The aim of this study was to estimate genetic parameters for milk yield (MY) in buffaloes using reaction norms. Model included the additive direct effect as random and contemporary group (herd and year of birth) were included as fixed effects and cow age classes (linear) as covariables. The animal additive direct random effect was modeled through linear Legendre polynomials on environment gradient (EG) standardized means. Mean trends were taken into account by a linear regression on Legendre polynomials of environmental group means. Residual variance was modeled trough 6 heterogeneity classes (EG). These classes of residual variance was formed : EG1: mean = 866,93 kg (621,68 kg-1011,76 kg); EG2: mean = 1193,00 kg (1011,76 kg-1251,49 kg); EG3: mean = 1309,37 kg (1251,49 kg -1393,20 kg); EG4: mean = 1497,59 kg (1393,20 kg-1593,53 kg); EG5: mean = 1664,78 kg (1593,53 kg -1727,32kg) e EG6: mean = 1973,85 kg (1727,32 kg -2422,19 kg).(Co) variance functions were estimated by restricted maximum likelihood (REML) using the GIBBS3F90 package. The heritability estimates for MY raised as the environmental gradient increased, varying from 0.20 to 0.40. However, in intermediate to favorable environments, the heritability estimates obtained with Considerable genotype-environment interaction was found for MY using reaction norms. For genetic evaluation of MY is necessary to consider heterogeneity of variances to model the residual variance.

Non-myrmecophilous lepidopteran larvae using plants bearing ant attractants such as extrafloral nectaries are good models for studying morphological and behavioural mechanisms against ant predation. Udranomia spitzi (Hesperiidae) is a butterfly whose larvae feed on leaves of Ouratea spectabilis (Ochnaceae), a plant with extrafloral nectaries. We described the early stages of U. spitzi, and used field observations and experiments to investigate the defensive strategies of caterpillars against predatory ants. Larvae pass through five instars and pupation occurs inside larval leaf shelters. Ant-exclusion experiments revealed that the presence of ants did not affect significantly caterpillar survival. Predation experiments showed that vulnerability to ant predation decreased with increase in larval size. The present study showed that predatory ants are not as relevant as demonstrated for other systems, and also illustrates how observational data and field experiments can contribute to a better understanding of the biology and ecology of a species of interest.

A new interaction between insects and carnivorous plants is reported from Brazil. Larvae of the predatory flower fly Toxomerus basalis (Diptera: Syrphidae: Syrphinae) have been found scavenging on the sticky leaves of several carnivorous sundew species (Drosera, Droseraceae) in Minas Gerais and São Paulo states, SE Brazil. This syrphid apparently spends its whole larval stage feeding on prey trapped by Drosera leaves. The nature of this plant-animal relationship is discussed, as well as the Drosera species involved, and locations where T. basalis was observed. 180 years after the discovery of this flower fly species, its biology now has been revealed. This is (1) the first record of kleptoparasitism in the Syrphidae, (2) a new larval feeding mode for this family, and (3) the first report of a dipteran that shows a kleptoparasitic relationship with a carnivorous plant with adhesive flypaper traps. The first descriptions of the third instar larva and puparium of T. basalis based on Scanning Electron Microscope analysis are provided.

This study examined how students leveraged different types of knowledge resources on an outdoor learning trail. We positioned the learning trail as an integral part of the curriculum with a pre- and post-trail phase to scaffold and to support students’ meaning-making process. The study was conducted with two classes of secondary two students. We coded two groups’ discourse to examine the use of knowledge resource types in the meaning-making process in an outdoor learning setting: contextual resource, new conceptual resource, prior knowledge resource, as well as the relationship among these knowledge resource types. Next, we also examined environmental interaction and integration in the students’ use of these knowledge resource types. Analysis showed that contextual resources are chiefly instrumental in fostering students’ capacity to harness new conceptual resource and to activate prior knowledge resource in interacting with and integrating the outdoor learning environment in the meaning-making process.

