Supportive breeding is an important tool in conservation management, but its long-term genetic consequences are not well understood. Among the factors that could affect the genetics of the offspring is sperm competition as a consequence of mixed-milt fertilizations - which is still a common practice in many hatcheries. Here, we measured and combined the relevant factors to predict the genetic consequences of various kinds of hatchery-induced sperm competition. We drew a random sample of male Coregonus zugensis (an Alpine whitefish) from a hatchery program and quantified their in vitro sperm potency by integrating sperm velocity during the first minute after activation, and their in vitro milt potency by multiplying sperm potency with milt volume and sperm cell density. We found that not controlling for sperm density and/or milt volume would, at a constant population size, decrease the variance effective number of male breeders N-em by around 40-50%. This loss would decrease with increasing population growth rates. Partial multifactorial breeding and the separate rearing of in total 799 batches of eggs revealed that neither sperm nor milt potency was significantly linked to egg survival. Sperm and milt potency was also not significantly correlated to other potential quality measures such as breeding tubercles or condition factor. However, sperm potency was correlated to male age and milt potency to male growth rate. Our findings suggest that hatchery-induced sperm competition not only increases the loss of genetic variation but may also induce artificial selection, depending on the fertilization protocol. By not equalizing milt volume in multi-male fertilization hatchery managers lose relatively more genetic variation and give fast-growing males a reproductive advantage, while equalizing milt volume reduces the loss of genetic variation and favors younger males who may have fast sperm to compensate for their subdominance at the spawning place. The objective of this work was to compare the morphometry of hatchery-reared and wild-caught mangrove crab (Ucides cordatus) megalopae. Ten U. cordatus megalopae of each group (hatchery-reared and wild-caught) were individually analyzed using a stereoscopic microscope equipped with an ocular micrometer. Length, width, and height of all megalopae were measured, and the size of body appendices was determined. The results indicate that the hatchery-reared megalopae are more robust than the wild ones. Furthermore, some significant differences in the size of certain appendices can be cues of the kind of alterations that hatchery-reared individuals experience.

After hatching, pullets are transported to brooding area and vaccinated. One day old chicks have not already developed thermoregulation ability; thus, brooding temperature variations may affect pullet quality leading to broiler meat production losses. This research aimed to calculate sensible heat loss in one day old pullets in hatching area and vaccination room. Ten one day old pullets were randomly selected from hatching area of a commercial hatchery. Infrared images were used to calculate bird surface temperature. Exposure areas for the two conditions were quantified, and both air temperature and wind speed was recorded. Total sensible heat loss was calculated as heat loss by radiation plus heat loss by convection. It was found that heat transfer occurs in different ways at different bird body parts. Total heat loss found for hatching baskets was equivalent to 0.81 J s-1 while for vaccination room was 1.16 J s-1. Pullet nutrition is based on energy loss from brooding to farm, and the overall pullet heat loss from hatchery to farm accepted is 13.95 J s-1. Thus, "starter feed" has relevant excess of energy input. These findings indicate that less energy can be used in initial feed, once heat loss is lower than assumed nowadays. Improved knowledge on these conditions may enhance broiler farm feeding strategies and economics during first rearing week.

Even though Bergey '5 Manual has been recognized globally as the guide to bacterial systematics, it has to be emphasized that descriptions given to a large extent are based on studies made with temperate isolates This leads one to conclude that any attempt to identify the tropical isolates with identification keys and tables generated from this information may lead to erroneous conclusions. And there is every possibility of the existence of genotypic and phenotypic variants or even nev. species in this part ofthe aquatic ecosystem. Applications ofa polythetic scheme of classification based on the principles of Numerical Taxonomy opens up exciting avenues for bringing to light, this possibility which otherwise would have been masked by the unidirectional approach as in monothetic schemes. Another added advantage of clustering a ‘natural’ bacterial population by numerical taxonomy, is the ease by which genotypic characterization could be performed on the clusters by selecting a representative from each cluster This helps overcome the practical impossibility of analyzing all the isolates in a pani:'_lar cluster. The genotypic characteizarion would either be mole °/o G-'rC. DNA-D.\_-X hybridization, DNA-RNA hybridization or DNA fingerprinting. Considering the requirement creating a broad base in the understanding of the family Vibrionaceae associated with the larvae ofM rosenbergii, the present work was undertaken to channelize every new information generated for developing appropriate managerial measures to protect the larvae from vibriosis during the unusually prolonged larval phase.

