Aim: The aim of this study was to assess the effect of different silver nanoparticles (SN) concentrations on the matrix composition and structure of Candida albicans and Candida glabrata biofilms. Methods and Results: Candida biofilms were developed in 6-well microtiter plates during 48 h. After, these biofilms were exposed to 13·5 or 54 μg SN ml-1 for 24 h. Then, extracellular matrices were extracted from biofilms and analysed chemically in terms of proteins, carbohydrates and DNA. To investigate the biofilm structure, scanning electron microscopy (SEM) and epifluorescence microscopy were used. SN interfered with the matrix composition of Candida biofilms tested in terms of protein, carbohydrate and DNA, except for the protein content of C. albicans biofilm. By SEM, Candida biofilms treated with SN revealed structural differences, when compared with the control groups. Further, SN showed a trend of agglomeration within the biofilms. Epifluorescence microscopy images suggest that SN induced damage on cell walls of the Candida isolates tested. Conclusions: In general, irrespective of concentration, SN affected the matrix composition and structure of Candida biofilms and these findings may be related to the mechanisms of biocide action of SN. Significance and Impact of the Study: This study reveals new insights about the behaviour of SN when in contact with Candida biofilms. SN may contribute to the development of therapies to prevent or control Candida infections. © 2012 The Society for Applied Microbiology.

This work evaluated the activity concentration of the radioelements K, eU and eTh in samples of granites from Rondônia State, Brazil. The statistical analysis of the data indicated that they fit lognormal distributions. The modal values correspond to about 11% for K, 29 ppm for eU and 85 ppm for eTh. Directsignificant correlations were found among the concentrations of the three radioelements, i.e. r = 0.71 (between K and eU), r = 0.72 (between K and eTh), andr = 0.72 (between eU and eTh), suggesting congruency of their accumulation in minerals occurring in the rocks analyzed. The activity concentration data allowed estimate the absorbed dose rate in air at 1 m above the ground, which also fits a lognormal distribution characterized by a mode of 2.7 mSv/y that is slightly higher than the average worldwide exposure of 2.4 mSv/y. The results obtained also allowed evaluate if the granites analyzed are radiometrically suitable as building and ornamental materials. © 2009 Sociedade Brasileira de Geofísica.

Coffea arabica L. is considered to be sensitive to low temperatures throughout its life cycle. In some Brazilian regions, seedling production occurs under shade conditions and during the winter, with average temperatures of around 10 °C. The formation and functioning of the photosynthetic apparatus are strongly controlled by temperature. This study aimed to assess the changes that occurred in pigment contents, lipid peroxidation and variables of chlorophyll a fluorescence during the greening process of coffee seedlings submitted to chilling. Results indicate that saturation of the photosynthetic activity of coffee seedlings occurred before saturation of the accumulation of chloroplastid pigments. Pigment accumulation during the greening process is far beyond the metabolic needs for the maintenance of photosynthetic activity, more specifically of photosystem II. Coffee seedlings attained a quantum yield equivalent to that of the control with approximately half the chlorophyll a and b contents and around 40% of the carotenoid. Low temperature decreases the metabolism of seedlings, consequently reducing free radical production and lipid peroxidation. The chilling temperature (10 °C) used inhibited the accumulation of chloroplast pigments, in turn altering the capacity of the photosynthetic tissue of etiolated coffee seedlings to capture and transfer photon energy to the photosystem II reaction centre. These alterations were better demonstrated by O-J-I-P chlorophyll a fluorescence transients, rather than F v/F m and F v/F 0 ratios. © 2009 Elsevier B.V. All rights reserved.

