PEO/PH共混体系的组份之间存在着氢键的相互作用,从偏光显微镜观察及熔点下降法测定,PEO/PH共混体系是相容体系,且PEO是在非晶区与PH相容,PH分子链不进入到PEO的晶格中,不引起晶胞参数的改变。对PEO/PH共混体系的等温结晶动力学研究表明,随共混体系中非晶组份PH含量的增加,体系的结晶生长方式由盘状生长转化为原纤状生长,成核方式由方式I(Kg=Kg(I)=4b. σσeTm/ΔHf.K)转化为方式II(Kg=Kg(Ii)=2b. σσeTm/ΔHf.K)析叠链表面自由能(σe)逐渐增大,体系的平衡溶点降低。在PEO/PH共混体系非等温结晶动力学的研究中,DSC实验表明,在常冷却速率下,PEO/PH共混体系符合Avrami方程所揭示的规律,为更好地反映非等温结晶特点,从Avrami方程和Ozawa方程出发,导出一个新的基本方程,根据这个方程,获得了描述非等温结晶过程的一些基本参数,在一定冷却速率下,随非晶组份PH含量的增加,东混体系的结晶速率降低;对于同一组成,冷却速率越大,体系结晶速率越快。WAXD和SAXS分析表明,随非晶组份PH含量的增加,PEO/PH共混体系的结晶度降低,长周期增大,过渡层厚略有变化,但变化很小。进一步表明,过渡层基本上是PEO的非晶相的贡献,PH不进入到PEO的晶格中,PEO是在非晶区与PH相容。

Hyperammonemia is a key factor in the pathogenesis of hepatic encephalopathy (HE) as well as other metabolic encephalopathies, such as those associated with inherited disorders of urea cycle enzymes and in Reye's syndrome. Acute HE results in increased brain ammonia (up to 5 mM), astrocytic swelling, and altered glutamatergic function. In the present study, using fluorescence imaging techniques, acute exposure (10 min) of ammonia (NH4+/NH3) to cultured astrocytes resulted in a concentration-dependent, transient increase in [Ca2+]i. This calcium transient was due to release from intracellular calcium stores, since the response was thapsigargin-sensitive and was still observed in calcium-free buffer. Using an enzyme-linked fluorescence assay, glutamate release was measured indirectly via the production of NADH (a naturally fluorescent product when excited with UV light). NH4+/NH3 (5 mM) stimulated a calcium-dependent glutamate release from cultured astrocytes, which was inhibited after preincubation with 1,2-bis(2-aminophenoxy)ethane-N,N,N',N'-tetraacetic acid acetoxymethyl ester but unaffected after preincubation with glutamate transport inhibitors dihydrokainate and DL-threo-beta-benzyloxyaspartate. NH4+/NH3 (5 mM) also induced a transient intracellular alkaline shift. To investigate whether the effects of NH4+/NH3 were mediated by an increase in pH(i), we applied trimethylamine (TMA+/TMA) as another weak base. TMA+/TMA (5 mM) induced a similar transient increase in both pH(i) and [Ca2+]i (mobilization from intracellular calcium stores) and resulted in calcium-dependent release of glutamate. These results indicate that an acute exposure to ammonia, resulting in cytosolic alkalinization, leads to calcium-dependent glutamate release from astrocytes. A deregulation of glutamate release from astrocytes by ammonia could contribute to glutamate dysfunction consistently observed in acute HE.

