第一部分:内蒙古锡林河流域草原植物种群和群落热值的时空变异研究 热值的研究是评价生态系统能量固定、传输和转化的基础,也是评价植物光合作用效率和植物营养值的有用参数。同种植物热值会随着植物部位、光照、养分条件、季节、土壤类型和气候条件的不同而发生变化。不同的种类和类群之间热值也存在差异。本项研究以内蒙古锡林河流域中段草原植物群落为对象,研究了植物种群和群落热值的时空变异规律。 对内蒙古羊草草原群落不同植物种群热值的时问动态研究结果表明,42种植物地上部分的热值在13.16土1.14 kJ.g-l和18.14土0.53 kJ.g-1之间变动,所有物种的平均热值为16.90土0,84 kJ.g-1,种间变异系数4.9%。小叶锦鸡儿(Caraganamicrophylla)具有最高的热值。禾草的平均热值高于杂草。根据生活型和生长型,草本物种被进一步分组,热值从高到低的排列顺序为:高禾草>豆科植物>矮禾草>其余杂草>半灌木>一二年生植物。 主要植物种群地下部分热值的分布范围为15.05-16.41 kJ.g-1。其中根茎型草地下部分热值较高。不同种类植物地下部分热值差异并不与地上部分一致。根茎型禾草地上、地下部分热值差异较小,而须根型植物差异较大。不同种群的植物地上部分热值随植物物候期的不同而波动,其变化规律是与植物种群本身的生物学特性相联系的。不同植物种群热值的年际波动规律有所不同,羊草(Leymuschinensis)、大针茅(Stipa grandis)和洽草(Koeloria cristata)的年际热值波动相关显著,但与生长季降水量和生长季累积日照时数之间无明显相关性。在某种程度上,植物热值的种内变化反映了植物生长状况的差异。 42种植物的热值和它们在群落中的相对生物量存在显著正相关关系。表现为优势种(17.74 kJ.g-1)>伴生种(17.24 kJ.g-l)>偶见种(16.65 kJ.g-1)。高热值的植物更具竞争力,在群落中通常占据优势地位,而低热值的植物竞争力通常较弱,构成草原群落的伴生种或偶见种。 以内蒙古锡林河流域3个草原群落类型(羊草典型草原,大针茅典型草原,羊草草甸草原)的放牧退化梯度系列(包括未退化,轻度退化,中度退化和重度退化4个强度)为研究对象,对主要植物种群和群落热值随草原类型和退化梯度的空间变异规律及热值与其他群落和土壤性质的相关性进行了研究。 结果表明,研究区出现的60个植物种平均热值为17.25土0.92 kJ.g-1,变异系数5.4%.热值大于18.00 kJ.g-1的高能植物包括3种优势高禾草(羊草、大针茅和羽茅(A. sibiricum))和一些有毒植物,热值小于17.00 kJ.g-1的低能值植物包括多数一年生杂草;热值在17.00-18.00 kJ.g-1之间的中能值植物包括大多数多年生杂草和矮禾草。 按照生活型分类,灌木的热值最高,多年生禾草显著高于一二年生植物,半灌木和多年生杂草介于二者之间。按照水分生态类型分类,旱生植物、中旱生、旱中生和中生植物之间在热值上没有明显差异。不同科之间热值存在显著差异,禾本科、豆科、菊科植物热值较高,藜科植物平均热值最低。 二因素方差分析结果表明,主要优势物种热值在不同草原类型之间存在显著差异,表现为羊草草甸草原>羊草典型草原>大针茅典型草原。对于大多数优势禾草,热值没有随退化梯度发生明显变化,洽草(K. cristata)、冰草(A.ctistatum)和所有优势杂草随退化程度的增强热值趋于下降。对于大多数优势物种,热值随不同草原类型的空间变异大于放牧退化所导致的空间变异。 不同草原类型的群落热值为羊草草甸草原>羊草典型草原>大针茅典型草原,群落平均热值表现出随退化强度的增加而下降的趋势,这主要归因于沿退化梯度不同物种构成比例的变化,即随退化程度的加剧,高能值植物在群落中的比例下降。其次是特定物种热值随退化梯度的变化。在同一草原区,放牧对群落热值的影响大于立地条件之间的差异。 群落和主要物种热值均表现出与某些群落特征和土壤性质的相关性。 关键词:内蒙古,锡林河流域,羊草草原;物种和群落热值,时空变异,退化梯度,草原类型,土壤性质 第二部分内蒙古羊草草原17年刈割演替过程中功能群组成动态及其对群落净初级生产力稳定性的影响 基于17年的野外实验数据,研究了内蒙古羊草草原群落刈割演替过程中的功能群组成动态,探索功能群组成变化与群落净初级生产力(ANPP)之间的关系,分析结构参数怎样影响功能参数。结果显示:在17年的割草演替过程中,群落的结构与功能均发生了变化。随着羊草群落刈割演替的进行,群落的功能群组成发生了显著变化,根茎禾草在群落中的优势地位相继被一二年生植物,高丛生禾草,矮丛生禾草所取代。到17年末,群落变成根茎禾草,矮丛生禾草,高丛生禾草共同建群的群落。在对照群落中ANPP与年降水量显著相关,但在刈割群落中二者则不相关。年降水量解释对照群落ANPP变异的62%,而连年的刈割干扰则是刈割群落中ANPP动态的主要驱动因子。群落净初级生产则显出对刈割干扰的抵抗能力,在刈割干扰的前几年,依靠群落内功能群组成的不断调节,保持相对稳定的水平,当刈割进行5年之后,群落结构的变化积累到一定程度,净初级生产迅速下降到一个较低的水平,此后依靠群落结构的不断调节来维持这一功能水平。因此,群落结构是以渐变的方式改变的,而群落功能的下降则是以跃变的形式完成的。群落依赖于结构的不断调整来保持功能的相对稳定,但结构变化到一定程度也会导致功能的衰退。 关键词:内蒙古,羊草草原;刈割演替;功能群组成;净初级生产;群落;稳定性

La fonte et l’effondrement du pergélisol riche en glace dans la région subarctique du Québec ont donné lieu à la formation de petits lacs (mares de thermokarst) qui émettent des gaz à effet de serre dans l’atmosphère tels que du dioxyde de carbone et du méthane. Pourtant, la composition de la communauté microbienne qui est à la base des processus biogéochimiques dans les mares de fonte a été très peu étudiée, particulièrement en ce qui concerne la diversité et l’activité des micro-organismes impliqués dans le cycle du méthane. L’objectif de cette thèse est donc d’étudier la diversité phylogénétique et fonctionnelle des micro-organismes dans les mares de fonte subarctiques en lien avec les caractéristiques de l’environnement et les émissions de méthane. Pour ce faire, une dizaine de mares ont été