An improved method of PCR in which the small segment of conchocelis is amplified directly without DNA extraction was used to amplify a RUBISCO intergenic spacer DNA fragment from nine species of red algal genus Porphyra (Bangiales, Rhodophyta), including Porphyra yezoensis (Jiangsu, China), P. haitanensis (Fujian, China), P. oligospermatangia (Qingdao, China), P. katadai (Qingdao, China), P. tenera (Qingdao, China), P. suborboculata (Fujian, China), P. pseudolinearis (Kogendo, Korea), P. linearis (Devon, England), and P. fallax (Seattle, USA). Standard PCR and the method developed here were both conducted using primers specific for the RUBISCO spacer region, after which the two PCR products were sequenced. The sequencing data of the amplicons obtained using both methods were identical, suggesting that the improved PCR method was functional. These findings indicate that the method developed here may be useful for the rapid identification of species of Porphyra in a germplasm bank. In addition, a phylogenetic tree was constructed using the RUBISCO spacer and partial rbcS sequence, and the results were in concordant with possible alternative phylogenies based on traditional morphological taxonomic characteristics, indicating that the RUBISCO spacer is a useful region for phylogenetic studies.

Process conditions for cell cultures derived from conchocelis of female red macroalga Porphyra haitanensis were optimized in an illuminated 0.3-l bubble-column photobioreactor, using CO2 in air as the sole carbon source during a 20-day cultivation period. It reached the highest growth rate when the initial cell density was 700 mg l(-1)(dry weight), the optional aeration rate was 1.2 v/v/min, inorganic nitrate concentration was 15 mM and inorganic phosphate concentration was 0.6 mM. This is the first reported bioreactor cultivation study of cell cultures derived from conchocelis of Porphyra haitanensis.

Penicillium herquei isolate GA4 was isolated from the infected Conchocelis of Porphyra yezoensis. A large-scale fermentation using yeast extract sucrose medium and repeated chromatography afforded a new symmetrical urea derivative, hualyzin (1). The structure was determined by detailed NMR spectroscopic investigations and MS fragmentation analysis.

R-phycoerythrin (R-PE) was purified from leafy gametophyte of Porphyra haitanensis T. J. Chang et B. F. Zheng (Bangiales, Rhodophyta) by a simple, scaleable procedure. Initially, phycobiliproteins were extracted by repeated freeze-thaw cycles, resulting in release from the algal cells by osmotic shock. Next, R-PE was recovered by applying the crude extract with a high concentration of (NH4)(2)SO4 salt directly to the expanded-bed columns loaded with phenyl-sepharose. An expanded-bed volume twice the settled-bed volume was maintained; then low (NH4)(2)SO4 concentration was used to develop the column. After two rounds of hydrophobic interaction chromatography (HIC), R-PE was purified by anion-exchange column. The method was also successful with free-living conchocelis of P. haitanensis. The purified R-PE was identified with electrophoresis, and absorption and fluorescence emission spectroscopy. The results were in agreement with those previously reported. The yield with a spectroscopic purity (OD565/OD280) higher than 3.2 (the ratio of A(565)/A(620) <= 0.02) was 1.4 mg . g(-1) of leafy gametophyte of P. haitanensis. For the free-living conchocelis of P. haitanensis extract, R-PE could be purified successfully with only one round of HIC. The yield with a spectroscopic purity (OD565/OD280) higher than 3.2 (the ratio of A(565)/A(620) <= 0.02) was 5.0 mg . g(-1) of free-living conchocelis of P. haitanensis. The method described here is a scaleable technology that allows a large quantity of R-PE to be recovered from the unclarified P. haitanensis crude extract. It is also a high protein recovery technology, reducing both processing costs and times, which enhances the value of this endemic Porphyra of China.