En una tesi anterior (Puig, 2004) es varen estudiar els aspectes mecànics vinculats al desfibratge i als consums energètics associats al procés de desintegració per acabar quantificant les forces implicades en el procés d'individualització dels components de diferents paperots. Aquest estudi, ha permès conèixer les condicions de desintegració que condueixen a un mínim consum energètic, aspecte complementari al reciclatge i destintatge del paperot però molt important per aconseguir un procés realment sostenible. Degut als requeriments del mercat, hi ha un altre aspecte o vessant realment important com és l'eliminació de tinta. Aquest procés, malgrat la complexitat industrial, es pot reduir o simplificar en dues etapes: la desintegració i la flotació. Ambdues, per raons d'eficàcia i condicionades pels agents químics que s'utilitzen, es realitzen en medi fortament alcalí (entorn d'un pH a 12). Aquest fet, genera una forta contaminació (DQO elevada) i per tant, és un procés poc respectuós amb el medi ambient. Els factors que condicionen treballar en medi alcalí són principalment, el despreniment de la tinta del suport, clarament accelerat en medi bàsic i fonamental per un bon destintatge i la utilització de tensioactius aniònics. Aquests tensioactius són sensibles al pH i la seva eficàcia disminueix en medi neutre o quasi neutre. Atès el que s'ha exposat, s'ha plantejat aquesta tesi que porta per títol "Aspectes Físics i Químics del Destintatge de Paper Revista mitjançant Tensioactius Catiònics", com un estudi del procés de destintatge de paper revista recuperat parant especial atenció a les variables físico-químiques i mecàniques del procés i a la seva influència en el despreniment i fragmentació de la tinta, així com en el procés d'eliminació de la tinta per flotació per tal d'avaluar la viabilitat d'un procés de destintatge en medi neutre enfront del destintatge alcalí convencional. S'ha utilitzat, com a paperot a destintar, el paper revista ja que aquest tipus de paper representa un percentatge elevat del paper recuperat a la Unió Europea. A més, aquest tipus de paper és utilitzat habitualment en les plantes de destintatge industrials per millorar les propietats de les suspensions destintades de paper de diari. La utilització de tensioactius catiònics és deguda a que la seva activitat no es veu afectada pel pH de la suspensió. Així, durant la comparació dels processos de destintatge en medi neutre i en medi alcalí, les eficàcies dels processos no es veuran influenciades pel grau d'ionització del tensioactiu. Donada l'absència d'estudis sobre la influència de les variables del procés en les mesures de concentració efectiva de tinta residual (ERIC) i ja que s'ha emprat aquest paràmetre per avaluar el despreniment i fragmentació de la tinta durant l'etapa de desintegració. En primer lloc, s'ha plantejat un estudi fonamental de la influència que poden tenir les variables del procés (granulometria, concentració de tinta i tipus de suport) en aquest paràmetre. Pel que fa a l'etapa de flotació, s'estudiarà l'efectivitat d'aquesta etapa, posant especial atenció a la capacitat del tensioactiu per eliminar tota la tinta despresa en l'etapa de desintegració. A més, donada la poca informació bibliogràfica referida als mecanismes de flotació amb tensioactius catiònics i la influència que té l'adsorció d'aquests agents per part de la suspensió paperera en l'etapa de flotació, s'ha dut a terme un estudi de l'adsorció dels tensioactius catiònics per part dels components d'una suspensió paperera. Per tant, la tesi realitzada té com a objectius:  Estudi fonamental de les mesures de ERIC (concentració efectiva de tinta residual) amb suports específics.  Estudi de la desintegració en medi neutre i comparació amb la desintegració clàssica.  Modelització del desfibratge de paperot a l'etapa de desintegració.  Estudi de l'eliminació de tinta per flotació en medi neutre i comparació amb la flotació clàssica.  Estudi del mecanisme d'adsorció de tensioactius en suspensions de cel·lulosa sintètica i de paperot.

The effect of ANG II on intracellular pH (pH(i)) recovery rate and AT(1) receptor translocation was investigated in transfected MDCK cells. The pHi recovery rate was evaluated by fluorescence microscopy using the fluorescent probe BCECF-AM. The human angiotensin II receptor isoform 1 (hAT(1)) translocation was analyzed by immunofluorescence and confocal microscope. Our data show that transfected cells in control situation have a pHi recovery rate of 0.219 +/- 0.017 pH U/min (n = 11). This value was similar to nontransfected cells [0.211 +/- 0.009 pH U/min (n = 12)]. Both values were significantly increased with ANG II (10(-9) M) but not with ANG II (10(-6) M). Losartan (10(-7) M) and dimethyl-BAPTA-AM (10(-7) M) decreased significantly the stimulatory effect of ANG II (10(-9) M) and induced an increase in Na+/H+ exchanger 1 (NHE-1) activity with ANG II (10(-6) M). Immunofluorescence studies indicated that in control situation, the hAT(1) receptor was predominantly expressed in cytosol. However, it was translocated to plasma membrane with ANG II (10(-9) M) and internalized with ANG II (10(-6) M). Losartan (10(-7) M) induced hAT(1) translocation to plasma membrane in all studied groups. Dimethyl-BAPTA-AM (10(-7) M) did not change the effect of ANG II (10(-9) M) on the hAT(1) receptor distribution but induced its accumulation at plasma membrane in cells treated with ANG II (10(-6) M). With ionomycin (10(-6) M), the receptor was accumulated in cytosol. The results indicate that, in MDCK cells, the effect of ANG II on NHE-1 activity is associated with ligand binding to AT(1) receptor and intracellular signaling events related to AT(1) translocation.