échantillonnées dans quatre vallées situées à travers un gradient de fonte du pergélisol, et disposant de différentes propriétés physico-chimiques. Selon les vallées, les mares peuvent être issues de la fonte de palses (buttes de tourbe, à dominance organique) ou de lithalses (buttes de sol à dominance minérale) ce qui influence la nature du carbone organique disponible pour la reminéralisation microbienne. Durant l’été, les mares étaient fortement stratifiées; il y avait un fort gradient physico-chimique au sein de la colonne d’eau, avec une couche d’eau supérieure oxique et une couche d’eau profonde pauvre en oxygène ou anoxique. Pour identifier les facteurs qui influencent les communautés microbiennes, des techniques de séquençage à haut débit ont été utilisées ciblant les transcrits des gènes de l’ARNr 16S et des gènes impliqués dans le cycle du méthane : mcrA pour la méthanogenèse et pmoA pour la méthanotrophie. Pour évaluer l’activité des micro-organismes, la concentration des transcrits des gènes fonctionnels a aussi été mesurée avec des PCR quantitatives (qPCR). Les résultats montrent une forte dominance de micro-organismes impliqués dans le cycle du méthane, c’est-à-dire des archées méthanogènes et des bactéries méthanotrophes. L’analyse du gène pmoA indique que les bactéries méthanotrophes n’étaient pas seulement actives à la surface, mais aussi dans le fond de la mare où les concentrations en oxygène étaient minimales; ce qui est inattendu compte tenu de leur besoin en oxygène pour consommer le méthane. En général, la composition des communautés microbiennes était principalement influencée par l’origine de la mare (palse ou lithalse), et moins par le gradient de dégradation du pergélisol. Des variables environnementales clefs comme le pH, le phosphore et le carbone organique dissous, contribuent à la distinction des communautés microbiennes entre les mares issues de palses ou de lithalses. Avec l’intensification des effets du réchauffement climatique, ces communautés microbiennes vont faire face à des changements de conditions qui risquent de modifier leur composition taxonomique, et leurs réponses aux changements seront probablement différentes selon le type de mares. De plus, dans le futur les conditions d’oxygénation au sein des mares seront soumises à des modifications majeures associées avec un changement dans la durée des périodes de fonte de glace et de stratification. Ce type de changement aura un impact sur l’équilibre entre la méthanogenèse et la méthanotrophie, et affectera ainsi les taux d’émissions de méthane. Cependant, les résultats obtenus dans cette thèse indiquent que les archées méthanogènes et les bactéries méthanotrophes peuvent développer des stratégies pour survivre et rester actives au-delà des limites de leurs conditions d’oxygène habituelles.

