Remorins form a superfamily of plant-specific plasma membrane/lipid-raft-associated proteins of unknown structure and function. Using specific antibodies, we localized tomato remorin 1 to apical tissues, leaf primordia and vascular traces. The deduced remorin protein sequence contains a predicted coiled coil-domain, suggesting its participation in protein-protein interactions. Circular dichroism revealed that recombinant potato remorin contains an alpha-helical region that forms a functional coiled-coil domain. Electron microscopy of purified preparations of four different recombinant remorins, one from potato, two divergent isologs from tomato, and one from Arabidopsis thaliana , demonstrated that the proteins form highly similar filamentous structures. The diameters of the negatively-stained filaments ranged from 4.6-7.4 nm for potato remorin 1, 4.3-6.2 nm for tomato remorin 1, 5.7-7.5 nm for tomato remorin 2, and 5.7-8.0 nm for Arabidopsis Dbp. Highly polymerized remorin 1 was detected in glutaraldehyde-crosslinked tomato plasma membrane preparations and a population of the protein was immunolocalized in tomato root tips to structures associated with discrete regions of the plasma membrane.

The microenvironment of the central nervous system is important for neuronal function and development. During the early stages of embryo development the cephalic vesicles are filled by embryonic cerebrospinal fluid, a complex fluid containing different protein fractions, which contributes to the regulation of the survival, proliferation and neurogenesis of neuroectodermal stem cells. The protein content of embryonic cerebrospinal fluid from chick and rat embryos at the start of neurogenesis has already been determined. Most of the identified gene products are thought to be involved in the regulation of developmental processes during embryogenesis. However, due to the crucial roles played by embryonic cerebrospinal fluid during brain development, the embryological origin of the gene products it contains remains an intriguing question. According to the literature most of these products are synthesised in embryonic tissues other than the neuroepithelium. In this study we examined the embryological origin of the most abundant embryonic cerebrospinal fluid protein fractions by means of slot-blot analysis and by using several different embryonic and extraembryonic protein extracts, immunodetected with polyclonal antibodies. This first attempt to elucidate their origin is not based on the proteins identified by proteomic methods, but rather on crude protein fractions detected by SDS-PAGE analysis and to which polyclonal antibodies were specifically generated. Despite some of the limitations of this study, i.e. that one protein fraction may contain more than one gene product, and that a specific gene product may be contained in different protein fractions depending on post-translational modifications, our results show that most of the analysed protein fractions are not produced by the cephalic neuroectoderm but are rather stored in the egg reservoir; furthermore, few are produced by embryo tissues, thus indicating that they must be transported from their production or storage sites to the cephalic cavities, most probably via embryonic serum. These results raise the question as to whether the transfer of proteins from these two embryo compartments is regulated at this early developmental stage.