鄂尔多斯高原是一个多层次、复杂的生态过渡带,具有复杂多样的环境条件、生态特点,因此也就具有复杂多样的植物与环境关系。本文从群落和景观两个尺度水平上研究鄂尔多斯高原植物或植被与环境关系及景观空间格局。利用鄂尔多斯高原野外植物群落样方调坦数据、微生境环境数据、气候数据,以典范对应分析(Canonical Correspondence Analysis, CCA)的方法分析了鄂尔多斯高原植物分布空间格局与环境要素的关系并对秋类环境要素对鄂尔多斯高原植物空间分布格局的贡献进行了定量分解;利用1:500 000 鄂尔多斯高原植被、土壤、土地利用、土地沙漠化类型等专题地图在GIS支持下分析了鄂尔多斯高原景观空间格局、并利用上述专题图数据加上鄂尔多斯高原气候数据库分析了土壤、土地利用、土地利用、土地沙漠化和气候等对鄂尔多斯高原植被空间分布格局的作用。通过分析,得到了以下主要结论: 1 在分析方法上、利用典范对应分析的方法,把植物分布的空间因素与环境因素分离的方法发展为植物分布空间格局不同类型影响因素作用的定量分离,提出了相应的概念模型和实现方法。 2 影响鄂尔多斯高原植物分布空间格局的主要微生境环境要素是基质类型、地下水位、覆沙厚度等,而影响鄂尔多斯高原分布空间格局的主要气候要素中,降水和干湿指标的作用大于温度和热量指标的作用。 3 通过对鄂尔多斯高原植物分布空间格局与环境关系的研究,以植物对微生境环境要素的反应为根据,把鄂尔多斯高原主要植物划分为4个大类群:梁地植物、沙地植物、草甸植物和耐盐植物。根据它们对气候要素的反应,把鄂尔多斯高原主要植物划分为典型草原植物、荒漠草原植物和草原化荒漠植物3大类。进一步,根据鄂尔多劳动保护高原植物与环境关系的研究,进行了鄂尔多斯高原植被功能型划分的尝试,得到了鄂尔多斯高原的12种主要植被功能型。 4 对鄂尔多斯高原植物分布的空间格局的影响环境因素的贡献作了定量地分解。分析结果显示:鄂尔多斯高原植物分布空间格局中有27.02%可由已知环境变量得到解释,其中21.56%与微生境环境要素相关,7.51%与气候要素的作用有关,而气候与微生境环境要素的耦合作用的份额为2.05%。根据植物生长是否直接受到地下水的影响,鄂尔多斯高原存在两大类型生态特点差异明显的生境类型:中性立地和隐域生境,对两大类型生境上影响植物空间分布格局的环境要素的作用也进行了定量分解。分析结果表明:对于植物生长不直接受地下水影响的中性立地,已知环境要素的作用可以解释植物空间分布格局总信息的29.36%,稍大于对总体上鄂尔多斯高原植物分布空间格局的解释,其中9.23%与气候要素相关,22.08%与微生境环境要素相关,而两种类型环境要素的耦合作用则占1.95%。对于植物生长直接受到地下水影响的隐域生境,所有已知环境要素对植物分布空间格局的贡献率为72.28%,其中气候要素的作用为30.31%,微生境环境要素的作用为49.08%,两类环境要素的耦合作用为7.11%。 5 描述景观空间格局的指数多种多样,这些能数在描述特定区域的景观空间格局时是有信息冗余的。本文对利用FRAGSTATS所获得的鄂尔多斯高原植被、土壤、土地利用、土地沙漠化等景观分量的20个景观指数实施了因子分析。通过因子分析,我们可以把描述鄂尔多斯高原景观空间格局的景观指数归并为以下8类:多样性指数、斑块多度指数、斑块类型丰富度指数、斑块面积指数、斑块形状指数、分形维数、空间配置指数和斑块面积变异指数。通过因子分析,还得到了这些景观指数对描述鄂尔多斯景观格局的共性特征:在描述鄂尔多斯高原景观空间格局时,作用最大的是多样性指数、斑块多度指数、面积加权平均斑块形状指数和面积加权平均分形维数,其次是斑块类型丰富度指数、平均分维指数、平均形状指数和斑块面积指数,而空间配置指数(扩散与毗连指数)和斑块面积变异指数的作用则比较微弱。 6 对鄂尔多斯高原景观指数的因子分析是非常有效和成功的。因子分析对鄂尔多斯高原植被、土壤、土地利用、土地沙漠化景观指数的分析分别得到了5-6个主要因子,可以表达原有20个景观指数所表达信息的91.1-96.0%,即可以反应鄂尔多斯高原景观空间格局的大部分信息。本文所进行的因子分析对因子进行了方差最大化(Varimax)正交旋转的处理,因子分析得到的每一个主要因子都有一个或几个与之相关性非常高的景观指数与之对应,因此,就可以用与因子分析所得主要因子相关性最高的景观指数代替该主要因子来表达鄂尔多斯高原的景观空间格局。另外还因为有些景观指数之间具有极高的相关系数,所以对因子分析所得到的景观指数可以进一步精减,最后利用因子分析成功地把原有20个景观指数减少到了11个。最后被选来描述鄂尔多斯高原景观格局的景观指数有下列11个:MSIEI(修正的Simpson均匀度指数)、AWMPFD(面积加权平均斑块分形维数)、AWMSI(面积加权平均形状指数)、NP(斑块数目)、PR(斑块类型丰富度)、MSI(平均形状指数)、MPFD(平均斑块分形维数)、MPS(平均斑块面积)、PSCV(斑块面积变异系数)、DLFD(双对数分形维数)和IJI(扩散与毗连指数)。 7 鄂尔多斯高原植被、土壤、土地利用在景观组成结构上具有一个共同特点,就是各种类型的面积差异极大,少数类型占有极大比重,而其余面积则很小。产生这一情形的原因主要与人为活动的强烈影响有关,表现在地带性的植被与土壤面积所占的比重不高,沙地、沙生植被与风沙土则占有很大比重。 8 以地带性植被和滩地隐域性植被表示的鄂尔多斯高原的原生植被仅占高原面积的不足30%,而以地带性土壤和滩地隐域性土壤表示的原生性土壤占鄂尔多斯高原总面积的近40%,说明土壤退化不如植被退化严重,或滞后于植被退化。 9 鄂尔多斯高原各景观指数的空间变化曲线,植被与土壤很相近,具有非常相似的格局;土地利用景观格局空间变化特征与植被、土壤等明显不同;土地沙漠化的景观格局空间变化曲线介植被曲线、土壤曲线与土地利用曲线之间,说明土地沙漠化不仅是一个受人为活动影响的过程,而且与自然过程密切相关。 10 鄂尔多斯高原景观格局的空间梯度变化表现出了东西向和南北向的梯度,但总体上以东西向的变化比较明显。 11 通过鄂尔多斯高原土壤类型、土地利用、土地沙漠化等景观要素、气候、空间要素与鄂尔多斯高原植被空间分布格局的CCA分析,探讨了它们之间的相互关系。以对鄂尔多斯高原植被组成数据的总方差解释的百分率为标准,土壤对鄂尔多斯植被分布的空间格局的作用最大,其方差贡献率可达44.28%,其次是土地利用与鄂尔多斯高原植被的关系也很密切,土地利用对鄂尔多斯高原植被空间分布格局的方差贡献率为22.45%,空间因素对鄂尔多斯高原植被空间分布格局的贡献率为17.51%,土地沙漠化对鄂尔多斯高原植被空间分布格局的贡献为15.65%,排在第四位;气候因素对鄂尔多斯高原植被空间格局的贡献率为11.95%,居第五位。 12 在气候要素对鄂尔多斯高原植被空间分布格局的作用中,降水与干湿指标的作用大于温度与热量指标的作用。这一点与利用野外调查样方的群落数据植物与气候关系的分析是完全一致的。CCA分析还表明鄂尔多斯高原植被空间格局的东西向变化大于南北向分异。 13 在群落和景观水平上,鄂尔多斯高原植物空间分布或植被格局的影响因素的作用具有相似的格局,即气候因子的作用明显地小于地质、土壤、水文等微生境环境要素(群落水平)或土壤(景观水平)的作用,并在这两个尺度上气候要素对植物空间分布或植被格局的定量解释份额上也是非常相近的,都仅有10%左右。气候因子对鄂尔多斯高原植物空间分布格局的这种弱的解释能力,从侧面说明了人为活动等非自然因素对鄂尔多斯高原植物空间分布格局的强烈作用。 14 在鄂尔多斯高原生态系统管理上,应协调人与自然的关系;加强鄂尔多斯高原的生物多样性保育,对于本区生态和经济对非常重要的滩地,应协调好对其开发利用与保护的关系;在鄂尔多斯高原土地沙漠化防治方面,应把调整人地关系与自然生态背景与条件相结合,如使用“三圈”模式等生态系统管理模式等。