Gradient-based approaches to direct policy search in reinforcement learning have received much recent attention as a means to solve problems of partial observability and to avoid some of the problems associated with policy degradation in value-function methods. In this paper we introduce GPOMDP, a simulation-based algorithm for generating a biased estimate of the gradient of the average reward in Partially Observable Markov Decision Processes (POMDPs) controlled by parameterized stochastic policies. A similar algorithm was proposed by Kimura, Yamamura, and Kobayashi (1995). The algorithm's chief advantages are that it requires storage of only twice the number of policy parameters, uses one free parameter β ∈ [0,1) (which has a natural interpretation in terms of bias-variance trade-off), and requires no knowledge of the underlying state. We prove convergence of GPOMDP, and show how the correct choice of the parameter β is related to the mixing time of the controlled POMDP. We briefly describe extensions of GPOMDP to controlled Markov chains, continuous state, observation and control spaces, multiple-agents, higher-order derivatives, and a version for training stochastic policies with internal states. In a companion paper (Baxter, Bartlett, & Weaver, 2001) we show how the gradient estimates generated by GPOMDP can be used in both a traditional stochastic gradient algorithm and a conjugate-gradient procedure to find local optima of the average reward. ©2001 AI Access Foundation and Morgan Kaufmann Publishers. Purpose: Nicardipine is a member of a family of calcium channel blockers named dihydropiridines that are known to be photolabile and may cause phototoxicity. It is therefore vital to develop analytical method which can study the photodegradation of nicardipine. Method: Forced acid degradation of nicardipine was conducted by heating 12 ml of 1 mg/ml nicardipine with 3 ml of 2.5 M HCl for two hours. A gradient HPLC medthod was developed using Agilent Technologies 1200 series quaternary system. Separation was achieved with a Hichrome (250 x 4.6 mm) 5 μm C18 reversed phase column and mobile phase composition of 70% A(100%v/v water) and 30% B(99%v/v acetonitrile + 1%v/v formic acid) at time zero, composition of A and B was then charged to 60%v/v A;40%v/v B at 10minutes, 50%v/v A; 50%v/v B at 30minutes and 70%v/v A; 30%v/v B at 35minutes. 20μl of 0.8mg/ml of nicardipine degradation was injected at room temperature (25oC). The gradient method was transferred onto a HPLC-ESI-MS system (HP 1050 series - AQUAMAX mass detector) and analysis conducted with an acid degradation concentration of 0.25mg/ml and 20μl injection volume. ESI spectra were acquired in positive ionisation mode with MRM 0-600 m/z. Results: Eleven nicardipine degradation products were detected in the HPLC analysis and the resolution (RS) between the respective degradants where 1.0, 1.2, 6.0, 0.4, 1.7, 3.7, 1.8, 1.0, and 1.7 respectively. Nine degradation products were identified in the ESI spectra with the respective m/z ratio; 171.0, 166.1, 441.2, 423.2, 455.2, 455.2, 331.1, 273.1, and 290.1. The possible molecular formulae for each degradants were ambiguously determined. Conclusion: A sensitive and specific method was developed for the analysis of nicardipine degradants. Method enables detection and quantification of nicardipine degradation products that can be used for the study of the kinetics of nicardipine degradation processes.

