Stigmaphyllom paralias is a herb belonging to the family Malpighiaceae that occurs in sand soil of brazilian " restinga". This is the first report regarding phytochemical study with this species. The hexane extract of the aerial parts of plant afforded the triterpenes friedelin, lupenone, 3-oxo-alpha-amirin and 3-oxo-beta-amirin, the mixture of alpha-amirinyl palmitate and stearate, lupeol and 3,4-seco-friedelan-3-oic acid. The AcOEt extract yielded the flavonoid luteolin-7-rutinoside. All compounds were characterized by analysis of spectrometric data and the fatty acids esterified with alpha-amirine were identified by GC/MS of methyl derivatives of transesterified products . This is the first natural occurence of 3,4-seco-friedelan-3-oic acid and the 13C NMR spectral data were inequivocally assigned by two-dimensional techniques. This work also permitted to correct the 13C NMR resonances attributed to methyl groups C-26 and C-27 of fridelin.

This work describes the phytochemical study of hexane extracts from the stem of Maprounea guianensis. Besides 3-oxo-21alpha-H-hop-22(29)-en (moretenone), beta-sitosterol, lupenone and lupeol, a mixture of dodecosyl, tetracosyl, hexacosyl, octacosyl and triacontyl ferulates was also isolated, as well as 3-beta-acetoxy-lup-20(29)-en-28-oic acid, 3beta-O-trans-p-coumaroyl-lup-20(29)-en-28-oic acid and 3beta-O-trans-p-coumaroyl-urs-12-en-28-oic acid. The structures of these compounds were established by spectroscopic analysis.

Three mixtures of triterpenes (maniladiol and breine; alpha and beta-amyrin; lupenone, alpha and beta-amyrinone) were isolated from Protium heptaphyllum March resin. The structural identification was based on NMR and mass spectrometry data. Lupenone, and alpha and beta-amyrinone were not reported before as constituents of this resin. The resin was submitted to methylation and acetylation reactions. The pure and derivatized resins and the mixtures (maniladiol and breine; alpha and beta-amyrin) were analyzed by TG and DSC. The TG curves revealed that the derivatization decreases the thermal stability of the resin. The DSC curves showed peaks that can be assigned to evaporation and phase transitions processes.

Eleven oleanane, ursane and lupane-type triterpenes were isolated from the leaves of Terminalia brasiliensis Camb, daturadiol (3b,6beta-dihydroxy-olean-12-ene), 3beta-hydroxy-30-norlupan-20-one, lupenone, beta-amyrenone, alpha-amyrenone, lupeol, beta-amyrin, alpha-amyrin, betulin, erythrodiol and uvaol, in addition to squalene, sitosterol and alpha-tocopherol. The structures of these compounds were identified by ¹H and 13C NMR spectral analysis and comparison with literature data.

Phytochemical investigation of a propolis sample from Alto Santo - Ceará, Brazil, allowed identification of triterpenes (lupeol, lupenone, germanicone, canaric acid) and flavonoids (quercetin, kaempferol and acacetin), which were identified by spectroscopic data (IR, MS, and NMR, including 2D techniques). This is the first report of canaric acid in propolis. Propolis extract and flavonoids showed antioxidant activity using a DPPH radical scavenging assay.

A new triterpene, 1-epi-castanopsol, besides eleven known compounds: sitosterol, stigmasterol, campesterol, lupeol, lupenone, simirane B, syringaresinol, scopoletin, isofraxidin, 6,7,8-trimethoxycoumarin and harman, were isolated from the wood of Simira glaziovii. The structures of the known compounds were defined by 1D, 2D ¹H, 13C NMR spectra data analyses and comparison with literature data. The detailed spectral data analyses allowed the definition of the structure of the new 1-epi isomer of castanopsol and performance of ¹H and 13C NMR chemical shift assignments.

The chemical investigation of hexane and ethanol extracts from the aerial parts of Vernonia scorpioides resulted in the isolation and characterization of a new polyacetylene lactone, rel-4-dihydro-4β-hydroxy-5a-octa-2,4,6-triynyl-furan-2-(5H)-one, along with the new ethyl 3,4-dihydroxy-6,8,10-triynyl-dodecanoate, and seven known compounds: taraxasteryl acetate, lupeyl acetate, lupeol, lupenone, β-sitosterol, stigmasterol and luteolin. The structure of all compounds was determined by spectrometric techniques (HR-ESI-MS, ¹H and 13C NMR and IV) and comparison with published spectral data.

