Caspase-3 is synthesized as a dormant proenzyme and is maintained in an inactive conformation by an Asp-Asp-Asp “safety-catch” regulatory tripeptide contained within a flexible loop near the large-subunit/small-subunit junction. Removal of this “safety catch” results in substantially enhanced autocatalytic maturation as well as increased vulnerability to proteolytic activation by upstream proteases in the apoptotic pathway such as caspase-9 and granzyme B. The safety catch functions through multiple ionic interactions that are disrupted by acidification, which occurs in the cytosol of cells during the early stages of apoptosis. We propose that the caspase-3 safety catch is a key regulatory checkpoint in the apoptotic cascade that regulates terminal events in the caspase cascade by modulating the triggering of caspase-3 activation.

Räjähdysvaaralliset tilat aiheuttavat lisävaatimuksia instrumentointisuunnittelulle. Tämän diplomityön tavoitteena on kartoittaa räjähdysvaarallisten tilojen standardien vaatimuksia instrumentoinnin suunnitteluun ja laitevalintoihin. Tarkemmin tarkastellaan luonnostaan vaaratonta räjähdyssuojausrakennetta, joita instrumentoinnin laitteilla tyypillisesti käytetään. Luonnostaan vaarattomassa räjähdyssuojausrakenteessa sähkölaitteen energiaa ja pintalämpötilaa rajoitetaan niin, ettei laite voi sytyttää räjähdysvaarallista seosta. Työssä perehdytään myös luonnostaan vaarattomia laitteita sisältäviin instrumentointipiireihin ja standardin niille asettamiin vaatimuksiin. Tällaisille piireille on tehtävä varmennustarkastelu, jossa todennetaan piirin täyttävän vaatimukset. Varmennusta varten diplomityön käytännönosuutena kehitetään varmennussovellus, joka suorittaa varmennuksen tietokannassa olevista arvoista.

In Australia, protection orders are a key legal response to domestic violence, and are often viewed as a way of providing for victim safety. For instance, recently the joint Australian and New South Wales Law Reform Commissions recommended that a common core purpose of all state and territory domestic violence legislation should be ‘to ensure or maximise the safety and protection of persons who fear or experience family violence’ (2010:Recommendation 7-4). Drawing and building upon prior research in Australia and the United States (‘US’), this paper uses comparative quantitative content analysis to assess the victim safety focus of domestic violence protection order legislation in each Australian state and territory. The findings of this analysis show that the Northern Territory, South Australia and Victoria ‘stand out’ from the other jurisdictions, having the highest victim safety focus in their legislation. However, there remains sizeable scope for improvement in all Australian jurisdictions, in terms of the victim safety focus of their legislative provisions and the considerations of legislative inconsistency between jurisdictions.

Future air traffic management concepts often involve the proposal of automated separation management algorithms that replaces human air traffic controllers. This paper proposes a new type of automated separation management algorithm (based on the satisficing approach) that utilizes inter-aircraft communication and a track file manager (or bank of Kalman filters) that is capable of resolving conflicts during periods of communication failure. The proposed separation management algorithm is tested in a range of flight scenarios involving during periods of communication failure, in both simulation and flight test (flight tests were conducted as part of the Smart Skies project). The intention of the conducted flight tests was to investigate the benefits of using inter-aircraft communication to provide an extra layer of safety protection in support air traffic management during periods of failure of the communication network. These benefits were confirmed.

