A aplicação antecipada de fertilizante potássico no cultivo de espécies de cobertura no sistema plantio direto (SPD) pode ser vantajosa para a lavoura comercial. O objetivo deste trabalho foi avaliar a produtividade de grãos e a acumulação de K na soja em função da aplicação antecipada de fertilizante potássico na instalação do milheto em relação com o K aplicado na semeadura da soja subseqüente no SPD. O experimento foi realizado na FCA-Unesp em Botucatu-SP, nas safras 2000/2001, 2001/2002 e 2002/2003. Utilizou-se um Latossolo Vermelho distroférrico de textura média, que estava sendo cultivado com soja e aveia-preta no SPD, por dois anos antes da instalação do experimento. O milheto (Pennisetum glaucum) foi semeado em setembro sobre a palhada de aveia-preta (Avena strigosa), e a soja (Glycine max) na primeira quinzena de dezembro, nos três anos agrícolas. Utilizou-se o delineamento experimental em blocos ao acaso no esquema fatorial 4 x 4, com quatro repetições, com 0, 30, 60 e 90 kg ha-1 de K2O no milheto, combinados com 0, 30, 60 e 90 kg ha-1 de K2O na soja. Coletaram-se plantas de soja aos 25, 50, 75 e 100 dias após a emergência, e os grãos no final do ciclo, para a determinação do acúmulo de K e da produtividade. A antecipação de 60 a 90 kg ha-1 de K2O na semeadura do milheto não comprometeu o acúmulo de K na lavoura de soja. As máximas produtividades de soja foram alcançadas no primeiro e segundo ano com doses de 85 a 90 kg ha-1 de K2O, que poderiam ser antecipadas totalmente na semeadura da gramínea de cobertura. A aplicação antecipada de KCl na semeadura do milheto minimizou a exportação de K pela colheita de grãos de soja.

Purpose We investigated the effects of weed control and fertilization at early establishment on foliar stable carbon (δ13C) and nitrogen (N) isotope (δ15N) compositions, foliar N concentration, tree growth and biomass, relative weed cover and other physiological traits in a 2-year old F1 hybrid (Pinus elliottii var. elliottii (Engelm) × Pinus caribaea var. hondurensis (Barr. ex Golf.)) plantation grown on a yellow earth in southeast Queensland of subtropical Australia. Materials and methods Treatments included routine weed control, luxury weed control, intermediate weed control, mechanical weed control, nil weed control, and routine and luxury fertilization in a randomised complete block design. Initial soil nutrition and soil fertility parameters included (hot water extractable organic carbon (C) and total nitrogen (N), total C and N, C/N ratio, labile N pools (nitrate (NO3 −) and ammonium (NH4 +)), extractable potassium (K+)), soil δ15N and δ13C. Relative weed cover, foliar N concentrations, tree growth rate and physiological parameters including photosynthesis, stomatal conductance, photosynthetic nitrogen use efficiency, foliar δ15N and foliar δ13C were also measured at early establishment. Results and discussion Foliar N concentration at 1.25 years was significantly different amongst the weed control treatments and was negatively correlated to the relative weed cover at 1.1 years. Foliar N concentration was also positively correlated to foliar δ15N and foliar δ13C, tree height, height growth rates and tree biomass. Foliar δ15N was negatively correlated to the relative weed cover at 0.8 and 1.1 years. The physiological measurements indicated that luxury fertilization and increasing weed competition on these soils decreased leaf xylem pressure potential (Ψxpp) when compared to the other treatments. Conclusions These results indicate how increasing N resources and weed competition have implications for tree N and water use at establishment in F1 hybrid plantations of southeast Queensland, Australia. These results suggest the desirability of weed control, in the inter-planting row, in the first year to maximise site N and water resources available for seedling growth. It also showed the need to avoid over-fertilisation, which interfered with the balance between available N and water on these soils.

Commercial cultivation of the dioecious brown macroalga Hizikia fusiformis (Harvey) Okamura in East Asia depends on the supply of young seedlings from regenerated holdfasts or from wild population. Recent development of synchronized release of male and female gametes in tumble culture provides a possibility of mass production of young seedlings via sexual reproduction. In this paper, we demonstrate that controlled fertilization can be efficiently realized in ambient light and temperature in a specially designed raceway tank in which the sperm-containing water has been recirculated. The effective fertilization time of eggs by sperm was found to be within six hours. Fast growth and development of the young seedlings relied on the presence of water currents. Velocity tests demonstrated that young seedlings of 2-3 mm in length could withstand a water current of 190 cm s(-1) stop without detachment. Culture experiments at 24 h postfertilization showed that elongation of both the seedlings and their rhizoids were not hampered by high irradiance up to 600 mu mol photons m(-2) stop s(-1) stop. However, growth was slightly retarded if cultured at a temperature of 16 degrees C compared to other culture temperatures of 22, 25 and 29 degrees C. No seedling detachment was observed after transfer of the young seedlings to raft cultivation in the sea after one and 1.5 months post-fertilization, indicating the feasibility of obtaining large quantity of seedlings in such a system.

