Objectives: This in vitro study assessed the effect of a 4% TiF4 varnish on demineralisation and remineralisation of sound enamel and artificial carious enamel lesions, respectively.Methods: Bovine sound and carious enamel (n = 110) were randomly allocated to each type of varnish: Duraphat (R))-D (NaF, 2.26%F, pH 4.5, Colgate-Brazil, n = 30), Duofluorid (R)-F (NaF, 2.71%F, pH 8.0, FGM-Brazil, n = 30), TiF4-T (2.45%F, pH 1.0, FGM-Brazil, n = 30) and no-fluoride-P (FGM-Brazil, pH 5.0, n = 20). For the formation of artificial enamel caries, half of the blocks were immersed in 32 mL buffer acetate solution (16 h), whereas the other half was maintained sound. The varnishes were applied onto the enamel surfaces. Thus, the samples were subjected to pH cycles (37 degrees C) for 7 days. The response variables tested were surface and cross-sectional hardness. Data were tested using Kruskal-Wallis test (p < 0.05).Results: All F varnishes significantly reduced demineralisation and increased remineralisation in comparison to placebo. The TiF4 did not significantly reduce the surface enamel softening when compared with the other F varnishes, but it decreased the loss of subsurface hardness to the same extent. In enamel blocks with previous artificial carious lesions, the TiF4 significantly improved the rehardening compared to the other varnishes up to 30 mu m depth.Conclusions: The TiF4 varnish was able to decrease the demineralisation and increase the remineralisation of previously sound and carious enamel, respectively. It was equally effective compared to NaF varnishes on reducing the demineralisation at subsurface, but it was more effective on improving the remineralisation at surface and subsurface. Objectives: Iron ions (Fe2+) have been shown to be cariostatic in many studies particularly by their ability to reduce bacterial metabolism. Nevertheless, the role of iron ions on dissolution of enamel is unexplored. The aim of the present study was therefore to investigate the protective effect of increasing concentrations (0-120 mmol/L) of Fe2+ on the dissolution of enamel.Design: Enamel powder was subjected to acetic acid made with increasing concentrations with respect to FeSO4 center dot 7H(2)O. In order to determine the amount of enamel dissolved, the phosphate released in the medium was analysed spectrophotometrically using the Fiske-Subarrow method. Data were tested using Kruskall-Wall and Dunn's tests (p < 0.05). The degree of protection was found to approach maximum at about 15 mmol/L Fe2+. Higher concentrations of Fe2+ did not have an extra effect on inhibition of dissolution of enamel powder. In the next step, the protective effect of 15 mmol/L Fe2+ against mineral dissolution of the bovine enamel was evaluated using a simple abiotic model system. Enamel blocks were exposed to a sequence of seven plastic vials, each containing 1 mL of 10 mmol/L acetic acid. The acid in vial 4 was made 15 mmol/L with respect to FeSO4 center dot 7H(2)O. The mineral dissolved during each challenge was thus determined by phosphate released as described above. Data were tested using two-way ANOVA (p < 0.05). Results: Lower demineralisation (around 45%) was found in vial 4 (with Fe) that continued stable until vial 7.Conclusions: Thus, our data suggest that Fe2+, can be effective on inhibition of dissolution of enamel and that this effect may be durable. AIMS The aims of this double-blind, controlled, crossover study were to assess the influence of food preservatives on in situ dental biofilm growth and vitality, and to evaluate their influence on the ability of dental biofilm to demineralize underlying enamel over a period of 14 days. MATERIALS AND METHODS Twenty volunteers wore appliances with six specimens each of bovine enamel to build up intra-oral biofilms. During four test cycles of 14 days, the subjects had to place the appliance in one of the assigned controls or active solutions twice a day for a minute: negative control 0.9 % saline, 0.1 % benzoate (BA), 0.1 % sorbate (SA) and 0.2 % chlorhexidine (CHX positive control). After 14 days, the biofilms on two of the slabs were stained to visualize vital and dead bacteria to assess biofilm thickness (BT) and bacterial vitality (BV). Further, slabs were taken to