La famille des gènes Hox code pour des facteurs de transcription connus pour leur contribution essentielle à l’élaboration de l’architecture du corps et ce, au sein de tout le règne animal. Au cours de l’évolution chez les vertébrés, les gènes Hox ont été redéfinis pour générer toute une variété de nouveaux tissus/organes. Souvent, cette diversification s’est effectuée via des changements quant au contrôle transcriptionnel des gènes Hox. Chez les mammifères, la fonction de Hoxa13 n’est pas restreinte qu’à l’embryon même, mais s’avère également essentielle pour le développement de la vascularisation fœtale au sein du labyrinthe placentaire, suggérant ainsi que sa fonction au sein de cette structure aurait accompagné l’émergence des espèces placentaires. Au chapitre 2, nous mettons en lumière le recrutement de deux autres gènes Hoxa, soient Hoxa10 et Hoxa11, au compartiment extra-embryonnaire. Nous démontrons que l’expression de Hoxa10, Hoxa11 et Hoxa13 est requise au sein de l’allantoïde, précurseur du cordon ombilical et du système vasculaire fœtal au sein du labyrinthe placentaire. De façon intéressante, nous avons découvert que l’expression des gènes Hoxa10-13 dans l’allantoïde n’est pas restreinte qu’aux mammifères placentaires, mais est également présente chez un vertébré non-placentaire, indiquant que le recrutement des ces gènes dans l’allantoïde précède fort probablement l’émergence des espèces placentaires. Nous avons généré des réarrangements génétiques et utilisé des essais transgéniques pour étudier les mécanismes régulant l’expression des gènes Hoxa dans l’allantoïde. Nous avons identifié un fragment intergénique de 50 kb capable d’induire l’expression d’un gène rapporteur dans l’allantoïde. Cependant, nous avons trouvé que le mécanisme de régulation contrôlant l’expression du gène Hoxa au sein du compartiment extra-embryonnaire est fort complexe et repose sur plus qu’un seul élément cis-régulateur. Au chapitre 3, nous avons utilisé la cartographie génétique du destin cellulaire pour évaluer la contribution globale des cellules exprimant Hoxa13 aux différentes structures embryonnaires. Plus particulièrement, nous avons examiné plus en détail l’analyse de la cartographie du destin cellulaire de Hoxa13 dans les pattes antérieures en développement. Nous avons pu déterminer que, dans le squelette du membre, tous les éléments squelettiques de l’autopode (main), à l’exception de quelques cellules dans les éléments carpiens les plus proximaux, proviennent des cellules exprimant Hoxa13. En contraste, nous avons découvert que, au sein du compartiment musculaire, les cellules exprimant Hoxa13 et leurs descendantes (Hoxa13lin+) s’étendent à des domaines plus proximaux du membre, où ils contribuent à générer la plupart des masses musculaires de l’avant-bras et, en partie, du triceps. De façon intéressante, nous avons découvert que les cellules exprimant Hoxa13 et leurs descendantes ne sont pas distribuées uniformément parmi les différents muscles. Au sein d’une même masse musculaire, les fibres avec une contribution Hoxa13lin+ différente peuvent être identifiées et les fibres avec une contribution semblable sont souvent regroupées ensemble. Ce résultat évoque la possibilité que Hoxa13 soit impliqué dans la mise en place de caractéristiques spécifiques des groupes musculaires, ou la mise en place de connections nerf-muscle. Prises dans leur ensemble, les données ici présentées permettent de mieux comprendre le rôle de Hoxa13 au sein des compartiments embryonnaires et extra-embryonnaires. Par ailleurs, nos résultats seront d’une importance primordiale pour soutenir les futures études visant à expliquer les mécanismes transcriptionnels soutenant la régulation des gènes Hoxa dans les tissus extra-embryonnaires.