Italy has a preeminent rank in kiwifruit industry, being the first exporter and the second largest producer after China. However, in the last years kiwifruit yields and the total cultivated area considerably decreased, due to the pandemic spread of the bacterial canker caused by Pseudomonas syringae pv. actinidiae (Psa). Several climatic conditions and cultural practices affect the development of the bacterial canker. This research work focused on the impact of agricultural practices and microclimate conditions on the incidence and epidemiology of Psa in the orchard. Therefore, the effect of fertilization, irrigation, use of bio-regulators, rootstock, training system and pruning were examined. The effect of different tunnel systems was analyzed as well, to study the plant-pathogen interaction. Considering the importance of insects as vectors in other pathosystems, the role of Metcalfa pruinosa in the spread of the bacterial canker was investigated in controlled conditions. In addition, quality and storage properties of fruits from infected plants were assessed. The study of all these aspects of the agronomic practices is useful to define a strategy to limit the bacterial diffusion in the orchard. Overall, excess nitrogen fertilization, water stress, stagnant water supplies, pruning before summer and the high number of Metcalfa pruinosa increased the Psa incidence. In contrast, tunnel covers may be useful for the control of the disease, with special attention to the kind of material.

The genus Methylobacterium comprises pink-pigmented facultative methylotrophic (PPFM) bacteria, known to be an important plant-associated bacterial group. Species of this group, described as plant-nodulating, have the dual capacity of producing cytokinin and enzymes, such as pectinase and cellulase, involved in systemic resistance induction and nitrogen fixation under specific plant environmental conditions. The aim hereby was to evaluate the phylogenetic distribution of Methylobacterium spp. isolates from different host plants. Thus, a comparative analysis between sequences from structural (16S rRNA) and functional mxaF (which codifies for a subunit of the enzyme methanol dehydrogenase) ubiquitous genes, was undertaken. Notably, some Methylobacterium spp. isolates are generalists through colonizing more than one host plant, whereas others are exclusively found in certain specific plant-species. Congruency between phylogeny and specific host inhabitance was higher in the mxaF gene than in the 16S rRNA, a possible indication of function-based selection in this niche. Therefore, in a first stage, plant colonization by Methylobacterium spp. could represent generalist behavior, possibly related to microbial competition and adaptation to a plant environment. Otherwise, niche-specific colonization is apparently impelled by the host plant.

Las NADPH oxidasas de plantas, denominadas “respiratory burst oxidase homologues” (RBOHs), producen especies reactivas del oxígeno (ROS) que median un amplio rango de funciones. En la célula vegetal, el ajuste preciso de la producción de ROS aporta la especificidad de señal para generar una respuesta apropiada ante las amenazas ambientales. RbohD y RbohF, dos de los diez genes Rboh de Arabidopsis, son pleiotrópicos y median diversos procesos fisiológicos en respuesta a patógenos. El control espacio-temporal de la expresión de los genes RbohD y RbohF podría ser un aspecto crítico para determinar la multiplicidad de funciones de estas oxidasas. Por ello, generamos líneas transgénicas de Arabidopsis con fusiones de los promoters de RbohD y