of choice for preparing gels of SA and CMC, and cold hydration for xanthan gels. Drying rates increased with low glycerol content, high temperature, and low relative humidity. The residual water content of the films increased with increasing glycerol content and high relative humidity and decreased at higher temperatures. Generally, temperature affected the drying rate to a greater extent than relative humidity. Glycerol significantly affected the toughness (increased) and rigidity (decreased) of CMC films. CMC films prepared at 45 degrees C and 6% RH produced suitable films at the fastest rate while films containing equal quantities of glycerol and CMC possessed an ideal balance between flexibility and rigidity.

Bovine rumen protein with two levels of residual lipids (1.9% or 3.8%) was subjected to thermoplastic extrusion under different temperatures and moisture contents. Protein Solubility in different buffers, disulphide cross-linking and molecular weight distribution were determined on the extrudates. After extrusion, samples with 1.9% residual lipids content had a higher concentration of protein insoluble by undetermined forces, irrespective of feed moisture and processing temperature used. Lipid content of 3.8% in the feed material resulted in more protein participating in the extrudate network through non-covalent interactions (hydrophobic and electrostatic) and disulphide bonds. A small dependency of the extrusion process on moisture and temperature and a marked dependency on lipid content, especially phospholipid, was observed, Electrophoresis under non-reducing conditions showed that protein extrusion with low feed moisture promoted high molecular breakdown inside the barrel, probably due to intense shear force, and further protein aggregation at the die end. Two high-performance liquid chromatographic methods for determination of residual monomer in dental acrylic resins are described. Monomers were detected by their UV absorbance at 230 nm, on a Nucleosil((R)) C-18 (5 mu m particle size, 100 angstrom pore size, 15 x 0.46 cm i.d.) column. The separation was performed using acetonitrile-water (55:45 v/v) containing 0.01% triethylamine (TEA) for methyl methacrylate and butyl methacrylate, and acetonitrile-water (60:40 v/v) containing 0.01% TEA for isobutyl methacrylate and 1,6-hexanediol dimethacrylate as mobile phases, at a flow rate of 0.8 mL/min. Good linear relationships were obtained in the concentration range 5.0-80.0 mu g/mL for methyl methacrylate, 10.0-160.0 mu g/mL for butyl methacrylate, 50.0-500.0 mu g/mL for isobutyl methacrylate and 2.5-180.0 mu g/mL for 1,6-hexanediol dimethacrylate. Adequate assay for intra- and inter-day precision and accuracy was observed during the validation process. An extraction procedure to remove residual monomer from the acrylic resins was also established. Residual monomer was obtained from broken specimens of acrylic disks using methanol as extraction solvent for 2 h in an ice-bath. The developed methods and the extraction procedure were applied to dental acrylic resins, tested with or without post-polymerization treatments, and proved to be accurate and precise for the determination of residual monomer content of the materials evaluated. A series of styrene-butadiene rubber (SBR) nanocomposites filledwith different particle sized kaolinites are prepared via a latex blending method. The thermal stabilities of these clay polymer nanocomposites (CPN) are characterized by a range of techniques including thermogravimetry (TG), digital photos, scanning electron microscopy (SEM) and Raman spectroscopy. These CPN show some remarkable improvement in thermal stability compared to that of the pure SBR. With the increase of kaolinite particle size, the residual char content and the average activation energy of kaolinite SBR nanocomposites all decrease; the pyrolysis residues become porous; the crystal carbon in the pyrolysis residues decrease significantly from 58.23% to 44.41%. The above results prove that the increase of kaolinite particle size is not beneficial in improving the thermal stability of kaolinite SBR nanocomposites.

Chitosan from prawn waste was used for the removal of mercury from solutions. Mercuric chloride solutions containing 250, 500, 1000, 10000 and 100000 ng of Hg super(+2)/ml were treated with chitosan samples of different particle size for different periods. The effect of initial concentration of mercury in the solution, particle size of chitosan and time of treatment on the adsorption of Hg super(+2) was studied. The residual mercury content after treatment for ten min. with chitosan of 40 mesh size from a solution of initial concentration 10000 ng/ml was 10 ng/ml whereas it was 50 ng/ml for chitosan of larger particle size (10-20 mesh). From solutions of lower concentrations complete removal of mercury was possible by chitosan treatment. Though the particle size and time of treatment have significant effect, the concentration of mercury in solution is more influential on the removal of mercury from solution.

