Determinaram-se os valores do hemograma de 187 eqüinos sadios da raça Pantaneira. Para a composição de seis grupos experimentais consideraram-se a faixa etária e o sexo. Verificaram-se 6,2± 1,2 a 8,9± 2,3 hemácias/µl (x10(6)), 32,0± 4,0 a 37,0± 2,8% de volume globular (VG), 11,5± 1,0 a 12,8± 1,6g de teor de hemoglobina/dl, 11,0± 3,2 a 17,0± 2,8 leucócitos/µl (x 10³), 4,9± 1,8 a 7,5± 1,5 neutrófilos segmentados/µl (x 10³) e 5,2± 2,0 a 8,6± 2,0 linfócitos/µl (x 10³). Observaram-se valores mais baixos na contagem de hemácias, e VG e valores mais altos do volume globular médio (VGM) em cavalos castrados (acima de 25 meses de idade). As contagens de leucócitos e neutrófilos segmentados em potros com até oito meses de idade apresentaram valores mais elevados que os encontrados em animais acima dos 25 meses de idade. Constatou-se que não houve influência do sexo nos parâmetros analisados.

Complete blood counts and hemoglobin isoform data were gathered from 36 specimens of the turtle species Phrynops geoffroanus from the northwestern region of São Paulo State, Brazil. They were collected in an urban area. The hemoglobin profiles were obtained after red blood cell lysis and by electrophoretic migration in alkaline pH, acid pH, and neutral pH buffer. The hemoglobin components were confirmed using high-performance liquid chromatography (HPLC). Erythrogram analysis included hematocrit, total hemoglobin concentration, total red blood cell count, and red blood cell indices. The leukogram included a total white blood cell count and a calculation of the percent values of neutrophils, lymphocytes, monocytes, basophils, eosinophils, heterophils, and azurophils. HPLC analysis revealed three hemoglobin components; the first with a concentration of 5.5%, the second was a major component with an average concentration of 67.1%, and the third with a concentration of 28.5%. The hematological profile obtained for these specimens allowed us to establish a pattern for P. geoffroanus in São Paulo State Northwestern region. The average hematocrit values were 22.5% for females and 24.0% for males. For total hemoglobin, we found average values of 6.66 g/dL in females and 7.22 g/dL in males. The number of white blood cells was 2725 x 103/μL for females and 2775 x 103/μL for males. There was a predominance of heterophils, eosinophils, and monocytes in both sexes. No significant differences were found between males and females for hematological profile. The hematological results were compared to literature data for other Chelonia. They were similar to what is known for fresh water turtles. © FUNPEC-RP.

Studies of the hemoglobin pattern in Brazilian reptiles are important for determining ecological and phylogenetic relationships, but they are scarce. Peripheral blood samples were obtained from 7 males and 18 females of Rhinoclemmys punctularia. The hematological profile was based on the total hemoglobin and hematocrit values. The hemoglobin profile was obtained using electrophoretic procedures at different pH, isoelectric focusing, globin chain electrophoresis, and HPLC. The hematocrit (31 ± 2%) and total hemoglobin (7.5 ± 0.2 g/dL) values did not indicate gender variations. Alkaline pH electrophoresis of the total blood samples treated with 1% saponin demonstrated the presence of four well-defined hemoglobin fractions, one major component (fraction I), showing cathodic migration and three others faster than fraction I with anodic migration. When the samples were precipitated with chloroform, only two hemoglobin fractions were observed, similar to fractions I and III from the first procedure. Isoelectric focusing and HPLC showed the same pattern. With acid and neutral pH electrophoresis, two fractions with anodic migration were observed. The globin chain identification at alkaline pH showed two fractions, but four fractions were observed at acidic pH, suggesting that different polypeptide chains are involved in the hemoglobin molecule. The chromatographic separation of the total blood sample demonstrated that the major fraction comprised 81.9% and the minor 18.1%. The results obtained demonstrated a similarity between these hemoglobin components and those of some Chelidae reported in the literature for both land and aquatic animals, reflecting the adaptation to environmental conditions. ©FUNPEC-RP.

The accelerated growth of finfish aquaculture has resulted in a series of health problems, including blood disorders by hemoparasites; there are scarce studies about these agents and their impact in actual intensive farming. The aim of this study was to identify and quantify the hemoparasites present in monocytes and erythrocytes in the blood of tilapia and their correlation with the hematological profile. Blood samples were collected from caudal vessels of 15 cage-reared Nile tilapia (Oreochromis niloticus), with 70 ± 10 g weight on average, from Itambaracá Municipality, Parana State, Brazil. The total red blood cells, mean corpuscular volume, hematocrit, total and differential white blood cells counts, and the number of thrombocytes were determined in blood smears stained with May-Grünwald-Giemsa-Wright and Quick Panoptic. The results showed the presence of pleomorphic cytoplasmic inclusions with corrugated appearance and basophilic staining, mainly in monocytes, suggesting Anaplasmataceae parasitemia and inclusions with the same morphological characteristics in erythrocytes of one Nile tilapia. The hematological analysis showed no significant difference (P < 0.05) between infected and not infected fish, and therefore, there was no correlation between parasitemia and hematological profile. These observations allow us to infer that the intracytoplasmic inclusions in monocytes and erythrocytes are compatible with the family Anaplasmataceae. There was no correlation between the blood profile and low level of parasitemia. © 2012 Springer-Verlag London.