RbohF a los genes delatores de la B-glucuronidasa y la luciferasa. Estas líneas fueron empleadas para revelar el patrón de expresión diferencial de RbohD y RbohF durante la respuesta inmune de Arabidopsis a la bacteria patógena Pseudomonas syringae pv. tomato DC3000, el hongo necrótrofo Plectosphaerella cucumerina y en respuesta a señales relacionadas con la respuesta inmune. Nuestros experimentos revelan un patrón de expresión diferencial de los promotores de RbohD y RbohF durante el desarrollo de la planta y en la respuesta inmune de Arabidopsis. Además hemos puesto de manifiesto que existe una correlación entre el nivel de actividad de los promotores de RbohD y RbohF con la acumulación de ROS y el nivel de muerte celular en respuesta a patógenos. La expression de RbohD y RbohF también es modulada de manera diferencial en respuesta a patrones moleculares asociados a patógenos (PAMPs) y por ácido abscísico (ABA). Cabe destacar que, mediante una estrategia de intercambio de promotores, hemos revelado que la región promotora de RbohD, es necesaria para dirigir la producción de ROS en respuesta a P. cucumerina. Adicionalmente, la activación del promotor de RbohD en respuesta al aislado de P. cucumerina no adaptado a Arabidopsis 2127, nos llevó a realizar ensayos de susceptibilidad con el doble mutante rbohD rbohF que han revelado un papel desconocido de estas oxidasas en resistencia no-huesped. La interacción entre la señalización dependiente de las RBOHs y otros componentes de la respuesta inmune de plantas podría explicar también las distintas funciones que median estas oxidasas en relación con la respuesta inmune. Entre la gran cantidad de señales coordinadas con la actividad de las RBOHs, existen evidencias genéticas y farmacológicas que indican que las proteínas G heterotriméricas están implicadas en algunas de las rutas de señalización mediadas por ROS derivadas de los RBOHs en respuesta a señales ambientales. Por ello hemos estudiado la relación entre estas RBOH-NADPH oxidasas y AGB1, la subunidad β de las proteínas G heterotriméricas en la respuesta inmune de Arabidopsis. Análisis de epistasis indican que las proteínas G heterotriméricas están implicadas en distintas rutas de señalización en defensa mediadas por las RBOHs. Nuestros resultados ilustran la relación compleja entre la señalización mediada por las RBOHs y las proteínas G heterotriméricas, que varía en función de la interacción planta-patógeno analizada. Además, hemos explorado la posible asociación entre AGB1 con RBOHD y RBOHF en eventos tempranos de la respuesta immune. Cabe señalar que experimentos de coímmunoprecipitación apuntan a una posible asociación entre AGB1 y la kinasa citoplasmática reguladora de RBOHD, BIK1. Esto indica un posible mecanismo de control de la función de esta NADPH oxidase por AGB1. En conjunto, estos datos aportan nuevas perspectivas sobre cómo, a través del control transcripcional o mediante la interacción con las proteínas G heterotriméricas, las NADPH oxidases de plantas median la producción de ROS y la señalización por ROS en la respuesta inmune. Nuestro trabajo ejemplifica cómo la regulación diferencial de dos miembros de una familia multigénica, les permite realizar distintas funciones fisiológicas especializadas usando un mismo mecanismo enzimático. ABSTRACT The plant NADPH oxidases, termed respiratory burst oxidase homologues (RBOHs), produce reactive oxygen species (ROS) which mediate a wide range of functions. Fine