Eukaryotic translation initiation factor 5A (eIF5A) has a unique character: the presence of an unusual amino acid, hypusine, which is formed by post-translational modifications. Even before the identification of hypusination in eIF5A, the correlation between hypusine formation and protein synthesis, shifting cell proliferation rates, had already been observed. Embryogenesis is a complex process in which cellular proliferation and differentiation are intense. In spite of the fact that many studies have described possible functions for eIF5A, its precise role is under investigation, and to date nothing has been reported about its participation in embryonic development. In this study we show that eIF5A is expressed at all mouse embryonic post-implantation stages with increase in eIF5A mRNA and protein expression levels between embryonic days E10.5 and E13.5. Immunohistochemistry revealed the ubiquitous presence of eIF5A in embryonic tissues and organs at E13.5 day. Interestingly, stronger immunoreactivity to eIF5A was observed in the stomodeum, liver, ectoderm, heart, and eye, and the central nervous system; regions which are known to undergo active differentiation at this stage, suggesting a role of eIF5A in differentiation events. Expression analyses of MyoD, a myogenic transcription factor, revealed a significantly higher expression from day E12.5 on, both at the mRNA and the protein levels suggesting a possible correlation to eIF5A. Accordingly, we next evidenced that inhibiting eIF5A hypusination in mouse myoblast C2C12 cells impairs their differentiation into myotubes and decreases MyoD transcript levels. Among the various possible embodiements of Advanced Therapies and in particular of Tissue Engineering the use of temporary scaffolds to regenerate tissue defects is one of the key issues. The scaffolds should be specifically designed to create environments that promote tissue development and not merely to support the maintenance of communities of cells. To achieve that goal, highly functional scaffolds may combine specific morphologies and surface chemistry with the local release of bioactive agents. Many biomaterials have been proposed to produce scaffolds aiming the regeneration of a wealth of human tissues. We have a particular interest in developing systems based in nanofibrous biodegradable polymers1,2. Those demanding applications require a combination of mechanical properties, processability, cell-friendly surfaces and tunable biodegradability that need to be tailored for the specific application envisioned. Those biomaterials are usually processed by different routes into devices with wide range of morphologies such as biodegradable fibers and meshes, films or particles and adaptable to different biomedical applications. In our approach, we combine the temporary scaffolds populated with therapeutically relevant communities of cells to generate a hybrid implant. For that we have explored different sources of adult and also embryonic stem cells. We are exploring the use of adult MSCs3, namely obtained from the bone marrow for the development autologous-based therapies. We also develop strategies based in extra-embryonic tissues, such as amniotic fluid (AF) and the perivascular region of the umbilical cord4 (Whartonâ s Jelly, WJ). Those tissues offer many advantages over both embryonic and other adult stem cell sourcess. These tissues are frequently discarded at parturition and its extracorporeal nature facilitates tissue donation by the patients. The comparatively large volume of tissue and ease of physical manipulation facilitates the isolation of larger numbers of stem cells. The fetal stem cells appear to have more pronounced immunomodulatory properties than adult MSCs. This allogeneic escape mechanism may be of therapeutic value, because the transplantation of readily available allogeneic human MSCs would be preferable as opposed to the required expansion stage (involving both time and logistic effort) of autologous cells. Topics to be covered: This talk will review our latest developments of nanostructured-based biomaterials and scaffolds in combination with stem cells for bone and cartilage tissue engineering.

Three species of Scorpions beloging to two different families were studied cytologically: a) Tityus mattogrossensis Borelli (Fam. Buthidae), - This species presents spermatogonia provided with 20 short chromosomes which orient at metaphase with their axis parallelly to the plane of the equator and move toward the poles without changing this position, from the stage pachytene to metaphase the bivalents become, as in Tityus bahiensis, progressivery shorter and thicker, without showing that chiasmata occured at any time. The paired chromosomes never open themselves, out to form loops as in orthodox meioses. As in Tityus bahiensis the bivalents are inserted In the spindle before reaching their maxim contraction. No diakinesis has been observed. The primary spermatocyte metaphases are provided, with 10 pairs of chromosones, two of which are larger and two smaller than the rest. The bivalents orient as in Tityus bahiensis with their length in the plane of the equator and separate parallelly. Spindle fibres are seen alongst their entire body. While, in Tityus bahiensis the ends of the chromosomes are pronouncedly turned to opposite poles at metaphase, nothing like this was observed in the present species. Only late in anaphase the chromosomes of Tityus mattogrossensis show a bending to the poles. The secondary spermatocytes present 10 short chromosomes, two being larger than, the others. Here, on the contrary, the chromosomes are strongly curved toward the poles since the beginning of anaphase. Some chromosomal anomalies have been noticed. Primary spermatocytes with 14 bivalents, some of which representing probably free fragments, were observed. Primary spermatocytes with 8 bivalents and one cross of 4 chromosomes were interpreted as resulting from breakages followed by translocations Primary spermatocytes with 9 bivalents, one of which being much longer than the longst of the normal plates, show that fusion by the extremities of two non homologous chromosomes on the onde side, and of their respective homologous in the same way on tre other, have occured. Orientation of bivalents with their body parallelly to the spindle axis and anaphasic bridges have been encountered. All in all points to the conclusion that the chromosomes of Tityus mattogrossesis, like those of Tityus bahiensia are provided with one kinetochore at each end. Ananteris balzani Thorell - (Fam. Buthidae). - This species which belongs to the same family as Tityus, is provided with 12 chromosomes (diploid). These studied in embryonic tissues, showed the same behavior as the somatic chromosomes of Tityus bahiensis. Bothrirus sp. (Bothriuridae). - Only spermatogonia were found in the testis, of the single male hitherto investigated. The chromosomes, in number of 36, are of different sizes but small and provided, as ordinarily, with a single kinetochore. They behave therefore in an orthodox manner in mitosis.

