Die Resonanzionisations-Massenspektrometrie (RIMS) ist sowohl für spektroskopische Untersuchungen seltener Isotope als auch für den Ultraspurennachweis langlebiger radioaktiver Elemente einsetzbar. Durch die mehrstufige resonante Anregung atomarer Energieniveaus mit anschließender Ionisation mit Laserlicht wird eine sehr hohe Elementselektivität und Ionisationseffizienz erreicht. Der nachfolgende massenselektive Ionennachweis liefert eine gute Isotopenselektivität zusammen mit einer effektiven Untergrundunterdrückung. Ein wichtiger Bestandteil der RIMS-Apparatur ist ein zuverlässig arbeitendes, leistungsstarkes Lasersystem für die Resonanzionisation. Im Rahmen dieser Arbeit wurde ein von einem hochrepetierenden Nd:YAG-Laser gepumptes, aus drei Titan-Saphir-Lasern bestehendes System fertig aufgebaut und in den Routinebetrieb überführt. Die Titan-Saphir-Laser liefern im Durchstimmbereich von 730 - 880 nm eine mittlere Leistung von bis zu 3 W pro Laser bei einer Linienbreite von 2 - 3 GHz. Sie lassen sich computergesteuert in ihren Wellenlängen durchstimmen. Die mittels Resonanzionisation erzeugten Ionen werden dann in einem Flugzeit-Massenspektrometer entsprechend ihrer Masse aufgetrennt und mit einem Kanalplattendetektor nachgewiesen.Als Voraussetzung für die isotopenselektive Ultraspurenanalyse von Plutonium wurden mit diesem Lasersystem die Isotopieverschiebungen eines effizienten, dreistufigen Anregungsschema für Plutonium bestimmt. Die Laserleistungen reichen zur vielfachen Sättigung der ersten beiden Anregungsschritte und zur zweifachen Sättigung des dritten Anregungsschritts aus.Außerdem wurden die Ionisationsenergien von Pu-239 und Pu-244 zur Untersuchung ihrer Isotopenabhängigkeit bestimmt. Die beiden Ionisationsenergien sind im Rahmen der erreichten Genauigkeit bei einem Meßwert von IP239-IP244 = 0,24(82) cm^-1 gleich.Die Nachweiseffizienz der RIMS-Apparatur für Plutonium wurde in Effizienzmessungen zu 10^-5 bestimmt. Durch die gute Untergrundunterdrückung ergab sich daraus eine Nachweisgrenze von 10^6 Atomen bei der Messung eines Plutoniumisotops. Die Bestimmung der Isotopenverhältnisse von Proben mit einer zertifizierten Isotopenzusammensetzung lieferte eine gute Übereinstimmung der Meßwerte mit den angegebenen Zusammensetzungen.Die RIMS-Apparatur wurde zur Bestimmung des Gehalts und der Isotopenzusammensetzung von Plutonium in Meerwasser- und Staubproben eingesetzt.Auf Grund der Isotopenzusammensetzung konnte gezeigt werden, daß das Plutonium bei den meisten Proben aus dem Fallout von oberirdischen Kernwaffentests stammte. Des weiteren wurde Plutonium in Urinproben bestimmt. Die Nachweisgrenzen lagen bei diesen Umweltproben bei 10^6 bis 10^7 Atomen Plutonium und damit um zwei Größenordnungen niedriger als die Nachweisgrenze für Pu-239 bei der alpha-Spektroskopie, der Standardmethode für den Plutoniumnachweis.

The coupling of kurtosis based-indexes and envelope analysis represents one of the most successful and widespread procedures for the diagnostics of incipient faults on rolling element bearings. Kurtosis-based indexes are often used to select the proper demodulation band for the application of envelope-based techniques. Kurtosis itself, in slightly different formulations, is applied for the prognostic and condition monitoring of rolling element bearings, as a standalone tool for a fast indication of the development of faults. This paper shows for the first time the strong analytical connection which holds for these two families of indexes. In particular, analytical identities are shown for the squared envelope spectrum (SES) and the kurtosis of the corresponding band-pass filtered analytic signal. In particular, it is demonstrated how the sum of the peaks in the SES corresponds to the raw 4th order moment. The analytical results show as well a link with an another signal processing technique: the cepstrum pre-whitening, recently used in bearing diagnostics. The analytical results are the basis for the discussion on an optimal indicator for the choice of the demodulation band, the ratio of cyclic content (RCC), which endows the kurtosis with selectivity in the cyclic frequency domain and whose performance is compared with more traditional kurtosis-based indicators such as the protrugram. A benchmark, performed on numerical simulations and experimental data coming from two different test-rigs, proves the superior effectiveness of such an indicator. Finally a short introduction to the potential offered by the newly proposed index in the field of prognostics is given in an additional experimental example. In particular the RCC is tested on experimental data collected on an endurance bearing test-rig, showing its ability to follow the development of the damage with a single numerical index.

