Diplomityön tavoitteena oli Karhulanvalimon valutoimitusprosessin ohjattavuuden kehittäminen. Asiakastyytyväisyyskyselyin on selvitetty valutoimitusprosessin nykytilaa ja kartoitettu kehityskohteita. Keskeisiksi kehityskohteiksi muodostuivat toimituskyvyn ja läpäisyajan kehittäminen. Kohdeyritystä on tutkittu ohjattavuusanalyysillä. Ohjattavuus analyysin perusteella parannuksen painopisteet suuntautuvat toimituskyvyn j a kapasiteetin hallinnan kehittämiseen. Työn keskeisiin aiheisiin liittyen on luotu kirjallisuuteen perustuva katsaus. Tärkeimpiä aihealueita ovat ohjattavuusanalyysi, varastojen hallinta ja tuotannon virtautus. Ennen varsinaisia kehitystoimenpiteitä on pyritty saamaan tietoa tämänhetkisen toiminnan tasosta ja pyritty löytämään parhaita toimintatapoja benchmarking-tyylistä toimintamallia soveltaen. Varastojen hallinnan kehittämistä työssä on lähestytty käytännönläheisesti. Varastoanalyyseihin perustuen on suoritettu kehitystoimenpiteitä ja luotu suunnitelmia sekä esitetty ehdotuksia varastojen hallinnan kehittämiseksi. Työssä on myös pyritty analysoimaan yksittäisiä tuotannon ohjattavuuden ongelmia ja pyritty näiden poistamiseen. Mikäli ohjattavuusongelman välitönpoistaminen ei ole mahdollista, on ongelmiin esitetty kehitysehdotuksia. Työssä on luotu suunnitelma läpäisyajan lyhentämiseksi tuotannon virtauttamiseen perustuen. Virtautuksessa on huomioitava materiaalien ja nimikkeiden suuri määrä, tuotannon layout ja tuotantovälineiden aiheuttamat rajoitukset. Layoutin muutostyössä on pyrittävä tiiviyteen. Diplomityön loppuosassa on tutkittu mahdollisuuksia Karhulan valimon ja sen pääasiakkaiden varastojen pienentämiseksi ja toiminnan kehittämiseksi toimituskykyä ja läpäisyaikaa vaarantamatta. Varastojen analysoinnin perusteella onmallinnettu varastoinnin taloudellista säästöpotentiaalia, vertailtu vaihtoehtoisia toimintamalleja ja esitetty kehitysehdotus sekä välttämättömiä toimenpiteitä varastoinnin kehittämiseksi.

Construction professional services (CPSs), such as architecture, engineering, and consultancy, are not only high value-added profit centers in their own right but also have a knock-on effect on other businesses, such as construction and the export of materials and machinery. Arguably, competition in the international construction market has shifted to these knowledge-intensive CPS areas. Yet CPSs represent a research frontier that has received scant attention. This research aims to enrich the body of knowledge on CPSs by examining strengths, weaknesses, opportunities, and threats (SWOT) of Chinese CPSs (CCPSs) in the international context. It does so by triangulating theories with quantitative and qualitative data gleaned from yearbooks, annual reports, interviews, seminars, and interactions with managers in major CCPS companies. It is found that CCPSs present both strengths and weaknesses in talents, administration systems, and development strategies in dealing with the external opportunities and threats brought about by globalization and market evolution. Low price, which has helped the Chinese construction business to succeed in the international market, is also a major CCPS strength. An opportunity for CCPSs is the relatively strong delivery capability possessed by Chinese contractors; by partnering with them CCPSs can better establish themselves in the international arena. This is probably the first ever comprehensive study on the performance of CCPSs in the international marketplace. The research is conducted at an opportune time, particularly when the world is witnessing the burgeoning force of Chinese businesses in many areas including manufacturing, construction, and, potentially, professional services. It adds new insights to the knowledge body of CPSs and provides valuable references to other countries faced with the challenge of developing CPS business efficiently in the international market.

