Um estudo foi conduzido com a finalidade de determinar a possibilidade de clonagem do cajueiro (Anacardium occidentale) por alporquia e a influência do AIB (ácido indolbutírico) nesse processo. Adotou-se delineamento experimental inteiramente casualizado, com 4 tratamentos, 10 alporques por parcela, repetidos por 4 vezes, num total de 160 alporques. Os tratamentos constaram das concentrações de AIB: 0 (testemunha), 1.000, 3.000 e 5.000 mg.kg-1. Foram avaliadas as percentagens de sobrevivência, calejamento e enraizamento, bem como número e comprimento médio de raízes. A maior percentagem de sobrevivência (67,5%) foi observada para a testemunha e concentração de 1.000 mg.kg-1, enquanto a melhor percentagem de enraizamento (82%) foi relacionada com o nível de 1.000 mg.kg-1. Para o número e comprimento médio de raízes, os melhores resultados foram concernentes à dose de 5.000 mg.kg-1. Não houve influência do AIB na clonagem do cajueiro por alporquia.

Rooted cutting propagation is widely used for maximising tree yield, quality and uniformity in conjunction with clonal selection. Some eucalypt species are deployed as rooted cuttings but many are considered to difficult to root. This study examined IBA effects on photoinhibition, root formation, mortality and root and shoot development in cuttings of Corymbia torelliana, C. citriodora and their hybrids. IBA had little or no effect on photoinhibition but it had strong, dose-dependent effects on root formation and mortality. IBA frequently increases primary root number of rooted cutting but it did not increase total root weight, length, surface area or volume, possibly because the highest doe (8g IBA/kg IBA/kg powder) caused leaf abscission and sometimes reduced leaf area (by 55-79%)or shoot dry weight (by 40-58%). An intermediate dose (3g IBA/kg powder) most consistnely improved root formation with little or no effect on mortality or shoot development. Across the F1 hybrid families this treatment increased the number of rooted cuttings by 72-121% and more than ddoubled the number of primary roots per rooted cutting (from 1.1-1.7 roots to 3.5-4.1 roots). This simple treatment will facilitate commercial multiplication of superior individuals or selected families of C. torelliana x C. citriodora through a vegetative propagation system.

Cytenamide forms a 1:1 solvate with butyric acid [systematic name: 5H-dibenzo[a,d]cycloheptatriene-5-carboxamide-butanoic acid (1/1)], C16H13NO center dot C4H8O2. The title compound crystallizes with one molecule of cytenamide and one of butyric acid in the asymmetric unit; these molecules are linked by N-H center dot center dot center dot O and O-H center dot center dot center dot O hydrogen bonds to form an R-2(2)(8) heterodimer motif. Pairs of adjacent motifs are further connected via N-H center dot center dot center dot O interactions to form a discrete centrosymmetric assembly.

Aloysia triphylla (L'Hérit) Britton is a perennial and bushy plant, with simple, entire, lanceolate and whorl shaped leaves and originally from South America. It is used as medicinal plant in Brazil with stomatic and sedative properties. The employment of stem cuttings for propagation of pre-selected plants, acquired great importance, because it eliminates the juvenile phase of seedlings, which can be produced in a shorter period of time. The rooting of stem cuttings is stimulated by auxin and, boric acid supply is essential for growth and development of initial rootlets. This micronutrient is required 48 hours after plant segments have been placed into auxin solution and it can be supplied any time, including the seedling growth period. The experiment was carried out in the Department of Plant Production, UNESP-Botucatu-SP-Brazil, with stem-cuttings of Aloysia triphylla (L'Hérit) Britton, Verbenaceae obtained from the Medicinal and Aromatic Plant Garden. The aim of the work was to verify the influence of growth regulators and boric acid on stem cutting rooting of this species. The 15 cm-stem cuttings, without leaves, were submerged during 24 hs in the following solutions: water; 150 mg.L-1 of IBA; 150 mg.L-1 of IBA+ Boric acid; 250 mg.L-1 of IBA; 250 mg.L-1 of IBA + Boric acid. The statistical design was entirely randomized with 5 treatments and 3 replications, totalizing 15 plots with 10 stem cuttings each. They were planted on propylene trays with vermiculite and kept under spraying condition during twenty five days. The best results were observed in treatment 250 mg.L -1 of IBA+ Boric acid on number of roots, length of roots, rooting percentage, fresh and dry weight of leaves when compared with all other treatments. We can conclude that this treatment is the most suitable for stem cutting rooting of this species.

The aim of this study was to investigate the ultrastructural characteristics of primordial follicles after culturing of sheep ovarian cortical slices in the presence of indol acetic acid (IAA), Epidermal Growth Factor (EGF), and FSH. To evaluate ultrastructure of primordial follicles cultured in MEM (control) or in MEM containing IAA, EGF, and FSH, fragments of cultured tissue were processes for transmission electron microscopy. Except in the control, primordial follicles cultured in supplemented media for 6d were ultrastructurally normal. They had oocyte with intact nucleus and the cytoplasm contained heterogeneous-sized lipid droplets and numerous round or elongated mitochondria with intact parallel cristae were observed. Rough endoplasmic reticulum (RER) was rarely found. The granulosa cells cytoplasm contained a great number of mitochondria and abundant RER. In conclusion, the presence of IAA, EGF, and FSH helped to maintain ultrastructural integrity of sheep primordial follicles cultured in vitro. © 2011 Evelyn Rabelo Andrade et al.

