Alloxan induced diabetic animal model was used to evaluate the antidiabetic effect of alkaloids extracted from the leaves of Aegis marine/ose. The alkaloid extract maintained the weight of animals near to that of control ones - whereas there was a decrease in the body weight of diabetic animals. A significant increase in blood glucose (342. 14 -+- 14.89 mg/dl) was seen in diabetic animals but in alkaloid treated group the blood glucose was lowered (90: 12 +_5.81 mg/dl). There was no decrease in blood urea arid sreum cholesterol in the alkaloid treated group of diabetic animals. The liver glycogen decreased in diabetic animals (1.27+.12 g/100g of wet tissue) and the treatment brought the glycogen level to that of control ones (2.51 +.75 g/100 g of wet tissue). The result show that the alkaloid extract has hypoglycaemic activity.

Berberine has been shown to have hypoglycaemic activity in several in vitro and in vivo models, although the mechanism of action is not fully known. Berberis lyceum Royle root produces high concentrations of berberine, and in traditional medicine, the whole extract of this plant is used widely to treat diabetes. The antidiabetic activity of the ethanol root extract of Berberis lyceum was compared with pure berberine in normal and alloxan-diabetic rats using similar doses of each. The concentration of berberine in the extract was determined to be 80% dry weight with only trace amounts of other alkaloids present. The purpose of the study was to investigate the effects of berberine and a whole extract of Berberis lyceum on blood glucose and other parameters associated with diabetes, to compare the effects of the crude extract with those of pure berberine and thus validate its use as a therapeutic agent, and finally to identify any contribution of the other components of the extract to these effects. Oral administration of 50 mg/kg of Berberis extract and berberine to normal and experimental diabetic rats produced a significant (p < 0.05) reduction in blood glucose levels from days 3-7 days of treatment. Significant effects were also observed on the glucose tolerance, glycosylated haemoglobin, serum lipid profiles and body weight of experimental animals. Berberis extract and berberine demonstrated similar effects on all parameters measured, and although the extract was comparable in efficacy to berberine, it did not produce any effects additional to those shown by pure berberine. The results support the use of the extract in traditional medicine, and demonstrate that apart from being a highly cost-effective means of treating with berberine, the total extract does not appear to confer any additional benefits or disadvantages compared with the pure compound. La pharmacopée Cris est riche en plantes médicinales et plusieurs d’entre elles sont étudiées par notre laboratoire pour leur potentiel antidiabétique. Certaines espèces ont démontré leur capacité à stimuler la protéine kinase activée par l’AMP (AMPK), une enzyme qui favorise la translocation de transporteurs de glucose à la membrane (effet hypoglycémiant). L’AMPK stimule également d’autres fonctions, telle l’oxydation des graisses, dans le but de rétablir l’énergie cellulaire. Ce projet a comme objectifs d’évaluer, premièrement, le stress métabolique induit par huit des extraits dans des cellules musculaires et des hépatocytes, effet qui serait responsable de l’activation de l’AMPK. Ce stress peut être déterminé en mesurant l’acidification du milieu extracellulaire ainsi que la déplétion du contenu en ATP des cellules suite aux traitements. Le deuxième objectif est de mesurer l’efficacité des extraits à réduire le contenu en gras (oxydation des graisses) et à ainsi normaliser la résistance à l’insuline dans des hépatocytes rendus insulino-résistants. Les hépatocytes sont rendus résistants à l’insuline (condition fortement lié à l’obésité) via traitement avec un acide gras saturé, le palmitate. Les résultats montrent que la majorité des extraits semble induire un stress métabolique de courte durée dans les cellules. Parmi les extraits, seul un a réussi à faire diminuer significativement le taux de triglycérides intracellulaire suite au traitement au palmitate sans toutefois améliorer la sensibilité à l’insuline. En conclusion, le potentiel hypoglycémiant des extraits serait du à leur capacité à affecter la respiration mitochondriale (stress métabolique). Toutefois, leur capacité à améliorer la sensibilité à l’insuline n’a pu être établie.