OBJECTIVES This study sought to describe the frequency and clinical impact of acute scaffold disruption and late strut discontinuity of the second-generation Absorb bioresorbable polymeric vascular scaffolds (Absorb BVS, Abbott Vascular, Santa Clara, California) in the ABSORB (A Clinical Evaluation of the Bioabsorbable Everolimus Eluting Coronary Stent System in the Treatment of Patients With De Novo Native Coronary Artery Lesions) cohort B study by optical coherence tomography (OCT) post-procedure and at 6, 12, 24, and 36 months. BACKGROUND Fully bioresorbable scaffolds are a novel approach to treatment for coronary narrowing that provides transient vessel support with drug delivery capability without the long-term limitations of metallic drug-eluting stents. However, a potential drawback of the bioresorbable scaffold is the potential for disruption of the strut network when overexpanded. Conversely, the structural discontinuity of the polymeric struts at a late stage is a biologically programmed fate of the scaffold during the course of bioresorption. METHODS The ABSORB cohort B trial is a multicenter single-arm trial assessing the safety and performance of the Absorb BVS in the treatment of 101 patients with de novo native coronary artery lesions. The current analysis included 51 patients with 143 OCT pullbacks who underwent OCT at baseline and follow-up. The presence of acute disruption or late discontinuities was diagnosed by the presence on OCT of stacked, overhung struts or isolated intraluminal struts disconnected from the expected circularity of the device. RESULTS Of 51 patients with OCT imaging post-procedure, acute scaffold disruption was observed in 2 patients (3.9%), which could be related to overexpansion of the scaffold at the time of implantation. One patient had a target lesion revascularization that was presumably related to the disruption. Of 49 patients without acute disruption, late discontinuities were observed in 21 patients. There were no major adverse cardiac events associated with this finding except for 1 patient who had a non-ischemia-driven target lesion revascularization. CONCLUSIONS Acute scaffold disruption is a rare iatrogenic phenomenon that has been anecdotally associated with anginal symptoms, whereas late strut discontinuity is observed in approximately 40% of patients and could be viewed as a serendipitous OCT finding of a normal bioresorption process without clinical implications. Wireless sensor networks (WSNs) have shown their potentials in various applications, which bring a lot of benefits to users from both research and industrial areas. For many setups, it is envisioned thatWSNs will consist of tens to hundreds of nodes that operate on small batteries. However due to the diversity of the deployed environments and resource constraints on radio communication, sensing ability and energy supply, it is a very challenging issue to plan optimized WSN topology and predict its performance before real deployment. During the network planning phase, the connectivity, coverage, cost, network longevity and service quality should all be considered. Therefore it requires designers coping with comprehensive and interdisciplinary knowledge, including networking, radio engineering, embedded system and so on, in order to efficiently construct a reliable WSN for any specific types of environment. Nowadays there is still a lack of the analysis and experiences to guide WSN designers to efficiently construct WSN topology successfully without many trials. Therefore, simulation is a feasible approach to the quantitative analysis of the performance of wireless sensor networks. However the existing planning algorithms and tools, to some extent, have serious limitations to practically design reliable WSN topology: Only a few of them tackle the 3D deployment issue, and an overwhelming number of works are proposed to place devices in 2D scheme. Without considering the full dimension, the impacts of environment to the performance of WSN are not completely studied, thus the values of evaluated metrics such as connectivity and sensing coverage are not sufficiently accurate to make proper decision. Even fewer planning methods model the sensing coverage and radio propagation by considering the realistic scenario where obstacles exist. Radio signals propagate with multi-path phenomenon in the real world, in which direct paths, reflected paths and diffracted paths contribute to the received signal strength. Besides, obstacles between the path of sensor and objects might block the sensing signals, thus create coverage hole in the application. None of the existing planning algorithms model the network longevity and packet delivery capability properly and practically. They often employ unilateral and unrealistic formulations. The optimization targets are often one-sided in the current works. Without comprehensive evaluation on the important metrics, the performance of planned WSNs can not be reliable and entirely optimized. Modeling of environment is usually time consuming and the cost is very high, while none of the current works figure out any method to model the 3D deployment environment efficiently and accurately. Therefore many researchers are trapped by this issue, and their algorithms can only be evaluated in the same scenario, without the possibility to test the robustness and feasibility for implementations in different environments. In this thesis, we propose a novel planning methodology and an intelligent WSN planning tool to assist WSN designers efficiently planning reliable WSNs. First of all, a new method is proposed to efficiently and automatically model the 3D indoor and outdoor environments. To the best of our knowledge, this is the first time that the advantages of image understanding algorithm are applied to automatically reconstruct 3D outdoor and indoor scenarios for signal propagation and network planning purpose. The experimental results indicate that the proposed methodology is able to accurately recognize different objects from the satellite images of the outdoor target regions and from the scanned floor plan of indoor area. Its mechanism offers users a flexibility to reconstruct different types of environment without any human interaction. Thereby it significantly reduces human efforts, cost and time spent on reconstructing a 3D geographic database and allows WSN designers concentrating on the planning issues. Secondly, an efficient ray-tracing engine is developed to accurately and practically model the radio propagation and sensing signal on the constructed 3D map. The engine contributes on efficiency and accuracy to the estimated results. By using image processing concepts, including the kd-tree space division algorithm and modified polar sweep algorithm, the rays are traced efficiently without detecting all the primitives in the scene. The radio propagation model iv is proposed, which emphasizes not only the materials of obstacles but also their locations along the signal path. The sensing signal of sensor nodes, which is sensitive to the obstacles, is benefit from the ray-tracing algorithm via obstacle detection. The performance of this modelling method is robust and accurate compared with conventional methods, and experimental results imply that this methodology is suitable for both outdoor urban scenes and indoor environments. Moreover, it