La tesi di laurea sperimentale tratta la produzione di membrane polimeriche ricoperte con coating grafenici mediante la tecnica del layer by layer per la separazione di miscele gassose. Il grafene utilizzato è stato prodotto in laboratorio mediante un mulino a sfere che ha portato contemporaneamente sia all’esfoliazione della grafite sia alla sua funzionalizzazione, mentre è stato utilizzato un ossido di grafene commerciale. I campioni sono stati preparati utilizzando sia membrane porose che non porose. Le membrane così preparate sono state testate al permeometro per verificarne la permeabilità a coppie di gas di interesse industriale, come H2/CO2.

Lo scopo di questo studio sperimentale è stato quello di determinare l’effetto dell’aggiunta di piccole quantità (1wt%) di grafene e ossido di grafene al poli(1-trimetilsilil-1-propino) (PTMSP). Il PTMSP è uno dei polimeri più promettenti per la separazione di gas tramite membrane polimeriche grazie al suo elevato volume libero (26%). Sono state studiate sia membrane spesse (60-180 micron) preparate per solvent casting che sottili (2-10 micron) preparate per spin coating supportate su un film poroso di polipropilene commerciale. L’ossido di grafene aumenta la permeabilità del PTMSP, mentre il grafene ha mostrato un comportamento variabile in funzione del protocollo di preparazione che è risultato dipendere fortemente dalla velocità di evaporazione del solvente. Le membrane così ottenute sono state testate al permeometro. È stata osservata una dipendenza della permeabilità in funzione dello spessore del film e del grado di invecchiamento. In particolare, la presenza di nanofiller riduce il grado di invecchiamento dei film di PTMSP. Nel caso specifico della coppia di gas permeanti He/CO2, i campioni hanno mostrato un comportamento intercambiabile di selettività all’He o alla CO2, modulabile in funzione della temperatura tra 30-60°C e del filler utilizzato.

A novel cellulose membrane was prepared by using amine oxides as the solvent and its mechanical performance was measured. Steady-state permeation rates of carbon dioxide, hydrogen, methane, nitrogen, oxygen, argon and helium in the homogeneous dense cellulose membrane were measured in the temperature range of 298-353 K and under gas pressures up to 1 MPa. The effect of swelling on hydrophilic membrane permeability was studied in some detail on the cellulose membrane. The difference in gas permeability between the "dry" cellulose membrane and the "water-swollen" cellulose membrane was investigated, and the gas permeability between the cellulose membrane and the Cellophane was compared. In this paper, the separation performance Of CO2 over H-2 in a "water-swollen" cellulose membrane is reported for the first time and the separation factor Of CO2/H-2 can be up to 15. NaA zeolite membrane was successfully synthesized on the porous alpha-Al2O3 support by microwave heating. The synthesis of NaA zeolite membrane in the microwave field only needs 15 min and the synthesis time is 10 times shorter than that by conventional heating. SEM characterization indicates that the zeolite crystals in the NaA zeolite membrane synthesized by microwave heating are uniform in size; the membrane thickness is about 4 mu m and is thinner than that of the NaA zeolite membrane synthesized by conventional heating. Gas permeation studies indicate that the permeances of the NaA zeolite membrane synthesized by microwave heating are 3-4 times higher than those of the NaA zeolite membrane synthesized by conventional heating, while their permselectivities are comparable.

The synthesis of NaA zeolite membrane on a porous alpha -Al2O3 support from clear solution and the evaluation of the perfection of the as-synthesized membrane by gas permeation were investigated. When an unseeded support was used, the NaA zeolite began to transform into other types of zeolites before a continuous NaA zeolite membrane formed. When the support was coated with nucleation seeds, not only the formation of NaA zeolite on the support surface was accelerated, but also the transformation of NaA zeolite into other types of zeolites was inhibited. A continuous NaA zeolite membrane can be formed. Perfection evaluation indicated that the NaA zeolite membrane with the synthesis time of 3 h showed the best perfection after a one-stage synthesis. The perfection of NaA zeolite membrane can be improved by employing the multi-stage synthesis method. The NaA zeolite membrane with a synthesis time of 2 h after a two-stage synthesis showed the best gas permeation performance, The permselectivity of H-2/n-C4H10 and O-2/N-2 were 19.1 and 1.8, respectively, higher than those of the corresponding Knudsen diffusion selectivity of 5.39 and 0.94, which showed the molecular sieving effect of NaA zeolite. However, the permeation of n-C4H10 also indicated that the NaA zeolite membrane had certain defects, the diameter of which were larger than the NaA zeolite channels. (C) 2001 Elsevier Science B.V. The attainable steady-state limiting currents and time responses of membrane-covered and membrane-independent gas sensors incorporating different electrode and electrolyte materials have been compared. A new design comprising a membrane-free microelectrode modified with a thin layer of a room temperature ionic liquid is considered. While the use of ionic liquid as electrolyte eliminates the need for a membrane and added supporting electrolyte, the slower diffusion of analyte within the more viscous medium results in slower time responses. Such sensors do, however, have potential application in more extreme operating conditions, such as high temperature and pressure, where traditional solvents would volatise.