determine mineral loss (ML), by quantitative light-induced laser fluorescence (QLF) and transversal microradiography (TMR), moreover the lesion depths (LD). RESULTS Nineteen subjects completed all test cycles. Use of SA, BA and CHX resulted in a significantly reduced BV compared to NaCl (p < 0.001). Only CHX exerted a statistically significant retardation in BT as compared to saline. Differences between SA and BA were not significant (p > 0.05) for both parameters. TMR analysis revealed the highest LD values in the NaCl group (43.6 ± 44.2 μm) and the lowest with CHX (11.7 ± 39.4 μm), while SA (22.9 ± 45.2 μm) and BA (21.4 ± 38.5 μm) lay in between. Similarly for ML, the highest mean values of 128.1 ± 207.3 vol% μm were assessed for NaCl, the lowest for CHX (-16.8 ± 284.2 vol% μm), while SA and BA led to values of 83.2 ± 150.9 and 98.4 ± 191.2 vol% μm, respectively. With QLF for both controls, NaCl (-33.8 ± 101.3 mm(2) %) and CHX (-16.9 ± 69.9 mm(2) %), negative values were recorded reflecting a diminution of fluorescence, while positive values were found with SA (33.9 ± 158.2 mm(2) %) and BA (24.8 ± 118.0 mm(2) %) depicting a fluorescence gain. These differences were non-significant (p > 0.05). CONCLUSION The biofilm model permited the assessment of undisturbed oral biofilm formation influenced by antibacterial components under clinical conditions for a period of 14 days. An effect of BA and SA on the demineralization of enamel could be demonstrated by TMR and QLF, but these new findings have to be seen as a trend. As part of our daily diet, these preservatives exert an impact on the metabolism of the dental biofilm, and therefore may even influence demineralization processes of the underlying dental enamel in situ.

Com a introdução do flúor como o principal agente anticariogênico e, talvez, um aumento do flúor na nossa cadeia alimentar, a fluorose dentária tornou-se um problema mundial. Os mecanismos que conduzem à formação do esmalte fluorótico são desconhecidos, mas devem envolver modificações nas reações físico-químicas básicas de desmineralização e remineralização do esmalte dentário. O aumento daquantidade de flúor no cristal apatita resulta no aumento dos parâmetros de rede. O objetivo deste trabalho é caracterizar o esmalte dentário humano saudável e fluorótico usando difração de raios X com luz síncrotron. Todos os perfis de espalhamento foram medidos na linha de difração de raios X (XRD1) do Laboratório Nacional de Luz Síncrotron, Campinas SP. Os experimentos foram realizados usando amostras em pó e em lâminas polidas. As amostras em pó foram analisadas a fim de obter a caracterização do esmalte dentário saudável. As lâminas foram analisadas em áreas do esmalte específicas identificadas como fluoróticas. Todos os perfis foram comparados com amostras de esmalte de controle e também com a literatura. A evidente similaridade entre os perfis de difração mostraram a analogia entre as estruturas do esmalte dentário e a hidroxiapatita padrão. Fica evidente que os perfis de difração do esmalte dentário das amostras em lâmina são diferentes daqueles obtidos para o esmalte em pó. As diferenças encontradas incluem variação na cristalinidade e orientação preferencial. Os valores encontrados para as distâncias interplanares para o esmalte de controle e fluorótico das amostras em lâmina não apresentaram diferenças estatisticamente significativas. Isto pode ser explicado pelo fato que a hidroxiapatita e a fluoropatita formam cristais com a mesma estrutura hexagonal, mesmo grupo de simetria e têm parâmetros de rede muito próximos, os quais a habilidade do sistema não foi suficiente para resolver. Finalmente, este trabalho mostra que a difração de raios X usando radiação síncrotron é uma técnica poderosa para o estudo da cristalografia e microestrutura do esmalte dentário e, ainda, pode ser igualmente aplicada no estudo de outros tecidos biológicos duros e de biomateriais sintéticos.

The process involves extraction of protein using 0.5% sodium hydroxide solution, bleaching the protein free mass with bleach liquor containing 0.3-0.5% available chlorine followed by demineralisation with 1.25 N hydrochloric acid in the cold and deacetylation using 1:1 (w/w) sodium hydroxide solution at 100°C for 2 hours.