can be applied to either GSM communication or ZigBee protocol by varying frequency parameter of the radio propagation model. Thirdly, WSN planning method is proposed to tackle the above mentioned challenges and efficiently deploy reliable WSNs. More metrics (connectivity, coverage, cost, lifetime, packet latency and packet drop rate) are modeled more practically compared with other works. Especially 3D ray tracing method is used to model the radio link and sensing signal which are sensitive to the obstruction of obstacles; network routing is constructed by using AODV protocol; the network longevity, packet delay and packet drop rate are obtained via simulating practical events in WSNet simulator, which to the best of our knowledge, is the first time that network simulator is involved in a planning algorithm. Moreover, a multi-objective optimization algorithm is developed to cater for the characteristics of WSNs. The capability of providing multiple optimized solutions simultaneously allows users making their own decisions accordingly, and the results are more comprehensively optimized compared with other state-of-the-art algorithms. iMOST is developed by integrating the introduced algorithms, to assist WSN designers efficiently planning reliable WSNs for different configurations. The abbreviated name iMOST stands for an Intelligent Multi-objective Optimization Sensor network planning Tool. iMOST contributes on: (1) Convenient operation with a user-friendly vision system; (2) Efficient and automatic 3D database reconstruction and fast 3D objects design for both indoor and outdoor environments; (3) It provides multiple multi-objective optimized 3D deployment solutions and allows users to configure the network properties, hence it can adapt to various WSN applications; (4) Deployment solutions in the 3D space and the corresponding evaluated performance are visually presented to users; and (5) The Node Placement Module of iMOST is available online as well as the source code of the other two rebuilt heuristics. Therefore WSN designers will be benefit from v this tool on efficiently constructing environment database, practically and efficiently planning reliable WSNs for both outdoor and indoor applications. With the open source codes, they are also able to compare their developed algorithms with ours to contribute to this academic field. Finally, solid real results are obtained for both indoor and outdoor WSN planning. Deployments have been realized for both indoor and outdoor environments based on the provided planning solutions. The measured results coincide well with the estimated results. The proposed planning algorithm is adaptable according to the WSN designer’s desirability and configuration, and it offers flexibility to plan small and large scale, indoor and outdoor 3D deployments. The thesis is organized in 7 chapters. In Chapter 1, WSN applications and motivations of this work are introduced, the state-of-the-art planning algorithms and tools are reviewed, challenges are stated out and the proposed methodology is briefly introduced. In Chapter 2, the proposed 3D environment reconstruction methodology is introduced and its performance is evaluated for both outdoor and indoor environment. The developed ray-tracing engine and proposed radio propagation modelling method are described in details in Chapter 3, their performances are evaluated in terms of computation efficiency and accuracy. Chapter 4 presents the modelling of important metrics of WSNs and the proposed multi-objective optimization planning algorithm, the performance is compared with the other state-of-the-art planning algorithms. The intelligent WSN planning tool iMOST is described in Chapter 5. RealWSN deployments are prosecuted based on the planned solutions for both indoor and outdoor scenarios, important data are measured and results are analysed in Chapter 6. Chapter 7 concludes the thesis and discusses about future works. vi Resumen en Castellano Las redes de sensores inalámbricas (en inglés Wireless Sensor Networks, WSNs) han demostrado su potencial en diversas aplicaciones que aportan una gran cantidad de beneficios para el campo de la investigación y de la industria. Para muchas configuraciones se prevé que las WSNs consistirán en decenas o cientos de nodos que funcionarán con baterías pequeñas. Sin embargo, debido a la diversidad de los ambientes para desplegar las redes y a las limitaciones de recursos en materia de comunicación de radio, capacidad de detección y suministro de energía, la planificación de la topología de la red y la predicción de su rendimiento es un tema muy difícil de tratar antes de la implementación real. Durante la fase de planificación del despliegue de la red se deben considerar aspectos como la conectividad, la cobertura, el coste, la longevidad de la red y la calidad del servicio. Por lo tanto, requiere de diseñadores con un amplio e interdisciplinario nivel de conocimiento que incluye la creación de redes, la ingeniería de radio y los sistemas embebidos entre otros, con el fin de construir de manera eficiente una WSN confiable para cualquier tipo de entorno. Hoy en día todavía hay una falta de análisis y experiencias que orienten a los diseñadores de WSN para construir las topologías WSN de manera eficiente sin realizar muchas pruebas. Por lo tanto, la simulación es un enfoque viable para el análisis cuantitativo del rendimiento de las redes de sensores inalámbricos. Sin embargo, los algoritmos y herramientas de planificación existentes tienen, en cierta medida, serias limitaciones para diseñar en la práctica una topología fiable de WSN: Sólo unos pocos abordan la cuestión del despliegue 3D mientras que existe una gran cantidad de trabajos que colocan los dispositivos en 2D. Si no se analiza la dimensión completa (3D), los efectos del entorno en el desempeño de WSN no se estudian por completo, por lo que los valores de los parámetros evaluados, como la conectividad y la cobertura de detección, no son lo suficientemente precisos para tomar la decisión correcta. Aún en menor medida los métodos de planificación modelan la cobertura de los sensores y la propagación de la señal de radio teniendo en cuenta un escenario realista donde existan obstáculos. Las señales de radio en el mundo real siguen una propagación multicamino, en la que los caminos directos, los caminos reflejados y los caminos difractados contribuyen a la intensidad de la señal recibida. Además, los obstáculos entre el recorrido del sensor y los objetos pueden bloquear las señales de detección y por lo tanto crear áreas sin cobertura en la aplicación. Ninguno de los algoritmos de planificación existentes modelan el tiempo de vida de la red y la capacidad de entrega de paquetes correctamente y prácticamente. A menudo se emplean formulaciones unilaterales y poco realistas. Los objetivos de optimización son a menudo tratados unilateralmente en los trabajos actuales. Sin una evaluación exhaustiva de los parámetros importantes, el rendimiento previsto de las redes inalámbricas de sensores no puede ser fiable y totalmente optimizado. Por lo general, el modelado del entorno conlleva mucho tiempo y tiene un coste muy alto, pero ninguno de los trabajos actuales propone algún método para modelar el entorno de despliegue 3D con eficiencia y precisión. Por lo tanto, muchos investigadores están limitados por este problema y sus