PEFCs employing Nafion-silica (Nafion-SiO2) and Nafion-mesoporous zirconium phosphate (Nafion-MZP) composite membranes are subjected to accelerated-durability test at 100 degrees C and 15% relative humidity (RH) at open-circuit voltage (OCV) for 50 h and performance compared with the PEFC employing pristine Nafion-1135 membrane. PEFCs with composite membranes sustain the operating voltage better with fluoride-ion-emission rate at least an order of magnitude lower than PEFC with pristine Nafion-1135 membrane. Reduced gas-crossover, fast fuel-cell-reaction kinetics and superior performance of the PEFCs with Nafion-SiO2 and Nafion-MZP composite membranes in relation to the PEFC with pristine Nafion-1135 membrane support the long-term operational usage of the former in PEFCs. An 8-cell PEFC stack employing Nafion-SiO2 composite membrane is also assembled and successfully operated at 60 degrees C without external humidification.

Gas permeability coefficients of a series of aromatic polyetherimides, which were prepared from 1,4-bis(3,4-dicarboxyphenoxy) benzene dianhydride (HQDPA) and various aromatic diamines, to H-2, CO2, O-2, N-2 and CH4 have been measured under 7 atm pressure and over the temperature range 30-150 degrees C. A significant change in permeability and permselectivity, which resulted from a systematic variation in chemical structure of the polyetherimides, was found. Generally, increases in permeability of the polyetherimides are accompanied by decreases in permselectivity. The order of decrease of the permeability coefficients is as follows: HQDPA-IPDA > HQDPA-DDS > HQDPA-MDA > HQDPA-ODA > HQDPA-DABP > HQDPA-BZD. However, HQDPA-DMoBZD and HQDPA-DMoMDA, with bulky methoxy side-groups on the aromatic rings of the diamine residue, display both high permeability coefficients and high permselectivity. The favourable gas separation property, excellent thermal and chemical stability, and high mechanical strength make HQDPA-DMoBZD and HQDPA-DMoMDA promising candidates for membrane-based gas separation applications.

The shell-side flow distribution of a parallel flow hollow-fiber gas permeator was characterized using a thermo-anemograph. The permeator has an internal diameter of 103 mm and contains 21000 fibers. The overall fiber packing fraction is 40.1%. Experimental results revealed that shell-side flow maldistributions exist in the operating conditions studied. The gas-flow velocity is the highest at the permeator center, but lowest near the shell wall. The effects of shell-side flow maldistribution on predicted permeator performances are discussed with a simple model. Model calculation results show that flow maldistributions can have considerable effect on permeation systems with relatively high separation factors and stage cuts. (C) 1998 Elsevier Science B.V.

This PhD thesis discusses the rationale for design and use of synthetic oligosaccharides for the development of glycoconjugate vaccines and the role of physicochemical methods in the characterization of these vaccines. The study concerns two infectious diseases that represent a serious problem for the national healthcare programs: human immunodeficiency virus (HIV) and Group A Streptococcus (GAS) infections. Both pathogens possess distinctive carbohydrate structures that have been described as suitable targets for the vaccine design. The Group A Streptococcus cell membrane polysaccharide (GAS-PS) is an attractive vaccine antigen candidate based on its conserved, constant expression pattern and the ability to confer immunoprotection in a relevant mouse model. Analysis of the immunogenic response within at-risk populations suggests an inverse correlation between high anti-GAS-PS antibody titres and GAS infection cases. Recent studies show that a chemically synthesized core polysaccharide-based antigen may represent an antigenic structural determinant of the large polysaccharide. Based on GAS-PS structural analysis, the study evaluates the potential to exploit a synthetic design approach to GAS vaccine development and compares the efficiency of synthetic antigens with the long isolated GAS polysaccharide. Synthetic GAS-PS structural analogues were specifically designed and generated to explore the impact of antigen length and terminal residue composition. For the HIV-1 glycoantigens, the dense glycan shield on the surface of the envelope protein gp120 was chosen as a target. This shield masks conserved protein epitopes and facilitates virus spread via binding to glycan receptors on susceptible host cells. The broadly neutralizing monoclonal antibody 2G12 binds a cluster of high-mannose oligosaccharides on the gp120 subunit of HIV-1 Env protein. This oligomannose epitope has been a subject to the synthetic vaccine development. The cluster nature of the 2G12 epitope suggested that multivalent antigen presentation was important to develop a carbohydrate based vaccine candidate. I describe the development of neoglycoconjugates displaying clustered HIV-1 related oligomannose carbohydrates and their immunogenic properties.