Primary cell cultures were obtained from eggs of Anopheles albimanus and Aedes taeniorhynchus mosquitoes, vectors of human malaria and of Venezuelan equine encephalitis virus, respectively. The cellular growth of the An. albimanus cells began four weeks after explanting the embryonic tissues in MK/VP12 medium, supplemented with 15% fetal bovine serum. The culture showed heterogeneous cellular morphology. With regard to the Ae. taeniorhynchus culture, growth occurred three weeks after initiating the culture in MM/VP12 medium. The majority of cells were small and round. Karyotypes were examined in the latter species.

Insect cell cultures are an important biotechnological tool for basic and applied studies. The objective of this work was to establish and characterise a new cell line from Culex quinquefasciatus embryonic tissues. Embryonated eggs were taken as a source of tissue to make explants that were seeded in L-15, Grace's, Grace's/L-15, MM/VP12, Schneider's and DMEM culture media with a pH range from 6.7-6.9 and incubated at 28ºC. The morphological, cytogenetic, biochemical and molecular characteristics of the cell cultures were examined by observing the cell shapes, obtaining the karyotypes, using a cellulose-acetate electrophoretic system and performing random amplified polymorphic DNA-polymerase chain reaction analysis, respectively. The Grace's/L-15 medium provided the optimal nutritional conditions for cell adhesion and proliferation. Approximately 40-60 days following the explant procedure, a confluent monolayer was formed. Cellular morphology in the primary cultures and the subcultures was heterogeneous, but in the monolayer the epithelioid morphology type predominated. A karyotype with a diploid number of six chromosomes (2n = 6) was observed. Isoenzymatic and molecular patterns of the mosquito cell cultures matched those obtained from the immature and adult forms of the same species. Eighteen subcultures were generated. These cell cultures potentially constitute a useful tool for use in biomedical applications.

Se evaluó la susceptibilidad de los cultivos celulares derivados de tejidos embrionarios de Aedes aegypti a la infección con Leishmania (L) chagasi y Leishmania (V) braziliensis, agentes etiológicos de leishmaniasis visceral americana y leishmaniasis cutánea, respectivamente. Metodología: Se seleccionaron células de A. aegypti mantenidas en una mezcla de medio de cultivo Grace/L15, suplementado con suero fetal bovino al 15%, albendazol 5,4 mg/ml y una mezcla de antibióticos, e incubadas a una temperatura promedio de 26 °C. Los cultivos celulares fueron inoculados con promastigotes metacíclicos de la cepa MH/CO/84/CI-044B de L. chagasi y la cepa HOM/BR752903 de L. braziliensis en una concentración de 10 parásitos por célula. Como control positivo de la infección se utilizó la línea celular J774. Resultados: Los registros más altos en el porcentaje de infección y en el número de amastigotes por células en los cultivos celulares A. aegypti y en la línea celular J774 se obtuvieron en los días 6 y 9 pos-infección. Los resultados mostraron interacción, internalización y maduración in vitro de las dos especies del parásito en las células de este insecto no vector de Leishmania. Las células de A. aegypti infectadas mostraron cambios en el área por la influencia de los parásitos, contrario a lo registrado en las células no infectadas (P<0,05). Conclusión: Los cultivos celulares de A. aegypti emergen como un nuevo modelo in vitro para el estudio del ciclo biológico de L. chagasi y L. braziliensis.