Porphyra katadae Miura var. hemiphylla Tseng et T. J. Chang, a species distributed around the Liaodong and Shandong Peninsulas of China, produces gametophytes from late winter to early spring. These are monoecious with male and female reproductive tissues in distinct halves or sectors. Vegetative tissues from sectors expected to differentiate into sexual tissue were cultured in the laboratory. Male and female reproductive organs, as well as conchocelis and blades, were differentiated from these tissues. The male and female reproductive tissues were in patches and mixed on the cultured tissue pieces. This was quite different from the wild-type sectored individuals. The F-1 conchospore germlings also produced monospores, carposporangia, spermatangia and conchocelis. These carposporangia and spermatangia were in patches and were mixed on the F-1 fronds. The results imply that P. katadae var. hemiphylla is possibly sex-differentiated rather than sex-determined. This is the first report of such a dimorphic life history in the genus Porphyra.

Scanning electron microscopy of the surfaces of the seaweeds Laminaria japonica, haploid Porphyra yezoensis, Ulva pertusa and the diploid conchocelis of P. yezoensis and P. haitanensis revealed Vibrio and Micrococcus to be abundant on the surfaces of U. pertusa and P. yezoensis. Vibrio, Flavobacterium, Pseudomonas, Staphylococcus, Bacillus, Corynebacterium and other genera were isolated from the surfaces of L. japonica.

Nets in traditional Porphyra mariculture are seeded with conchospores derived from the conchocelis phase, and spend a nursery period in culture tanks or calm coastal waters until they reach several centimeters in length. Some species of Porphyra can regenerate the foliose phase directly through asexual reproduction, which suggests that the time, infrastructure, and costs associated with conchocelis culture might be avoided by seeding nets with asexual spores. Here, we present work from a short-term mariculture study using nets seeded with asexual spores (neutral spores) of a native Maine species of Porphyra. Porphyra umbilicalis (L.) Kutzing was selected for this proof of concept research because of its reproductive biology, abundance across seasons in Maine, and evidence of its promise as a mariculture crop. We studied the maturation, release, and germination of the neutral spores to develop an appropriate seeding protocol for nets, followed by development of a nursery raceway to provide an easily manipulated environment for the seeded nets. Neutral spores were produced throughout the year on the central Maine coast,however, there was a temporal variability in the number and survival of released neutral spores, depending upon thallus position in the intertidal zone. Small thalli were strictly vegetative, but most thalli reproduced by neutral spores- sexual reproduction was absent. Neutral spores germinated quickly at 10 and 15 'C, but germination was delayed at 5 degrees C. Unlike some algal zygotes and spores, neutral spores of R umbilicalis required light to germinate; however, irradiances of 25 and 100 mu mol photons M-2 S-1 were equally sufficient for germination. Rafts of seeded nets were deployed in Cobscook Bay, Maine, at two distances from salmon aquaculture pens and at a control site on a nearby, fallow aquaculture site (no salmon). There was no difference in nitrogen content of harvested thalli; however, both the density and the surface area of harvested thalli were different among the sites. Cladosporium sp. isolate N5 was isolated as a dominant fungus from the healthy conchocelis of Porphyra yezoensis. In the re-infection test, it did not cause any pathogenic symptoms in the alga. Twenty-one cultural conditions were chosen to test its antimicrobial activity in order to obtain the best condition for large-scale fermentation. Phenylacetic acid, p-hydroxyphenylethyl alcohol, and L-beta-phenyllactic acid were isolated from the crude extract as strong antimicrobial compounds and they are the first reported secondary metabolites for the genus Cladosporium. In addition, the Cladosporium sp. produced the reported Porphyra yezoensis growth regulators phenylacetic acid and p-hydroxyphenylacetic acid. No cytotoxicity was found in the brine shrimp lethality test, which indicated that the environmental-friendly Cladosporium sp. could be used as a potential biocontrol agent to protect the alga from pathogens.