tuning this ROS production provides the signaling specificity to the plant cell to produce the appropriate response to environmental threats. RbohD and RbohF, two of the ten Rboh genes present in Arabidopsis, are pleiotropic and mediate diverse physiological processes in response to pathogens. One aspect that may prove critical to determine the multiplicity of functions of RbohD and RbohF is the spatio-temporal control of their gene expression. Thus, we generated Arabidopsis transgenic lines with RbohD- and RbohF-promoter fusions to the β-glucuronidase and the luciferase reporter genes. These transgenics were employed to reveal RbohD and RbohF promoter activity during Arabidopsis immune response to the pathogenic bacterium Pseudomonas syringae pv tomato DC3000, the necrotrophic fungus Plectosphaerella cucumerina and in response to immunity-related cues. Our experiments revealed a differential expression pattern of RbohD and RbohF throughout plant development and during Arabidopsis immune response. Moreover, we observed a correlation between the level of RbohD and RbohF promoter activity, the accumulation of ROS and the amount of cell death in response to pathogens. RbohD and RbohF gene expression was also differentially modulated by pathogen associated molecular patterns and abscisic acid. Interestingly, a promoter-swap strategy revealed the requirement for the promoter region of RbohD to drive the production of ROS in response to P. cucumerina. Additionally, since the RbohD promoter was activated during Arabidopsis interaction with a non-adapted P. cucumerina isolate 2127, we performed susceptibility tests to this fungal isolate that uncovered a new role of these oxidases on non-host resistance. The interplay between RBOH-dependent signaling with other components of the plant immune response might also explain the different immunity-related functions mediated by these oxidases. Among the plethora of signals coordinated with RBOH activity, pharmacological and genetic evidence indicates that heterotrimeric G proteins are involved in some of the signaling pathways mediated by RBOH–derived ROS in response to environmental cues. Therefore, we analysed the interplay between these RBOH-NADPH oxidases and AGB1, the Arabidopsis β-subunit of heterotrimeric G proteins during Arabidopsis immune response. We carried out epistasis studies that allowed us to test the implication of AGB1 in different RBOH-mediated defense signaling pathways. Our results illustrate the complex relationship between RBOH and heterotrimeric G proteins signaling, that varies depending on the type of plant-pathogen interaction. Furthermore, we tested the potential association between AGB1 with RBOHD and RBOHF during early immunity. Interestingly, our co-immunoprecipitation experiments point towards an association of AGB1 and the RBOHD regulatory kinase BIK1, thus providing a putative mechanism in the control of the NADPH oxidase function by AGB1. Taken all together, these studies provide further insights into the role that transcriptional control or the interaction with heterotrimeric G-proteins have on RBOH-NADPH oxidase-dependent ROS production and signaling in immunity. Our work exemplifies how, through a differential regulation, two members of a multigenic family achieve specialized physiological functions using a common enzymatic mechanism.