Ultra-endurance exercise, such as an Ironman triathlon, induces muscle damage and a systemic inflammatory response. As the resolution of recovery in these parameters is poorly documented, we investigated indices of muscle damage and systemic inflammation in response to an Ironman triathlon and monitored these parameters 19 days into recovery. Blood was sampled from 42 well-trained male triathletes 2 days before, immediately after, and 1, 5 and 19 days after an Ironman triathlon. Blood samples were analyzed for hematological profile, and plasma values of myeloperoxidase (MPO), polymorphonuclear (PMN) elastase, cortisol, testosterone, creatine kinase (CK) activity, myoglobin, interleukin (IL)-6, IL-10 and high-sensitive C-reactive protein (hs-CRP). Immediately post-race there were significant (P < 0.001) increases in total leukocyte counts, MPO, PMN elastase, cortisol, CK activity, myoglobin, IL-6, IL-10 and hs-CRP, while testosterone significantly (P < 0.001) decreased compared to prerace. With the exception of cortisol, which decreased below prerace values (P < 0.001), these alterations persisted 1 day post-race (P < 0.001; P < 0.01 for IL-10). Five days post-race CK activity, myoglobin, IL-6 and hs-CRP had decreased, but were still significantly (P < 0.001) elevated. Nineteen days post-race most parameters had returned to prerace values, except for MPO and PMN elastase, which had both significantly (P < 0.001) decreased below prerace concentrations, and myoglobin and hs-CRP, which were slightly, but significantly higher than prerace. Furthermore, significant relationships between leukocyte dynamics, cortisol, markers of muscle damage, cytokines and hs-CRP after the Ironman triathlon were noted. This study indicates that the pronounced initial systemic inflammatory response induced by an Ironman triathlon declines rapidly. However, a low-grade systemic inflammation persisted until at least 5 days post-race, possibly reflecting incomplete muscle recovery.

Objective: The present study was undertaken to evaluate the antitumor and antioxidant status of ethanol extract of Terminalia catappa leaves against Ehrlich ascites carcinoma (EAC) in Swiss albino mice. Materials and Methods: The leaves powder was extracted with Soxhlet apparatus and subjected to hot continuous percolation using ethanol (95% v/v). Tumor bearing animals was treated with 50 and 200 mg/kg of ethanol extract. EAC induced in mice by intraperitoneal injection of EAC cells 1 x 10(6) cells/mice. The study was assed using life span of EAC-bearing hosts, hematological parameters, volume of solid tumor mass and status of antioxidant enzymes such as lipid peroxidation (LPO), reduced glutathione (GSH), superoxide dismutase (SOD) and catalase (CAT) activities. Total phenolics and flavonoids contents from the leaves extract were also determined. Results: Total phenolics and flavonoids contents from the leaves extract were found 354.02 and 51.67 mg/g extract. Oral administration of ethanol extract of T. catappa (50 and 200 mg/kg) increased the life span (27.82% and 60.59%), increased peritoneal cell count (8.85 +/- 0.20 and 10.37 +/- 0.26) and significantly decreased solid tumor mass (1.16 +/- 0.14 cm(2)) at 200 mg/kg as compared with EAC-tumor bearing mice (P < 0.01). Hematological profile including red blood cell count, white blood cell count, hemoglobin (11.91 +/- 0.47 % g) and protein estimation were found to be nearly normal levels in extract-treated mice compared with tumor bearing control mice. Treatment with T. catappa significantly decreased levels of LPO and GSH, and increased levels of SOD and CAT activity (P < 0.01). Conclusion: T. catappa exhibited antitumor effect by modulating LPO and augmenting antioxidant defense systems in EAC bearing mice. The phenolic and flavonoid components in this extract may be responsible for antitumor activity.

Tetrasomy, pentasomy, and hexasomy 8 (polysomy 8) are relatively rare compared to trisomy 8. Here we report on a series of 12 patients with acute myeloid leukemia (AML), myelodysplastic syndrome (MDS), or myeloproliferative disorder (MPD) associated with polysomy 8 as detected by conventional cytogenetics and fluorescence in situ hybridization (FISH). In an attempt to better characterize the clinical and hematological profile of this cytogenetic entity, our data were combined with those of 105 published patients. Tetrasomy 8 was the most common presentation of polysomy 8. In 60.7% of patients, polysomy 8 occurred as part of complex changes (16.2% with 11q23 rearrangements). No cryptic MLL rearrangements were found in cases in which polysomy 8 was the only karyotypic change. Our study demonstrates the existence of a polysomy 8 syndrome, which represents a subtype of AML, MDS, and MPD characterized by a high incidence of secondary diseases, myelomonocytic or monocytic involvement in AML and poor overall survival (6 months). Age significantly reduced median survival, but associated cytogenetic abnormalities did not modify it. Cytogenetic results further demonstrate an in vitro preferential growth of the cells with a high level of aneuploidy suggesting a selective advantage for polysomy 8 cells.