Electrostatic discharges have been identified as the most likely cause in a number of incidents of fire and explosion with unexplained ignitions. The lack of data and suitable models for this ignition mechanism creates a void in the analysis to quantify the importance of static electricity as a credible ignition mechanism. Quantifiable hazard analysis of the risk of ignition by static discharge cannot, therefore, be entirely carried out with our current understanding of this phenomenon. The study of electrostatics has been ongoing for a long time. However, it was not until the wide spread use of electronics that research was developed for the protection of electronics from electrostatic discharges. Current experimental models for electrostatic discharge developed for intrinsic safety with electronics are inadequate for ignition analysis and typically are not supported by theoretical analysis. A preliminary simulation and experiment with low voltage was designed to investigate the characteristics of energy dissipation and provided a basis for a high voltage investigation. It was seen that for a low voltage the discharge energy represents about 10% of the initial capacitive energy available and that the energy dissipation was within 10 ns of the initial discharge. The potential difference is greatest at the initial break down when the largest amount of the energy is dissipated. The discharge pathway is then established and minimal energy is dissipated as energy dissipation becomes greatly influenced by other components and stray resistance in the discharge circuit. From the initial low voltage simulation work, the importance of the energy dissipation and the characteristic of the discharge were determined. After the preliminary low voltage work was completed, a high voltage discharge experiment was designed and fabricated. Voltage and current measurement were recorded on the discharge circuit allowing the discharge characteristic to be recorded and energy dissipation in the discharge circuit calculated. Discharge energy calculations show consistency with the low voltage work relating to discharge energy with about 30-40% of the total initial capacitive energy being discharged in the resulting high voltage arc. After the system was characterised and operation validated, high voltage ignition energy measurements were conducted on a solution of n-Pentane evaporating in a 250 cm3 chamber. A series of ignition experiments were conducted to determine the minimum ignition energy of n-Pentane. The data from the ignition work was analysed with standard statistical regression methods for tests that return binary (yes/no) data and found to be in agreement with recent publications. The research demonstrates that energy dissipation is heavily dependent on the circuit configuration and most especially by the discharge circuit's capacitance and resistance. The analysis established a discharge profile for the discharges studied and validates the application of this methodology for further research into different materials and atmospheres; by systematically looking at discharge profiles of test materials with various parameters (e.g., capacitance, inductance, and resistance). Systematic experiments looking at the discharge characteristics of the spark will also help understand the way energy is dissipated in an electrostatic discharge enabling a better understanding of the ignition characteristics of materials in terms of energy and the dissipation of that energy in an electrostatic discharge.