OBJECTIVE To assess macular hole surgery in patients with end-stage choroideremia with regard to anatomic closure and visual outcome. DESIGN Retrospective, interventional case series. PARTICIPANTS Thirty adult male patients with a diagnosis of advanced choroideremia were reviewed and underwent spectral domain optical coherence tomography (OCT) as part of the screening process for a gene therapy clinical trial. From within that cohort, 3 were identified as having a full-thickness macular hole (FTMH). METHODS A 23-gauge pars plana vitrectomy was performed with peeling of the inner limiting membrane and gas tamponade. Preoperative best-corrected visual acuity ranged from perception of light to 6/24. MAIN OUTCOME MEASURES Prevalence of FTMH in advanced choroideremia, morphologic phenotype characteristics of FTMH in OCT, pre- and postoperative best-corrected visual acuity, and closure rate after surgery. RESULTS The prevalence of FTMH in advanced choroideremia in our cohort was 10%. One hole was associated with significant macular schisis, presumed to be attributable to degeneration of outer retinal layers. Anatomic closure was achieved in all 3 patients and confirmed with spectral domain OCT. Gas tamponade lasted approximately twice as long as might be expected compared with standard FTMH surgery. Objective visual acuity did not improve; however, perceived vision improved in all patients. CONCLUSIONS Although FTMH in choroideremia is a rare finding, it could potentially mask central progression of the disease. Regular screening may help to diagnose holes at an earlier stage when the visual prognosis after surgery may be better. Standard macular hole surgery seems to be effective in gaining anatomic closure, which would be significant for patients who subsequently require macula detachment for subretinal gene therapy. The work of this thesis has been focused on the characterisation of inorganic membranes for the hydrogen purification from steam reforming gas. Composite membranes based on porous inorganic supports coated with palladium silver alloys and ceramic membranes have been analysed. A brief resume of theoretical laws governing transport of gases through dense and porous inorganic membranes and an overview on different methods to prepare inorganic membranes has been also reported. A description of the experimental apparatus used for the characterisation of gas permeability properties has been reported. The device used permits to evaluate transport properties in a wide range of temperatures (till 500°C) and pressures (till 15 bar). Data obtained from experimental campaigns reveal a good agreement with Sievert law for hydrogen transport through dense palladium based membranes while different transport mechanisms, such as Knudsen diffusion and Hagen-Poiseuille flow, have been observed for porous membranes and for palladium silver alloy ones with pinholes in the metal layer. Mixtures permeation experiments reveal also concentration polarisation phenomena and hydrogen permeability reduction due to carbon monoxide adsorption on metal surface.

Membrane-based separation processes are acquiring, in the last years, an increasing importance because of their intrinsic energetic and environmental sustainability: some types of polymeric materials, showing adequate perm-selectivity features, appear rather suitable for these applications, because of their relatively low cost and easy processability. In this work have been studied two different types of polymeric membranes, in view of possible applications to the gas separation processes, i.e. Mixed Matrix Membranes (MMMs) and high free volume glassy polymers. Since the early 90’s, it has been understood that the performances of polymeric materials in the field of gas separations show an upper bound in terms of permeability and selectivity: in particular, an increase of permeability is often accompanied by a decrease of selectivity and vice-versa, while several inorganic materials, like zeolites or silica derivates, can overcome this limitation. As a consequence, it has been developed the idea of dispersing inorganic particles in polymeric matrices, in order to obtain membranes with improved perm-selectivity features. In particular, dispersing fumed silica nanoparticles in high free volume glassy polymers improves in all the cases gases and vapours permeability, while the selectivity may either increase or decrease, depending upon material and gas mixture: that effect is due to the capacity of nanoparticles to disrupt the local chain packing, increasing the dimensions of excess free volume elements trapped in the polymer matrix. In this work different kinds of MMMs were fabricated using amorphous Teflon® AF or PTMSP and fumed silica: in all the cases, a considerable increase of solubility, diffusivity and permeability of gases and vapours (n-alkanes, CO2, methanol) was observed, while the selectivity shows a non-monotonous trend with filler fraction. Moreover, the classical models for composites are not able to capture the increase of transport properties due to the silica addition, so it has been necessary to develop and validate an appropriate thermodynamic model that allows to predict correctly the mass transport features of MMMs. In this work, another material, called poly-trimethylsilyl-norbornene (PTMSN) was examined: it is a new generation high free volume glassy polymer that, like PTMSP, shows unusual high permeability and selectivity levels to the more condensable vapours. These two polymer differ each other because PTMSN shows a more pronounced chemical stability, due to its structure double-bond free. For this polymer, a set of Lattice Fluid parameters was estimated, making possible a comparison between experimental and theoretical solubility isotherms for hydrocarbons and alcoholic vapours: the successfully modelling task, based on application of NELF model, offers a reliable alternative to direct sorption measurement, which is extremely time-consuming due to the relevant relaxation phenomena showed by each sorption step. For this material also dilation experiments were performed, in order to quantify its dimensional stability in presence of large size, swelling vapours.