Samenausbreitung und Regeneration von Bäumen sind wichtig für den langfristigen Bestand von Baum- und Frugivorengemeinschaften in tropischen Regenwäldern. Zunehmende Rohdung und Degradation gefährden den Ablauf dieser mutualistischen Prozesse in diesem Ökosystem. Um den Einfluss von kleinräumiger menschlicher Störung auf die Frugivorengemeinschaft und die zentralen Ökosystemprozesse Samenausbreitung und Regeneration zu erforschen, habe ich 1) die Frugivorengemeinschaft und die Samenausbreitungsrate von Celtis durandii (Ulmaceae) und 2) den Zusammenhang zwischen Baumarten mit fleischigen Früchten, Frugivoren und der Etablierung von Keimlingen dieser Baumarten in unterschiedlich stark gestörten Flächen dreier ostafrikanischer tropischer Regenwälder untersucht. Insgesamt konnte ich 40 frugivore Vogel- und Primatenarten in den drei untersuchten Waldgebieten nachweisen. Auf gering gestörten Flächen wurden mehr Frugivore als auf stark gestörten Flächen aufgenommen. Auch die Beobachtungen an C. durandii ergaben mehr frugivore Besucher in Bäumen auf gering gestörten als auf stark gestörten Flächen. Dies führte zu einer marginal signifikant höheren Samenausbreitungsrate auf den gering gestörten Flächen. Diese Ergebnisse waren auf regionaler Ebene in allen drei untersuchten Wäldern konsistent. Dies zeigt, dass kleinräumige Störung einen umfassenderen negativen Einfluss auf Frugivore und ihre Funktion als Samenausbreiter hat als zuvor angenommen. Bei der Vegetationserfassung nahm ich 131 verschiedene Baumarten mit fleischigen Früchten in den drei Regenwäldern auf. Kleinräumige menschliche Störung erhöhte den Artenreichtum dieser Baumarten marginal signifikant, hatte jedoch keinen direkten Einfluss auf die Frugivorendichte und den Artenreichtum von Keimlingen dieser Baumarten. Der Artenreichtum von Baumarten mit fleischigen Früchten zeigte einen marginal signifikant positiven Einfluss auf die Frugivorendichte, allerdings nicht auf die Keimlinge. Allerdings führte die Dichte der Frugivoren zu signifikant erhöhtem Artenreichtum der Keimlinge. Folglich scheint kleinräumige Störung die Keimlingsetablierung indirekt durch erhöhten Baumartenreichtum und erhöhte Frugivorendichte zu beeinflussen. Die Frugivorendichte hatte einen größeren Einfluss auf die Waldregeneration als kleinräumige Störung und Baumartenreichtum. Demnach scheint kleinräumige menschliche Störung sowohl positive als auch negative Effekte auf Samenausbreitung und Regeneration zu haben. Somit sind weitere Studien notwendig, die den Einfluss von kleinräumiger menschlicher Störung auf Mutualismen tropischer Regenwälder aufklären.