La Comunidad de Madrid ha tenido un pasado minero importante que ha dejado su vestigio en el territorio. Actualmente la explotación de minerales no metálicos y de rocas industriales y ornamentales continúa, pero no así la de la minería metálica que en el pasado fue abundante. Hoy en día todas estas minas están abandonadas, dando lugar a zonas degradadas con un elevado riesgo para la seguridad de las personas y de los animales terrestres que habitan por sus inmediaciones, y necesitan ser intervenidas atendiendo, además, a su integración o recuperación ecológica y paisajística en el territorio. El principal propósito de esta tesis es conocer la situación actual que presentan las minas abandonadas de metales y proponer unos modelos de actuación para la rehabilitación e integración de los espacios mineros abandonados de la Comunidad de Madrid, cuya ventaja también estriba en su posible extrapolación a otras aéreas de gestión con una problemática similar. Partiendo de una selección de cincuenta y siete minas de interior metálicas abandonadas, se ha hecho un diagnostico en función de su seguridad (riesgo), interés cultural, arqueológico e histórico y por su afección a espacios protegidos, resultando que en todas, excepto en tres de ellas, es preciso llevar a cabo medidas protectoras y de restauración e integración en el medio ambiente. El conjunto de minas catalogadas de alto riesgo para la seguridad son veintitrés, y sobre ellas se ha realizado un Análisis Clúster, en el que además de los criterios de gestión formulados: seguridad, protección del patrimonio minero-industrial e integración ecológico-paisajística, se han incorporado otros modificadores como distancia a poblaciones, caminos, pistas y vías pecuarias, y accesibilidad. Con los resultados de este análisis se obtienen una clasificación por grupos de las minas en relación con las características intrínsecas de las explotaciones preseleccionadas y la tipología de problemas que presentan, y a partir de ella se plantean soluciones viables que se concretan en la redacción de una serie de anteproyectos tipo. Una de las principales aportaciones es que se trata de un modelo de inventariocaracterización- actuación extrapolable a otros entornos similares con problemáticas parecidas. Las propuestas de actuación que figuran en los anteproyectos tipo se proponen como medidas aplicables en situaciones similares. La tesis además también incorpora una base de datos georreferenciada que permite la localización de las explotaciones mineras abandonadas, el acceso rápido a sus características y a la propuesta de restauración correspondiente. ABSTRACT The Community of Madrid had an important mining past that left its traces in the territory. Currently the explotation of non-metallic minerals and industrial and ornamental rocks continues, but not so much with the metal deposits that were abundant in the past. Today these mines are abandoned, resulting in degraded areas with a high risk to the safety of nearby people and animals that inhabit its vicinity, and they need to be intervened, by tending to their environment integration or ecological and landscape recovery. The main objective of this thesis is to know the current situation of abandoned metal mines and propose an action model for the rehabilitation and integration of abandoned mining areas of the Community of Madrid, whose advantage also is its possible extrapolation to other management areas with similar problems. From a selection of fifty-seven abandoned metal mines, a diagnosis has been made, based on safety (risk), cultural, archaeological and historical interests and how it can be related to protected areas, resulting that all but three of them need measures of protection, restoration and integration to the environment. The set of mines that are classified as a high security risk are twenty-three, and it has been made on them an Cluster Analysis, as in addition to the management formulated criteria, such as safety, protection of mining and industrial heritage and eco-landscape integration, it has been incorporated other modifying variables, such as distance to populations, roads, tracks and livestock paths, and accessibility. With the results of this analysis, a classification of similar groups of mines in relation with the intrinsic characteristics of the preselected mines and typology problems, and from it a set of viable solutions will be specified, being then drafted on a series of proposals. One of the main contributions of this research is that it consists of an inventorycharacterization- action model, which can be extrapolated to other similar environments with similar problems. The action measures that are contained in the proposals are defined as applicable to similar situations. The thesis also incorporates a geo-referenced database that allows finding the location of abandoned mines, thus giving quick access to its features and its corresponding restoration proposal.

While knowledge about standardization of skin protection against ultraviolet radiation (UVR) has progressed over the past few decades, there is no uniform and generally accepted standardized measurement for UV eye protection. The literature provides solid evidence that UV can induce considerable damage to structures of the eye. As well as damaging the eyelids and periorbital skin, chronic UV exposure may also affect the conjunctiva and lens. Clinically, this damage can manifest as skin cancer and premature skin ageing as well as the development of pterygia and premature cortical cataracts. Modern eye protection, used daily, offers the opportunity to prevent these adverse sequelae of lifelong UV exposure. A standardized, reliable and comprehensive label for consumers and professionals is currently lacking. In this review we (i) summarize the existing literature about UV radiation-induced damage to the eye and surrounding skin; (ii) review the recent technological advances in UV protection by means of lenses; (iii) review the definition of the Eye-Sun Protection Factor (E-SPF®), which describes the intrinsic UV protection properties of lenses and lens coating materials based on their capacity to absorb or reflect UV radiation; and (iv) propose a strategy for establishing the biological relevance of the E-SPF. © 2013 John Wiley & Sons A/S.

This paper discusses the development and the application of a safety performance indicator which measures the intrinsic safety of a country's vehicle fleet related to fleet composition. The indicator takes into account both the ‘relative severity’ of individual collisions between different vehicle types, and the share of those vehicle types within a country's fleet. The relative severity is a measure for the personal damage that can be expected from a collision between two vehicles of any type, relative to that of a collision between passenger cars. It is shown how this number can be calculated using vehicle mass only. A sensitivity analysis is performed to study the dependence of the indicator on parameter values and basic assumptions made. The indicator is easy to apply and satisfies the requirements for appropriate safety performance indicators. It was developed in such a way that it specifically scores the intrinsic safety of a fleet due to its composition, without being influenced by other factors, like helmet wearing. For the sake of simplicity, and since the required data is available throughout Europe, the indicator was applied to the relative share of three of the main vehicle types: passenger cars, heavy goods vehicles and motorcycles. Using the vehicle fleet data from 13 EU Member States and Norway, the indicator was used to rank the countries’ safety performance. The UK was found to perform best in terms of its fleet composition (value is 1.07), while Greece has the worst performance with the highest indicator value (1.41).