The great potential for the culture of non-penaeid prawns, especially Macrobrachium rosenbergii in brackish and low saline areas of Indian coastal zone has not yet been fully exploited due to the non availability of healthy seed in adequate numbers and that too in the appropriate period. In spite of setting up several prawn hatcheries around the country to satiate the ever growing demands for the seed of the giant fresh water prawn, the supply still remains fear below the requirement mainly due to the mortality of the larvae at different stages of the larval cycle. In a larval rearing system of Macrobrachium rosenbergii, members of the family Vibrionaceae were found to be dominant flora and this was especially pronounced during the times of mortality However, to develop any sort of prophylactic and therapeutic measures, the pathogenic strains have to be segregated from the lot. This would never be possible unless they were clustered based on the principles of numerical taxonomy It is with these objectives and requirements that the present work involving phenotypic characterization of the isolates belonging to the family Vibrionaceae and working out the numerical taxonomy, determination of mole % G+C ratio, segregation of the pathogenic strains and screening antibiotics as therapeutics at times of emergency, was carried out.

A packed bed bioreactor (PBBR) was developed for rapid establishment of nitrification in brackish water hatchery systems in the tropics. The reactors were activated by immobilizing ammonia-oxidizing (AMONPCU- 1) and nitrite-oxidizing (NIONPCU-1) bacterial consortia on polystyrene and low-density polyethylene beads, respectively. Fluorescence in situ hybridization demonstrated the presence of autotrophic nitrifiers belong to Nitrosococcus mobilis, lineage of b ammonia oxidizers and nitrite oxidizer Nitrobacter sp. in the consortia. The activated reactors upon integration to the hatchery system resulted in significant ammonia removal (P\0.01) culminating to its undetectable levels. Consequently, a significantly higher percent survival of larvae was observed in the larval production systems. With spent water the reactors could establish nitrification with high percentage removal of ammonia (78%), nitrite (79%) and BOD (56%) within 7 days of initiation of the process. PBBR is configured in such a way to minimize the energy requirements for continuous operation by limiting the energy inputs to a single stage pumping of water and aeration to the aeration cells. The PBBR shall enable hatchery systems to operate under closed recirculating mode and pave the way for better water management in the aquaculture industry.

For establishing nitrification in prawn (non-penaeid, salinity 10–15 ppt) and shrimp (penaeid, salinity 30–35 ppt) larval production systems, a stringed bed suspended bioreactor (SBSBR) was designed, fabricated, and validated. It was fabricated with 5 mm polystyrene and low density polyethylene beads as the substrata for ammonia and nitrite oxidizing bacterial consortia, respectively, with an overall surface area of 684 cm2. The reactors were activated in a prototype activator and were transported in polythene bags to the site of testing. Performance of the reactors activated with the nitrifying bacterial consortia AMONPCU-1 (ammonia oxidizers for non-penaeid culture) and NIONPCU-1 (nitrite oxidizers for non-penaeid culture) was evaluated in a Macrobrachium rosenbergii larval rearing system and those activated with AMOPCU-1 (ammonia oxidizers for penaeid culture) and NIOPCU-1 (nitrite oxidizers for penaeid culture) in a Penaeus monodon seed production system. Rapid setting up of nitrification could be observed in both the static systems which resulted in a higher relative per cent survival of larvae