Sickle Cell disease is a generic term for a group of genetic disorders characterized by the predominance of hemoglobin S. These disorders include Sickle Cell anemia, the Sickle Cell beta Thalassemia syndromes and Hemoglobinopathies in which hemoglobin S is in association with another abnormal hemoglobin, such as hemoglobin S/C. The Sickle Cell trait (hemoglobin AS) associated with Alpha Thalassemia presents alterations in the red blood cells morphology, usually absent in the heterozygous for this hemoglobin variant. The interaction between hemoglobin Sand alpha Thalassemia has been described as one of the factors responsible for the improvement in the clinical picture of homozygous of hemoglobin S (Sickle Cell Anemia), decreasing the number of episodes of pain. The genetic mechanisms of this influence are evaluated using molecular analyses of the human globin genes. With the objective of verifying the presence of alpha Thalassemia in heterozygous of hemoglobin S, with anemia, sent to the Laboratory of Hemoglobins, Department of Biology, UNESP, São José do Rio Preto, SP, we analyzed 1002 blood samples with Sickle Cell trait, in the period from 1990 to 1998. The samples were picked with EDTA 5% as anticoagulant, after previous authorization of the carriers. Appropriated counseling and management requires definitive diagnosis. For the laboratorial diagnosis the blood samples were submitted to electrophoretic procedures in alkaline and acid pH and cytological evaluation of hemoglobin H. The electrophoretic procedures confirmed the presence of hemoglobin AS. The cytological evaluation evidenced the presence of alpha Thalassemia. Of this total analyzed, 16(1,59%) blood samples presented the association between hemoglobin AS and alpha Thalassemia and two individuals belonged of the same family. Our results addressed us to suggest to the routine laboratories, that is important to accomplish the research of alpha Thalassemia among the Sickle Cell trait, with anemia, to verify the interaction with alpha Thalassemia, supplying to the carriers a important information on its hematological profile, genetic pattern of hemoglobinopathies and the appropriated counseling. A dengue é causada pelo Vírus da dengue (VDEN - Flaviviridae, Flavivirus) e é considerada a arbovirose mais difundida no mundo. Atualmente, não há um animal que sirva como modelo para estudos sobre a patogênese do VDEN. Para investigar a susceptibilidade da espécie Callithrix penicillata ao VDEN foram inoculados amostras do VDEN-3 e do VDEN-2 de isoladas de casos humanos fatais. Para tal, vinte e dois animais foram infectados com VDEN-3 (3,23 × 103 PFU/mL) e, sessenta dias após a infecção (d.p.i.), 11 deles foram secundariamente infectados com VDEN-2 (4,47 × 104 PFU/mL). Sangue, plasma e soro, foram coletados diariamente durante os primeiros sete dias e em 15, 20, 45 e 60 d.p.i.. Foram investigados a produção de anticorpos IgM e inibidores da hemaglutinação, assim como foram análisados parâmetros hematologicos e bioquímicos e o perfil sérico de citocinas (IL-6, TNF-, IL-2, IFN-α, IL-4 e IL-5). A infecção primária (VDEN-3) revelou anticorpos IgM (15- 20 d.p.i.), anticorpos IH (15-60 d.p.i.), aumento dos níveis de TNF-α e IFN-γ e diminuição da IL-5. Na infecção secundária (VDEN-2), foi detectado anticorpos IgM (15-20 d.p.i.), anticorpos IH (15-60 d.p.i.) e diminuição dos níveis de IL-6, TNF-α, de IFN-γ e IL-5. Além disso, foi observado em ambas infecções leucopenia, neutropenia, linfocitopenia, monocitopenia, trombocitopenia, e aumentou da AST. O aumento dos níveis de INF-γ e TNF- α indicaram a ativação da resposta inflamatória, com a diferenciação para a resposta celular e supressão da proliferação de citocinas características da resposta humoral. A presença de anticorpos neutralizantes pode ter ocasionado a supressão da resposta inflamatória na infecção secundária o que pode ter levado a ausência de sinais de Febre Hemorrágica do Dengue/Sindrome do Choque do Dengue (FHD/SCD) durante a infecção secundária (VDEN- 2). Os resultados indicam que o primatas não humanos da espécie Callithrix penicillata demonstraram susceptibilidade à cepas de VDEN de origem humana, sugerindo que esses animais são um bom modelo para estudos da resposta imunológica da Febre do Dengue (FD), assim como para a avaliação de uma vacina tetravalente.