Ampelozizyphus amazonicus Ducke is a tree commonly found in the Amazon region and an extract of its stem bark is popularly used as an antimalarial and anti-inflammatory agent and as an antidote to snake venom. Ursolic acid; five lupane type triterpenes: betulin, betulinic acid, lupenone, 3ß-hydroxylup-20(29)-ene-27,28-dioic acid, and 2a,3ß-dihydroxylup-20(29)-ene-27,28-dioic acid, and three phytosteroids: stigmasterol, sitosterol and campesterol, have been isolated from stem extracts of A. amazonicus Ducke. Their structures were characterized by spectral data including COSY and HMQC. In an in vitro biological screening of the isolated compounds, 3ß-hydroxylup-20(29)-ene-27,28-dioic acid was cytotoxic against the SKBR-3 human adenocarcinoma cell line (1 to 10 mg/mL), while 2a,3ß-dihydroxylup-20(29)-ene-27,28-dioic acid exhibited cytotoxicity against both SKBR-3 human adenocarcinoma and C-8161 human melanoma tumor cell lines (>0.1 mg/mL). In the present study, different extracts and some fractions of this plant were also investigated for trypanocidal activity due to the presence of pentacyclic triterpenes. The triterpene classes are potent against Trypanosoma cruzi. The bioassays were carried out using blood collected from Swiss albino mice by cardiac puncture during the parasitemic peak (7th day) after infection with the Y strain of T. cruzi. The results obtained showed that A. amazonicus is a potential source of bioactive compounds since its extracts and fractions isolated from it exhibited in vitro parasite lysis against trypomastigote forms of T. cruzi at concentrations >100 µg/mL. Fractions containing mainly betulin, lupenone, 3ß-hydroxylup-20(29)-ene-27,28-dioic acid, and 2a,3ß-dihydroxylup-20(29)-ene-27,28-dioic acid showed more activity than crude extracts.

Ethanol extracts of four propolis samples (E1-E4) from Manaus (Brazilian Amazon) were analysed by HPLC/DAD/ESI-MS/MS and GC/EIMS. The major constituents of E2 and E4 were analysed by NMR ((1)H and (13)C) and ESI/MS/MS. The main constituents of E2 and E4 are polyprenylated benzophenones: 7-epi-nemorosone, 7-epi-clusianone (major E4 constituents), xanthochymol and gambogenone (major E2 constituents), making up a chemical profile so far unreported for Brazilian propolis. Aristhophenone, methyl insigninone, 18-ethyloxy-17-hydroxy-17,18-dihydroscrobiculatone B, and derivatives of dimethyl weddellianone A and B, propolones, and a scrobiculatone derivative, were detected as minor constituents. Triterpenoids (beta-amyrins, beta-amyrenone, lupeol and lupenone) were ubiquitous and predominant in El and E3. The extracts E2 and E4 were highly active against the cariogenic bacteria Streptococcus mitis, Streptococcus mutans and Streptococcus salivarius. E2 was more active than E4, probably due to a higher content of 2-epi-nemorosone, while the latter was richer in di-hydroxylated compounds. (C) 2010 Elsevier Ltd. All rights reserved.

Recently, marine organisms have attracted attention because of the complexity and potent biological activity from your secondary metabolites. Our planet has 80% it surface covered by oceans and seas, therefore, housing a wide number of different forms of life, among them, the sponges. These sessile and filtrating animals, according to numerous researches, come showing like true chemistry factories. The substances from these animals, sometimes show as news targets to therapeutics agents, and some countries has already use them for treatment of some diseases. Further of the secondary metabolites, the polysaccharides of marine origin also have been target of studies, because the presence of the sulfates groups in its molecules. Polysaccharides with differents biological activities have been related in a large number of researches. Actually, many studies show the sponges as source of promising medicine. These studies inspire new researches, because the few number of sponges species studied until now. Because of that, the present work shows the chemistry prospection of the sponge Callyspongia vaginalis. Chromatographic methods in silica gel allowed the isolations of two secondary metabolites: the known β- sitosterol and a ceramide, no reported in the genus Callyspongia, previously. The analysis of the their lipid extracts show different kinds of fatty acids with a variety of chain length (saponifiable fraction), and others metabolites like Lupenone and stigmasterol, also unprecedented in the genus. The Polysaccharide characterization and the elucidation of the secondary metabolites acquired through of chromatography analysis (CC, molecular exclusion) and spectrometric (NMR 1H and 13C, mass, IR), respectively and comparison with literature data