Alloxan induced animal model was used to evaluate the potential antidiabetic effect of A . marmelose leaf extract. The diabetic animals were given insulin injection and another group A . marmelose leaf extract orally. It maintained the weight of the animals near to the control rats but a significant decrease in weight was noted it diabetic animals without any treatment . The blood elucose level in 'treated animals were near to that of control ones. Also a significantly increased glucose tolerance was observed in animals orally given the leaf extract prior to the experiment . A significant decrease in liver glycogen ( 1.24 -.07 s!/ 100 g of wet tissue) was observed in diabetic rats which was brought to almost the normal level (I.84- . 14 g; 100 g) with Icaf extract treatment. Blood urea and serum cholesterol increased ( 62.66 - 3.50 and 192.67± 13.64 mg' dl) significantly in alloxxan diabetic rats . The leaf extract treatment decreased the blood urea and serum . cholesterol (37.83 - 3.97 and 99?0±8.43 mg/dl ) to that of control ones . A similar effect was seen with insulin treatment . The results indicate that the active principle in .4. marmelose leaf extract has similar hypoglycaemic activity to insulin treatment.

Dentre as numerosas terapias para minimizar as complicações diabéticas, os antioxidantes e flavonoides são testados na clínica médica. Foi analisado o efeito da naringerina sobre os parâmetros bioquímicos em ratos diabéticos induzidos por estreptozotocina (STZ - 60mg/kg, i.p.). Ratos machos foram divididos em 4 grupos: G1: controle não tratado; G2: ratos normais que receberam naringerina; G3: diabéticos não tratados; G4: ratos diabéticos que receberam naringerina. Naringerina (50mg/kg, i.p.), decresceu a hiperglicemia e a hiperlipidemia em ratos diabéticos. A concentração sérica de insulina em ratos tratados tendeu aumentar. A naringerina preveniu as alterações, provocadas pela estreptozotocina, na atividade hepática e cardíaca de ALT, AST e LDH, indicando o efeito protetor da naringerina sobre estes tecidos, contra toxicidade provocada pela STZ. O nível de glicogênio nos tecidos cardíaco e hepático elevou com a naringerina em ratos diabéticos. A naringerina melhorou o metabolismo da glicose e de lipídios e preveniu as complicações diabéticas.

The purpose of the present study was to discover the relative potency of onion, Allium cepa, with respect to its hypoglycaemic and hypolipidaemic effects on the diabetic situation, and the association of these effects with the potential against oxidative stress. Male Wistar rats were divided into four groups. A normal control ( group A), and a non-diabetic group ( group B) were treated daily with 1 ml A. cepa solution (0.4 g A. cepa/rat). Groups C and D were made diabetic by an intraperitoneal injection of streptozotocin (STZ) ( 60 mg/kg body weight) in citrate buffer ( pH 6.3). These animals ( groups C and D) were the STZ diabetic control and STZ diabetic rats with onion intake, respectively. Onion increased the fasting serum high-density lipoprotein levels, and demonstrated alleviation of hyperglycaemia in STZ diabetic rats. The hypoglycaemic and hypolipidaemic actions of A. cepa were associated with antioxidant activity, since onion decreased superoxide dismutase activities while no increased lipid hydroperoxide and lipoperoxide concentrations were observed in diabetic rats treated with A. cepa.

In the present study a detailed investigation on the alterations of dopamine and its receptors in the brain regions of streptozotocin induced diabetic and insulin induced hypoglycaemic rats were carried out. Glutamate receptor, NMDARI gene expression in the hypoglycaemic and hyperglycaemic brain was also studied. EEG recording in hypoglycaemic and hyperglycaemic will be carried out to measure brain activity. in vitro studies on glucose uptake and insulin secretion, with and without specific antagonists were carried out to confirm the specific receptor subtypes - DA D1, DA D2 and NMDA involved in the functional regulation during hyperglycaemic and hypoglycaemic brain damage. The molecular studies on the brain damage through dopaminergic and glutamergic receptors will elucidate the therapeutic role in the corrective measures of the damage to the brain during hypoglycaemia and hyperglycaemia. This has importance in the management of diabetes and antidiabetic treatment for better intellectual functioning of the individual.

Two hundred and eighty-five botanically identifide plant materials have been extracted with 50 percent ethanol and the extracts put through a wide biological screen of 61 tests. These include tests for antibacterial, antifungal, anthelminthic, antiprotozoal, anticancer, antifertillity, hypoglycaemic and a wide range of pharmacological activities.