Il presente lavoro di tesi si è focalizzato sullo studio e sulla ottimizzazione di un sistema integrato, che utilizzi la reazione di oxy-reforming del metano al fine di produrre syngas che venga trattato attraverso la water-gas shift al fine di abbattere il contenuto di CO e al tempo stesso aumentare la resa in H2. Con l’obiettivo di ottenere H2 ad elevata purezza (>99%) da poter essere inviato direttamente a celle a combustible ed in impianti di piccola taglia con possibile delocalizzazione della produzione industriale di energia elettrica e termica “pulita”, la miscela reale uscente dal processo di oxy-reforming è stata processata tramite successiva water-gas shift direttamente all’interno di una membrana ceramica al Pd selettiva nella separazione di H2. L’innovativià di questo progetto di studio è data da diversi parametri quali: 1) l’impiego dell’oxy-reforming in alternativa al normale steam-reforming del CH4, che permette di condurre il processo a temperature decisamente inferiori (700-750°C), utilizzando un minor quantitativo di vapore (S/C = 0.7); 2) l’utilizzo di due nuove formulazioni di catalizzatore di WGS per alte temperature, capace di operare in un unico stadio conversioni di CO ottenibili industrialmente solo attraverso i convenzionali due due stadi di reazione (e due diverse formulazioni di catalizzatori a base di Fe/Cr e Cu); 3) l’utilizzo di supporti ceramici con membrana a base di Pd, capaci di ospitare al loro interno un catalizzatore eterogeneo per la reazione di WGS a 400°C, rendendo quindi possibile la produzione e contemporanea separazione di H2 con un ulteriore effetto positivo poiché la membrana rimuovendo H2 dalla zona di reazione favorisce il superamento dell’equilibrio termodinamico per la conversione del CO, abbassandone il contenuto nel flusso uscente dei gas reazione e rendendo non più necessari sistemi aggiuntivi di separazione quali PSA o PROXY.

I processi di separazione sono tra quelli che consumano più energia nelle nazioni maggiormente industrializzate, questo rende necessaria la ricerca continua di tecnologie e materiali che siano in grado di aumentarne l’efficienza. Uno dei principali campi di studio di questa ricerca riguarda la separazione di gas, in cui le membrane polimeriche si candidano come principale alternativa innovativa ai metodi classici. Questo lavoro di tesi mira all’analisi delle variabili che governano il trasposto di gas attraverso membrane polimeriche di tipo vetroso, alla comprensione dei fenomeni destabilizzanti quali aging e plasticizzazione in termini di cause ed effetti e alla primaria rielaborazione dei dati di letteratura relativi agli stessi fenomeni per verificare analiticamente quanto studiato nella parte precedente. Per fare ciò, operativamente, ci si è serviti del supporto di articoli scientifici che trattano il tema, andando ad estrapolare dati e informazioni rilevanti e delle indicazioni del professor Minelli che da anni è impegnato nella ricerca su questi materiali. Dopo una prima parte introduttiva riguardante le tecnologie a membrana e le loro applicazioni, si passa allo studio approfondito della teoria e alla discussione dei due fenomeni, ai quali viene dedicato un capitolo ciascuno per capirne nel dettaglio le implicazioni che comportano nell’utilizzo di tecnologie a membrane polimeriche. Nella rielaborazione ci si sofferma poi sui film sottili, nell’ordine del micron, che corrispondo alle dimensioni tipiche delle membrane utilizzate per la separazione di gas. Ci si focalizza su miscele contenenti anidride carbonica, gas di estremo interesse sia perché è uno dei maggiori contributori al fenomeno del surriscaldamento globale, sia perché è uno dei principali responsabili della plasticizzazione di un polimero, per via della sua buona solubilità. Si conclude l’elaborato con una breve analisi sulle possibili contromisure riguardanti aging e plasticizzazione.