The presence of heparin and a mixture of penicillamine, hypotaurine, and epinephrine (PHE) solution in the in vitro fertilization (IVF) media seem to be a prerequisite when bovine spermatozoa are capacitated in vitro, in order to stimulate sperm motility and acrosome reaction. The present study was designed to determine the effect of the addition of heparin and PHE during IVF on the quality and penetrability of spermatozoa into bovine oocytes and on subsequent embryo development. Sperm quality, evaluated by the integrity of plasma and acrosomal membranes and mitochondrial function, was diminished (P < 0.05) in the presence of heparin and PHE. Oocyte penetration and normal pronuclear formation rates, as well as the percentage of zygotes presenting more than two pronuclei, was higher (P < 0.05) in the presence of heparin and PHE. No differences were observed in cleavage rates between treatment and control (P > 0.05). However, the developmental rate to the blastocyst stage was increased in the presence of heparin and PHE (P > 0.05). The quality of embryos that reached the blastocyst stage was evaluated by counting the inner cell mass (ICM) and trophectoderm (TE) cell numbers and total number of cells; the percentage of ICM and TE cells was unaffected (P > 0.05) in the presence of heparin and PHE (P < 0.05). In conclusion, this study demonstrated that while the supplementation of IVF media with heparin and PHE solution impairs spermatozoa quality, it plays an important role in sperm capacitation, improving pronuclear formation, and early embryonic development. © 2013 The Society for In Vitro Biology.

The initial growth and mineral nutrition of the physic nut (Jatropha curcas L.) as a function of nitrogen (N) fertilization was investigated. The transplanting of seedlings was carried out in plastic pots filled with 50 dm3 of a Rhodic Hapludox, under a plastic greenhouse. In addition to a control treatment, the dosages of 0, 40, 80, 120, and 160 mg dm−3 N were tested. The results demonstrated that N fertilization for the cultivation of physic nut could be added as top dressing from 60 days after planting with a dosage of 65 mg dm−3. A SPAD index of 46 can be used as a nutritional reference to its initial development. Furthermore, the results suggested that the order of nutrient accumulation by the physic nut plants is as follows: potassium (K) > N > magnesium (Mg) > calcium (Ca) > phosphorus (P) > sulfur (S) > iron (Fe) > manganese (Mn) > boron (B) > zinc (Zn) > copper (Cu).

Outbreaks of crown-of-thorns starfish (COTS), Acanthaster planci, contribute to major declines of coral reef ecosystems throughout the Indo-Pacific. As the oceans warm and decrease in pH due to increased anthropogenic CO2 production, coral reefs are also susceptible to bleaching, disease and reduced calcification. The impacts of ocean acidification and warming may be exacerbated by COTS predation, but it is not known how this major predator will fare in a changing ocean. Because larval success is a key driver of population outbreaks, we investigated the sensitivities of larval A. planci to increased temperature (2-4 °C above ambient) and acidification (0.3-0.5 pH units below ambient) in flow-through cross-factorial experiments (3 temperature × 3 pH/pCO2 levels). There was no effect of increased temperature or acidification on fertilization or very early development. Larvae reared in the optimal temperature (28 °C) were the largest across all pH treatments. Development to advanced larva was negatively affected by the high temperature treatment (30 °C) and by both experimental pH levels (pH 7.6, 7.8). Thus, planktonic life stages of A. planci may be negatively impacted by near-future global change. Increased temperature and reduced pH had an additive negative effect on reducing larval size. The 30 °C treatment exceeded larval tolerance regardless of pH. As 30 °C sea surface temperatures may become the norm in low latitude tropical regions, poleward migration of A. planci may be expected as they follow optimal isotherms. In the absence of acclimation or adaptation, declines in low latitude populations may occur. Poleward migration will be facilitated by strong western boundary currents, with possible negative flow-on effects on high latitude coral reefs. The contrasting responses of the larvae of A. planci and those of its coral prey to ocean acidification and warming are considered in context with potential future change in tropical reef ecosystems.