algoritmos sólo se pueden evaluar en el mismo escenario, sin la posibilidad de probar la solidez y viabilidad para las implementaciones en diferentes entornos. En esta tesis, se propone una nueva metodología de planificación así como una herramienta inteligente de planificación de redes de sensores inalámbricas para ayudar a los diseñadores a planificar WSNs fiables de una manera eficiente. En primer lugar, se propone un nuevo método para modelar demanera eficiente y automática los ambientes interiores y exteriores en 3D. Según nuestros conocimientos hasta la fecha, esta es la primera vez que las ventajas del algoritmo de _image understanding_se aplican para reconstruir automáticamente los escenarios exteriores e interiores en 3D para analizar la propagación de la señal y viii la planificación de la red. Los resultados experimentales indican que la metodología propuesta es capaz de reconocer con precisión los diferentes objetos presentes en las imágenes satelitales de las regiones objetivo en el exterior y de la planta escaneada en el interior. Su mecanismo ofrece a los usuarios la flexibilidad para reconstruir los diferentes tipos de entornos sin ninguna interacción humana. De este modo se reduce considerablemente el esfuerzo humano, el coste y el tiempo invertido en la reconstrucción de una base de datos geográfica con información 3D, permitiendo así que los diseñadores se concentren en los temas de planificación. En segundo lugar, se ha desarrollado un motor de trazado de rayos (en inglés ray tracing) eficiente para modelar con precisión la propagación de la señal de radio y la señal de los sensores en el mapa 3D construido. El motor contribuye a la eficiencia y la precisión de los resultados estimados. Mediante el uso de los conceptos de procesamiento de imágenes, incluyendo el algoritmo del árbol kd para la división del espacio y el algoritmo _polar sweep_modificado, los rayos se trazan de manera eficiente sin la detección de todas las primitivas en la escena. El modelo de propagación de radio que se propone no sólo considera los materiales de los obstáculos, sino también su ubicación a lo largo de la ruta de señal. La señal de los sensores de los nodos, que es sensible a los obstáculos, se ve beneficiada por la detección de objetos llevada a cabo por el algoritmo de trazado de rayos. El rendimiento de este método de modelado es robusto y preciso en comparación con los métodos convencionales, y los resultados experimentales indican que esta metodología es adecuada tanto para escenas urbanas al aire libre como para ambientes interiores. Por otra parte, se puede aplicar a cualquier comunicación GSM o protocolo ZigBee mediante la variación de la frecuencia del modelo de propagación de radio. En tercer lugar, se propone un método de planificación de WSNs para hacer frente a los desafíos mencionados anteriormente y desplegar redes de sensores fiables de manera eficiente. Se modelan más parámetros (conectividad, cobertura, coste, tiempo de vida, la latencia de paquetes y tasa de caída de paquetes) en comparación con otros trabajos. Especialmente el método de trazado de rayos 3D se utiliza para modelar el enlace de radio y señal de los sensores que son sensibles a la obstrucción de obstáculos; el enrutamiento de la red se construye utilizando el protocolo AODV; la longevidad de la red, retardo de paquetes ix y tasa de abandono de paquetes se obtienen a través de la simulación de eventos prácticos en el simulador WSNet, y según nuestros conocimientos hasta la fecha, es la primera vez que simulador de red está implicado en un algoritmo de planificación. Por otra parte, se ha desarrollado un algoritmo de optimización multi-objetivo para satisfacer las características de las redes inalámbricas de sensores. La capacidad de proporcionar múltiples soluciones optimizadas de forma simultánea permite a los usuarios tomar sus propias decisiones en consecuencia, obteniendo mejores resultados en comparación con otros algoritmos del estado del arte. iMOST se desarrolla mediante la integración de los algoritmos presentados, para ayudar de forma eficiente a los diseñadores en la planificación de WSNs fiables para diferentes configuraciones. El nombre abreviado iMOST (Intelligent Multi-objective Optimization Sensor network planning Tool) representa una herramienta inteligente de planificación de redes de sensores con optimización multi-objetivo. iMOST contribuye en: (1) Operación conveniente con una interfaz de fácil uso, (2) Reconstrucción eficiente y automática de una base de datos con información 3D y diseño rápido de objetos 3D para ambientes interiores y exteriores, (3) Proporciona varias soluciones de despliegue optimizadas para los multi-objetivo en 3D y permite a los usuarios configurar las propiedades de red, por lo que puede adaptarse a diversas aplicaciones de WSN, (4) las soluciones de implementación en el espacio 3D y el correspondiente rendimiento evaluado se presentan visualmente a los usuarios, y (5) El _Node Placement Module_de iMOST está disponible en línea, así como el código fuente de las otras dos heurísticas de planificación. Por lo tanto los diseñadores WSN se beneficiarán de esta herramienta para la construcción eficiente de la base de datos con información del entorno, la planificación práctica y eficiente de WSNs fiables tanto para aplicaciones interiores y exteriores. Con los códigos fuente abiertos, son capaces de comparar sus algoritmos desarrollados con los nuestros para contribuir a este campo académico. Por último, se obtienen resultados reales sólidos tanto para la planificación de WSN en interiores y exteriores. Los despliegues se han realizado tanto para ambientes de interior y como para ambientes de exterior utilizando las soluciones de planificación propuestas. Los resultados medidos coinciden en gran medida con los resultados estimados. El algoritmo de planificación x propuesto se adapta convenientemente al deiseño de redes de sensores inalámbricas, y ofrece flexibilidad para planificar los despliegues 3D a pequeña y gran escala tanto en interiores como en exteriores. La tesis se estructura en 7 capítulos. En el Capítulo 1, se presentan las aplicaciones de WSN y motivaciones de este trabajo, se revisan los algoritmos y herramientas de planificación del estado del arte, se presentan los retos y se describe brevemente la metodología propuesta. En el Capítulo 2, se presenta la metodología de reconstrucción de entornos 3D propuesta y su rendimiento es evaluado tanto para espacios exteriores como para espacios interiores. El motor de trazado de rayos desarrollado y el método de modelado de propagación de radio propuesto se describen en detalle en el Capítulo 3, evaluándose en términos de eficiencia computacional y precisión. En el Capítulo 4 se presenta el modelado de los parámetros importantes de las WSNs y el algoritmo de planificación de optimización multi-objetivo propuesto, el rendimiento se compara con los otros algoritmos de planificación descritos en el estado del arte. La herramienta inteligente de planificación de redes de sensores inalámbricas, iMOST, se describe en el Capítulo 5. En el Capítulo 6 se llevan a cabo despliegues reales de acuerdo a las soluciones previstas para los escenarios interiores y exteriores, se miden los datos importantes y se analizan los resultados. En el Capítulo 7 se concluye la tesis y se discute acerca de los trabajos futuros.