During maturation, pollen undergoes a period of dehydration accompanied by the accumulation of compatible solutes.Solute import across the pollen plasma membrane, which occurs via proteinaceous transporters, is required to support pollen development and also for subsequent germination and pollen tube growth. Analysis of the free amino acid composition of various tissues in tomato revealed that the proline content in flowers was 60 times higher than in any other organ analyzed. Within the floral organs, proline was confined predominantly to pollen, where it represented >70 of total free amino acids. Uptake experiments demonstrated that mature as well as germinated pollen rapidly take up proline. To identify proline transporters in tomato pollen, we isolated genes homologous to Arabidopsis proline transporters. LeProT1 was specifically expressed both in mature and germinating pollen, as demonstrated by RNA in situ hybridization. Expression in a yeast mutant demonstrated that LeProT1 transports proline and γ-amino butyric acid with low affinity and glycine betaine with high affinity. Direct uptake and competition studies demonstrate that LeProT1 constitutes a general transporter for compatible solutes.

Feed characteristics may influence the bacterial community composition and metabolic activities in the pig gastrointestinal tract, known to be associated with positive effects on the gut. Use of mash feed is associated with reduced Salmonella excretion, but little is known of its effect on the Escherichia coli population or of the mechanism of action. Our objectives were to assess the effect of feed texture combined with feed particle size on VFA profiles and levels, total E. coli count, and the presence of genes encoding virulence factors of pathogenic E. coli strains in the digestive tract along with their impact on pig performance of fattening pigs. Pigs (n = 840) on a commercial farm received mash or pellet diets of different particle sizes during the fattening period. Caecal and colon contents from 164 pigs were sampled at the slaughterhouse for enumeration of E. coli by quantitative PCR (qPCR) and for VFA quantification by capillary gas chromatography. The yccT gene was used to enumerate total E. coli. Improved pig performances associated with pellet texture and a 500-μm size were observed. Caecal (P = 0.02) and colon (P < 0.01) propionic acid concentrations were lower for pigs receiving pellet rather than mash feed. Similarly, caecal (P = 0.01) and colon (P < 0.001) butyric acid concentrations were also lower for pigs receiving pellet rather than mash feed, as determined by capillary gas chromatography. Moreover, caecal (P = 0.03) and colon (P < 0.001) butyric acid concentrations were higher for pigs receiving a feed with a 1,250-μm particle size rather than a 500-μm particle size. On the other hand, total caecal and colon E. coli levels were higher for pigs receiving pellet feed than for those receiving mash feed. For total E. coli enumeration, caecal (P < 0.01) and colon (P < 0.01) yccT gene copies were higher for pigs receiving pellet rather than mash feed. No effect of particle size on fatty acid concentrations or on E. coli numbers was observed. Virulence gene quantification revealed no trend. Taken together, results showed that mash feed is associated with lower growth performance but with favorable intestinal changes linked to VFA levels and E. coli reduction in the intestine.

The consumption of exotic fruits has been showing accentuated increase and the cultivated area is in expansion, generating demand for adequate culturing techniques. The Malay apple (Syzygium malaccense), with probable origin in India, has a fruit widely known and appreciated in the North and Northeasthern Brazilian states. The Malay apple tree is extremely tall and has a long juvenile period when propagated by seed, making its vegetative multiplication is desirable, to anticipate the productive period and decrease its size, and also to obtain uniform orchards. The experiment was conducted at UNESP/FCAV, Jaboticabal Campus, using Malay apple herbaceous cuttings subjected to treatments with indol butyric acid (IBA) (0; 1,000; 3,000 and 5,000 mgL(-1)) and cuttings with and without basal incision. The variables analyzed were percentage of survival and rooting of the cuttings, number and mean length of roots per cutting. The experiment was conducted under CRB on a factorial scheme (4 X 2) with 4 replicates constituted by 10 cuttings each. Data were analyzed by Tukey's mean test at 5% probability. The vegetative propagation by rooting of herbaceous cuttings of the Malay apple is possible, however, both IBA treatments and basal incision have not shown significant effect on the analyzed variables.