Using low cost portable devices that enable a single analytical step for screening environmental contaminants is today a demanding issue. This concept is here tried out by recycling screen-printed electrodes that were to be disposed of and by choosing as sensory element a low cost material offering specific response for an environmental contaminant. Microcystins (MCs) were used as target analyte, for being dangerous toxins produced by cyanobacteria released into water bodies. The sensory element was a plastic antibody designed by surface imprinting with carefully selected monomers to ensure a specific response. These were designed on the wall of carbon nanotubes, taking advantage of their exceptional electrical properties. The stereochemical ability of the sensory material to detect MCs was checked by preparing blank materials where the imprinting stage was made without the template molecule. The novel sensory material for MCs was introduced in a polymeric matrix and evaluated against potentiometric measurements. Nernstian response was observed from 7.24 × 10−10 to 1.28 × 10−9 M in buffer solution (10 mM HEPES, 150 mM NaCl, pH 6.6), with average slopes of −62 mVdecade−1 and detection capabilities below 1 nM. The blank materials were unable to provide a linear response against log(concentration), showing only a slight potential change towards more positive potentials with increasing concentrations (while that ofthe plastic antibodies moved to more negative values), with a maximum rate of +33 mVdecade−1. The sensors presented good selectivity towards sulphate, iron and ammonium ions, and also chloroform and tetrachloroethylene (TCE) and fast response (<20 s). This concept was successfully tested on the analysis of spiked environmental water samples. The sensors were further applied onto recycled chips, comprehending one site for the reference electrode and two sites for different selective membranes, in a biparametric approach for “in situ” analysis.

Interaction of the estrogen receptor/ligand complex with a DNA estrogen response element is known to regulate gene transcription. In turn, specific conformations of the receptor-ligand complex have been postulated to influence unique subsets of estrogen-responsive genes resulting in differential modulation and, ultimately, tissue-selective outcomes. The estrogen receptor ligands raloxifene and tamoxifen have demonstrated such tissue-specific estrogen agonist/antagonist effects. Both agents antagonize the effects of estrogen on mammary tissue while mimicking the actions of estrogen on bone. However, tamoxifen induces significant stimulation of uterine tissue whereas raloxifene does not. We postulate that structural differences between raloxifene and tamoxifen may influence the conformations of their respective receptor/ligand complexes, thereby affecting which estrogen-responsive genes are modulated in various tissues. These structural differences are 4-fold: (A) the presence of phenolic hydroxyls, (B) different substituents on the basic amine, (C) incorporation of the stilbene moiety into a cyclic benzothiophene framework, and (D) the imposition of a carbonyl “hinge” between the basic amine-containing side chain and the olefin. A series of raloxifene analogs that separately exemplify each of these differences have been prepared and evaluated in a series of in vitro and in vivo assays. This strategy has resulted in the development of a pharmacophore model that attributes the differences in effects on the uterus between raloxifene and tamoxifen to a low-energy conformational preference imparting an orthogonal orientation of the basic side chain with respect to the stilbene plane. This three-dimensional array is dictated by a single carbon atom in the hinge region of raloxifene. These data indicate that differences in tissue selective actions among benzothiophene and triarylethylene estrogen receptor modulators can be ascribed to discrete ligand conformations.

Genes containing the interferon-stimulated response element (ISRE) enhancer have been characterized as transcriptionally responsive primarily to type I interferons (IFN alpha/beta). Induction is due to activation of a multimeric transcription factor, interferon-stimulated gene factor 3 (ISGF3), which is activated by IFN alpha/beta but not by IFN gamma. We found that ISRE-containing genes were induced by IFN gamma as well as by IFN alpha in Vero cells. The IFN gamma response was dependent on the ISRE and was accentuated by preexposure of cells to IFN alpha, a treatment that increases the abundance of ISGF3 components. Overexpression of ISGF3 polypeptides showed that the IFN gamma response depended on the DNA-binding protein ISGF3 gamma (p48) as well as on the 91-kDa protein STAT91 (Stat1 alpha). The transcriptional response to IFN alpha required the 113-kDa protein STAT113 (Stat2) in addition to STAT91 and p48. Mutant fibrosarcoma cells deficient in each component of ISGF3 were used to confirm that IFN gamma induction of an ISRE reporter required p48 and STAT91, but not STAT113. A complex containing p48 and phosphorylated STAT91 but lacking STAT113 bound the ISRE in vitro. IFN gamma-induced activation of this complex, preferentially formed at high concentrations of p48 and STAT91, may explain some of the overlapping responses to IFN alpha and IFN gamma.

An unstructured mesh �nite volume discretisation method for simulating di�usion in anisotropic media in two-dimensional space is discussed. This technique is considered as an extension of the fully implicit hybrid control-volume �nite-element method and it retains the local continuity of the ux at the control volume faces. A least squares function recon- struction technique together with a new ux decomposition strategy is used to obtain an accurate ux approximation at the control volume face, ensuring that the overall accuracy of the spatial discretisation maintains second order. This paper highlights that the new technique coincides with the traditional shape function technique when the correction term is neglected and that it signi�cantly increases the accuracy of the previous linear scheme on coarse meshes when applied to media that exhibit very strong to extreme anisotropy ratios. It is concluded that the method can be used on both regular and irregular meshes, and appears independent of the mesh quality.
