To explore whether an environment of weightlessness will cause damage to the reproductive system of animals, we used the tail-suspension model to simulate microgravity, and investigated the effect of microgravity on the tissue structure and function of the testis in sexually mature male rats. Forty-eight male Wistar rats weighing 200-250 g were randomly assigned to three groups (N = 16 each): control, tail traction, and tail suspension. After the rats were suspended for 7 or 14 days, morphological changes of testis were evaluated by histological and electron microscopic methods. The expression of HSP70, bax/bcl-2 and AR (androgen receptor) in testis was measured by immunohistochemistry. Obvious pathological lesions were present in the testis after the rats were suspended for 7 or 14 days. We detected overexpression of HSP70 and an increase of apoptotic cells, which may have contributed to the injury to the testis. The expression of AR, as an effector molecule in the testis, was significantly decreased in the suspended groups compared to control (P < 0.01). We also observed that, with a longer time of suspension, the aforementioned pathological damage became more serious and some pathological injury to the testis was irreversible. The results demonstrated that a short- or medium-term microgravity environment could lead to severe irreversible damage to the structure of rat testis.

Immune dysfunction is encountered during spaceflight. Various aspects of spaceflight, including microgravity, cosmic radiation, and both physiological and psychological stress, may perturb immune function. We sought to understand the impact of microgravity alone on the cellular mechanisms critical to immunity. Clinostatic RWV bioreactors that simulate aspects of microgravity were used to analyze the response of human PBMC to polyclonal and oligoclonal activation. PHA responsiveness in the RWV bioreactor was almost completely diminished. IL-2 and IFN-$\gamma$ secretion was reduced whereas IL-1$\beta$ and IL-6 secretion was increased, suggesting that monocytes may not be as adversely affected by simulated microgravity as T cells. Activation marker expression (CD25, CD69, CD71) was significantly reduced in RWV cultures. Furthermore, addition of exogenous IL-2 to these cultures did not restore proliferation. Antigen specific T cell activation, including the mixed-lymphocyte reaction, tetanus toxoid responsiveness, and Borrelia activation of a specific T cell line, was also suppressed in the RWV bioreactor.^ The role of altered culture conditions in the suppression of T cell activation were considered. Potential reduced cell-cell and cell-substratum interactions in the RWV bioreactor may play a role in the loss of PHA responsiveness. However, PHA activation in Teflon culture bags that limit cell-substratum interactions was not affected. Furthermore, increasing cell-population density, and therefore cell-cell interactions, in the RWV cultures did not help restore PHA activation. However, placing PBMC within small collagen beads did partially restore PHA responsiveness. Finally, activation of purified T cells with crosslinked CD2/CD28 or CD3/CD28 antibody pairs, which does not require costimulation through cell-cell contact, was completely suppressed in the RWV bioreactor suggesting a defect internal to the T cell.^ Activation of both PBMC and purified T cells with PMA and ionomycin was unaffected by RWV culture, indicating that signaling mechanisms downstream of PKC activation and calcium flux are not sensitive to simulated microgravity. Furthermore, sub-mitogenic doses of PMA alone but not ionomycin alone restored PHA responsiveness of PBMC in RWV culture. Thus, our data indicate that during polyclonal activation in simulated microgravity, there is a specific dysfunction within the T cell involving the signaling pathways upstream of PKC activation. ^

The experimental results obtained in experiment “STACO” made on board the Spacelab D-2 are re-visited, with image-analysis tools not then available. The configuration consisted of a liquid bridge between two solid supporting discs. An expected breakage occurred during the experiment. The recorded images are analysed and the measured behaviour compared with the results of a three dimensional model of the liquid dynamics, obtaining a much better fit than with linear models

The behaviour of confined liquids on board an orbiting spacecraft is mainly driven by surface tension phenomena, which cause an apparently anomalous response of the liquid when compared with the behaviour that can be observed on an Earth laboratory provided that the amount of liquid is high enough. The reason is that in an orbiting spacecraft the different inertial forces acting on the bulk of the liquid are almost zero, causing thus capillary forces to be the dominant ones. Of course, since gravity forces are proportional to the liquid volume, whereas surface tension forces are proportional to the liquid surface, there are situations on Earth where capillarity can be the dominant effect, as it happens when very small volume liquid samples are considered. However, work with small size samples may require the use of sophisticated optical devices. Leaving aside the neutral buoyancy technique, a way of handling large liquid interfaces is by using drop towers, where the sample falls subjected to the action of Earth's gravity. This approach is suitable when the characteristic time of the problem under consideration is much smaller than the drop time. In this work the transformation of an out-of-use chimney into a drop tower is presented. Because of the miniaturization, hardiness and low cost of current electronic devices, a drop tower can be used as an inexpensive tool for undergraduate students to experimentally analyse a large variety of surface tension driven phenomena.

The dynamics of inviscid, axisymmetric liquid bridges permits a simplified treatment if the bridge is long enough. Under such condition the evolution of the liquid zone is satisfactorily explained through a non-linear one-dimensional model. In the case of breaking, the one-dimensional model fails when the neck radius of the liquid column is close to zero; however, the model allows the calculation of the time variation of the liquid-bridge interface as well as of the fluid velocity field and, because the last part of the evolution is not needed, the overall results such as the breaking time and the volume of each of the two drops resulting after breakage can be calculated. In this paper numerical results concerning the behavior of clinical liquid bridges subjected to a small axial gravitational field are presented.

A feature of stability diagrams of liquid bridges between unequal disks subjected to small axial gravity forces is that, for each separation of disks, there is a value of microgravity for which an absolute minimum volume limit is reached. The dependence of such microgravity values on the liquid bridge geometry has been experimentally checked by using the neutral buoyancy technique, experimental results being in complete agreement with theoretical ones. Analytical background assuring the experimental procedure used is presented, and a second order analytical expression for the equilirium interface is also calculated.

This paper deals with the dynamics of liquid bridges when subjected to an oscillatory microgravity field. The analysis has been performed by using a one-dimensional slice model, already used in liquid bridge problems, which allows to calculate not only the resonance frequencies of a wide range of such fluid configurations but also the dependence of the dynamic response of the liquid bridge on the frequency on the imposed perturbations. Theoretical results are compared with experimental ones obtained aboard Spacelab-Dl, the agreement between theoretical and experimental results being satisfactory

Besides space laboratories for in-orbit experimentation, Earth based facilities for laboratory experimentation are of paramount importance for the enhancement on liquid bridge knowledge. In spite of the constraints imposed by simulated microgravity (which force to work either with very small size liquid bridges or by using the Plateau tank technique, amongst other techniques), the availability and accessibility of Earth facilities can circumvent in many cases the drawbacks associated with simulated microgravity conditions. To support theoretical and in orbit experimental studies on liquid bridges under reduced gravity conditions, several ground facilities were developed at IDR. In the following these ground facilities are briefly described, and main results obtained by using them are cited.
