Background and Aims The frequency at which males can be maintained with hermaphrodites in androdioecious populations is predicted to depend on the selfing rate, because self-fertilization by hermaphrodites reduces prospective siring opportunities for males. In particular, high selfing rates by hermaphrodites are expected to exclude males from a population. Here, the first estimates are provided of the mating system from two wild hexaploid populations of the androdioecious European wind-pollinated plant M. annua with contrasting male frequencies.Methods Four diploid microsatellite loci were used to genotype 19-20 progeny arrays from two populations of M. annua, one with males and one without. Mating-system parameters were estimated using the program MLTR.Key Results Both populations had similar, intermediate outcrossing rates (t(m) = 0.64 and 0.52 for the population with and without males, respectively). The population without males showed a lower level of correlated paternity and biparental inbreeding and higher allelic richness and gene diversity than the population with males.Conclusions The results demonstrate the utility of new diploid microsatellite loci for mating system analysis in a hexaploid plant. It would appear that androdioecious M. annua has a mixed-mating system in the wild, an uncommon finding for wind-pollinated species. This study sets a foundation for future research to assess the relative importance of the sexual system, plant-density variation and stochastic processes for the regulation of male frequencies in M. annua over space and time.

The objective of this work was to estimate outcrossing rates between Haden and Tommy Atkins mango cultivars, using AFLP and microsatellite markers. Progenies of an isolated 'Haden' plant, identified in a 'Tommy Atkins' commercial orchard, in Petrolina, PE, Brazil, were analyzed. Total DNA was isolated from the progeny leaves and used for AFLP and microsatellite reactions. Multilocus outcrossing rates (t m) were estimated by direct count of AFLP or microsatellite markers and by the mLTR software. Outcrossing rates ranged from 0.85 to 0.87 with the analysis based on seven AFLP markers, and from 0.83 to 0.91 based on three microsatellite primers. No unexpected band patterns were observed for 'Haden' and 'Tommy Atkins'. The estimates obtained with the mLTR software were close to those obtained by direct AFLP and microsatellite allele counting, which indicates that the multilocus model was appropriate for this kind of study. The microsatellites mMiCIR005, mMiCIR030, and mMiCIR036 can be used to elucidate the origin of 'Haden' and 'Tommy Atkins' seedlings.

O umbuzeiro é uma das espécies mais importantes do semiárido brasileiro, devido à sua capacidade de produzir frutos em ambiente de estresse hídrico. O objetivo deste estudo foi estimar taxas de polinização cruzada em umbuzeiro (
), considerando-se a frequência de heterozigotos observada e esperada nas populações maternal e de descendentes (M1), respectivamente, e a estimativa de multilocos (M2), de forma a orientar programas de preservação e melhoramento genético da espécie. Amostras de DNA extraído de uma população maternal de 96 plantas, estabelecidas no campo em 1991, em Petrolina, PE, bem como DNA de um indivíduo descendente de cada planta-mãe, foram analisadas para 16 bandas polimórficas das combinações de primer de AFLPAAA_CTG e AAA_CTC. No M1, considerou-se a frequência de heterozigotos estimada pela raiz quadrada de 1 menos a frequência de ausência de fragmentos AFLP, tanto para a população maternal (frequência observada) quanto para a população de descendentes (frequência esperada), enquanto no M2 a estimativa foi obtida por estimativa multilocos (
m). As estimativas de
no M1 variaram de 1,74 a 0,50, com média de 1,063. Como valores de
acima de 1,0 são biologicamente inaceitáveis, ficou demonstrado que M1 não foi apropriado para estimar
. As frequências de óvulos e pólen para 15 locos de AFLP foram iguais, indicando boa adequação do modelo de acasalamento misto no M2. A taxa
m obtida pelo M2 foi de 0,719, próxima de estimativas prévias com isoenzimas em outras populações da espécie. Os resultados indicam que o umbuzeiro é predominantemente de polinização cruzada e há necessidade de amostras amplas para preservar a variabilidade genética da espécie.