Buckler sorrel (Rumex induratus Boiss. & Reut.) is an underutilized leafy vegetable with peculiar sensory properties and potential as a gourmet food. In the food industry, different packaging methods have been used for shelf-life extension, but it is important to know how the quality of minimally processed vegetable is affected by these treatments. Recently, nitrogen and argon have been used for food packaging. Nitrogen is low soluble in water and other food constituents and does not support the growth of aerobic microbes. In turn, argon is biochemically active and appears to interfere with enzymatic oxygen receptor sites. In this study, modified atmospheres enriched with nitrogen and argon were evaluated for shelf-life extension of buckler sorrel leaves. Wild samples were gathered in Bragança, Portugal, considering local consumers’ sites and criteria. Healthy and undamaged leaves were selected, rinsed in tap water, and a portion was immediately analyzed (non-stored control). The remaining fresh material was packaged in polyethylene bags under nitrogen- and argon-enriched atmospheres and a conventional control atmosphere (air). All packaged samples were stored at 4 ºC for 12 days and then analyzed. The headspace gas composition was monitored during storage. Different quality attributes were evaluated, including visual (colour), nutritional (macronutrients, individual sugars and fatty acids) and bioactive (hydrophilic and lipophilic molecules and antioxidant properties) parameters. Different statistical tools were used; the one-way analysis of variance (ANO VA) was applied for analyse the differences among treatments and a linear discriminant analysis (LDA ) was used to evaluate the effects on the overall postharvest quality. The argon-enriched atmosphere better prevent the samples yellowing. The proximate composition did not change significantly during storage. Samples in control atmosphere revealed higher protein and ash contents and lower levels of lipids. The non-stored control samples presented the higher amounts of fructose, glucose and trehalose. The storage time increased the palmitic acid levels and decreased the content in α-linolenic and linoleic acids. The γ- e δ-tocopherols were higher after the 12 days of cold storage. Probably, the synthesis of these lipophilic compounds was a plant strategy to fight against the abiotic stress induced by storage. Higher levels of total phenolics and flavonoids and increased reducing power and β-carotene bleaching inhibition capacity were also found in the stored control samples. Once again, this result may be attributed to the intrinsic plant-protection mechanisms. Overall, the argon atmosphere was more suitable for quality preservation and shelf-life extension of buckler sorrel.

The Accelerator Driven Subcritical Reactor (ADSR) is one of the reactor designs proposed for future nuclear energy production. Interest in the ADSR arises from its enhanced and intrinsic safety characteristics, as well as its potential ability to utilize the large global reserves of thorium and to burn legacy actinide waste from other reactors and decommissioned nuclear weapons. The ADSR concept is based on the coupling of a particle accelerator and a subcritical core by means of a neutron spallation target interface. One of the candidate accelerator technologies receiving increasing attention, the Fixed Field Alternating Gradient (FFAG) accelerator, generates a pulsed proton beam. This paper investigates the impact of pulsed proton beam operation on the mechanical integrity of the fuel pin cladding. A pulsed beam induces repetitive temperature changes in the reactor core which lead to cyclic thermal stresses in the cladding. To perform the thermal analysis aspects of this study a code that couples the neutron kinetics of a subcritical core to a cylindrical geometry heat transfer model was developed. This code, named PTS-ADS, enables temperature variations in the cladding to be calculated. These results are then used to perform thermal fatigue analysis and to predict the stress-life behaviour of the cladding. © 2011 Elsevier Ltd. All rights reserved.