This thesis is concerned with the development of hydrogels that adhere to skin and can be used for topical or trans dermal release of active compounds for therapeutic or cosmetic use. The suitability of a range of monomers and initiator systems for the production of skin adhesive hydro gels by photopolymerisation was explored and an approximate order of monomer reactivity in aqueous solution was determined. Most notably, the increased reactivity of N-vinyl pyrrolidone within an aqueous system, as compared to its low rate of polymerisation in organic solvents, was observed. The efficacy of a series of photoinitiator systems for the preparation of sheet hydrogels was investigated. Supplementary redox and thermal initiators were also examined. The most successful initiator system was found to be Irgacure 184, which is commonly used in commercial moving web production systems that employ photopolymerisation. The influence of ionic and non-ionic monomers, crosslinking systems, water and glycerol on the adhesive and dynamic mechanical behaviour of partially hydrated hydrogel systems was examined. The aim was to manipulate hydrogel behaviour to modify topical and transdermal delivery capability and investigated the possibility of using monomer combinations that would influence the release characteristics of gels by modifying their hydrophobic and ionic nature. The copolymerisation of neutral monomers (N-vinyl pyrrolidone, N,N-dimethyl acrylamide and N-acryloyl morpholine) with ionic monomers (2-acrylamido-2-methylpropane sulphonic acid; sodium salt, and the potassium salt of 3-sulphopropyl acrylate) formed the basis of the study. Release from fully and partially hydrated hydrogels was studied, using model compounds and a non-steroidal anti-inflammatory drug, Ibuprofen. Release followed a common 3-stage kinetic profile that includes an initial burst phase, a secondary phase of approximate first order release and a final stage of infinitesimally slow release such that the compound is effectively retained within the hydrogel. Use of partition coefficients, the pKa of the active and a knowledge of charge-based and polar interactions of polymer and drug were complementary in interpreting experimental results. In summary, drug ionisation, hydrogel composition and external release medium characteristics interact to influence release behaviour. The information generated provides the basis for the optimal design of hydrogels for specific dermal release applications and some understanding of the limitations of these systems for controlled release applications.

OBJECTIVES: Our analysis assessed the impact of information on patients' preferences in prescription versus over-the-counter (OTC) delivery systems. METHODS: A contingent valuation (CV) study was implemented, randomly assigning 534 lay people into the receipt of limited or extended information concerning new influenza drugs. In each information arm, people answered two questions: the first asked about willingness to pay (WTP) for the new prescription drug; the second asked about WTP for the same drug sold OTC. RESULTS: We show that WTP is higher for the OTC scenario and that the level of information plays a significant role in the evaluation of the OTC scenario, with more information being associated with an increase in the WTP. In contrast, the level of information provided has no impact on WTP for prescription medicine. Thus, for the kind of drug considered here (i.e. safe, not requiring medical supervision), a switch to OTC status can be expected to be all the more beneficial, as the patient is provided with more information concerning the capability of the drug. CONCLUSIONS: Our results shed light on one of the most challenging issues that health policy makers are currently faced with, namely the threat of a bird flu pandemic. Drug delivery is a critical component of pandemic influenza preparedness. Furthermore, the congruence of our results with the agency and demand theories provides an important test of the validity of using WTP based on CV methods.

Managing a construction project supply chain effectively and efficiently is extremely difficult due to involvement of numerous sectors that are supported by ineffective communication system. An efficient construction supply chain system ensures the delivery of materials and other services to construction site while minimising costs and rewarding all sectors based on value added to the supply chain. The advancement of information, communication and wireless technologies is driving construction companies to deploy supply chain management strategies to seek better outputs. As part of the emerging wireless technologies, contextaware computing capability represents the next generation of ICT to the construction services. Conceptually, context-awareness could be integrated with Web Services in order to ensure the delivery of pertinent information to construction site and enhance construction supply chain collaboration. An initial study has indicated that this integrated system has the potential of serving and improving the construction services delivery through access to context-specific data, information and services on as-needed basis.