To address daily fluctuations in electricity demands, the quantities of water passing through the turbines of hydropower plants can vary significantly (up to fourfold) during a 24-h cycle. This study evaluates the effects of hourly variations in water discharges on the limnological conditions observed in two below-dam river stretches. The study reservoirs, Capivara and Taquaruçu, are the 9th and 10th reservoirs in a cascade of dams in the Paranapanema River in south-east Brazil. The reservoirs exhibit different trophic conditions, water retention times, thermal regimes and spillway positions. Capivara Reservoir is deeper, meso-eutrophic, with a high water retention time and hypolimnetic discharges (32 m) varying between 500 and 1400 m3 s-1. In contrast, Taquaruçu Reservoir is relatively shallow, oligo-mesotrophic, and has a low retention time, with water discharges varying between 500 and 2000 m3 s-1. Its turbine water intake zone also is more superficial (7 m). For two periods of the year, winter and summer, profiles of limnological measurements were developed in the lacustrine (above-dam site) zones of the reservoirs, as well as in the downstream river stretches (below-dam site). In both cases, the sampling was carried out at 4-h intervals over a complete nictemeral cycle. The results demonstrated that the reservoir operating regime (water discharge variations) promoted significant differences in the conditions of the river below the dams, especially for water velocity, turbidity, and nutrient and suspended solids concentrations. The reservoir physical characteristics, including depth, thermal stratification and outlet structure, are also key factors influencing the limnology and water quality at the below-dam sampling sites. In the case of Capivara Reservoir, for example, the low dissolved oxygen concentration (<5.0 mg L-1) in its bottom water layer was transferred to the downstream river stretches during the summer. These study results demonstrated that it is important to continue such investigations as a means of verifying whether or not these high-amplitude/low-frequency variations could negatively affect the downstream river biota. © 2009 The Authors. Metaultramafic bodies tectonically emplaced within the metasedimentary sequence of the Araxá Group are associated with an ophiolitic melange in southeast Goiás. In the region of Crominia - Mairipotaba, they occur as lenticular bodies aligned E-W. Cumulate textures and geochemical data indicate that the parent rocks had harzburgitic to dunitic compositions. Relicts of primary crystals of olivine and orthopyroxene are suggestive of amphibolite facies metamorphic re-equilibration fabrics, even though the paragenesis and mineral associations of these metaultramafic rocks are typical of greenschist facies (T < 550°C and P = 5.5 kbar). The chromitites exhibit massive to breccioid structure and pull-apart texture, with chromite crystals around 0.5 mm in size. Chromite concentrations in the chromitite levels reach 70 to 85% by volume of the rock. The crystals are dispersed in the matrix, which is composed essentially of serpentine, and subordinately of chlorite and talc. The textures and geochemical data (Cr 2O 3 x TiO 2 and Mg x Cr ratios present in the chromitite) are similar to those observed in ophiolitic complexes. Hence, they correspond to allochthonous bodies (Alpine type) associated with an ophiolitic mélange.

Soil flushing is an alternative remediation technology for soils contaminated with heavy metals, which the main contaminant removing process consists in percolating an extraction solution. This work aimed to use the response surface methodology to point out combinations among the parameters of the extraction solution (Na2EDTA concentration, volume e pH) in order to reduce the concentration of copper in a sandy soil to risk levels lower than the intervention levels for exposure scenarios adopted by the Environmental Company of Sao Paulo State. Thus, a series of tests in leaching columns were carried out using a Fluvisol artificially contaminated (1257,3 mg kg-1). The tests were conducted in triplicate and setup a central composite rotatable design with 15 different parameters combinations of the extraction solution and one replicate in the center point. Using 5% significance level, the adjusted model (R2 = 0,98) indicated combinations of Na2EDTA concentration, pH and volume of the extraction solution which allow reduction of copper concentration below levels reported by environmental agency of Sao Paulo State for industrial, residential, agricultural or maximum exposure scenarios.

The Thermal Corridor of Uruguay River is located in the triple border of Argentine, Brazil and Uruguay, and shows an intense economic thermal tourism activity, mainly based on groundwaters from Guarani Aquifer System (GAS). Recent studies have pointed out the occurrence of high concentrations of arsenic (>10 μg/L) in GAS groundwater in this area. The complex geological and hydrogeological framework in the area is associated to the Paraná Basin geological evolution south of the Assuncion-Rio Grande Arch; it encompass paleozoic marine sequences and continental sequences permian/eotriassic to mesozoic in age, which are covered by basaltic lavas of Serra Geral Formation. Iron oxide coatings have been described in sandstones of Buena Vista and Sanga do Cabral formations, which underlie GAS units. Arsenic occurrence is associated to sodium bicarbonate groundwater with pH values over 8.0. Arsenic is released to groundwater by desorption from iron oxides/hydroxides, as result of the higher pH of these waters, indicating that arsenic is released from the units that underlie GAS units. Increase in chromium and uranium concentrations are also related to high pH groundwaters, thus indicating special care on using groundwater from this region.

