OBJETIVO: Avaliar os parâmetros de cães anestesiados com diferentes protocolos de fármacos dissociativos por infusão intravenosa contínua. MÉTODOS: Foram utilizados 30 cães, machos e fêmeas, clinicamente sadios, distribuídos aleatoriamente em três grupos (G1,G2 e G3) (*)). em G1 utilizou-se levomepromazina como medicação pré-anestésica (MPA), midazolam-cetamina pela via intravenosa em bolus para indução e midazolam-cetamina em infusão intravenosa contínua por 60 minutos para manutenção. em G2 procedeu-se da mesma forma que em G1 elevando-se, porém, a dose de midazolam durante a manutenção. em G3 repetiu-se o tratamento empregado em G2, acrescentando-se a xilazina à manutenção. Após a indução, iniciou-se imediatamente a manutenção anestésica, realizando-se aferições, 15 minutos depois da MPA, em intervalos de 10 minutos, durante a manutenção (M0 a M7). RESULTADOS: em G3 ocorreu bradicardia, bloqueio átrio-ventricular, bradipnéia e hipoxemia e em G1 e G2, discreta hipotensão. CONCLUSÃO: A via intravenosa contínua apresentou vantagens quanto a: não oscilação dos parâmetros e redução no período de recuperação anestésica. A elevação da dose de midazolam resultou em discretas variações paramétricas, estas, acentuadas pelo uso da xilazina, que causou hipoxemia, bradiarritmia, diminuição da freqüência respiratória e volume minuto.

A laparoscopia ainda é pouco utilizada como ferramenta para técnicas de reprodução assistida em cervídeos sul-americanos, não havendo informações sobre seus efeitos e protocolos anestésicos seguros para sua realização. Objetivaramse avaliar as possíveis alterações na freqüência cardíaca (FC), respiratória (FR), saturação de oxihemoglobina (SpO2) e temperatura retal (TR) durante a laparoscopia para visualização dos órgãos reprodutivos de seis fêmeas de veado-catingueiro (Mazama gouazoubira) anestesiadas com a associação cetamina (5mg/kg), xilazina (0,3mg/kg), midazolam (0,5mg/kg) e isofluorano. Cada animal, após anestesiado, foi posicionado em decúbito dorsal para realização de duas laparoscopias com insuflação abdominal de CO2 (14,2 ± 2,39mmHg; M ± EPM) com intervalo de 40 dias. Para avaliar os principais eventos da laparoscopia, esta foi dividida em três períodos: animal sem insuflação abdominal (P1), com insuflação abdominal (P2) e insuflação abdominal com os quadris elevados a 45º (P3). O controle foi realizado após 40 dias da última laparoscopia, para isto, cada animal foi novamente anestesiado e mantido em decúbito dorsal por um período de tempo igual ao tempo médio de duração das anestesias realizadas nas laparoscopias, sem que o procedimento laparoscópico fosse realizado. O tempo de anestesia dos controles foi também dividido em P1, P2 e P3, respeitando o tempo médio de duração de cada um destes períodos das laparoscopias. Para análise dos dados foi usado o teste de análise de variância (ANOVA) seguido do teste de Tukey e valores de P<0,05 considerados significativos. Não houve diferença significativa nos parâmetros estudados em nenhum dos períodos estabelecidos para o controle e laparoscopia. Porém, a FR média entre P1 (38,8 ± 4,42) e P3 (32,7 ± 4,81) e a TR média entre P1 (38,2ºC ± 0,17), P2 (37,6ºC ± 0,19) e P3 (37,0ºC ± 0,21) variaram significativamente, independente da laparoscopia. Tais dados permitiram concluir que a laparoscopia não promoveu alterações significativas nos parâmetros avaliados, embora o protocolo anestésico utilizado tenha contribuído para redução da temperatura retal resultando em risco de hipotermia durante a anestesia.

The auditory brainstem response (ABR) is a test widely used to assess the integrity of the brain stem. Although it is considered to be an auditory-evoked potential that is influenced by the physical characteristics of the stimulus, such as rate, polarity and type of stimulus, it may also be influenced by the change in several parameters. The use of anesthetics may adversely influence the value of the ABR wave latency. One of the anesthetics used for e ABR assessment, especially in animal research, is the ketamine/xylazine combination. Our objective was to determine the influence of the ketamine/xylazine anesthetic on the ABR latency values in adult gerbils. The ABRs of 12 adult gerbils injected with the anesthetic were collected on three consecutive days, or a total of six collections, namely: pre-collection and A, B, C, D, and E collections. Before each collection the gerbil was injected with a dose of ketamine (100 mg/kg)/xylazine (4 mg/kg). For the capture of the ABR, 2000 click stimuli were used with rarefaction polarity and 13 stimuli per second, 80 dBnHL intensity and in-ear phones. A statistically significant difference was observed in the latency of the V wave in the ABR of gerbils in the C and D collections compared to the pre-, A and E collections, and no difference was observed between the pre-, A, B, and E collections. We conclude that the use of ketamine/xylazine increases the latency of the V wave of the ABR after several doses injected into adult gerbils; thus clinicians should consider the use of this substance in the assessment of ABR.