The present study aimed at the utlisation of microbial organisms for the
production of good quality chitin and chitosan. The three strains used for the
study were Lactobacillus plantarum, Lactobacililus brevis and Bacillus subtilis.
These strains were selected on the basis of their acid producing ability to reduce
the pH of the fermenting substrates to prevent spoilage and thus caused
demineralisation of the shell. Besides, the proteolytic enzymes in these strains
acted on proteinaceous covering of shrimp and thus caused deprotenisation of
shrimp shell waste. Thus the two processes involved in chitin production can be
affected to certain extent using bacterial fermentation of shrimp shell.Optimization parameters like fermentation period, quantity of inoculum,
type of sugar, concentration of sugar etc. for fermentation with three different
strains were studied. For these, parameters like pH, Total titrable acidity (TTA),
changes in sugar concentration, changes in microbial count, sensory changes
etc. were studied.Fermentation study with Lactobacillus plantarum was continued with 20%
w/v jaggery broth for 15 days. The inoculum prepared yislded a cell
concentration of approximately 108 CFU/ml. In the present study, lactic acid and
dilute hydrochloric acid were used for initial pH adjustment because; without
adjusting the initial pH, it took more than 5 hours for the lactic acid bacteria to
convert glucose to lactic acid and during this delay spoilage occurred due to
putrefying enzymes active at neutral or higher pH. During the fermentation study,
pH first decreased in correspondence with increase in TTA values. This showed
a clear indication of acid production by the strain. This trend continued till their
proteolytic activity showed an increasing trend. When the available sugar source
started depleting, proteolytic activity also decreased and pH increased. This was
clearly reflected in the sensory evaluation results. Lactic acid treated samples
showed greater extent of demineralization and deprotenisation at the end of
fermentation study than hydrochloric acid treated samples. It can be due to the
effect of strong hydrochloric acid on the initial microbial count, which directly
affects the fermentation process. At the end of fermentation, about 76.5% of ash was removed in lactic acid treated samples and 71.8% in hydrochloric acid
treated samples; 72.8% of proteins in lactic acid treated samples and 70.6% in
hydrochloric acid treated samples.The residual protein and ash in the fermented residue were reduced to
permissible limit by treatment with 0.8N HCI and 1M NaOH. Characteristics of
chitin like chitin content, ash content, protein content, % of N- acetylation etc.
were studied. Quality characteristics like viscosity, degree of deacetylation and
molecular weight of chitosan prepared were also compared. The chitosan
samples prepared from lactic acid treated showed high viscosity than HCI treated
samples. But degree of deacetylation is more in HCI treated samples than lactic
acid treated ones. Characteristics of protein liquor obtained like its biogenic
composition, amino acid composition, total volatile base nitrogen, alpha amino
nitrogen etc. also were studied to find out its suitability as animal feed
supplement.Optimization of fermentation parameters for Lactobacillus brevis
fermentation study was also conducted and parameters were standardized. Then
detailed fermentation study was done in 20%wlv jaggery broth for 17 days. Also
the effect of two different acid treatments (mild HCI and lactic acid) used for initial
pH adjustment on chitin production were also studied. In this study also trend of
changes in pH. changes in sugar concentration ,microbial count changes were
similar to Lactobacillus plantarum studies. At the end of fermentation, residual
protein in the samples were only 32.48% in HCI treated samples and 31.85% in
lactic acid treated samples. The residual ash content was about 33.68% in HCI
treated ones and 32.52% in lactic acid treated ones. The fermented residue was
converted to chitin with good characteristics by treatment with 1.2MNaOH and
1NHCI.Characteristics of chitin samples prepared were studied and extent of Nacetylation