Achieving higher particles energies and beam powers have long been the main focus of research in accelerator technology. Since Accelerator Driven Subcritical Reactors (ADSRs) have become the subject of increasing interest, accelerator reliability and modes of operation have become important matters that require further research and development in order to accommodate the engineering and economic needs of ADSRs. This paper focuses on neutronic and thermo-mechanical analyses of accelerator-induced transients in an ADSR. Such transients fall into three main categories: beam interruptions (trips), pulsed-beam operation, and beam overpower. The concept of a multiple-target ADSR is shown to increase system reliability and to mitigate the negative effects of beam interruptions, such as thermal cyclic fatigue in the fuel cladding and the huge financial cost of total power loss. This work also demonstrates the effectiveness of the temperature-to-reactivity feedback mechanisms in ADSRs. A comparison of shutdown mechanisms using control rods and beam cut-off highlights the intrinsic safety features of ADSRs. It is evident that the presence of control rods is crucial in an industrial-scale ADSR. This paper also proposes a method to monitor core reactivity online using the repetitive pattern of beam current fluctuations in a pulsed-beam operation mode. Results were produced using PTS-ADS, a computer code developed specifically to study the dynamic neutronic and thermal responses to beam transients in subcritical reactor systems. © 2012 Elsevier B.V.

Desde janeiro de 2009 que passou a ser obrigatório a implementação das Medidas de Autoproteção (MA) em edifícios e recintos, conforme o Regulamento Técnico de Segurança contra Incêndio em Edifícios (RT-SCIE). Pretendemos com este trabalho apresentar um estudo descritivo, para o quinquénio de 2008 a 2012, sobre a implementação das MA num complexo escolar ocupado por cerca de 300 funcionários e 2000 alunos/crianças com idades compreendidas entre os 3 e os 18 anos. É importante salientar que o complexo está classificado na 4.ª categoria de risco de incêndio e é constituído por vários edifícios, na sua maioria catalogados na Utilização-Tipo IV (de acordo com o Decreto-Lei 220/2008). As MA foram materializadas em várias etapas: 1. Diagnóstico das condições de segurança; 2. Registos de Segurança; 3. Plano de Segurança Interno (PSI); 4. Ações de sensibilização em SCIE para todos os ocupantes; 5. Ações de formação específica em SCIE para a Equipa de Segurança; 6. Realização de Simulacros de Evacuação com periodicidade anual. Com base no trabalho anteriormente desenvolvido e com o que se encontra presentemente em curso, foram estabelecidos os seguintes objetivos: • Caraterizar e analisar as MA implementadas no Complexo Escolar; • Avaliar o contributo das ações de formação em SCIE para a adoção dos procedimentos de prevenção e de emergência, por parte dos elementos da Equipa de Segurança; • Avaliar a eficácia dos Simulacros na preparação para a emergência; • Identificar dificuldades e propor oportunidades de melhoria que facilitem a evacuação de emergência. Em termos de metodologia, optámos pela Observação Direta Não Participante, mediante a utilização de Listas de Verificação preenchidas durante a realização dos Simulacros, que constituem uma ferramenta essencial de monitorização da eficácia do PSI. Apesar da dimensão, da complexidade e destas instalações escolares serem muito anteriores ao RT-SCIE, as conclusões preliminares apontam para uma bem sucedida implementação das MA.

The purpose of this study envisaged the quantification of skin and respiratory exposures occasioned by work conditions during pesticide spraying of citrus fruits using hand gun sprayers; the evaluation of the efficiency of individual and group protection measures for the workers; the determination of workers' most exposed body regions; and the classification of work conditions, with and without the tested work-safety protection measures as recommended for the registered pesticides used to control the main pests and diseases that attack these types of trees and fruits. The AZR protection equipment proved to be the most efficient for the tractor driver, when spraying using pistol sprayers. The two sets of individual protection equipment that were checked also proved to be efficient. The most exposed regions of the tractor driver's body were the thighs, the front of the legs, the feet and hands. The most exposed regions of the individual sprayer working on foot were the hands and feet.