In a typical protocol for attaching DNA to a gold electrode, thiolated DNA is incubated with the electrode at neutral pH overnight. Here we report fast adsorption of non-thiolated DNA oligomers on gold electrodes at acidic pH (i.e., pH ~3.0). The peak-to-peak potential difference and the redox peak currents in typical cyclic voltammetry of [Fe(CN)6]3- are investigated to monitor the attachment. Compared with incubation at neutral pH, the lower pH can significantly promote the adsorption processes, enabling efficient adsorption even in 30min. The adsorption rate is DNA concentration-dependent, while the ionic strength shows no influence. Moreover, the adsorption is base-discriminative, with a preferred order of A>C≫G, T, which is attributed to the protonation of A and C at low pH and their higher binding affinity to gold surface. The immobilized DNA is functional and can hybridize with its complementary DNA but not a random DNA. This work is promising to provide a useful time-saving strategy for DNA assembly on gold electrodes, allowing fast fabrication of DNA-based biosensors and devices. © 2013 Elsevier Inc.

The pH(i) (intracellular pH) is an important physiological parameter which is altered during hypoxia and ischaemia, pathological conditions accompanied by a dramatic decrease in pH(i). Sensors of pH(i) include ion transport systems which control intracellular Ca2+ gradients and link changes in pH(i) to functions as diverse as proliferation and apoptosis. The annexins are a protein family characterized by Ca2+-dependent interactions with cellular membranes. Additionally, in vitro evidence points to the existence of pH-dependent, Ca(2+)-independent membrane association of several annexins. We show that hypoxia promotes the interaction of the recombinant annexin A2-S100A10 (p11) and annexin A6 with the plasma membrane. We have investigated in vivo the influence of the pH(i) on the membrane association of human annexins A1, A2, A4, A5 and A6 tagged with fluorescent proteins, and characterized this interaction for endogenous annexins present in smooth muscle and HEK (human embryonic kidney)-293 cells biochemically and by immunofluorescence microscopy. Our results show that annexin A6 and the heterotetramer A2-S100A10 (but not annexins A1, A4 and A5) interact independently of Ca2+ with the plasma membrane at pH 6.2 and 6.6. The dimerization of annexin A2 within the annexin A2-S100A10 complex is essential for the pH-dependent membrane interaction at this pH range. The pH-induced membrane binding of annexins A6 and A2-S100A10 might have consequences for their functions as membrane organizers and channel modulators.

An increase in carbon dioxide (CO2) and protons (H+) are the primary signals for breathing. Cells that sense changes in CO2/H+ levels and increase breathing accordingly are located in a region of the caudal medulla oblongata called the retrotrapezoid nucleus (RTN). Specifically, select RTN neurons are intrinsically pH sensitive and send excitatory projections to the respiratory rhythm generator to drive breathing. Glial cells in the RTN are thought to contribute to this respiratory drive, possibly by releasing ATP in response to increases in CO2/H+ levels. However, pH sensitivity of RTN glial cells has yet to be determined. Therefore, the goal of my thesis is to determine if acutely dissociated RTN cells can respond to changes in pH in isolation. To make this determination I used ratiometric fluorescent microscopy to measure intracellular calcium in dissociated RTN cells during changes in bath pH. I found that a small percentage of RTN cells (16%) respond to bath acidification from pH 7.3 to pH 6.9 with an increase in fluorescence indicating an increase in intracellular calcium. Preliminary electrophysiological findings suggest that responsive cells are unable to make action potentials, thus suggesting their identity to be glia. These results indicate that a subset of pH sensitive cells in the RTN are intrinsically pH sensitive and that glia cells may possibly play a role in central chemoreception.