Lorsque l’anesthésie par inhalation ne peut être utilisée chez le rat, la combinaison de kétamine et de xylazine est l’alternative la plus fréquemment utilisée. Les doses administrées peuvent varier selon le protocole expérimental. En présence de fièvre, d’infections ou de processus tumoral accompagné de fièvre, la pharmacocinétique de ces drogues peut être modifiée. Ce projet porte sur l’évaluation des changements physiologiques, hématologiques, biochimiques et pharmacocinétiques chez le rat Sprague Dawley lors d’anesthésie avec le mélange kétamine-xylazine suite à l’administration de trois doses différentes de lipopolysaccharide (LPS). Après l’administration de LPS, une anesthésie à la kétamine-xylazine fut induite chez des rats Sprague Dawley. Des prélèvements sanguins périodiques ainsi que des mesures des paramètres physiologiques furent effectués afin d’évaluer l’effet du LPS sur la pharmacocinétique des deux drogues ainsi que sur les paramètres biochimiques et hématologiques. Les différentes doses de LPS ont causé certaines modifications notamment en produisant une baisse marquée de la saturation en oxygène et de l’albumine sérique, une augmentation de la durée d’anesthésie ainsi que des lésions hépatiques mineures. Les paramètres pharmacocinétiques de la kétamine furent peu altérés par l’administration de LPS tandis qu’une diminution de la clairance et une augmentation de l’aire sous la courbe (AUC) furent observées pour la xylazine dans les groupes ayant reçu les doses moyenne et élevée de LPS. Ces résultats montrent que les doses de xylazine doivent être adaptées en présence de LPS pour permettre une anesthésie de courte durée et des changements physiologiques et biochimiques moindres lorsqu’elle est administrée avec de la kétamine.

Chez les animaux de laboratoire, même si les anesthésiques par inhalation sont généralement plus sécuritaires que les injectables, leur utilité est souvent restreinte lorsqu’un protocole expérimental exige une autre approche. Des combinaisons d’anesthésiques contenant de la kétamine sont considérées comme l’option de choix pour les anesthésies injectables chez les rats. Le vieillissement entraîne des changements dégénératifs au niveau de la structure et la fonction des organes, modifiant souvent à la pharmacocinétique des drogues. Ce projet porte sur l’évaluation des changements pharmacodynamiques (physiologiques, biochimiques et histologiques) et pharmacocinétiques, lors d’une combinaison anesthésique de kétamine-­‐xylazine chez le rat Sprague-­‐Dawley vieillissant. Une anesthésie à la kétamine-­‐xylazine fut induite chez des rats Sprague-­‐Dawley de différents âges. Afin d’évaluer l’effet du vieillissement sur le métabolisme des deux drogues, des prélèvements sanguins périodiques pour l’analyse de la pharmacocinétique ainsi que des mesures des paramètres physiologiques, biochimiques et une histopathologie furent effectués. Le vieillissement a causé certaines modifications notamment en produisant une diminution de la saturation d’oxygène, une baisse marquée de la fréquence cardiaque et respiratoire, une hypoalbuminémie ainsi qu’une augmentation de la durée d’anesthésie. Les paramètres pharmacocinétiques de la kétamine et de la xylazine furent grandement affectés par le vieillissement causant une augmentation progressive significative de l’aire sous la courbe (AUC) et du temps de demi-­‐vie, ainsi qu’une diminution de la clairance. À la lumière de ces résultats, les doses de kétamine et de xylazine doivent être adaptées chez les rats vieillissants pour permettre une anesthésie de durée raisonnable et un réveil sans complications.

This survey was conducted with the objective of developing and adapting diagnostic techniques in neurology for non-human primates, of the species Alouatta guariba Geoffroy Saint-Hilaire, 1812 (howler monkey) healthy and kept in captivity. Physical-chemical analyses were carried out in cerebrospinal fluid and its pressure measurement in the cisterna magna. Eight animals were used for the analysis of cerebrospinal fluid. In all procedures, were used chemical restraint and anesthesia with ketamine, xylazine and association of midazolam and inhalational anesthesia with isoflurane for carrying out the spinal tap. The cerebrospinal fluid analysis showed average values: protein: 16.92±9.84; glucose: 131.25mg/dL±106.7; pH: 8.37±0.69; nucleated cells: 0.5/mm³±0.75; red blood cells: 49.37/mm³±111.76 and CSF cisternal pressure: 7.37cm H2O±1.77. This study demonstrated the safety and efficacy of cerebrospinal fluid collection in the cisterna magna of Alouatta guariba, and the reference values for the specie.