It is well established that nitrate is a potent inhibitor of nodulation and nitrogen fixation in legumes. The objective of this study was to demonstrate the relative insensitivity of these processes to nitrate with Calopogonium mucunoides, a tropical South American perennial legume, native to the cerrado (savannah) region. It was found that nodule number was reduced by about half in the presence of high levels of nitrate (15 mM) but nodule growth (total nodule mass per plant) and nitrogen fixation (acetylene reduction activity and xylem sap ureide levels) were not affected. Other sources of N (ammonium and urea) were also without effect at these concentrations. At even higher concentrations (30 mM), nitrate did promote significant inhibition (ca. 50%) of acetylene reduction activity, but no significant reduction in xylem sap ureides was found. The extraordinary insensitivity of nodulation and N2 fixation of C. mucunoides to nitrate suggests that this species should be useful in studies aimed at elucidating the mechanisms of nitrate inhibition of these processes. © 2010 Springer Science+Business Media B.V.

Soil and subsoil pollution is not only significant in terms of environmental loss, but also a matter of environmental and public health. Solid, liquid and gaseous residues are the major soil contamination agents. They originate from urban conglomerates and industrial areas in which it is impossible to emphasize the chemical, petrochemical and textile industry; thermoelectric, mining, and ironmaster activities. The contamination process can thus be defined as a compound addition to soil, from what qualitative and or quantitative manners can modify soil's natural characteristics and use, producing baneful and deteriorative effects on human health. Studies have shown that human exposition to high concentration of some heavy metals found on soil can cause serious health problems, such as pulmonary or kidney complications, liver and nervous system harm, allergy, and the chronic exposition that leads to death. The present study searches for the correlation among soil contamination, done through a geochemical baseline survey of an industrial contamination area on the shoreline of Sao Paulo state. The study will be conducted by spatial analysis using Geographical Information Systems for mapping and regression analysis. The used data are 123 soil samples of percentage concentration of heavy metals. They were sampled and spatially distributed by geostatistics methods. To verify if there is a relation between heavy metals soil pollution and morbidity an executed correlation and regression analysis will be done using the pollution registers as the independent variables and morbidity as dependable variables. It is expected, by the end of the study, to identify the areas relation between heavy metals soil pollution and morbidity, moreover to be able to provide assistance in terms of new methodologies that could facilitate soil pollution control programs and public health planning. © 2010 WIT Press.

Fossil fuels such as diesel are being gradually replaced by biodiesel, a renewable energy source, cheaper and less polluting. However, little is known about the toxic effects of this new energy source on aquatic organisms. Thus, we evaluated biochemical biomarkers related to oxidative stress in Nile tilapia (Oreochromis niloticus) after two and seven exposure days to diesel and pure biodiesel (B100) and blends B5 and B20 at concentrations of 0.01 and 0.1mLL -1. The hepatic ethoxyresorufin-O-deethylase activity was highly induced in all groups, except for those animals exposed to B100. There was an increase in lipid peroxidation in liver and gills in the group exposed to the higher concentration of B5. All treatments caused a significant increase in the levels of 1-hydroxypyrene excreted in the bile after 2 and 7d, except for those fish exposed to B100. The hepatic glutathione-S-transferase increased after 7d in animals exposed to the higher concentration of diesel and in the gill of fish exposed to the higher concentration of pure diesel and B5, but decreased for the two tested concentrations of B100. Superoxide dismutase, catalase and glutathione peroxidase also presented significant changes according to the treatments for all groups, including B100. Biodiesel B20 in the conditions tested had fewer adverse effects than diesel and B5 for the Nile tilapia, and can be suggested as a less harmful fuel in substitution to diesel. However, even B100 could activate biochemical responses in fish, at the experimental conditions tested, indicating that this fuel can also represent a risk to the aquatic biota. © 2011 Elsevier Ltd.

The addition of phosphoric acids and their (by)products became a common practice in areas like metal treatment, detergent production, water and effluent treatment, as well by food, beverage and pharmaceutical industries. The phosphoric acids exhibit different radionuclides activity concentration, being important to evaluate the implications for the human and animal health. The 238U concentration in almost all raw acid phosphoric are within the worldwide range and the mean exposure rate for the filtration cake is 10 nGy/h, which is mainly attributed to 40K. The results obtained for total and (bio)available uranium concentration in filtration cake indicate that only 40% is (bio)available for plants. The radionuclides present in phosphoric acid food grade and filtration cake do not raise their concentration in human food chain or soils to harmful levels, consequently, not offering hazard to the ecosystem and animal or human health.