was about 84% in HCI treated chitin and 85%in lactic acid treated
ones assessed from FTIR spectrum. Chitosan was prepared from these samples
by usual chemical method and its extent of solubility, degree of deacetylation,
viscosity and molecular weight etc were studied. The values of viscosity and
molecular weight of the samples prepared were comparatively less than the
chitosan prepared by Lactobacillus plantarum fermentation. Characteristics of protein liquor obtained were analyzed to determine its quality and is suitability as
animal feed supplement.Another strain used for the study was Bacillus subtilis and fermentation
was carried out in 20%w/v jaggery broth for 15 days. It was found that Bacillus
subtilis was more efficient than other Lactobacillus species for deprotenisation
and demineralization. This was mainly due to the difference in the proteolytic

BACKGROUND: Chemical chitin extraction generates large amounts of wastes and increases partial deacetylation of the product. Therefore, the use of biological methods for chitin extraction is an interesting alternative. The effects of process conditions on enzyme assisted extraction of chitin from the shrimp shells in a systematic way were the focal points of this study. RESULTS: Demineralisation conditions of 25C, 20 min, shells-lactic acid ratio of 1:1.1 w/w; and shells-acetic acid ratio of 1:1.2 w/w, the maximum demineralisation values were 98.64 and 97.57% for lactic and acetic acids, respectively. A total protein removal efficiency of 91.10% by protease from Streptomyces griseus with enzyme-substrate ratio 55 U/g, pH 7.0 and incubation time 3 h is obtained when the particle size range is 50-25 μm, which was identified as the most critical factor. The X-ray diffraction and 13C NMR spectroscopy analysis showed that the lower percent crystallinity and higher degree of acetylation of chitin from enzyme assisted extraction may exhibit better solubility properties and less depolymerisation in comparison with chitin from the chemical extraction. CONCLUSION: The present work investigates the effects of individual factors on process yields, and it has shown that, if the particle size is properly controlled a reaction time of 3 h is more than enough for deproteination by protease. Physicochemical analysis indicated that the enzyme assisted production of chitin seems appropriate to extract chitin, possibly retaining its native structure.

Purpose: The aim of this in situ double-blind randomised crossover study was to investigate the effect of calcium (Ca) pre-rinse on the composition of plaque and on enamel prior to the use of fluoride (F) dentifrice. Materials and Methods: During four phases (14 days each) of this study, 10 volunteers had agreed to wear dental appliances containing two healthy bovine enamel blocks. A fresh solution containing 20% weight/volume (w/v) sucrose was dripped on the enamel blocks ex vivo for 5 min three times a day. Subsequently, the appliances were replaced in the mouth, and the volunteers rinsed their mouth with 10 mL of a Ca (150 mmol/L) or a placebo rinse (1 min). In sequence, a slurry (1:3 w/v) of F (1030 ppm) or placebo dentifrice was dripped onto the blocks ex vivo for 1 min. During this time, the volunteers brushed their teeth with the respective dentifrice. The appliances were replaced in the mouth, and the volunteers rinsed their mouth with water. The plaque formed on the blocks was analysed for F and Ca. The enamel demineralisation as well as the incorporation of F on enamel was evaluated by cross-sectional microhardness and alkali-soluble F analysis, respectively. Data were tested using analysis of variance (P < 0.05). Results: The Ca pre-rinse prior to the use of the F dentifrice led to a three- and sixfold increase in the plaque F and Ca concentrations, respectively. It also did not have any additive effect on the F content on the enamel and the demineralisation of the enamel, in comparison with the use of F dentifrice alone. Conclusions: A Ca lactate rinse used prior to the F dentifrice was able to change the mineral content in the plaque, but it was unable to prevent enamel demineralisation.