In the months of January 2001 and 2002, female cachara Pseudoplatystoma fasciatum were selected during their first and second gonadal maturation (2 years and 7 months old and 3 years and 7 months old, respectively) with an of oocyte diameter of 937.5 mum (82.5% with central nuclei and 17.5% with peripheral nuclei). Nine females in first maturation received two doses of carp pituitary extract (CPE), 0.5 mg/kg and 5.0 mg/kg; seven received two doses of human chorionic gonadotropin (hCG), 5 and 10 IU/g; five received doses of 0.5 CPE mg/kg and 5 hCG IU/g (CPE+hCG); and four received 0.9% saline (saline). Nine females from CPE and seven from hCG presented oocytes with the same diameter at the moment of oocyte release (100% with germinal vesicle breakdown and fertilization rate of 53.44 +/- 18.3 and 54.81 +/- 11.8%; larvae number of 165,330 +/- 94.1 and 158,570 +/- 20.6, respectively). The five females from CPE+hCG did not respond to the hormonal treatment. The four females from the saline group did not ovulate. In January 2002, 6 of 15 selected females that were going through the second reproductive cycle received CPE (five received hCG and four received saline), showing oocyte diameters similar to the ones in the first maturation. At stripping, CPE females had an oocyte diameter of 1062.5 mum (the hCG females had oocyte diameters ranging from 937.5 to 1125.0 mum; fertilization rates of 56.08 +/- 30.9 and 81.90 +/- 17.3%; 364,547 +/- 244 and 633,129 +/- 190, larvae, respectively). Hatchery fish stocking for stock enhancement has been operated at a massive and global scale. However, the use of hatchery fish as a means of stock enhancement is highly controversial, and little is known about its effects on wild stock and consequences for stock enhancement. Here we review the scientific literature on this subject in order to address a fundamental - question is hatchery stocking a help or harm for wild stock and stock enhancement? We summarized 266 peer-reviewed papers that were published in the last 50 years, which describe empirical case studies on ecology and genetics of hatchery stocks and their effects on stock enhancement. Specifically, we asked whether hatchery stock and wild stock differed in fitness and the level of genetic variation, and whether stocking affected population abundance. Seventy studies contained comparisons between hatchery and wild stocks, out of which 23 studies showed significantly negative effects of hatchery rearing on the fitness of stocked fish, and 28 studies showed reduced genetic variation in hatchery populations. None of these studies suggested a positive genetic effect on the fitness of hatchery-reared individuals after release. These results suggest that negative effects of hatchery rearing are not just a concern but undeniably present in many aquaculture species. In a few cases, however, no obvious effect of hatchery rearing was observed, and a positive contribution of hatchery stock to the abundance of fish populations was indicated. These examples suggest that there is a chance to improve hatchery practices and mitigate the negative effects on wild stocks, although scientific data supporting the positive effect on stock enhancement are largely missing at this moment. Technically, microsatellite-based parentage assignments have been proven as a useful tool for the evaluation of reproductive fitness in natural settings, which is a key for stock enhancement by hatchery-based stocking. When salmonid fish that have been raised in hatcheries spawn in the wild, they often produce fewer surviving adult offspring than wild fish. Recent data from steelhead (Oncorhynchus mykiss) in the Hood River (Oregon, USA) show that even one or two generations of hatchery culture can result in dramatic declines in fitness. Although intense domestication selection could cause such declines, it is worth considering alternative explanations. One possibility is heritable epigenetic changes induced by the hatchery environment. Here, we show, using methylation-sensitive amplified fragment length polymorphism, that hatchery and wild adult steelhead from the Hood River do not appear to differ substantially in overall levels of genomic methylation. Thus, although altered methylation of specific DNA sites or other epigenetic processes could still be important, the hatchery environment does not appear to cause a global hypo- or hypermethylation of the genome or create a large number of sites that are differentially methylated.

This study evaluated the use of electroshock as in alternative to traditional techniques for immobilizing and euthanizing hatchery fish. We used a commercially available electroanesthesia unit at the U.S. Fish and Wildlife Service's Carson National Fish Hatchery (Carson, Washington) to euthanize adult spring Chinook salmon Oncorhynchus tshawytscha and to son and collect gametes of fish at maturation. During euthanization by electroshock, the response of each fish was observed, Muscular and vertebral hemorrhaging wits quantified, and electrical settings were optimized accordingly. During gamete collection, fish were either electroshocked or exposed to tricaine methanesulfortate (MS-222); hemorrhaging, egg viability. egg size and quantity, and resultant fry quality were examined for each treatment group. Electroshocked fish had a higher likelihood Of injury during gamete collection than did fish exposed to MS-222. On average, each electroshocked fish had less than two hemorrhages oil both fillets examined. The size of each hemorrhage was less than 0.10% of the fillet surface. Fecundity and egg and fry quality were not affected by either immobilization method. Electroshock was a viable and efficient means of euthanizing adult spring Chinook salmon or sorting the fish and collecting their gametes. However, equipment settings must be optimized based on site-specific (e.g., water conductivity) and species-specific (e.g., fish size and seasonal state of maturation) factors.