Wireless Sensor Networks (WSNs) have been an exciting topic in recent years. The services offered by a WSN can be classified into three major categories: monitoring, alerting, and information on demand. WSNs have been used for a variety of applications related to the environment (agriculture, water and forest fire detection), the military, buildings, health (elderly people and home monitoring), disaster relief, and area or industrial monitoring. In most WSNs tasks like processing the sensed data, making decisions and generating emergency messages are carried out by a remote server, hence the need for efficient means of transferring data across the network. Because of the range of applications and types of WSN there is a need for different kinds of MAC and routing protocols in order to guarantee delivery of data from the source nodes to the server (or sink). In order to minimize energy consumption and increase performance in areas such as reliability of data delivery, extensive research has been conducted and documented in the literature on designing energy efficient protocols for each individual layer. The most common way to conserve energy in WSNs involves using the MAC layer to put the transceiver and the processor of the sensor node into a low power, sleep state when they are not being used. Hence the energy wasted due to collisions, overhearing and idle listening is reduced. As a result of this strategy for saving energy, the routing protocols need new solutions that take into account the sleep state of some nodes, and which also enable the lifetime of the entire network to be increased by distributing energy usage between nodes over time. This could mean that a combined MAC and routing protocol could significantly improve WSNs because the interaction between the MAC and network layers lets nodes be active at the same time in order to deal with data transmission. In the research presented in this thesis, a cross-layer protocol based on MAC and routing protocols was designed in order to improve the capability of WSNs for a range of different applications. Simulation results, based on a range of realistic scenarios, show that these new protocols improve WSNs by reducing their energy consumption as well as enabling them to support mobile nodes, where necessary. A number of conference and journal papers have been published to disseminate these results for a range of applications.

Disease, injury, and age problems compromise human quality of life and continuously motivate the search for new and more efficacious therapeutic approaches. The field of Tissue Regeneration and Engineering has greatly evolved over the last years, mainly due to the combination of the important advances verified in Biomaterials Science and Engineering with those of Cell and Molecular Biology. In particular, a new and promising area arose – Nanomedicine – that takes advantage of the extremely small size and especial chemical and physical properties of Nanomaterials, offering powerful tools for health improvement. Research on Stem Cells, the self-renewing progenitors of body tissues, is also challenging to the medical and scientific communities, being expectable the appearance of new and exciting stem cell-based therapies in the next years. The control of cell behavior (namely, of cell proliferation and differentiation) is of key importance in devising strategies for Tissue Regeneration and Engineering. Cytokines, growth factors, transcription factors and other signaling molecules, most of them proteins, have been identified and found to regulate and support tissue development and regeneration. However, the application of these molecules in long-term regenerative processes requires their continuous presence at high concentrations as they usually present short half-lives at physiological conditions and may be rapidly cleared from the body. Alternatively, genes encoding such proteins can be introduced inside cells and be expressed using cell’s machinery, allowing an extended and more sustained production of the protein of interest (gene therapy). Genetic engineering of stem cells is particularly attractive because of their self-renewal capability and differentiation potential. For Tissue Regeneration and Engineering purposes, the patient’s own stem cells can be genetically engineered in vitro and, after, introduced in the body (with or without a scaffold) where they will not only modulate the behavior of native cells (stem cell-mediated gene therapy), but also directly participate in tissue repair. Cells can be genetically engineered using viral and non-viral systems. Viruses, as a result of millions of years of evolution, are very effective for the delivery of genes in several types of cells, including cells from primary sources. However, the risks associated with their use (like infection and immunogenic reactions) are driving the search for non-viral systems that will efficiently deliver genetic material into cells. Among them, chemical methods that are promising and being investigated use cationic molecules as carriers for DNA. In this case, gene delivery and gene expression level remain relatively low when primary cells are used. The main goal of this thesis was to develop and assess the in vitro potential of polyamidoamine (PAMAM) dendrimers based carriers to deliver genes to mesenchymal stem cells (MSCs). PAMAM dendrimers are monodispersive, hyperbranched and nanospherical molecules presenting unique characteristics that make them very attractive vehicles for both drug and gene delivery. Although they have been explored for gene delivery in a wide range of cell lines, the interaction and the usefulness of these molecules in the delivery of genes to MSCs remains a field to be explored. Adult MSCs were chosen for the studies due to their potential biomedical applications (they are considered multipotent cells) and because they present several advantages over embryonic stem cells, such as easy accessibility and the inexistence of ethical restrictions to their use. This thesis is divided in 5 interconnected chapters. Chapter I provides an overview of the current literature concerning the various non-viral systems investigated for gene delivery in MSCs. Attention is devoted to physical methods, as well as to chemical methods that make use of polymers (natural and synthetic), liposomes, and inorganic nanoparticles as gene delivery vectors. Also, it summarizes the current applications of genetically engineered mesenchymal stem cells using non-viral systems in regenerative medicine, with special focus on bone tissue regeneration. In Chapter II, the potential of native PAMAM dendrimers with amine termini to transfect