Calcifying echinoid larvae respond to changes in seawater carbonate chemistry with reduced growth and developmental delay. To date, no information exists on how ocean acidification acts on pH homeostasis in echinoderm larvae. Understanding acid-base regulatory capacities is important because intracellular formation and maintenance of the calcium carbonate skeleton is dependent on pH homeostasis. Using H(+)-selective microelectrodes and the pH-sensitive fluorescent dye BCECF, we conducted in vivo measurements of extracellular and intracellular pH (pH(e) and pH(i)) in echinoderm larvae. We exposed pluteus larvae to a range of seawater CO(2) conditions and demonstrated that the extracellular compartment surrounding the calcifying primary mesenchyme cells (PMCs) conforms to the surrounding seawater with respect to pH during exposure to elevated seawater pCO(2). Using FITC dextran conjugates, we demonstrate that sea urchin larvae have a leaky integument. PMCs and spicules are therefore directly exposed to strong changes in pH(e) whenever seawater pH changes. However, measurements of pH(i) demonstrated that PMCs are able to fully compensate an induced intracellular acidosis. This was highly dependent on Na(+) and HCO(3)(-), suggesting a bicarbonate buffer mechanism involving secondary active Na(+)-dependent membrane transport proteins. We suggest that, under ocean acidification, maintained pH(i) enables calcification to proceed despite decreased pH(e). However, this probably causes enhanced costs. Increased costs for calcification or cellular homeostasis can be one of the main factors leading to modifications in energy partitioning, which then impacts growth and, ultimately, results in increased mortality of echinoid larvae during the pelagic life stage.

Varttuminen vietnamilaisena Suomessa: 12 vuoden seurantajakso – Vietnamilaisten hyvinvointi ja sosiokulttuurinen sopeutuminen lapsena/nuorena sekä nuorena aikuisena Tämä tutkimus oli määrällinen pitkittäistutkimus lapsena tai nuorena vuosina 1979-1991 Suomeen saapuneiden vietnamilaisten akkulturaatiosta (kulttuurin muutoksista), psyykkisestä hyvinvoinnista ja sosiokulttuurisesta sopeutumisesta. Tutkimukseen osallistui ensimmäisessä vaiheessa (vuonna 1992) 97 satunnaisesti valittua vietnamilaista peruskoululaista ympäri maata, joita verrattin suomalaisiin luokkatovereihin. Seurantavaiheeseen (vuonna 2004) osallistui 59 ensimmäisessä vaiheessa mukana ollutta vietnamilaista, nyt iältään 20 – 31 -vuotiaita. Tutkimuksen tavoitteena oli selvittää mitkä tekijät ennustivat akkulturaation lopputuloksia, samalla huomioiden iän ja ympäristön (kontekstin) vaikutukset psyykkiseen hyvinvointiin ja sosiokulttuuriseen sopeutumiseen. Yksittäiset akkulturaatiodimensiot (kieli, arvot ja identiteetti) osoittautuivat tärkeämmiksi psyykkiselle hyvinvoinnille ja sosiokulttuuriselle sopeutumiselle kuin etniset, kansalliset tai kaksikulttuuriset profiilit, joissa yhdistyivät ao. kieli, arvot ja identiteetti. Identiteettimuutosta tapahtui (etniseen) vietnamilaiseen suuntaan ajan kuluessa, kun taas arvomuutosta tapahtui (kansalliseen) suomalaiseen suuntaan. Sekä suomen että vietnamin kielen taito lisääntyivät ajan myötä, millä oli myönteisiä vaikutuksia sekä psyykkiseen hyvinvointiin että sosiokulttuuriseen sopeutumiseen. Lähtötilanteen psyykkinen hyvinvointi ennusti hyvinvointia (masennuksen puutetta ja itsetuntoa) aikuisena, mutta sosiokulttuurinen sopeutuminen (koulumenestys) lapsena tai nuorena ei ennustanut kouluttautumista aikuisena. Parempi suomen kielen taito ja vähemmän identifioitumista suomalaiseksi aikuisena sekä masentuneisuuden puute ja vähemmän koettua syrjintää lapsena tai nuorena erottelivat psyykkisesti paremmin voivat aikuiset (ei-masentuneet) heistä, jotka olivat masentuneita. Parempaa kouluttautumista aikuisena ennustivat toisaalta vähemmän koettua syrjintää lapsena tai nuorena ja toisaalta aikuisena parempi suomen kielen taito, suurempi kansallisten (suomalaisten) itsenäisyysarvojen kannattaminen, mutta kuitenkin vähemmän identifioitumista suomalaisiin. Koetun syrjinnän merkitys psyykkiselle hyvinvoinnille, erityisesti lapsena tai nuorena, sekä sen pitkäaikaisvaikutukset psyykkiselle hyvinvoinnille ja sosiokulttuuriselle sopeutumiselle aikuisena osoittavat tarpeen puuttua varhain psyykkisiin ongelmiin sekä parantaa etnisten ryhmien välisiä suhteita. The polarographic behaviour of glycine, α-alanine, β-alanine, valine, aspartic acid, glutamic acid and asparagine complexes of lead has been studied at various pH values and in presence of (1) NaOH, (2) Na2CO3 and (3) NH4 NO3+NH4OH. All the polarographic waves have been found to be reversible. 2. Experiments conducted on the effect of variation of pH, i.e., 7phic measurements for 18 complexes. 6. The suitability of incorporating amino acids in base solutions for the polarographic estimation of lead has been tested.