The presence of cover crop straw and early application of total N at sowing may provide significant changes in the microbial population, reflecting on the N dynamics in the soil and in upland rice plants. This study aimed at determining the effect of the early application of nitrogen doses as mineral N and microbial biomass carbon in the soil, as well as in the activity of nitrate reductase, and grain yield of upland rice plants cultivated under notillage system (NTS). A randomized blocks design, in a split-plot scheme, with four replications, was used. The treatments consisted of N doses (0 kg ha-1, 40 kg ha-1, 80 kg ha-1 and 120 kg ha-1) and the presence or absence of U. brizantha cover straw. Maintaining the straw on the soil surface reduces the ammonium levels and increases the microbial biomass carbon content of the soil. The application of increasing doses of N in the soil provides increases in the levels of nitrate and ammonium in the soil up to 28 days after emergence. The activity of the nitrate reductase enzyme in the plants increases and the contents of ammonium and nitrate in the soil decrease with the crop development. The number of panicles and grain yield of upland rice increase with the increase of the nitrogen fertilization, but decrease in the presence of U. brizantha straw. Thus, it is recommend the use of early N fertilization in upland rice crop.

In semi-arid areas such as western Nebraska, interest in subsurface drip irrigation (SDI) for corn is increasing due to restricted irrigation allocations. However, crop response quantification to nitrogen (N) applications with SDI and the environmental benefits of multiple in-season (IS) SDI N applications instead of a single early-season (ES) surface application are lacking. The study was conducted in 2004, 2005, and 2006 at the University of Nebraska-Lincoln West Central Research and Extension Center in North Platte, Nebraska, comparing two N application methods (IS and ES) and three N rates (128, 186, and 278 kg N ha(-1)) using a randomized complete block design with four replications. No grain yield or biomass response was observed in 2004. In 2005 and 2006, corn grain yield and biomass production increased with increasing N rates, and the IS treatment increased grain yield, total N uptake, and gross return after N application costs (GRN) compared to the ES treatment. Chlorophyll meter readings taken at the R3 corn growth stage in 2006 showed that less N was supplied to the plant with ES compared to the IS treatment. At the end of the study, soil NO3-N masses in the 0.9 to 1.8 m depth were greater under the IS treatment compared to the ES treatment. Results suggested that greater losses of NO3-N below the root zone under the ES treatment may have had a negative effect on corn production. Under SDI systems, fertigating a recommended N rate at various corn growth stages can increase yields, GRN, and reduce NO3-N leaching in soils compared to concentrated early-season applications.

ENGLISH: The anchoveta, Cetengraulis mysticetus (Günther), is an important bait fish used to capture tunas in the Eastern Tropical Pacific Ocean. Contributions to the early life history of this species in the Gulf of Panama were made by Simpson (1959), who was able to identify deductively the planktonic egg of the anchoveta from 10 other anchovy eggs concurrently present. He also reared these planktonic eggs in the laboratory and described the resultant larvae to the age of 48 hours after hatching. Because of the lack of differences among the anchovy larvae, this description does not permit the identification of anchoveta larvae from those of other engraulid species. Furthermore, while adult specimens are easily recognized, up to the present it has not been possible to extend the identification of the juvenile anchoveta to specimens smaller than about 25 mm. The purpose of this study, therefore, was to identify anchoveta from the time of hatching to about 25 mm. SPANISH: La anchoveta, Cetengraulis mysticetus (Günther), es un importante pez de carnada que se emplea en la captura de los atunes en el Océano Pacífico Oriental Tropical. Simpson (1959) logró identificar deductivamente el huevo planctónico de la anchoveta al separarlo de otros diez huevos de anchoas que se encuentran al mismo tiempo, contribuyendo de esta manera a conocer los primeros estados de la historia natural de esta especie en el Golfo de Panamá. El también estableció un criadero en el laboratorio con estos huevos planctónicos y describió las larvas resultantes hasta la edad de 48 horas después de la eclosión. Debido a que no hay diferencias entre las larvas de las anchoas, esta descripción no permite identificar las larvas de la anchoveta de las otras especies de engráulidos. Más aun, a pesar de que los especímenes adultos son fácilmente reconocibles, hasta ahora no ha sido posible identificar la anchoveta juvenil de menos de unos 25 mm. Consecuentemente, el propósito del presente estudio ha sido el de identificar al anchoveta desde el momento de la eclosión hasta que tiene unos 25 mm.

Early embryogenesis in metazoa is controlled by maternally synthesized products. Among these products, the mature egg is loaded with transcripts representing approximately two thirds of the genome. A subset of this maternal RNA pool is degraded prior to the transition to zygotic control of development. This transfer of control of development from maternal to zygotic products is referred to as the midblastula transition (or MBT). It is believed that the degradation of maternal transcripts is required to terminate maternal control of development and to allow zygotic control of development to begin. Until now this process of maternal transcript degradation and the subsequent timing of the MBT has been poorly understood. I have demonstrated that in the early embryo there are two independent RNA degradation pathways, either of which is sufficient for transcript elimination. However, only the concerted action of both pathways leads to elimination of transcripts with the correct timing, at the MBT. The first pathway is maternally encoded, is triggered by egg activation, and is targeted to specific classes of mRNAs through cis-acting elements in the 3' untranslated region (UTR}. The second pathway is activated 2 hr after fertilization and functions together with the maternal pathway to ensure that transcripts are degraded by the MBT. In addition, some transcripts fail to degrade at select subcellular locations adding an element of spatial control to RNA degradation. The spatial control of RNA degradation is achieved by protecting, or masking, transcripts from the degradation machinery. The RNA degradation and protection events are regulated by distinct cis-elements in the 3' untranslated region (UTR). These results provide the first systematic dissection of this highly conserved process in development and demonstrate that RNA degradation is a novel mechanism used for both temporal and spatial control of development.