To formulate therapeutic proteins into polymeric devices the protein is typically in the solid state, which can be achieved by the process of freeze-drying. However, freeze-drying not only risks denaturing the protein but it can adversely affect the cure characteristics of protein-loaded silicone elastomers. This study demonstrates that a variation in the parameters of the freeze-dryer can significantly affect the residual moisture content of freeze-dried BSA, which in turn has an effect on the bulk density and flow properties of the BSA. The bulk density and flow properties of the BSA subsequently affect the cure characteristics of BSA-loaded silicone elastomers. An increase in the residual moisture content results in the freeze-dried BSA having a decreased bulk density and poor flow properties which can have a detrimental effect on the cure characteristics of a freeze-dried BSA-loaded silicone elastomer. © 2012 Wiley Periodicals, Inc. This study describes the preclinical development of a matrix-type silicone elastomer vaginal ring device designed to provide controlled release of UC781, a non-nucleoside re- verse transcriptase inhibitor. Testing of both human- and macaque-sized rings in a sink condition in vitro release model demonstrated continuous UC781 release in quantities consid- ered sufficient to maintain vaginal fluid concentrations at levels 82–860-fold higher than the in vitro IC50 (2.0 to 10.4 nM) and therefore potentially protect against mucosal trans- mission of HIV. The 100-mg UC781 rings were well tolerated in pig-tailed macaques, did not induce local inflammation as determined by cytokine analysis and maintained median con- centrations in vaginal fluids of UC781 in the range of 0.27 to 5.18 mM during the course of the 28-day study. Analysis of residual UC781 content in rings after completion of both the in vitro release and macaque pharmacokinetic studies revealed that 57 and 5 mg of UC781 was released, respectively. The pharmacokinetic analysis of a 100-mg UC781 vaginal ring in pig-tailed macaques showed poor in vivo–in vitro correlation, attributed to the very poor solubility of UC781 in vaginal fluid and resulting in a dissolution-controlled drug release mecha- nism rather than the expected diffusion-controlled mechanism.