Objectives: The aim of this in vitro study was to assess the effects of saliva substitutes (modified with respect to calcium, phosphates, and fluorides) in combination with a high-concentrated fluoride toothpaste on demineralised dentin.Methods: Before and after demineralisation of bovine dentin specimens (subsurface lesions; 37 degrees C, pH 5.0, 5 d), one-quarter of each specimen's surface was covered with nail varnish (control of sound/demineralised tissue). Subsequently, specimens were exposed to original Saliva natura (saturation with respect to octacalciumphosphate [S(OCP)]: 0.03; SN 0), or to three lab-produced Saliva natura modifications (S(OCP): 1, 2, and 3; SN 1-3) for 2 and 5 weeks (37 degrees C). An aqueous solution (S(OCP): 2.5) served as positive control (PC). Two times daily (2 min each), Duraphat toothpaste (5000 ppm F(-); Colgate)/saliva substitute slurry (ratio 1:3) was applied gently. Differences in mineral losses (Delta Delta Z) and lesion depths (Delta LD) between values before and after exposure were microradiographically evaluated.Results: After both treatment periods specimens immersed in SN 0 revealed significantly higher mineral losses (lower Delta Delta Z values) and lesion depths (lower Delta LD) compared to PC (p < 0.05; ANOVA). After 5 weeks, specimens stored in SN 1 and 2 showed significantly higher mineral losses compared to PC (p < 0.05), while those stored in SN 3 showed similar results (p > 0.05). No differences in mineral loss could be observed between SN 2 and 3 (p > 0.05).Conclusions: Under the conditions of this limited protocol, the combination of Saliva natura solutions slightly saturated with respect to OCP in combination with a high-concentrated fluoride toothpaste enabled remineralisation of dentin in vitro. Design: During four phases of 14 days, 10 volunteers wore appliances containing two artificially demineralised bovine enamel blocks. Three times a day, they rinsed with 10 mL, of Ca (150 mM) or placebo rinse (1 min). A slurry (1:3, w/v) of F (1030 ppm) or placebo dentifrice was dripped onto the blocks. During I min, the volunteers brushed their teeth with the respective dentifrice. The appliance was replaced into the mouth and the volunteers rinsed with water. The biofilm formed on the blocks was analysed for F and Ca. Enamel alterations were evaluated by the percentage of surface microhardness change (%SMHC), cross-sectional microhardness (% mineral volume) and alkali-soluble F analysis. Data were analysed by ANOVA (p < 0.05). Results: the use of the Ca pre-rinse before the F dentifrice produced a six- and four-fold increase in biofilm F and Ca concentrations, respectively. For enamel, the remineralisation was significantly improved by the Ca pre-rinse when compared to the other treatments. There was a significantly higher concentration of alkali-soluble F in enamel when the F dentifrice was used, but the Ca pre-rinse did not have any significant additive effect. Conclusions: According to our protocol, the Ca pre-rinse significantly increased biofilm F concentration and, regardless the use of F dentifrice, significantly enhanced the remineralisation of artificially demineralised enamel. Objectives: This in situ/ex vivo study evaluated whether a rinse with an iron solution could reduce wear and the percentage of microhardness change of human enamel and dentine submitted to erosion followed by brushing after 1 or 30 min.Design: During 2 experimental 5-day crossover phases (wash-out period of 10 days), 10 volunteers wore intraoral palatal devices, with 12 specimens (6 of enamel and 6 of dentine) arranged in 3 horizontal rows (4 specimens each). In one phase, the volunteers immersed the device for 5 min in 150 mL of cola drink, 4 times a day. Immediately after immersion, no treatment was performed in one row. The other row was brushed after 1 min using a fluoride dentifrice and the device was replaced into mouth. After 30 min, the remaining row was brushed. In the other phase, the procedures were repeated, but after immersion the volunteers rinsed for 1 min with 10 mL of a 10 mM ferrous sulphate solution. Changes in surface microhardness (%SMH) and wear (profilometry) of enamel and dentine were measured. Data were tested using ANOVA and Tukey's tests (p < 0.05).Results: the enamel presented more wear than dentine, under all experimental conditions. The iron solution caused a significant reduction on the %SMH in enamel, and a significant reduction on the wear in dentine, regardless the other conditions.Conclusions: Rinsing with an iron solution after an erosive attack, followed or not by an abrasive episode, may be a viable alternative to reduce the loss of dental structure. A Lhassa Apso is presented in emergency after having been injured by another dog. It was suffering from a mandibular fracture. The clinical exam revealed generalized and excessive mobility of the whole dentition. Radiographs showed generalized jaw bone demineralisation. A blood sample was analysed and revealed chronic renal disease. We came to the conclusion that this dog was suffering from secondary renal hyperparathyroidism. The hypocalcified bone was so thin that a slight trauma induced the mandibular fracture.