During embryo implantation, invasive trophoblast cells mediate embryo invasion into the decidualized stroma, forming a rich network of lacunae that connect the embryonic tissues to the maternal blood vessels. Placentation is probably guided by the composition and organization of the endometrial extracellular matrix. Certain pathological conditions that occur during pregnancy, including diabetes, have been linked to abnormal placental morphology and consequent fetal morbidity. We used immunoperoxidase techniques to identify members of the collagen, proteoglycan and glycoprotein families in the various compartments of the rat placenta and to determine whether experimentally induced diabetes affects placental morphology and alters the distribution of these molecules during pregnancy. Single injections of alloxan (40 mg kg(-1) i.v.) were used to induce diabetes on day 2 of pregnancy in Wistar rats. Placentas were collected on days 14, 17, and 20. Type I and III collagen, as well as the proteoglycans decorin and biglycan, were found to be distributed throughout the placentas of control and diabetic rats. In both groups, laminin expression decreased at the end of pregnancy. In contrast, fibronectin was detected in the labyrinth region of diabetic rats at all gestational stages studied, whereas it was detected only at term pregnancy in the placentas of control rats. These results show for the first time that some extracellular matrix molecules are modulated during placental development. However, as diabetic rats presented increased fibronectin deposition exclusively in the labyrinth region, we speculate that diabetes alters the microenvironment at the maternal-fetal interface, leading to developmental abnormalities in the offspring.

The ligand-controlled retinoic acid (RA) receptors and retinoid X receptors are important for several physiological processes, including normal embryonic development, but little is known about how their ligands, all-trans and 9-cis RA, are generated. Here we report the identification of a stereo-specific 9-cis retinol dehydrogenase, which is abundantly expressed in embryonic tissues known to be targets in the retinoid signaling pathway. The membrane-bound enzyme is a member of the short-chain alcohol dehydrogenase/reductase superfamily, able to oxidize 9-cis retinol into 9-cis retinaldehyde, an intermediate in 9-cis RA biosynthesis. Analysis by nonradioactive in situ hybridization in mouse embryos shows that expression of the enzyme is temporally and spatially well controlled during embryogenesis with prominent expression in parts of the developing central nervous system, sensory organs, somites and myotomes, and several tissues of endodermal origin. The identification of this enzyme reveals a pathway in RA biosynthesis, where 9-cis retinol is generated for subsequent oxidation to 9-cis RA.

The transcriptional activation potential of proteins can be assayed in chimeras containing a heterologous DNA-binding domain that mediates their recruitment to reporter genes. This approach has been widely used in yeast and in transient mammalian cell assays. Here, we applied it to assay the transactivation potential of proteins in transgenic Drosophila embryos. We found that a chimera between the DNA-binding bacterial LexA protein and the transactivation domain from yeast GAL4 behaved as a potent synthetic activator in all embryonic tissues. In contrast, a LexA chimera containing Drosophila Fos (Dfos) required an unexpected degree of context to function as a transcriptional activator. We provide evidence to suggest that this context is provided by Djun and Mad (a Drosophila Smad), and that these partner factors need to be activated by signaling from Jun N-terminal kinase and decapentaplegic, respectively. Because Dfos behaves as an autonomous transcriptional activator in more artificial assays systems, our data suggest that context-dependence of transcription factors may be more prevalent than previously thought.

Members of the IRF family mediate transcriptional responses to interferons (IFNs) and to virus infection. So far, proteins of this family have been studied only among mammalian species. Here we report the isolation of cDNA clones encoding two members of this family from chicken, interferon consensus sequence-binding protein (ICSBP) and IRF-1. The predicted chicken ICSBP and IRF-1 proteins show high levels of sequence similarity to their corresponding human and mouse counterparts. Sequence identities in the putative DNA-binding domains of chicken and human ICSBP and IRF-1 were 97% and 89%, respectively, whereas the C-terminal regions showed identities of 64% and 51%; sequence relationships with mouse ICSBP and IRF-1 are very similar. Chicken ICSBP was found to be expressed in several embryonic tissues, and both chicken IRF-1 and ICSBP were strongly induced in chicken fibroblasts by IFN treatment, supporting the involvement of these factors in IFN-regulated gene expression. The presence of proteins homologous to mammalian IRF family members, together with earlier observations on the occurrence of functionally homologous IFN-responsive elements in chicken and mammalian genes, highlights the conservation of transcriptional mechanisms in the IFN system, a finding that contrasts with the extensive sequence and functional divergence of the IFNs.