Viroids and most viral satellites have small, noncoding, and highly structured RNA genomes. How they cause disease symptoms without encoding proteins and why they have characteristic secondary structures are two longstanding questions. Recent studies have shown that both viroids and satellites are capable of inducing RNA silencing, suggesting a possible role of this mechanism in the pathology and evolution of these subviral RNAs. Here we show that preventing RNA silencing in tobacco, using a silencing suppressor, greatly reduces the symptoms caused by the Y satellite of cucumber mosaic virus. Furthermore, tomato plants expressing hairpin RNA, derived from potato spindle tuber viroid, developed symptoms similar to those of potato spindle tuber viroid infection. These results provide evidence suggesting that viroids and satellites cause disease symptoms by directing RNA silencing against physiologically important host genes. We also show that viroid and satellite RNAs are significantly resistant to RNA silencing-mediated degradation, suggesting that RNA silencing is an important selection pressure shaping the evolution of the secondary structures of these pathogens.

1. Some of the most damaging invasive plants are dispersed by frugivores and this is an area of emerging importance in weed management. It highlights the need for practical information on how frugivores affect weed population dynamics and spread, how frugivore populations are affected by weeds and what management recommendations are available. 2. Fruit traits influence frugivore choice. Fruit size, the presence of an inedible peel, defensive chemistry, crop size and phenology may all be useful traits for consideration in screening and eradication programmes. By considering the effect of these traits on the probability, quality and quantity of seed dispersal, it may be possible to rank invasive species by their desirability to frugivores. Fruit traits can also be manipulated with biocontrol agents. 3. Functional groups of frugivores can be assembled according to broad species groupings, and further refined according to size, gape size, pre- and post-ingestion processing techniques and movement patterns, to predict dispersal and establishment patterns for plant introductions. 4. Landscape fragmentation can increase frugivore dispersal of invasives, as many invasive plants and dispersers readily use disturbed matrix environments and fragment edges. Dispersal to particular landscape features, such as perches and edges, can be manipulated to function as seed sinks if control measures are concentrated in these areas. 5. Where invasive plants comprise part of the diet of native frugivores, there may be a conservation conflict between control of the invasive and maintaining populations of the native frugivore, especially where other threats such as habitat destruction have reduced populations of native fruit species. 6. Synthesis and applications. Development of functional groups of frugivore-dispersed invasive plants and dispersers will enable us to develop predictions for novel dispersal interactions at both population and community scales. Increasingly sophisticated mechanistic seed dispersal models combined with spatially explicit simulations show much promise for providing weed managers with the information they need to develop strategies for surveying, eradicating and managing plant invasions. Possible conservation conflicts mean that understanding the nature of the invasive plant-frugivore interaction is essential for determining appropriate management.