A produção de mudas de caramboleira é um dos fatores limitantes à expansão comercial da cultura, devido ao tempo que estas levam para serem formadas e iniciarem a produção. Uma boa alternativa pode estar na otimização dos métodos de propagação vegetativa, através de estudos relativos aos processos e fatores envolvidos no enraizamento de estacas. Este trabalho foi desenvolvido com o objetivo de estudar o enraizamento de estacas apicais e basais de caramboleira, tratadas com ácido indolbutírico (IBA), em condições de nebulização intermitente. Estacas apicais e basais de caramboleira foram coletadas de ramos de plantas-matrizes da cultivar B-10 e submetidas à aplicação de cinco concentrações de IBA (0; 1.000; 3.000; 5.000 e 7000 mg.L-1), em imersão por 10 segundos, para avaliar a capacidade de sobrevivência, enraizamento e número médio de raízes/estaca. Posteriormente, as estacas foram colocadas em caixas de madeira contendo vermiculita média como substrato e mantidas em casa de vegetação, sob nebulização intermitente, durante 70 dias. O delineamento experimental utilizado foi em esquema fatorial 2 x 5, com 4 repetições e 10 estacas/parcela. As estacas apicais obtiveram melhores resultados para porcentagem de sobrevivência (49,26%), enraizamento (34,84%) e número médio de raízes (20,51), mostrando-se superiores às basais. O uso de IBA não influenciou em nenhuma das variáveis analisadas.

O trabalho objetivou avaliar a propagação da figueira com estacas semilenhosas, empregando-se concentrações de ácido indolbutírico, em diferentes épocas de coleta. O material propagativo utilizado foi proveniente de figueiras da cv. Roxo de Valinhos, com 5 anos de idade. As estacas foram retiradas por ocasião da poda hibernal, no final dos meses de agosto, setembro e outubro, as quais foram tratadas com AIB preparado em pó, nas seguintes concentrações: 0 (testemunha); 2.500; 5.000; 7.500 e 10.000 mg kg-1. Posteriormente, foram colocadas para enraizar em bandejas de polipropileno, tendo como substrato vermiculita e mantidas sob nebulização intermitente por 70 dias. Decorrido esse período, avaliaram-se a porcentagem de estacas enraizadas (%), o comprimento da maior raiz (cm) e a massa seca das raízes (g). O delineamento experimental utilizado foi o inteiramente casualizado, em esquema fatorial 3 x 5. Observou-se que estacas provenientes do tratamento-testemunha e da poda de agosto apresentaram baixa porcentagem de enraizamento (20%), menor comprimento e massa seca de raiz, necessitando de tratamento com AIB na concentração de 2.500 mg kg-1, que aumentou significativamente esta porcentagem (90%). Estacas oriundas da poda de setembro e outubro não necessitaram de tratamento com AIB para conseguirem um elevado enraizamento. O mês de setembro foi o mais viável para a coleta de estacas de figueira, pois houve maior porcentagem de enraizamento das estacas (95-100%). As estacas tratadas com 7.500 e 10.000 mg kg-1 de AIB apresentaram as mais baixas porcentagens de enraizamento nos meses de agosto e outubro.

A propagação do pessegueiro no Brasil baseia-se na enxertia de cultivares-copa em porta-enxertos propagados por sementes, e uma alternativa de propagação para esta frutífera poderia ser a estaquia. O objetivo deste trabalho foi avaliar o potencial de enraizamento de estacas semilenhosas de cultivares de pessegueiro através da aplicação de 2,6-di-hidroxiacetofonona (2,6-DHAP) antes do tratamento com AIB (ácido indolbutírico). As estacas foram preparadas a partir de ramos coletados das cultivares Delicioso Precoce, Jóia 1 e Okinawa, em dezembro de 2001, para serem tratadas na base com 2,6-DHAP (0 e 300mg L-1), por 4h, em aeração e depois com AIB (200mg L-1 e 2500mg L-1). As estacas foram plantadas em bandejas de poliestireno expandido com vermiculita fina e colocadas em casa de nebulização, por 45 dias. A aplicação de 300mg L-1 de 2,6-DHAP antes da aplicação de 200mg L-1 de AIB em estacas de 'Okinawa' proporcionou aumentos nos resultados das características de maior relevância para a propagação por estacas e pode ser uma técnica interessante para a estaquia em pessegueiro.

A propagação do pessegueiro (Prunus persica) no Brasil é baseada na enxertia sobre porta-enxertos oriundos de sementes. Outros métodos de propagação de frutíferas poderiam ser utilizados para o pessegueiro visando obtenção de materiais de melhor qualidade. Neste trabalho objetivou-se avaliar o efeito do 2,6-di-hidroxiacetofenona aplicado previamente ao ácido indol-butírico em estacas semi-lenhosas de pessegueiro da cultivar Okinawa preparadas com diferentes tipos de corte basal. Foram coletados ramos do porta-enxerto Okinawa em dezembro de 2001 para o preparo das estacas sem folhas, com 12cm de comprimento, 7mm de diâmetro, quatro gemas e diferentes tipos de cortes basais (corte longitudinal, corte lateral e corte da casca) tratado-as na base com 0 e 300mg L-1 de 2,6-di-hidroxiacetofenona por 4h em aeração (oxigenação) e depois com 2500mg L-1 ácido indol-butírico por 5s. As estacas foram plantadas em bandejas de poliestireno expandido (72 células) com vermiculita fina e colocadas em casa de nebulização por 45 dias. A aplicação de 300mg L-1 de 2,6-DHAP e a realização de lesões nas bases das estacas foram eficientes para aumentar o enraizamento das estacas semi-lenhosas do pessegueiro Okinawa , demonstrando que estas técnicas podem ser utilizadas para a propagação de pessegueiro por estaquia.