The free living conchocelis of Porphyra yezoensis Ueda was treated with N-methyl-N-nitro-N-nitrosoguanidine to induce pigmentation mutants. The artificial green pigmentation mutant of P. yezoensis conchocelis, which was composed entirely of green cells, was isolated through visualization with the unaided eye. The acquired green conchocelis was further developed into a green gametophytic blade. This mutant was relatively stable in color in both gametophytic blade and conchocelis phases. The gametophytic blade mutant was successively cultivated for commerce at some Porphyra farms in Rudong, China, and few wild type or sectorially variegated gametophytic blade occurred, indicating that the green mutant has commercial value. The green mutant was characterized as having lower phycoerythrin and higher phycocyanin content, and SDS-PAGE suggested that phycoerythrin was missing the gamma-subunit in comparison to the wild type. The wild type and the green mutant showed a clear difference in 02 evolution rates in white, green, yellow, and red light, which might be due to the qualitative and quantitative changes of phycoerythrin, and the quantitative difference of phycocyanin. Bangia fuscopurpurea (Rhodophyta) was cultivated in Putian (Fujian province, China). The characteristics of the life history concerned with cultivation were investigated and the cultivation procedure was presented. The gametophytic phase (thallus) and the sporophytic phase (conchocelis) occurred alternately in the life history of B. fuscopurpurea. Young thalli produced archeospores, and the number depended on the environmental factors. Temperature affected the number of archeospore release and percent of germination, and photo flux density (PFD) mainly affected the time of spore release and germination. Thalli matured from December to February and developed into the conchocelis phase through sexual reproduction. The conchocelis grown in shells had three developmental stages: vegetative conchocelis, conchosporangiall formation and conchospore formation. Pit-connections were present in the first 2 stages but absent after conchospore formation. Vegetative conchocelis and conchosporangial. branches can transform into each other. However, conchospores only developed into the gametophytic phase. Cultivation of B. fuscopurpurea was based on characterization of the life history, consisting of 3 steps: zygotospores collection, indoor cultivation of conchocelis and outdoor cultivation of thalli. Young thalli that developed from conchospores produced numerous archeospores before December. Over 90% of the crop was from the development of archeospores. The results indicated that conchosporelings were a good source of archeospores, and the development of the large quantity of archeospores acted as a more prevailing means to increase the population size. The color mutations in Bangiaceae were investigated by treating the blades, conchocelis and conchospores phase of Bangia sp., Porphyra yezoensis, and P. haitanensis sampled in China with mutagen N-methyl-N'-nitro-N-nitrosoguanidine (MNNG). A high percentage of mutation in different expression characteristics in all three phases were shown within optimum mutagen concentrations. Among mutagenized blades, mutations occurred on single cells, which is a direct outcome of mutation of haploid cells. The mutation of mutagenized conchocelis resulted in a two-step process: low-level expression in conchocelis phase, and high-level expression in progeny, explaining that mutation took place in diploid cells. The mutations of conchospores were expressed immediately at germination of spores, indicating a change in ploidy. This paper reports the process of meiosis and its effect on frond development, and the relation between color mutations and morphological characteristics expressed by mutations in Bangiaceae.

本研究分为三个部分:1.以坛紫菜(Porphyra haitanesis Chang et Zheng)的叶状体和丝状体为研究对象,比较坛紫菜叶状体和丝状体的光合色素、色素蛋白的组成,并提取纯化藻红蛋白、藻蓝蛋白、藻胆体及类囊体膜和光系统。研究结果表明坛紫菜叶状体和丝状体色素及色素蛋白的含量不同,藻红蛋白是主要的色素蛋白,坛紫菜叶状体和丝状体的藻红蛋白的含量分别为2.9mg藻红蛋白/g鲜重、4.2mg藻红蛋白/g鲜重,这表明坛紫菜叶状体和丝状体藻红蛋白含量丰富,是提取藻红蛋白很好的材料。藻胆体的性质差异不大,但类囊体膜差异显著,从坛紫菜叶状体中分离到了两种不同的类囊体膜带,光系统Ⅰ(PSⅠ)和PSⅡ分别结合在两条类囊体膜带上,但从坛紫菜丝状体中也分离到两条类囊体膜带,它们的光谱性质和蛋白组成相似,仅放氧速率和DCIP活性有差异,从坛紫菜丝状体中我们仅分离到PSⅡ。坛紫菜叶状体PSⅡ有5种外在蛋白(33、20、Cytc 550、15、12kDa蛋白),而坛紫菜丝状体外在蛋白仅有4条,缺少12kDa蛋白。2. 以在中国江苏部分地区进行了大规模的商业化栽培的突变体条斑紫菜(Porphyra yezoensis Ueda)和野生型条斑紫菜为研究对象,比较其色素及色素蛋白组成、对不能光质的利用率及藻胆体的组成。条斑紫菜和突变型条斑紫菜对不同的光质利用效果有差异,在白光的照射下,野生型紫菜的放氧速率最大,而突变型紫菜在黄光照射下的放氧速率最大。条斑紫菜野生型与突变型色素含量上有明显的差异,突变型紫菜的藻红蛋白含量明显减少而藻蓝蛋白的含量增加。通过杂交的方法证实诱变所获得条斑紫菜突变体为细胞质突变,但是突变型紫菜却发生了由细胞核编码的γ亚基的缺失,这表明突变型紫菜藻红蛋白含量和性质发生了明显的变化。3. 为了找出淡水红藻-深紫美芒藻(Compsopogon coeruleus (Balbis) Montagne)分布狭窄及生物产量低的原因,本文对深紫美芒藻在不同的盐离子浓度下的放氧速率及藻胆体色素组成和结构上进行研究。结果显示:微量的NaCl(0.1mM)促进深紫美芒藻放氧,而深紫美芒藻在较高的NaCl(1、10mM), NaH2PO4 (0.1、1、10mM)和 NH4NO3(0.1、1、10mM)溶液中却没有检测到氧气的产生。这与深紫美芒藻生长的环境一致即深紫美芒藻生活在低盐浓度、低营养的泉水中。深紫美芒藻的藻胆体是由藻红蛋白、藻蓝蛋白及别藻蓝蛋白组成,上面结合α、β和γ亚基,含有藻红胆素、藻篮胆素,但缺乏缺少藻尿胆素。