植物与昆虫的互作关系是个长期进化的过程,虫害给农业生产带来巨大损失。本研究以甘蓝型油菜(Brassica napus)为例,研究了不同环境条件和遗传背景下外源基因的表达与效用,同时利用蛋白质组技术,研究了虫害损伤模拟条件下植物可能存在的内源抗性机制。甘蓝型油菜中转入了人工合成的Bt(Bacillus thuringiensis)杀虫基因,能使植物产生抗虫蛋白抵御虫害。我们在湖北湖南两个实验点进行了大田实验,按植株生长发育的4个不同时期从转基因植株的叶片上采样,研究抗虫蛋白在植物体内的表达动态。植株顶部第三片展开叶的Bt毒蛋白浓度在结荚期前随植物生长而不断增加,而在结荚期出现或增或减的现象。采样叶片的可溶性总蛋白浓度含量一直呈增加的趋势,直到结荚以后出现含量的明显降低。同时,收集了转基因油菜与湘油15号在田间自然杂交形成的杂交后代种子用于栽培,用GFP仪检测杂交后代的绿色荧光蛋白(green fluorescent protein),并用聚合酶链式反应(polymerase chain reaction, PCR)检测并确认带有转基因的杂交植株。为了检测带有转基因的杂交后代油菜中Bt毒蛋白的杀虫效率,用对Bt毒蛋白敏感的试虫品系——初孵棉铃虫幼虫(Helicoverpa armigera)进行杀虫活性检测实验。结果表明,携带Bt基因的杂交湘油及其转基因亲本对试虫的体重增长量均产生了负面影响,可以推断在调查取样的植株生长发育阶段,转基因杂交后代与其转基因亲本植株的杀虫效率没有显著差异。转基因植物及其杂交后代中抗虫蛋白的持续表达及田间带有转基因的自播植物的出现会使害虫产生耐受抗性的潜在可能性增加。 相对于人为增加的抗虫基因,植物在长期对抗昆虫的过程中也进化形成了自我防御机制,能够产生特异的抗性蛋白来应对昆虫的取食。本研究用机械损伤模拟害虫取食,对比了油菜受到物理损伤前后可溶性总蛋白的含量变化并试图通过蛋白质组学技术来检测可能发生变化的蛋白质。Bradford定量测定发现,同一植株同一叶片损伤前后可溶性总蛋白含量差异显著,损伤后蛋白表达量显著增高。蛋白质组双向凝胶电泳及其差异分析显示,损伤前后有8个蛋白质点发生明显的上调或下调。选择其中2个差异蛋白点经过MALDI-TOF质谱鉴定,它们分别是Rubisco小亚基前体以及果糖-1,6-二磷酸醛缩酶和粪卟啉-3-氧化酶的混合物,这些蛋白质在其他植物的抗逆研究中也有报道,它们可能在油菜叶片应答机械损伤过程中对维持植物的生理功能也有重要作用。

BACKGROUND: In response to concerns expressed by workers at a public meeting, we analyzed the mortality experience of workers who were employed at the IBM plant in Endicott, New York and died between 1969-2001. An epidemiologic feasibility assessment indicated potential worker exposure to several known and suspected carcinogens at this plant. METHODS: We used the mortality and work history files produced under a court order and used in a previous mortality analysis. Using publicly available data for the state of New York as a standard of comparison, we conducted proportional cancer mortality (PCMR) analysis. RESULTS: The results showed significantly increased mortality due to melanoma (PCMR = 367; 95% CI: 119, 856) and lymphoma (PCMR = 220; 95% CI: 101, 419) in males and modestly increased mortality due to kidney cancer (PCMR = 165; 95% CI: 45, 421) and brain cancer (PCMR = 190; 95% CI: 52, 485) in males and breast cancer (PCMR = 126; 95% CI: 34, 321) in females. CONCLUSION: These results are similar to results from a previous IBM mortality study and support the need for a full cohort mortality analysis such as the one being planned by the National Institute for Occupational Safety and Health.