Part I. The cellular slime mold Dictyostelium discoideum is a simple eukaryote which undergoes a multi-cellular developmental process. Single cell myxamoebae divide vegetatively in the presence of a food source. When the food is depleted or removed, the cells aggregate, forming a migrating pseudoplasmodium which differentiates into a fruiting body containing stalk and spore cells. I have shown that during the developmental cycle glycogen phosphorylase, aminopeptidase, and alanine transaminase are developmentally regulated, that is their specific activities increased at a specific time in the developmental cycle. Phosphorylase activity is undetectable in developing cells until mid-aggregation whereupon it increases and reaches a maximum at mid-culmination. Thereafter the enzyme disappears. Actinomycin D and cycloheximide studies as well as studies with morphologically aberrant and temporally deranged mutants indicate that prior RNA and concomitant protein synthesis are necessary for the rise and decrease in activity and support the view that the appearance of the enzyme is regulated at the transcriptional level. Aminopeptidase and alanine transaminase increase 3 fold starting at starvation and reach maximum activity at 18 and 5 hours respectively.

The cellular DNA s of D. discoideum were characterized by CsC1 buoyant density gradient centrifugation and by renaturation kinetics. Whole cell DNA exhibits three bands in CsCl: ρ = 1.676 g/cc (nuclear main band), 1.687 (nuclear satellite), and 1.682 (mitochondrial). Reassociation kinetics at a criterion of Tm -23°C indicates that the nuclear reiterated sequences make up 30% of the genome (Cot1/2 (pure) 0.28) and the single-copy DNA 70% (Cot1/2(pure) 70). The complexity of the nuclear genome is 30 x 109 daltons and that of the mitochondrial DNA is 35-40 x 106 daltons (Cot1/2 0.15). rRNA cistrons constitute 2.2% of nuclear DNA and have a ρ = 1.682.

RNA extracted from 4 stages during developmental cycle of Dictyostelium was hybridized with purified single-copy nuclear DNA. The hybrids had properties indicative of single-copy DNA-RNA hybrids. These studies indicate that there are, during development, qualitative and quantitative changes in the portion of the single-copy of the genome transcribed. Overall, 56% of the genome is represented by transcripts between the amoeba and mid-culmination stages. Some 19% are sequences which are represented at all stages while 37% of the genome consists of stage specific sequences.

Part II. RNA and protein synthesis and polysome formation were studied during early development of the surf clam Spisula solidissima embryos. The oocyte has a small number of polysomes and a low but measurable rate of protein synthesis (leucine-3H incorporation). After fertilization, there is a continual increase in the percentage of ribosomes sedimenting in the polysome region. Newly synthesized RNA (uridine-5-3H incorporation) was found in polysomes as early as the 2-cell stage. During cleavage, the newly formed RNA is associated mainly with the light polysomes.

RNA extracted from polysomes labeled at the 4-cell stage is polydisperse, nonribosomal, and non-4 S. Actinomycin D causes a reduction of about 30% of the polysomes formed between fertilization and the 16-cell stage.

In the early cleavage stages the light polysomes are mostly affected by actinomycin.

In order to study the early developmental stages of Nandus nandus an experiment was conducted, where eggs and milt were obtained from the laboratory reared N nandus by stripping after 15 hours of 150 mg/kg body weight of carp PG extract injection. Then the eggs were fertilized in the laboratory and subsequent developmental stages were studied. First cleavage (two cell), four cell, eight cell, sixteen cell and multi cell stages were found 30, 50, 70, 105 and 160 minutes after fertilization respectively. Morula, early gastrula, middle gastrula, late gastrula and yolk plug stages were found 5, 8, 9, 11 and 13 hours after fertilization respectively. Hatching occurred within 20±2 hours after fertilization, and larvae were measured 1.60 mm in diameter. After one hour of hatching two melanophore bands were found at the caudal region of the body of the larvae. Eyes were first observed in l 0 hours, pectoral and pelvic fin buds appeared in 22 hours and well developed in 38 hours old larvae. Mouth cleft and brain lobes were visible when the larvae were 34 and 38 hours old respectively. Myomeres partially appeared in 16 hours, which were clearly visible in 74 hours old larvae. Larvae started wandering and searching for food after 56 hours of hatching. The yolk sac was completely absorbed when larvae became 62 hours old.