The present study aimed at the utlisation of microbial organisms for the production of good quality chitin and chitosan. The three strains used for the study were Lactobacillus plantarum, Lactobacililus brevis and Bacillus subtilis. These strains were selected on the basis of their acid producing ability to reduce the pH of the fermenting substrates to prevent spoilage and thus caused demineralisation of the shell. Besides, the proteolytic enzymes in these strains acted on proteinaceous covering of shrimp and thus caused deprotenisation of shrimp shell waste. Thus the two processes involved in chitin production can be affected to certain extent using bacterial fermentation of shrimp shell.Optimization parameters like fermentation period, quantity of inoculum, type of sugar, concentration of sugar etc. for fermentation with three different strains were studied. For these, parameters like pH, Total titrable acidity (TTA), changes in sugar concentration, changes in microbial count, sensory changes etc. were studied.Fermentation study with Lactobacillus plantarum was continued with 20% w/v jaggery broth for 15 days. The inoculum prepared yislded a cell concentration of approximately 108 CFU/ml. In the present study, lactic acid and dilute hydrochloric acid were used for initial pH adjustment because; without adjusting the initial pH, it took more than 5 hours for the lactic acid bacteria to convert glucose to lactic acid and during this delay spoilage occurred due to putrefying enzymes active at neutral or higher pH. During the fermentation study, pH first decreased in correspondence with increase in TTA values. This showed a clear indication of acid production by the strain. This trend continued till their proteolytic activity showed an increasing trend. When the available sugar source started depleting, proteolytic activity also decreased and pH increased. This was clearly reflected in the sensory evaluation results. Lactic acid treated samples showed greater extent of demineralization and deprotenisation at the end of fermentation study than hydrochloric acid treated samples. It can be due to the effect of strong hydrochloric acid on the initial microbial count, which directly affects the fermentation process. At the end of fermentation, about 76.5% of ash was removed in lactic acid treated samples and 71.8% in hydrochloric acid treated samples; 72.8% of proteins in lactic acid treated samples and 70.6% in hydrochloric acid treated samples. The residual protein and ash in the fermented residue were reduced to permissible limit by treatment with 0.8N HCI and 1M NaOH. Characteristics of chitin like chitin content, ash content, protein content, % of N- acetylation etc. were studied. Quality characteristics like viscosity, degree of deacetylation and molecular weight of chitosan prepared were also compared. The chitosan samples prepared from lactic acid treated showed high viscosity than HCI treated samples. But degree of deacetylation is more in HCI treated samples than lactic acid treated ones. Characteristics of protein liquor obtained like its biogenic composition, amino acid composition, total volatile base nitrogen, alpha amino nitrogen etc. also were studied to find out its suitability as animal feed supplement.Optimization of fermentation parameters for Lactobacillus brevis fermentation study was also conducted and parameters were standardized. Then detailed fermentation study was done in 20%wlv jaggery broth for 17 days. Also the effect of two different acid treatments (mild HCI and lactic acid) used for initial pH adjustment on chitin production were also studied. In this study also trend of changes in pH. changes in sugar concentration ,microbial count changes were similar to Lactobacillus plantarum studies. At the end of fermentation, residual protein in the samples were only 32.48% in HCI treated samples and 31.85% in lactic acid treated samples. The residual ash content was about 33.68% in HCI treated ones and 32.52% in lactic acid treated ones. The fermented residue was converted to chitin with good characteristics by treatment with 1.2MNaOH and 1NHCI.Characteristics of chitin samples prepared were studied and extent of Nacetylation was about 84% in HCI treated chitin and 85%in lactic acid treated ones assessed from FTIR spectrum. Chitosan was prepared from these samples by usual chemical method and its extent of solubility, degree of deacetylation, viscosity and molecular weight etc were studied. The values of viscosity and molecular weight of the samples prepared were comparatively less than the chitosan prepared by Lactobacillus plantarum fermentation. Characteristics of protein liquor obtained were analyzed to determine its quality and is suitability as animal feed supplement.Another strain used for the study was Bacillus subtilis and fermentation was carried out in 20%w/v jaggery broth for 15 days. It was found that Bacillus subtilis was more efficient than other Lactobacillus species for deprotenisation and demineralization. This was mainly due to the difference in the proteolytic nature of the strains. About 84% of protein and 72% of ash were removed at the end of fermentation.

Perchlorate-reducing bacteria fractionate chlorine stable isotopes giving a powerful approach to monitor the extent of microbial consumption of perchlorate in contaminated sites undergoing remediation or natural perchlorate containing sites. This study reports the full experimental data and methodology used to re-evaluate the chlorine isotope fractionation of perchlorate reduction in duplicate culture experiments of Azospira suillum strain PS at 37 degrees C (Delta Cl-37(Cr)--ClO4-) previously reported, without a supporting data set by Coleman et al. [Coleman, M.L., Ader, M., Chaudhuri, S., Coates,J.D., 2003. Microbial Isotopic Fractionation of Perchlorate Chlorine. Appl. Environ. Microbiol. 69, 4997-5000] in a reconnaissance study, with the goal of increasing the accuracy and precision of the isotopic fractionation determination. The method fully described here for the first time, allows the determination of a higher precision Delta Cl-37(Cl)--ClO4- value, either from accumulated chloride content and isotopic composition or from the residual perchlorate content and isotopic composition. The result sets agree perfectly, within error, giving average Delta Cl-37(Cl)--ClO4- = -14.94 +/- 0.15%omicron. Complementary use of chloride and perchlorate data allowed the identification and rejection of poor quality data by applying mass and isotopic balance checks. This precise Delta Cl-37(Cl)--ClO4-, value can serve as a reference point for comparison with future in situ or microcosm studies but we also note its similarity to the theoretical equilibrium isotopic fractionation between a hypothetical chlorine species of redox state +6 and perchlorate at 37 degrees C and suggest that the first electron transfer during perchlorate reduction may occur at isotopic equilibrium between art enzyme-bound chlorine and perchlorate.