The aim of this study was evaluate the effects of three anesthetic combinations, ketamine-midazolam, ketamine-xylazine and tiletamine-zolazepam, on IOP in rabbits. In a experimental, blind, randomized, crossover study, six rabbits were anesthetized with each of 3 treatments in random order. Groups KM (ketamine, 30 mg/kg + midazolam, 1 mg/kg); KX (ketamine, 30 mg/kg + xylazine, 3 mg/kg); and TZ (tiletamine + zolazepam, 20 mg/kg). The drugs were mixed in the same syringe injected intramuscularly (IM) into the quadriceps muscle. IOP was measured before drug administration (baseline) and at 5-minute intervals for 30 minutes. The data were analyzed by a 2-way repeated measures ANOVA followed by Bonferroni test. All groups had significant decreases in IOP compared to baseline (p < 0.001). There was no difference between groups at any of the time points assessed (p > 0.05). Administration of either ketamine-midazolam, ketamine-xylazine, or tiletamine-zolazepam similarly decrease IOP in rabbits within 30 minutes of injection.

In surgical animal studies anesthesia is used regularly. Several reports in the literature demonstrate respiratory and cardiovascular side effects of anesthesiologic agents. The aim of this study was to compare two frequently used anesthesia cocktails (ketamine/xylazine [KX] versus medetomidine/climazolam/fentanyl [MCF]) in skin flap mouse models. Systemic blood values, local metabolic parameters, and surgical outcome should be analyzed in critical ischemic skin flap models. Systemic hypoxia was found in the animals undergoing KX anesthesia compared with normoxia in the MCF group (sO(2): 89.2% +/- 2.4% versus 98.5% +/- 1.2%, P < 0.01). Analysis of tissue metabolism revealed impaired anaerobic oxygen metabolism and increased cellular damage in critical ischemic flap tissue under KX anesthesia (lactate/pyruvate ratio: KX 349.86 +/- 282.38 versus MCF 64.53 +/- 18.63; P < 0.01 and glycerol: KX 333.50 +/- 83.91 micromol/L versus MCF 195.83 +/- 29.49 micromol/L; P < 0.01). After 6 d, different rates of flap tissue necrosis could be detected (MCF 57% +/- 6% versus KX 68% +/- 6%, P < 0.01). In summary we want to point out that the type of anesthesia, the animal model and the goal of the study have to be well correlated. Comparing the effects of KX and MCF anesthesia in mice on surgical outcome was a novel aspect of our study.

Sequential studies of osteopenic bone disease in small animals require the availability of non-invasive, accurate and precise methods to assess bone mineral content (BMC) and bone mineral density (BMD). Dual-energy X-ray absorptiometry (DXA), which is currently used in humans for this purpose, can also be applied to small animals by means of adapted software. Precision and accuracy of DXA was evaluated in 10 rats weighing 50-265 g. The rats were anesthetized with a mixture of ketamine-xylazine administrated intraperitoneally. Each rat was scanned six times consecutively in the antero-posterior incidence after repositioning using the rat whole-body software for determination of whole-body BMC and BMD (Hologic QDR 1000, software version 5.52). Scan duration was 10-20 min depending on rat size. After the last measurement, rats were sacrificed and soft tissues were removed by dermestid beetles. Skeletons were then scanned in vitro (ultra high resolution software, version 4.47). Bones were subsequently ashed and dissolved in hydrochloric acid and total body calcium directly assayed by atomic absorption spectrophotometry (TBCa[chem]). Total body calcium was also calculated from the DXA whole-body in vivo measurement (TBCa[DXA]) and from the ultra high resolution measurement (TBCa[UH]) under the assumption that calcium accounts for 40.5% of the BMC expressed as hydroxyapatite. Precision error for whole-body BMC and BMD (mean +/- S.D.) was 1.3% and 1.5%, respectively. Simple regression analysis between TBCa[DXA] or TBCa[UH] and TBCa[chem] revealed tight correlations (n = 0.991 and 0.996, respectively), with slopes and intercepts which were significantly different from 1 and 0, respectively.(ABSTRACT TRUNCATED AT 250 WORDS)

Fourten Weddell seals (Leptonychotes weddellii) and two crabeater seals (Lobodon carcinophaga) were immobilised at Drescher Inlet (Riiser Larsen Ice Shelf), eastern Weddell Sea coast, between January and February 1990 using a combination of ketamine, xylazine, and diazepam. Eleven Weddell seals were drugged once, and two and one were drugged two and three times each, coming to a total of 18 immobilisation procedures. Another 16 seals were immobilised between January and February 1992. Ten seals were drugged once, and three and two were drugged two and three times each, coming to a total of 25 immobilisation procedures. Narcoses were terminated with yohimbine.