MSCs is evaluated. The level of transfection achieved with the dendrimers is, in a first step, studied using a plasmid DNA (pDNA) encoding for the β-galactosidase reporter gene. The effect of dendrimer’s generation, cell passage number, and N:P ratio (where N= number of primary amines in the dendrimer; P= number of phosphate groups in the pDNA backbone) on the level of transfection is evaluated, being the values always very low. In a second step, a pDNA encoding for bone morphogenetic protein-2, a protein that is known for its role in MSCs proliferation and differentiation, is used. The BMP-2 content produced by transfected cells is evaluated by an ELISA assay and its effect on the osteogenic markers is analyzed through several classical assays including alkaline phosphatase activity (an early marker of osteogenesis), osteocalcin production, calcium deposition and mineralized nodules formation (late osteogenesis markers). Results show that a low transfection level is enough to induce in vitro osteogenic differentiation in MSCs. Next, from Chapter III to Chapter V, studies are shown where several strategies are adopted to change the interaction of PAMAM dendrimers with MSCs cell membrane and, as a consequence, to enhance the levels of gene delivery. In Chapter III, generations 5 and 6 of PAMAM dendrimers are surface functionalized with arginine-glycine-aspartic acid (RGD) containing peptides – experiments with dendrimers conjugated to 4, 8 and 16 RGD units were performed. The underlying concept is that by including the RGD integrin-binding motif in the design of the vectors and by forming RGD clusters, the level of transfection will increase as MSCs highly express integrins at their surface. Results show that cellular uptake of functionalized dendrimers and gene expression is enhanced in comparison with the native dendrimers. Furthermore, gene expression is dependent on both the electrostatic interaction established between the dendrimer moiety and the cell surface and the nanocluster RGD density. In Chapter IV, a new family of gene delivery vectors is synthesized consisting of a PAMAM dendrimer (generation 5) core randomly linked at the periphery to alkyl hydrophobic chains that vary in length and number. Herein, the idea is to take advantage of both the cationic nature of the dendrimer and the capacity of lipids to interact with biological membranes. These new vectors show a remarkable capacity for internalizing pDNA, being this effect positively correlated with the –CH2– content present in the hydrophobic corona. Gene expression is also greatly enhanced using the new vectors but, in this case, the higher efficiency is shown by the vectors containing the smallest hydrophobic chains. Finally, chapter V reports the synthesis, characterization and evaluation of novel gene delivery vectors based on PAMAM dendrimers (generation 5) conjugated to peptides with high affinity for MSCs membrane binding - for comparison, experiments are also done with a peptide with low affinity binding properties. These systems present low cytotoxicity and transfection efficiencies superior to those of native dendrimers and partially degraded dendrimers (Superfect®, a commercial product). Furthermore, with this biomimetic approach, the process of gene delivery is shown to be cell surface receptor-mediated. Overall, results show the potential of PAMAM dendrimers to be used, as such or modified, in Tissue Regeneration and Engineering. To our knowledge, this is the first time that PAMAM dendrimers are studied as gene delivery vehicles in this context and using, as target, a cell type with clinical relevancy. It is shown that the cationic nature of PAMAM dendrimers with amine termini can be synergistically combined with surface engineering approaches, which will ultimately result in suitable interactions with the cytoplasmic membrane and enhanced pDNA cellular entry and gene expression. Nevertheless, the quantity of pDNA detected inside cell nucleus is always very small when compared with the bigger amount reaching cytoplasm (accumulation of pDNA is evident in the perinuclear region), suggesting that the main barrier to transfection is the nuclear membrane. Future work can then be envisaged based on the versatility of these systems as biomedical molecular materials, such as the conjugation of PAMAM dendrimers to molecules able to bind nuclear membrane receptors and to promote nuclear translocation.

Therapeutic RNAs, especially siRNAs, are a promising approach for treating diseases like cancer, neurodegenerative disorders and viral infections. Their application, however, is limited due to a lack of safe and efficient delivery systems. Nanosized carriers with the ability to either complex or entrap RNA species are a promising option. rn rn rnSuch a carrier has to meet a lot of requirements, some of which are even partly contradictive. Understanding and controlling the interplay between the different demands would advance a strategic design at an early stage of therapeutic development. rn rn This work is centered around a systematic evaluation of polyplexes, such carriers that are able to complex siRNA due to electrostatic interactions. Six structurally and chemically diverse candidates, poly-L-lysine brushes, block copolymers, cationic peptides, cationic lipids, nanohydrogels, and manganese oxide particles, were tested in a simultaneous fashion. The assays, mostly based on fluorescently labeled siRNA, ranged from the evaluation of polyplex formation and stability to in vitro parameters like cellular uptake and knockdown capability. The analysis from several perspectives offered insight into the interplay between the specifications of one polyplex. Assessing the different carriers under exactly the same experimental conditions also allowed conclusions about favourable traits and starting points for further optimization. This comparative approach also revealed weaknesses of some of the conventional protocols, which were therefore contrasted with alternative methods. In addition, in vitro knockdown assays were optimized and the impact of fluorescently labeled siRNA on knockdown efficiency was assessed. rn rn rn A second class of carriers, which share the ability to entrap siRNA inside their matrix, are briefly addressed. Nanocapsules, dextran particles and liposomes were assessed for basic features like siRNA encapsulation and knockdown capability. rn rn rn rn In an approach towards targeted delivery of RNA, liposomes were endowed with mitochondriotropic tags. Despite successful functionalization, no colocalization between the liposomal cargo and mitochondria was so far observed, which makes further optimization necessary.