红毛菜(Bangia Lyngb.)属于红藻门,与紫菜属同属红毛菜科,其味道和营养都优于紫菜。目前红毛菜栽培产业已在我国福建莆田展开,但栽培技术还有待提高。海藻栽培技术的发展和成熟依赖于对其生长发育过程的认识。本研究针对红毛菜发育过程及相关光合生理展开,并初步探讨了一采自山西娘子关泉淡水红毛菜群体(FWB)的系统地位。 色素突变标记的壳孢子萌发特征表明最初两次分裂产生的4细胞决定了完整植株的形态建成。成熟植株,为雌雄异体。雌性生殖器果胞的标志性分化结构为原始受精丝,环境因子是促发原始受精丝发展的外部因素,其膨大程度随受精的延迟而增大。原孢子是主要的无性生殖孢子类型,在不良环境中,藻体也会形成内生孢子或休眠包囊,或者藻体断裂后重新形成完整的植株。 红毛菜的生长发育很大程度上受环境因子的控制。高温不利于配子体的发育,15-20 ºC比较适宜。红毛菜无性繁殖的最适温度-光照组合为20 ºC-8 h,有性繁殖为15 ºC-12h。 不同发育阶段,PSII实际光合效率(Y(II))与细胞的健康状况以及光合器官完整性及其在细胞内的分布有关,而与细胞的类型关系不大。健康的假根细胞、已分化未成熟的精子以及果孢子细胞均具有很高的Y(II)。色素体由中间位变为围周位,中央大液泡(营养藻丝)和大小纤维囊泡(成熟孢子与精子)的产生,使得细胞Y(II)降低。刚放散的壳孢子Y(II)很低,说明在壳孢子由贝壳基质释放到自由水体过程,光合作用受到一定程度抑制;而2h后,Y(II)开始恢复,rbcL的转录水平非常高,为孢子的萌发储备物质和能量需求。 在失水和低盐胁迫下,藻体均维持较高的Y(II)。干出处理至藻体重量不再变化,复水后Y(II)可回复初始水平。海生红毛菜在100%淡水培养基中(约20ºC)培养7天后,部分雄性藻体依然活着。从而体现了红毛菜位居高潮带的生理优势。 FWB终生行无性繁殖,藻体形态与发生以及染色体数目(4条)与海生群体没有区别。而rbcL-rbcS Spacer序列显示,红毛菜海生群体(无性和有性)具有完全相同的序列,而FWB与它们有5bp差异,但是与欧洲、北美地区的淡水群体仅1bp不同,初步说明所有淡水红毛菜群体具有共同的原始起源。