Organic soils in peatlands store a great proportion of the global soil carbon pool and can lose carbon via the atmosphere due to degradation. In Germany, most of the greenhouse gas (GHG) emissions from organic soils are attributed to sites managed as grassland. Here, we investigated a land use gradient from near-natural wetland (NW) to an extensively managed (GE) to an intensively managed grassland site (GI), all formed in the same bog complex in northern Germany. Vertical depth profiles of δ13C, δ15N, ash content, C / N ratio and bulk density as well as radiocarbon ages were studied to identify peat degradation and to calculate carbon loss. At all sites, including the near-natural site, δ13C depth profiles indicate aerobic decomposition in the upper horizons. Depth profiles of δ15N differed significantly between sites with increasing δ15N values in the top soil layers paralleling an increase in land use intensity owing to differences in peat decomposition and fertilizer application. At both grassland sites, the ash content peaked within the first centimetres. In the near-natural site, ash contents were highest in 10–60 cm depth. The ash profiles, not only at the managed grassland sites, but also at the near-natural site indicate that all sites were influenced by anthropogenic activities either currently or in the past, most likely due to drainage. Based on the enrichment of ash content and changes in bulk density, we calculated the total carbon loss from the sites since the peatland was influenced by anthropogenic activities. Carbon loss at the sites increased in the following order: NW < GE < GI. Radiocarbon ages of peat in the topsoil of GE and GI were hundreds of years, indicating the loss of younger peat material. In contrast, peat in the first centimetres of the NW was only a few decades old, indicating recent peat growth. It is likely that the NW site accumulates carbon today but was perturbed by anthropogenic activities in the past. Together, all biogeochemical parameters indicate a degradation of peat due to (i) conversion to grassland with historical drainage and (ii) land use intensification.

NADPH:protochlorophyllide oxidoreductase is a key enzyme for the light-induced greening of etiolated angiosperm plants. In barley, two POR proteins exist termed PORA and PORB that have previously been proposed to structurally and functionally cooperate in terms of a higher molecular mass light-harvesting complex named LHPP, in the prolamellar body of etioplasts [Nature 397 (1999) 80]. In this study we examined the expression pattern of LHPP during seedling etiolation and de-etiolation under different experimental conditions. Our results show that LHPP is developmentally expressed across the barley leaf gradient. We further provide evidence that LHPP operates both in plants that etiolate completely before being exposed to white light and in plants that etiolate only partially and begin light-harvesting as soon as traces of light become available in the uppermost parts of the soil. As a result of light absorption, in either case LHPP converts Pchlide a to chlorophyllide (Chlide) a and in turn disintegrates. The released Chlide a, as well as Chlide b produced upon LHPP’s light-dependent dissociation, which leads to the activation of the PORA as a Pchlide b-reducing enzyme, then bind to homologs of water-soluble chlorophyll proteins of Brassicaceae. We propose that these proteins transfer Chlide a and Chlide b to the thylakoids, where their esterification with phytol and assembly into the photosynthetic membrane complexes ultimately takes place. Presumably due to the tight coupling of LHPP synthesis and degradation, as well as WSCP formation and photosynthetic membrane assembly, efficient photo-protection is conferred onto the plant.

Brassicales species rich in glucosinolates are used for biofumigation, a process based on releasing enzymatically toxic isothiocyanates into the soil. These hydrolysis products are volatile and often reactive compounds. Moreover, glucosinolates can be degraded also without the presence of the hydrolytic enzyme myrosinase which might contribute to bioactive effects. Thus, in the present study the stability of Brassicaceae plant-derived and pure glucosinolates hydrolysis products was studied using three different soils ( model biofumigation). In addition, the degradation of pure 2-propenyl glucosinolate was investigated with special regard to the formation of volatile breakdown products. Finally, the influence of pure glucosinolate degradation on the bacterial community composition was evaluated using denaturing gradient gel electrophoresis of 16S rRNA gene amplified from total community DNA. The model biofumigation study revealed that the structure of the hydrolysis products had a significant impact on their stability in the soil but not the soil type. Following the degradation of pure 2-propenyl glucosinolate in the soils, the nitrile as well as the isothiocyanate can be the main degradation products, depending on the soil type. Furthermore, the degradation was shown to be both chemically as well as biologically mediated as autoclaving reduced degradation. The nitrile was the major product of the chemical degradation and its formation increased with iron content of the soil. Additionally, the bacterial community composition was significantly affected by adding pure 2-propenyl glucosinolate, the effect being more pronounced than in treatments with myrosinase added to the glucosinolate. Therefore, glucosinolates can have a greater effect on soil bacterial community composition than their hydrolysis products.