Phytochemical investigations of the stems of a specimen of Alibertia macrophylla led to the isolation and characterization of the new diterpene ent-kaurane-2β,3α,16α-triol (1), along with triterpenes 2-8, iridoids 9-12, and phenolic acids 13-15. The structure of 1 was established based on spectroscopic studies (1H- and 13C-NMR, IR, and HR-ESI-MS). This is the first report of the isolation of a diterpene from the Alibertia genus in Rubiaceae. © 2007 Verlag Helvetica Chimica Acta AG.

Estudos fitoquímicos com as cascas do caule e com as folhas de Croton palanostigma Klotzsch (Euphorbiaceae) levaram ao isolamento do novo diterpeno clerodânico 8-epicordatina (2), além de éster metílico do ácido 12-oxohardwickiico (3), aparisthmano, cordatina (1), ácido ent-trachiloban-18-óico, óxido de ent-13-epimanoila, óxido de ent-3-oxo-13-epimanoila, óxido de ent-3β-hidroxi-13-epimanoila, sitosterol, estigmasterol, estigmastan-3-ona, 6β-hidroxiestigmast-4-en-3-ona, 6β-hidroxiestigmasta-4,22-dien-3-ona, estigmast-4-en-3-ona, estigmasta-4,22-dien-3-ona, ácido 3-O-acetilaleuritolico, 11α-hidroxiurs-12-en-3-ona, α-amirenona, 24-metilenocicloartenona e lupenona. Estas substâncias foram isoladas através de procedimentos fitoquímicos usuais e suas estruturas foram deduzidas por estudos espectroscópicos, incluindo experimentos em 2D. Adicionalmente, a estrutura cristalina de 8-epicordatina (2) foi determinada por difração de raios-X. Cálculos teóricos de RMN ao nível B3PW91/DGDZVP foram usados para confirmação dos assinalamentos dos deslocamentos químicos dos hidrogênios H-7α e H-7β de 8-epicordatina.

Os objetivos deste trabalho foram isolar, identificar e caracterizar a atividade alelopática de substâncias químicas produzidas por Acacia mangium, além de determinar as variações na atividade das substâncias em função da variação do pH da solução. A atividade alelopática foi avaliada em bioensaios de germinação (25 ºC de temperatura e fotoperíodo de 12 horas) e crescimento de radícula e hipocótilo (25 ºC de temperatura e fotoperíodo de 24 horas) das plantas daninhas malícia (Mimosa pudica) e mata-pasto (Senna obtusifolia). Avaliou-se a interferência do pH (3,0 e 9,0) da solução na atividade alelopática das substâncias sobre a germinação das sementes da espécie malícia. Os triterpenoides lupenona (3-oxolup-20(29)-eno) e lupeol (3β-hidroxilup-20(29)-eno), obtidos das folhas caídas da planta doadora, isolados e em par, evidenciaram baixo efeito alelopático inibitório da germinação de sementes e do crescimento do hipocótilo, especialmente do primeiro, cujos efeitos não ultrapassaram o valor de 2,0%. Os efeitos promovidos sobre o crescimento da radícula foram de maior magnitude, atingindo valores superiores a 40%, com destaque para as inibições promovidas pela substância lupenona. Isoladamente, as substâncias promoveram efeitos superiores aos efetivados pelas substâncias analisadas em pares, indicando a existência de antagonismo. O pH da solução influenciou a atividade alelopática das substâncias; para lupenona os efeitos foram mais intensos em pH ácido, enquanto para lupeol os melhores resultados foram verificados em condições alcalinas, mostrando que este fator é ponto importante a ser considerado em trabalhos de campo.