虾青素是一种极具开发潜力的类胡萝卜素,在食品、饲料、化妆品、医药等领域有着广阔的应用前景。利用雨生红球藻在特定条件下大量积累虾青素的能力进行生产是最具发展潜力的虾青素开发途径,受到了国内外学者的广泛关注。 随着雨生红球藻实验室研究工作的逐步成熟,人们开始将目光转向该藻的室外规模化培养。受雨生红球藻自身生物学特性和规模培养用光生物反应器设计的限制,在规模化培养过程中存在很多科学和技术问题,集中表现在如防治生物污染困难、培养规模难以扩大等方面。需要构建新型可规模放大的光生物反应器并研究其相关性能,探讨规模化培养过程中出现的某些生物学问题,为雨生红球藻规模化开发进一步提供应用基础。 通过多年的工作,首先构建了用于微藻培养的三种新型的光生物反应器(即柱式、管道、生物幕光生物反应器)。其中,上述柱式反应器结构简单、成本低廉,能够有效提高培养液的混合效果;管道反应器理论上可突破管道长度限制,大大超过传统管道反应器的培养体积;生物幕反应器从原理上解决了生物污染的问题,可以使红球藻细胞累积的虾青素含量大大提高。在此基础上,将上述三种光生物反应器有机搭配在一起,组成了从藻种、扩大培养、虾青素积累、到敌害生物防治和藻体收获等一整套工艺流程。该工艺技术能够较好的控制红球藻培养过程中的温度、光线、溶氧和pH等相关指标在适宜的范围内。并成功实现红球藻的规模化生产,藻粉质量优良。 本研究进一步分析了光线在雨生红球藻藻液中的光衰减特性。通过分析和拟合,研究了光强、光径、藻密度之间的关系,得出光线通过雨生红球藻藻液时的光衰减方程:I=I0exp[-(0.113+0.94OD750)L]。并利用此方程,结合两种规模化培养光生物反应器(柱式和管道)的构造得出了两种反应器的平均光强方程,分别为Iavg=I0[2αr0+3exp(-αr0)-(1+αr0)exp(-2αr0)-2]/(α2r02)和Iavg=2I0[αr0+exp(-αr0)-1]/(α2r02),其中α= 0.113+0.94OD750,R2=0.9893。利用这两个方程可以在规模化培养红球藻过程中,适时根据太阳光强度的变化调整反应器的入射光强,保证红球藻处于适宜的光照条件下。 另外,本研究还探讨了雨生红球藻在不同温度和光照条件下的混合营养生长。结果表明,在较高的温度24℃和较弱的光强24EM-2S-1条件下,红球藻维持较高的生长速率,可获得较高生物量。室外雨生红球藻的规模化混合营养培养实验表明,在较为寒冷的冬季和春季,混合营养培养能够大幅度提高雨生红球藻的生长速率和最终生物量,是一种非常有前途的规模化红球藻培养方式。 在上述工作基础上,比较了本技术与传统的开放池技术在不同季节规模化培养雨生红球藻生产虾青素的效果,前者比后者的年培养天数大约提高了3倍,在每个批次培养中细胞生长所需时间缩短了20%,藻生物量提高了至少1.75倍,虾青素累积所需时间缩短了40%,虾青素含量提高了4倍,全年培养红球藻生产虾青素的效率有很大提高(理论上共提高了35倍),较为成功地构建了一套雨生红球藻规模化培养模式。