As células-tronco podem ser isoladas tanto de tecidos embrionários quanto de tecidos provenientes de um organismo adulto. Este projeto teve por objetivo caracterizar, descrever as células derivadas da região uterina e da cinta placentária junção materno/fetal da placenta de carnívoros domésticos (cães e gatos), e verificar a sua capacidade de pluripotência. Os úteros gestantes e não gestantes foram obtidos em campanhas de castrações e de controle populacional de cães e gatos, na cidade de Pirassununga/SP. Foram coletados 24 úteros gravídicos de animais hígidos, em diferentes idades gestacionais. O material foi dividido em três fases distintas da gestação, ou seja inicio que compreende de 8 a 20 dias de gestação; meio de 21 a 30 dias de gestação e final de 31 a 60 dias de gestação. O material foi coletado de fêmeas caninas e felinas, quatro úteros de cada fase, totalizando 12 úteros de cães e 12 de felinos. Coletamos também 8 úteros de fêmeas nulíparas (4 de cadelas e 4 de gatas) e 8 úteros com um mês pós parto (4 de cadelas e 4 de gatas). As amostras foram fixadas em paraformoldeido tamponado a 4% para a análise histológica e de imunohistoquimica. Para a padronização da imunohistoquimica inúmeros testes de marcação e diluição dos anticorpos utilizados nesta pesquisa foram realizados, todo protocolo aqui descrito foi padronizado pela primeira vez. Nas análises de imunohistoquimica avaliamos a expressão de marcadores associados a células-tronco pluripotentes Nanog, Oct4 e Sox2. Nas cadelas, as marcações foram positivas em todas as fases, gestacionais e não gestacionais. A detecção dessas proteínas nesta espécie ficaram padronizadas, destacando algumas diferenças quantitativas durante alguns períodos da gestação. Foi observado que o Oct4 na cadela, mostra uma diferença significativa (p=0,0064), entre as fases de início e meio da gestação e entra o início e a fase de termo. Quando comparados os resultados das análises imunohistoquimicas utilizando os três anticorpos entre si, nos três períodos gestacionais ficou evidente uma diferença (p=0,0005) somente relativa a proteína Nanog com Oct4. Nas gatas apenas foi possível padronizar o protocolo do Nanog e do Sox2, sendo a marcação feita com Oct4 negativa. Nesta espécie foi possível observar uma diferença da proteína Nanog (p=0,0006) quando comparada na fase inicial para a fase do meio e início da gestação para a fase termo. No que se refere as fêmeas nulíparas e fêmeas pós-parto destaca-se a ausência de diferenças quando comparados os anticorpos na fase pós parto tanto em cadelas quanto em gatas. Na fase nulípara foram observadas diferenças somente na cadela (p=0,0018) para os três anticorpos. Desta forma, a caracterização de células de origem placentária com característica de células tronco pode abrir um leque de possibilidades para obtenção destas células de forma mais ética, uma vez que este material é descartado na castrações. Foi possível a identificação das células que expressão proteínas pluripotentes em diferentes idades gestacionais, tanto na região de cinta placentária como no útero. Apesar de semelhantes, as espécies aqui estudadas apresentaram diferenças na realização do protocolo da imunihistoquímica. Pesquisas relacionadas com as células-tronco do endométrio vêm crescendo, principalmente porque estas células podem ser facilmente obtidas, a partir de fontes descartadas, sem entraves éticos. Desta forma tem o potencial de serem uma nova fonte para o desenvolvimento na terapêutica como terapia celular