Cellular delivery involving the transfer of various drugs and bio-active molecules (peptides, proteins and DNAs, etc.) through the cell membrane into cells has attracted increasing attention because of its importance in medicine and drug delivery. This topic has been extensively reviewed. The direct delivery of drugs and biomolecules, however, is generally inefficient and suffering from problems such as enzymic degradation of DNAs. Therefore, searching for efficient and safe transport vehicles (carriers) to delivery genes or drugs into cells has been challenging yet exciting area of research. In past decades, many carriers have been developed and investigated extensively which can be generally classified into four major groups: viral carriers, organic cationic compounds, recombinant protiens and inorganic nanoparticles. Many inorganic materials, such as calcium phosphate, gold, carbon materials, silicon oxide, iron oxide and layered double hydroxide (LDH), have been studied. Inorganic nanoparticles show low toxicity and promise for controlled delivery properties, thus presenting a new alternative to viral carriers and cationic carriers. Inorganic nanoparticles generally possess versatile properties suitable for cellular delivery, including wide availability, rich functionality, good biocompatibility, potential capability of targeted delivery (e.g. selectively destroying cancer cells but sparing normal tissues) and controlled release of carried drugs. This paper reviews the latest advances in inorganic nanoparticle applications as cellular delivery carriers and highlights some key issues in efficient cellular delivery using inorganic nanoparticles. Critical proper-ties of inorganic nanoparticles, surface functionalisation (modification), uptake of biomolecules, the driving forces for delivery, and release of biomolecules will be reviewed systematically. Selected examples of promising inorganic nanoparticle delivery systems, including gold, fullerences and carbon nanotubes, LDH and various oxide nanoparticles in particular their applications for gene delivery will be discussed. The fundamental understanding of properties of inorganic nanoparticles in relation to cellular delivery efficiency as the most paramount issue will be highlighted. (c) 2005 Elsevier Ltd. The controlled co-delivery of multiple agents to the lung offers potential benefits to patients. This study investigated the preparation and characterisation of highly respirable spray-dried powders displaying the sustained release of two chemically distinct therapeutic agents. Spray-dried powders were produced from 30% (v/v) aqueous ethanol formulations that contained hydrophilic (terbutaline sulphate) and hydrophobic (beclometasone dipropionate) model drugs, chitosan (as a drug release modifier) and leucine (aerosolisation enhancer). The influence of chitosan molecular weight on spray-drying thermal efficiency, aerosol performance and drug release profile was investigated. Resultant powders were physically characterised: with in vitro aerosolisation performance and drug release profile investigated by the Multi-Stage Liquid Impinger and modified USP II dissolution apparatus, respectively. It was found that increased chitosan molecular weight gave increased spray-drying thermal efficiency. The powders generated were of a suitable size for inhalation—with emitted doses over 90% and fine particle fractions up to 72% of the loaded dose. Sustained drug release profiles were observed in dissolution tests for both agents: increased chitosan molecular weight associated with increased duration of drug release. The controlled co-delivery of hydrophilic and hydrophobic entities underlines the capability of spray drying to produce respirable particles with sustained release for delivery to the lung. Cationic liposomes of dimethyldioctadecylammonium bromide (DDA) incorporating the glycolipid trehalose 6,6-dibehenate (TDB) forms a promising liposomal vaccine adjuvant. To be exploited as effective subunit vaccine delivery systems, the physicochemical characteristics of liposomes were studied in detail and correlated with their effectiveness in vivo, in an attempt to elucidate key aspects controlling their efficacy. This research took the previously optimised DDA-TDB system as a foundation for a range of formulations incorporating additional lipids of 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC) or 1,2-distearoyl-sn-glycero-3-phosphocholine (DSPC), by incrementally replacing the cationic content within DDA-TDB or reducing the total DDA-TDB dose upon its substitution, to ascertain the role of DDA and the effect of DDA-TDB concentration in influencing the resultant immunological performance upon delivery of the novel subunit TB vaccine, Ag85B–ESAT-6-Rv2660c (H56 vaccine). With the aim of using the DPPC based systems for pulmonary vaccine delivery and the DSPC systems for application via the intramuscular route, initial work focused on physicochemical characterisation of the systems with incorporation of DPPC or DSPC displaying comparable physical stability, morphological structure and levels of antigen retention to that of DDA-TDB. Thermodynamic analysis was also conducted to detect main phase transition temperatures and subsequent in vitro cell culture studies demonstrated a favourable reduction in cytotoxicity, stimulation of phagocytic activity and macrophage activation in response to the proposed liposomal immunoadjuvants. Immunisation of mice with H56 vaccine via the proposed liposomal adjuvants showed that DDA was an important factor in mediating resultant immune responses, with partial replacement or substitution of DDA-TDB stimulating Th1 type cellular immunity characterised by elevated levels of IgG2b antibodies and IFN-? and IL-2 cytokines, essential for providing protective efficacy against TB. Upon increased DSPC content within the formulation, either by DDA replacement or reduction of DDA and TDB, responses were skewed towards Th2 type immunity with reduced IgG2b antibody levels and elevated IL-5 and IL-10 cytokine production, as resultant immunological responses were independent of liposomal zeta potential. The role of the cationic DDA lipid and the effect of DDA-TDB concentration were appreciated as the proposed liposomal formulations elicited antigen specific antibody and cellular immune responses, demonstrating the potential of cationic liposomes to be utilised as adjuvants for subunit vaccine delivery. Furthermore, the promising capability of the novel H56 vaccine candidate in eliciting protection against TB was apparent in a mouse model.

Current treatment strategies for the treatment of brain tumor have been hindered primarily by the presence of highly lipophilic insurmountable blood-brain barrier (BBB). The purpose of current research was to investigate the efficiency of engineered biocompatible polymeric nanoparticles (NPs) as drug delivery vehicle to bypass the BBB and enhance biopharmaceutical attributes of anti-metabolite methotrexate (MTX) encapsulated NPs. The NPs were prepared by solvent diffusion method using cationic bovine serum albumin (CBA), and characterized for physicochemical parameters such as particle size, polydispersity index, and zeta-potential; while the surface modification was confirmed by FTIR, and NMR spectroscopy. Developed NPs exhibited zestful relocation of FITC tagged NPs across BBB in albino rats. Further, hemolytic studies confirmed them to be non-toxic and biocompatible as compared to free MTX. In vitro cytotoxicity assay of our engineered NPs on HNGC1 tumor cells proved superior uptake in tumor cells; and elicited potent cytotoxic effect as compared to plain NPs and free MTX solution. The outcomes of the study evidently indicate the prospective of CBA conjugated poly (D,L-lactide-co-glycolide) (PLGA) NPs loaded with MTX in brain cancer bomber with amplified capability to circumvent BBB.