This paper introduces an indirect estimate for the coefficients of distribution, hydrodynamic dispersion and retardation for contaminants commonly encountered in sanitary landfills and their liners, such as Cu2+ and K+; this estimate is based on the relationship between concentration and certain physical characteristics of typical Brazilian soils. The results of previous studies investigating the migration of contaminants were used to develop mathematical expressions from multiple non-linear regressions. Using minimal squares regression, this transport was linked to various combinations of contaminant concentration and both structural and textural characteristics of the porous medium. Various combinations of characteristics and concentrations were investigated, with a mathematical expression obtained for each. The relationship between percentage of clay and the contaminant content proved to be the most closely correlated with actual transport parameters, with coefficients close to one.

Improper use of pesticides can lead to significant environmental impacts such as, contamination of environmental compartments, being the aquatic compartments the most vulnerable. In this context, the spatialization of pesticides concentrations estimative in groundwater provides important insights for decision making in managing and monitoring pesticides use. This study aimed to spatialize estimatives of groundwater contamination by Tebuthiuron, from different irrigation depths in the Rio Pardo basin, Pardinho-SP, Brazil. The simulations were performed using the ARAquá computer system, considering 0mm, 200 mm and 400 mm annual irrigation depths. Geostatistical techniques were used to obtain the spatial distribution of the simulated estimative. Tebuthiuron maps estimating concentration in groundwater were obtained by Kriging interpolation method, and indicated the areas with high potential for groundwater contamination. Considering all the simulations, it was concluded that there was no risk of groundwater contamination by Tebuthiuron in the study area.

The efficiency of monitoring fertigation by means of porous capsule extractors is directly related to its correct placement, since the nutrients applied through fertigation have different mobility throughout the soil profile. The objective of this study was to evaluate the distributions of NO3, P, and K in the soil solution, when applied in citrus in sandy soil through drip fertigation systems, aiming to define the proper placement of the solution extractors. In order to measure the concentration of ions in the soil solution, the extractors were installed at four different distances (5, 15, 25, and 35 cm) and depths (15, 30, 60, and 90 cm) with an emitter located under the projection of the tree canopy. The experiment was conducted in a Valencia orange orchard on citrumelo Swingle rootstock, in Reginópolis/SP. The experimental design was randomized blocks and the treatment arrangement was a 4 x 4 factorial design with five repetitions. Sixteen soil solution extractors were installed per block, with a total of 80 extractors in the experiment. According to the obtained results, to determine P and K, it is recommended to install the solution extractor at 15 cm horizontal and 30 cm depth from the emitter. For NO3, the extractor recommended placement is 25 cm horizontal and 30 cm depth. The soil solution extractor proved to be a sensitive tool, capable of determining the ions mobility in the wet bulb.

The micro and macro structures of the caudal vertebrae and muscles of Dendrophidion dendrophis and Mastigodryas bifossatus were described using histological slides, dissections, radiographs, and clearing and double staining of intact, broken, and healed tails. To analyze the relationship among the frequencies of healed tails we established two populations of D. dendrophis and six groups of M. bifossatus. We found that fractures of the tail in the two species are intervertebral, and there are no morphological and/or structural mechanisms that facilitate the urotomy, which is classified as non-specialized pseudoautotomy. The caudal vertebrae of D. dendrophis and M. bifossatus show minor differences in the shape of the condyle, cotyle, and border of the neural spine, and in the size and orientation of the hemapophysis and pleurapophysis. The absence of bleeding at the moment of tail breakage may indicate the presence of sphincters in the veins and arteries of D. dendrophis. The distal part of the last vertebrae retained in healed tails of D. dendrophis and M. bifossatus participates in the healing processes as a possible source of calcium in the formation of a calcified cap. We found high frequency of tail breakage in both species, which occurs in almost the entire length of the tail, with no specific areas of concentration. There was no difference in the frequencies of healed tails among males and females of different populations of D. dendrophis and M. bifossatus. Juveniles have lower breakage frequencies than adults in both species, except for populations of M. bifossatus from the Cerrado and Pampa. © 2013 by the American Society of Ichthyologists and Herpetologists.
